facilities.Two new Ru(II)-based complexes containing 2-thiouracil derivatives, known as 2-thiouracil (2TU) and 6-methyl-2-thiouracil (6m2TU), were synthesized using cis,trans-[RuCl2(PPh3)2(bipy)] as a precursor. The obtained compounds with a general formula trans-[Ru(2TU)(PPh3)2(bipy)]PF6 (1) and trans-[Ru(6m2TU)(PPh3)2(bipy)]PF6 (2) were characterized by analytical techniques such as NMR, UV-vis, and IR spectroscopies, elementary analysis, mass spectrometry, and single-crystal X-ray diffraction. Moreover, the investigation of the complexes-DNA interaction were carried out using spectrophotometric titrations and showed that the complexes present a weak interaction with this biomolecule. The compounds were evaluated against HL-60, K-562, HepG2, and B16-F10 cancer cells and against noncancer cells (PBMCs). The results of the biological assay revealed that complex 2 is more promising than complex 1. Finally, the present study suggests that complexes 1 and 2 causes cell death by apoptosis, significantly increasing the percentage of apoptotic HL-60 cells, in which the compounds altered the cell cycle, reducing the cells in G1/G0, G2/M, and S phases

Há cerca de quatro anos, em 2016, foi criado o Comitê Fiocruz pela Acessibilidade e Inclusão de Pessoas com Deficiência, uma ação articulada por profissionais de diferentes unidades da instituição que
levaram a Vice-Presidência de Ambiente, Atenção e Promoção da Saúde (VPAAPS) da Fiocruz à proposta de aglutinar os esforços que estavam dispersos no Campus de Manguinhos. Dessa primeira reunião, nasceu um grupo de trabalho que redigiu o Termo de Referência apresentado e aprovado no Conselho Deliberativo da Fundação, dando início à trajetória consolidada por um plano de ação e diferentes eventos, pelos quais o tema da acessibilidade e inclusão tem ajudado a Fiocruz a transformar gradativamente sua abordagem e cultura institucional

Trypanosoma cruzi promotes transcriptomic remodeling of the JAK/STAT signaling and cell cycle pathways in Myoblasts
No full textChagas disease is responsible for more than 10,000 deaths per year and about 6 to 7 million infected people worldwide. In its chronic stage, patients can develop mega-colon, mega-esophagus, and cardiomyopathy. Differences in clinical outcomes may be determined, in part, by the genetic background of the parasite that causes Chagas disease. Trypanosoma cruzi has a high genetic diversity, and each group of strains may elicit specific pathological responses in the host. Conflicting results have been reported in studies using various combinations of mammalian host—T. cruzi strains. We previously profiled the transcriptomic signatures resulting from infection of L6E9 rat myoblasts with four reference strains of T. cruzi (Brazil, CL, Y, and Tulahuen). The four strains induced similar overall gene expression alterations in the myoblasts, although only 21 genes were equally affected by all strains. Cardiotrophin-like cytokine factor 1 (Clcf1) was one of the genes found to be consistently upregulated by the infection with all four strains of T. cruzi. This cytokine is a member of the interleukin-6 family that binds to glycoprotein 130 receptor and activates the JAK/STAT signaling pathway, which may lead to muscle cell hypertrophy. Another commonly upregulated gene was tyrosine 3-monooxygenase/tryptophan 5-monooxygenase activation protein theta (Ywhaq, 14-3-3 protein 2), present in the Cell Cycle Pathway. In the present work, we reanalyzed our previous microarray dataset, aiming at understanding in more details the transcriptomic impact that each strain has on JAK/STAT signaling and Cell Cycle pathways. Using Pearson correlation analysis between the expression levels of gene pairs in biological replicas from each pathway, we determined the coordination between such pairs in each experimental condition and the predicted protein interactions between the significantly altered genes by each strain. We found that although these highlighted genes were similarly affected by all four strains, the downstream genes or their interaction partners were not necessarily equally affected, thus reinforcing the idea of the role of parasite background on host cell transcriptome. These new analyses provide further evidence to the mechanistic understanding of how distinct T. cruzi strains lead to diverse remodeling of host cell transcriptome

Salud mental en la seguridad pública: estudios, enfoques y tendencias de la producción del conocimiento de la temática
No full textObjetivo:sistematizar estudos brasileiros sobre saúde mental no trabalho de segurança pública no período de 2007 a 2017. Metodologia:buscas em bancos de produção acadêmica nacionais e internacionais de acesso gratuito na internet. Resultados:deparou-se com o predomíniode estudos voltados a servidores ligados à polícia militar e a leituras de gênero e suas repercussões na saúde das trabalhadoras, além de dados epidemiológicos relacionados ao estresse profissional. Conclusão:a noção de valorização do trabalhador desse segmento ainda é recente, com predomínio de estudos segmentados que não se aprofundam em uma abordagem sistêmica, o que abre oportunidade de novos estudos.Objective:organize Brazilian studies on mental health in the workplace of public security from 2007 to 2017. Methodology:research on national and international free access academic databases. Results:predominance of studies focused on police officers from the Brazilian military police and on gender studies and their repercussions on the female workers' health, in addition to epidemiological data related to professional stress. Conclusion:the notion of worker's valorization in this sector is still recent and showssuperficial scientific production, which opens opportunities for further studies.Objetivo:sistematizar los estudios brasileños sobre salud mental en el trabajo de seguridad pública entre los años de 2007 a 2017. Metodología:búsqueda en base de datos de producción académicos nacionales e internacionales. Resultados:predominaron los estudios dirigidos a los empleados de la policía militar y las lecturas de género y sus repercusiones en la salud de los trabajadores, además de datos epidemiológicos y relacionados con el estrés profesional. Conclusión:la noción de valorar a las cuestiones generales de los trabajadores en ese segmento es aún reciente, con prioridad de estudios que no profundizan el enfoque sistémico, pero que abre oportunidades para futuros estudios
Microsatellite variation revealed panmictic pattern for Triatoma brasiliensis (Triatominae: Reduviidae) in rural northeastern Brazil: the control measures implications
No full textBackground: Triatoma brasiliensis Neiva, 1911 is the main vector of Trypanosoma cruzi in the caatinga of Northeastern Brazil. Despite of its epidemiological relevance, there are few studies on its genetic variability. Using microsatellite markers, we characterized the variability and dynamics of infestation and reinfestation of T. brasiliensis after residual insecticide spraying in five surveys conducted in a well-defined rural area located in the municipality of Tauá, Ceará, between 2009 and 2015. We evaluated: (1) general variability among local of captures; (2) variability along the time analysis (2009, 2010 and 2015); (3) and reinfestation process. Results: On the analysis (1) global and pairwise FST values suggested absence of clusters among the area. AMOVA indicated that total variation is mainly represented by individual differences. Absence of clustering indicates a panmitic unit, with free gene flow. For (2), Pairwise FST indicated alterations in the genetic profile of the triatomines along the time. (3) Analysis of the reinfestation process showed that the domiciliary units investigated had different sources of infestation despite of its proximity. Conclusions: Observed homogeneity can be explained by the great dispersal capacity of T. brasiliensis, overlapping the different environments. Persistent house infestation in Tauá may be attributed to the occurrence of postspraying residual foci and the invasion of triatomines from their natural habitats
