Reliability Sensitivity Analysis of Dropped Objects Hitting on the Pipeline at Seabed
Nowadays, as oil industry gradually moves towards deep sea fields with water depth more than 1000 meters, they are subjected to several threats which can cause failure of the pipeline, of which the accidentally-dropped objects have become the leading external risk factor for subsea developments. In this thesis, a sample field layout introduced in Det Norske Veritas (DNV) guide rules is selected as the study case with 100 m water depth. Six different groups of dropped objects are used in this paper. The conditional hit probability for long/flat shaped objects will be calculated with the methods from both DNV rules and an in-house tool Dropped Objects Simulator (DROBS). The difference between the results will be discussed. Meanwhile, the sensitivity analysis on mass, collision area , the volume, added mass coefficient and drag coefficient of the objects are calculated
CpeF is the Bilin Lyase that Ligates the Doubly Linked Phycoerythrobilin on Phycoerythrin in the Cyanobacterium Fremyella Diplosiphon
Phycoerythrin (PE) is a green light-absorbing protein present in the light-harvesting complex of cyanobacteria and red algae. The spectral characteristics of PE are due to its prosthetic groups, or phycoerythrobilins (PEBs), that are covalently attached to the protein chain by specific bilin lyases. Only two PE lyases have been identified and characterized so far, and the other bilin lyases are unknown. Here, using in silico analyses, markerless deletion, biochemical assays with purified and recombinant proteins, and site-directed mutagenesis, we examined the role of a putative lyase-encoding gene, cpeF, in the cyanobacterium Fremyella diplosiphon. Analyzing the phenotype of the cpeF deletion, we found that cpeF is required for proper PE biogenesis, specifically for ligation of the doubly linked PEB to Cys-48/Cys-59 residues of the CpeB subunit of PE. We also show that in a heterologous host, CpeF can attach PEB to Cys-48/Cys-59 of CpeB, but only in the presence of the chaperone-like protein CpeZ. Additionally, we report that CpeF likely ligates the A ring of PEB to Cys-48 prior to the attachment of the D ring to Cys-59. We conclude that CpeF is the bilin lyase responsible for attachment of the doubly ligated PEB to Cys-48/Cys-59 of CpeB and together with other specific bilin lyases contributes to the post-translational modification and assembly of PE into mature light-harvesting complexes
How Many Consumers Prefer an Expert Over a Non-Expert For an Iphone Ad?
Apple\u27s iconic iPhone is a product that only needs a new introduction when the next annual models are introduced and marketed prior to release. However, our team is curious how many consumers are more influenced to buy an iPhone from an ad featuring an iPhone expert in comparison to a non-expert. We\u27ll have 2 separate advertisements with different spokespersons, one featuring a tech expert and the other featuring an influential celebrity. Each ad will promote the new features from the upcoming iPhones word-for-word. Our team will then show these ads and conduct surveys with participants asking them which one do they prefer and what factor(s) contributed most to their answers. We want to see how many consumers are shifting away from Apple\u27s earliest marketing strategies and have began valuing more promotions from knowledgeable people
Drinking Decisions: Twentieth-Century Marketing and Tradition in New Orleans Alcoholic Beverage Trends
Over the past twenty years, the national beverage industry adapted to a growing interest in historic cocktails and classic recipes. Among the many rediscovered classics, New Orleans’ own century-old recipes, like the Sazerac cocktail, garnered praise, national attention, and consumer embrace – even legislative endorsement. However, for most of the past forty years, the city retained a reputation as a place for wild abandon doused in alcoholic beverages of mediocre pedigree. Rather than dismiss the evolution of drinking trends from elegant, classic recipes to indulgent, high-proof booze-bombs as an inherent choice of local drinkers, this paper explores evidence in historic menus and the scholarship in New Orleans tourism marketing. From a careful examination of bar guides, advertisements, newspaper articles, menus and reviews, it is apparent that New Orleans did not eschew its appreciation for traditional, old-time cocktail customs. Rather, two parallel stories unfold; locals continued to demand beverages from previous generations, while business owners recognized the need to accommodate visitors wanting to experience the city’s liberal relationship with liquor. Though the local community is not always harmonious with the dependency on tourism, local bar operators continually offered traditional, historic drink options while also catering to the needs of tourists who chose New Orleans for the escapist experience the city marketed

‘Habituated to Drunkenness’: Opinions of New Orleanians about Prohibition as Revealed through Letters to the Editor of The Times-Picayune, 1918-1922
Both popular and scholarly observers have portrayed New Orleans as a city both supported and burdened by its image as a diverse cultural other within the American South, historically tolerant of certain sins of the flesh. This image has been used by proponents and critics alike in order to push their respective agenda regarding the Crescent City. This thesis will not seek to discredit this image that is based largely on fact. However, using Prohibition as a case study, this thesis will use letters to the editor to uncover attitudes of New Orleanians in opposition to this reputation to reveal alternative and historically silenced voices of New Orleans, since for instance people of a certain age, gender, or ethnicity were silenced in the halls of government. This paper will reveal the opinions of New Orleanians regarding Prohibition and what these opinions can tell us about New Orleans’s image
Let Me Take a #Shelfie: An Assemblage Explored Through Framing
As emerging technologies rapidly change the way that individuals socially interact, researchers can look back to older theories of social organization in order to apply traditional concepts to new ways of being. A #shelfie is a popular hashtag used on Instagram, a social media application, typically used with a post that consist of any visual media containing book(s) or item(s) related to literature in a physical space within, around, and/or upon a piece of furniture. This thesis is centered around the examination of a data collection that gathered top #shelfie posts on Instagram for the purpose of visual content and textual analysis.
I argue that users are performing and constructing identities using curated #shelfies that actually span beyond the original typical bookshelf content, that this particular content is being utilized mostly by users identifying as women, and that it highlights areas of multiple framing occurring at the same time through Instagram and the objects being posted. My thesis is anchored within a multi-disciplinary framework that utilizes Erving Goffman’s theories of self and framing, cultural materialism as framing from scholars such as Daniel Miller, and that this analysis can be understood in a Deleuzian lens by examining how assemblage theory can help to navigate what #shelfie is, how it was produced, and what it may mean for future studies of digital media and the self. StackCBpred: A Stacking based Prediction of Protein-Carbohydrate Binding Sites from Sequence
Carbohydrate-binding proteins play vital roles in many vital biological processes and study of these interactions, at residue level, are useful in treating many critical diseases. Analyzing the local sequential environments of the binding and non-binding regions to predict the protein-carbohydrate binding sites is one of the challenging problems in molecular and computational biology. Prediction of such binding sites, directly from sequences, using computational methods, can be useful to fast annotate the binding sites and guide the experimental process. Because the number of carbohydrate-binding residues is significantly lower than non-carbohydrate-binding residues, most of the methods developed are biased towards over predicting the non-carbohydrate-binding residues. Here, we propose a balanced predictor, called StackCBPred, which utilizes features, extracted from evolution-driven sequence profile, called the position-specific scoring matrix (PSSM) and several predicted structural properties of amino acids to effectively train a stacking-based machine learning method for the accurate prediction of protein-carbohydrate binding sites
“To be men, not destroyers”: Developing Dabrowskian Personalities in Ezra Pound’s The Cantos and Neil Gaiman’s American Gods
Kazimierz Dabrowski’s psychological theory of positive disintegration is a lesser known theory of personality development that offers an alternative critical perspective of literature. It provides a framework for the characterization of postmodern protagonists who move beyond heroic indoctrination to construct their own self-organized, autonomous identities. Ezra Pound’s The Cantos captures the speaker-poet’s extensive process of inner conflict, providing a unique opportunity to track the progress of the hero’s transformation into a personality, or a man. American Gods is a more fully realized portrayal of a character who undergoes the complete paradigmatic collapse of positive disintegration and deliberate self-derived self-revision in a more distilled linear fashion. Importantly, using a Dabrowskian lens to re-examine contemporary literature that has evolved to portray how the experience of psychopathology leads to metaphorical death—which may have any combination of negative or positive outcomes—has not only socio-cultural significance but important personal implications as well
