Repetitive neuromuscular magnetic stimulation as a novel treatment option in pediatric headache disorders
Neuromodulation using rNMS is a promising therapeutic approach for headache disorders (3-5, 14, 56). Besides tension-type headache and post-traumatic headache, migraine in particular is a common headache syndrome that is associated with a high prevalence in pediatric patients. Non-pharmacological, non-invasive prophylactic treatments are urgently needed, especially for children and adolescents (8, 10, 14).
rNMS has already been applied in preliminary studies in adult migraine patients with mTrP in the shoulder-neck area and promising results have been achieved with regard to headache symptoms and the muscular component (1, 3). In order to pave the way for the implementation of this non-invasive therapeutic modality in the pediatric population, a prospective study and a retrospective data analysis regarding the use of rNMS in children and adolescents with primary headache disorders were conducted (4, 5).
This dissertation included publications regarding the use of rNMS targeting the shoulder and neck muscles in children and adolescents with episodic migraine and mTrP in the trapezius muscle, as well as in pediatric patients with migraine, tension-type headache and post-traumatic headache. The feasibility, tolerability, acceptance, and clinical effects of rNMS in relation to headache symptoms and at the muscular level were investigated (4, 5, 73).
For the prospective study, 14 patients were treated in six rNMS sessions, resulting in a total number of 84 rNMS sessions. To evaluate the adherence rate and feasibility of rNMS, standardized questionnaires were used to record the satisfaction and tolerability of the patients and their caregivers as well as the side effects during the intervention. Headache frequency, intensity and medication intake were recorded using a headache calendar from the German Society for Migraine and Headache Disorders. Muscular effects were evaluated by means of physiotherapeutic examination and algometry on the trapezius muscle. Questionnaires were completed to assess migraine-associated disability and quality of life. The acceptance, feasibility, and safety of rNMS was demonstrated. In addition, promising results were shown with regard to reduced headache frequency, medication use, reduced muscular hypersensitivity, and impairment in everyday life due to the headache disorder (4).
The retrospective analysis achieved similar results with regard to the acceptance, safety, and tolerability of the therapeutic approach. Promising results were also achieved in the present analysis with regard to the central and muscular effects (5, 73).
In summary, rNMS as a neuromodulatory treatment method for pediatric migraine and other headache disorders appears to be a practicable, well-tolerated, and well-accepted approach. The clinical effects of rNMS as a pediatric treatment method needs to be further evaluated in future, sham-controlled and larger-scale studies

tRNA modifications as molecular fossils of a protocellular translation system
Centuries of debate on the topics of the origin of life on Earth resulted in the current viewpoint that small organic molecules formed by cosmic or atmospheric phenomena would slowly accumulate and react under prebiotic conditions on the early Earth to form essential precursors that are basic building blocks of primitive biopolymers. Short biopolymers resulting from the random chemical oligomerization of these building blocks could then be selected by their inhabiting environments, and those with the fittest physicochemical properties to accumulate and enrich would polymerize to acquire higher complexity in structures and functions, a process now regarded as ‘chemical evolution’. Among the three component biopolymers of life, namely DNA, RNA and peptides, RNA possesses the capabilities to transfer genetic information by base pairing and perform a wide range of catalytic activities, when folded into ribozymes. Hence, the ‘RNA world’ theory indicates a key period of the chemical evolution in which RNA molecules served as a key hereditary molecule and evolved rapidly to catalyze rudimentary biochemical reactions that progressively shaped the modern biochemistry. However, the RNA world model faces several unanswered questions and one of those being the origin of translation, a process where RNAs are decoded to form peptides and evolved beyond the RNA world. Translation is characterized by two features - a template-directed peptide synthesis and(in) a defined genetic code dictionary. Attempting to address the mystery of the origin of translation, we looked into non-canonical nucleotides that are today found in the tRNAs and rRNAs of the translation machinery, which are highly conservative among all lifeforms on Earth and are considered as ‘molecular fossils’. Since many of them were found to form parallelly with the canonical nucleic acid molecules in various prebiotic reaction pathways, they were likely to be incorporated into early RNA oligonucleotides and served important roles in the early functionalization and survival of RNA.
In this thesis, we designed the chemical synthesis of these modified RNAs and investigated their physicochemical properties. We built a model in which these modifications could perform template-directed peptide synthesis and acquire elementary chemoselectivity by liposome interactions
O contrato de Escrow em alienações de participação societária
Die Doktorarbeit beginnt mit einer Darstellung der wichtigsten Merkmale des Escrow-Agreements und seiner Anwendung in der Praxis. Anschließend werden die wichtigsten rechtlichen Merkmale des Escrow-Agreements im U.S.-amerikanischen Recht analysiert. Anhand der identifizierten Funktionen des Escrow-Agreements im US-amerikanischen Recht wird ein Vergleich der rechtlichen Behandlung des Escrow-Agreements im deutschen und brasilianischen Recht vorgenommen, beginnend mit seiner (1) Umschreibung. Danach geht die Arbeit in die Fragen (2) der wesentlichen Merkmale für die Qualifizierung eines Vertrags als Escrow-Agreement, sowie (3) der üblichen Gestaltungsformen des Escrow-Agreements, (4) seiner Rechtsnatur, (5) der Besitzverhältnisse zwischen den Parteien des Escrow-Agreements, (6) der Auswirkungen der Vertragsverbindung mit dem zugrundeliegenden Hauptgeschäft auf das Escrow-Agreement, (7) des Innenverhältnisses des Escrow-Agreements, also der Rechtsbeziehungen zwischen den Parteien und (8) der rechtlichen Behandlung des Escrow-Gegenstands in der Zwangsvollstreckung durch Drittgläubiger der Parteien. Schließlich führt die Doktorarbeit einige konkrete Anwendungen der allgemeinen Regeln des Escrow-Agreements auf M&A-Transaktionen aus. Die Hauptthese der Doktorarbeit im deutschen Recht lautet, dass das Escrow-Agreement eine typische Geschäftsbesorgung, genauer gesagt eine Treuhand, darstellt, auf die die Regeln der notariellen Verwahrung (§ 57 ff. BeurkG) analog angewendet werden können. Im brasilianischen Recht ist die Hauptthese, dass die Rechtsnatur des Escrow-Agreements die eines atypischen fiduziarischen Geschäfts (negócio fiduciário) bildet, auf das bestimmte Regeln der Geschäftsbesorgung (contrato de mandato) (Art. 653 ff. CC) analog angewendet werden können
Quantum gas microscopy of interacting quantum matter with artificial gauge fields
Analog quantum simulation with ultracold atoms in optical lattices represents a powerful approach to investigate strongly correlated quantum many-body systems, whose complexity renders classical simulation infeasible. This thesis reports key advances achieved with a newly developed quantum gas microscope based on ultracold cesium atoms, specifically tailored to explore exotic quantum phenomena in large-scale systems with unprecedented tunability, precision, and control.
First, we introduce a method based on unsupervised neural networks to reconstruct lattice occupations at atomic separations significantly smaller than the optical resolution limit. The unsupervised approach, trained directly on experimental fluorescence images, accurately captures the full imaging process including complex correlated emission processes arising in the short-spacing regime. We demonstrate that this method provides a high-fidelity detection, enabling quantum simulations in previously inaccessible optical regimes.
Secondly, we expand quantum gas microscopy beyond simple density measurements by implementing a robust and scalable detection technique for off-diagonal observables, such as local currents and kinetic energies. By partitioning a two-dimensional lattice system into an array of isolated double wells using an optical superlattice, we demonstrate direct measurement and manipulation of coherences with both large-scale global and local control. This enhancement broadens the experimental capabilities of quantum gas microscopes, allowing novel exploration of many-body systems with important applications in topological matter, non-equilibrium dynamics and superconductivity.
Lastly, we experimentally study interacting many-body states in the presence of an artificial gauge field on optical flux ladders. In particular, we realize the strongly interacting Mott-Meissner phase -- a state combining interaction-induced localization with chiral currents induced by the gauge field -- in large-scale bosonic flux ladders with 48 sites at half filling. By combining the local readout of currents with a flexible interaction control, we reveal the emerging equilibrium particle currents with full spatial resolution and study their interaction dependence, providing direct experimental evidence of the predicted Mott-Meissner phase. These results constitute a significant experimental advance towards the large-scale realization of interacting topological matter, and more broadly, they provide new avenues for the analog quantum simulation with ultracold atoms.Die analoge Quantensimulation mit ultrakalten Atomen in optischen Gittern bietet einen leistungsfähigen Ansatz zur Untersuchung stark korrelierter quantenmechanischer Vielteilchensysteme, deren Komplexität klassische Simulationen unmöglich macht. Diese Dissertation berichtet über wesentliche Fortschritte, die mithilfe eines neu entwickelten Quantengasmikroskops auf Basis ultrakalter Cäsiumatome erzielt wurden. Die Plattform wurde speziell entworfen, um exotische Quantenmaterie in großskaligen Systemen mit bislang unerreichter Abstimmbarkeit, Präzision und Kontrolle zu erforschen.
Zunächst stellen wir eine Methode basierend auf unüberwacht trainierten neuronalen Netzen vor, um die atomare Besetzung in optischen Gittern in einem Regime zu rekonstruieren, in dem der Gitterabstand deutlich unterhalb der optischen Auflösungsgrenze liegt. Das unüberwachte Training direkt auf experimentelle Fluoreszenzbilder erfasst den gesamten Abbildungsprozess akkurat, einschließlich komplexer korrelierter Emissionsprozesse, die insbesondere im Bereich kurzer atomarer Abstände auftreten. Wir zeigen, dass diese Methode eine hochpräzise Detektion ermöglicht und so den Weg für Quantensimulationen in bisher unerreichbaren Regimen ebnet.
Des Weiteren ergänzen wir die Detektions- und Kontrollfähigkeiten der Quantengasmikroskopie über reine Dichtemessungen hinaus, indem wir eine robuste und skalierbare Detektionstechnik für nichtdiagonale Observablen wie lokale Ströme und kinetische Energien implementieren. Durch die Unterteilung eines zweidimensionalen Gittersystems in eine Anordnung isolierter Doppelpotentialtöpfe mittels eines optischen Supergitters demonstrieren wir die direkte Messung und Manipulation von Kohärenzen sowohl mit globaler als auch lokaler Kontrolle in großen Systemen. Diese Erweiterung verbessert die experimentellen Fähigkeiten von Quantengasmikroskopen erheblich und ermöglicht neuartige Untersuchungen von Vielteilchensystemen mit wichtigen Anwendungen in topologischer Materie, Nichtgleichgewichtsdynamik und Supraleitung.
Abschließend untersuchen wir wechselwirkende Vielteilchenphasen in Gegenwart eines künstlichen Magnetfeldes auf optischen Flussleitern. Insbesondere realisieren wir die Mott-Meissner-Phase -- einen Zustand, der wechselwirkungsinduzierte Lokalisierung mit chiralen Strömen kombiniert, welche durch das Magnetfeld erzeugt werden -- in großskaligen bosonischen Flussleitern mit 48 Gitterplätzen bei halber Füllung. Durch die Kombination der lokalen Messung von Teilchenströmen mit einer flexiblen Kontrolle der Wechselwirkungsstärke enthüllen wir die entstehenden Gleichgewichtsströme mit vollständiger räumlicher Auflösung und untersuchen deren Abhängigkeit von der Wechselwirkungsstärke. Diese Ergebnisse liefern einen direkten experimentellen Nachweis der vorhergesagten Mott-Meissner-Phase und stellen so einen bedeutenden Fortschritt in Richtung der großskaligen Realisierung wechselwirkender topologischer Materie dar. Zusammen mit den vorher genannten Ergebnissen eröffnen wir damit eine Vielzahl neuer Wege für die analoge Quantensimulation mit ultrakalten Atomen
