Public Archaeology in Italy: communication onsite and online
In Italia, l’archeologia pubblica ha subìto negli ultimi anni un accelerazione, grazie ad una maggiore consapevolezza da parte degli archeologi sulla necessità di un dialogo con il pubblico. Sempre più progetti di ricerca archeologica investono risorse nell’organizzazione di iniziative volte a coinvolgere la comunità, sia onsite che online.
Ma per valutare effettivamente l’impatto di queste iniziative c’è bisogno di delineare una metodologia condivisa, assente attualmente in Italia: manca una progettualità, una pianificazione, una riflessione critica sull’argomento.
Il mio lavoro, inserito all’interno del progetto di “Archeologia dei paesaggi di Ravenna” dell’Università di Bologna, ha l’obiettivo di individuare le strategie comunicative migliori per coinvolgere il pubblico verso tematiche storico-archeologiche, ricorrendo in particolare all’utilizzo dei social media, e di individuare una metodologia efficace per la realizzazione e valutazione delle iniziative di archeologia pubblica nell’ambito della ricerca universitaria.
Essendoci la necessità di una pianificazione, ho deciso di considerare il mio progetto come se fosse un’azienda con un prodotto o servizio da portare sul mercato. In tal caso, l’azienda deve ricorrere alla stesura di un piano marketing, un documento che formalizza le analisi e le conseguenti decisioni di marketing per un determinato prodotto/mercato, in un determinato arco temporale/spaziale al fine di conseguire gli obiettivi prefissati.In Italy, public archaeology has accelerated in recent years, thanks to a greater awareness by archaeologists of the need for dialogue with the public. More and more archaeological research projects are investing resources in the organization of initiatives aimed at involving the community, both onsite and online.
But in order to effectively assess the impact of these initiatives, it is necessary to outline a shared methodology, currently absent in Italy: there is a lack of planning, planning and critical reflection on the subject.
My work, inserted within the project of "Archaeology of the landscapes of Ravenna" of the University of Bologna, aims to identify the best communication strategies to engage the public towards historical issues-the use of social media, and to identify an effective methodology for the implementation and evaluation of public archaeology initiatives in the field of university research.
Since there was a need for planning, I decided to consider my project as a company with a product or service to bring to the market. In this case, the company must resort to the drafting of a marketing plan, a document that formalizes the analysis and the resulting marketing decisions for a given product/ market, in a certain time/space in order to achieve the objectives set
Nuclear shape instability caused by lamin A deregulation promotes invasiveness in pediatric bone sarcomas: from nucleo-cytoskeleton dynamics to novel therapeutic opportunities
Osteosarcoma (OS) and Ewing sarcoma (EWS) are the two most frequent primary bone tumors, in which metastases remain the most relevant adverse prognostic factor. Lamin A is the main constituent of the nuclear lamina, with a fundamental role in maintaining the connection between nucleus and cytoskeleton (through LINC complex proteins interactions), and its alterations can be implicated in tumor progression.
We investigated how nucleo-cytoskeleton dynamics is influenced by lamin A modulation in OS and EWS, demonstrating that both these cancer models had low levels of lamin A, which are linked to a significantly more marked nuclear misshaping. In our in vitro studies, reduced levels of lamin A promoted migratory abilities in these tumors. Moreover, these findings were corroborated by gene expression analyses on EWS patient samples, showing that LMNA levels were significantly lower in metastatic lesions compared to primary tumors and that patients with low LMNA had a significant worse overall survival. We also found that LMNA expression significantly impaired EWS metastases formation in vivo.
We demonstrated that low lamin A expression was linked to a severe mislocalization of LINC complex proteins, thus disrupting nucleo-cytoskeleton interactions, with a corresponding gain in malignant properties, which resulted in increased invasiveness. Lamin A overexpression or its accumulation by a statin-based pharmacological treatment allowed us to reconstitute a functional nucleo-cytoskeleton interplay, which resulted in significant downmodulation of ROCK2 and YAP, two crucial drivers of EWS aggressiveness.
Our study demonstrated that lamin A is a favorable mediator of nuclear shape stability in bone sarcomas, and its modulation rescues LINC complex protein localization and regulates mechano-signaling pathways, thus promoting a less aggressive cancer phenotype. We also identified statins, already employed in clinical practice, as a tool capable to increase lamin A levels, and to reconstitute functional nucleo-cytoskeletal dynamics, resulting in reduced cellular migration
The frozen elephant trunk technique for the treatment of complex aortic arch and descending thoracic aorta pathologies: long term outcomes and distal reintervention
Background: The frozen elephant trunk(FET) technique is one of the last evolution in the treatment of complex pathologies of the aortic arch and the descending thoracic aorta.Materials and methods: Between January 2007 and March 2021, a total of 396 patients underwent total aortic arch replacements with the FET technique in our centre.The main indications were thoracic aortic aneurysm(n=104,28.2%), chronic aortic dissection(n=224,53.4%) and acute aortic dissection(n=68, 18.4%). We divided the population in two groups according the position of the distal anastomosis (zone 2 vs zone 3) and the length of the stent graft ( 150 mm): conservative group (Zone 2 anastomosis + stent length 150mm, n. 141). Results: The overall 30-day mortality rate was 13%(48/369); the risk factor analysis showed that an aggressive approach was neither a risk factor for major complication (permanent dialysis, tracheostomy, bowel malperfusion and permanent paraplegia) neither for 30-day mortality. The survival rate at 1, 5,10 and 15 years was 87.7%,75%,61.3% and 58.4% respectively. During the follow up, an aortic reintervention was performed in 122 patients (38%), 5 patients received a non-aortic cardiac surgery. Freedom from aortic reintervention at 1-,5- and 10-year was 77%,54% and 44% respectively. The freedom from aortic reintervention was higher in the ‘aggressive’ group (62.5%vs40.0% at 5 years, log-rank=0.056). An aggressive approach was not protective for aortic reintervention at follow up and for death at follow up. Conclusions: The FET technique represents a feasible and efficient option in the treatment of complex thoracic aortic pathologies. An aortic reintervention after FET is very common and the decision-making approach should consider and balance the higher risk of an aggressive approach in terms of post-operative complication versus the higher risk of a second aortic reintervention at follow-up
Endoscopic treatment of anastomotic leak with vacuum-assisted therapy (EVAT) after colo-rectal surgery: first stage vs second stage treatment
La fistola anastomotica è una delle complicanze più temute nella chirurgia colo-rettale. Le anastomosi colo-rettali basse , le colo-anali e le pouch anali hanno un rischio più elevato di sviluppare una fistola anastomotica . La terapia endoluminale a pressione negativa (Endosponge®) è stata proposta come strategia di trattamento, tuttavia, la tempistica migliore in cui attuare la procedura rimane ancora poco definita. Lo scopo dello studio è confrontare i risultati ottenuti con l'Endosponge® come trattamento di prima linea rispetto a quelli in cui è stato applicato a seguito del fallimento di ulteriori trattamenti.
Lo studio retrospettivo monocentrico ha incluso pazienti con fistola anastomotica trattati con Endosponge® in un periodo di tempo compreso tra novembre 2019 e novembre 2022. L'Endosponge® è stato applicato come prima linea o come salvataggio. Il dispositivo è stato applicato nella sede della deiscenza e periodicamente sostituito fino alla guarigione. La risoluzione del leak anastomotico è stata confermata con esame endoscopico.
Dei 25 pazienti inclusi, 9 sono stati sottoposti a Endosponge® come trattamento di prima linea, mentre 16 sono stati sottoposti a Endosponge® di salvataggio. La deiscenza anastomotica è stata diagnosticata dopo un intervallo di tempo mediano di 14 giorni (range 10-413) nel primo gruppo e di 38 giorni (range 11-362) nel secondo (p=0,82). L'Endosponge® è stato applicato dopo 7 giorni (range 1-60) dalla diagnosi di fistola anastomotica nel primo gruppo e dopo 76 giorni (range 6-780) nel secondo gruppo (p=0,058). La risoluzione della fistola anastomotica è stata ottenuta in una percentuale di casi maggiore nel primo gruppo rispetto al secondo 88,9% vs 37,6% (p =0,033).
Lo studio conferma l'efficacia dell'Endosponge® nel trattamento delle fistole anastomotiche colorettali basse quando utilizzato precocemente e come trattamento di prima linea.Background: Anastomotic leak is one of the most challenging complications after colorectal surgery. Low colorectal, coloanal and ileal pouch-anal anastomoses are at highest risk of developing anastomotic leak. Endoluminal-Vacuum-Assisted Therapy (EVAT) has been proposed as an effective treatment, however, the best timing of the procedure remains unclear. The aim of the study was to compare the outcomes of EVAT as a first-stage vs. second-stage (following other operative procedures) treatment.
Methods: Retrospective study including patients undergoing EVAT after confirmed anastomotic leak in a single centre between November 2019 and November 2022. EVAT was applied either as a first-stage treatment or a second-stage treatment based on the individual case. The device was positioned in the cavity and replaced until the gap was reduced in size and covered by granulating tissue. The resolution of the anastomotic leak was confirmed at the endoscopy .
Results: Of the 25 patients who were included, 9 underwent first-stage EVAT, while 16 underwent second-stage EVAT. The anastomotic leak was diagnosed at a median of 14 days (range 10-413) after surgery for the first-stage group and 38 days (range 11-362) in the second-stage group (p=0.82). The time of EVAT from the onset of the leak was 7 days (1-60) in the first-stage group and 76 (6-780) days in the second-stage group (p=0.058). The resolution of the anastomotic leak was significantly higher in the first-stage EVAT group 88,9% vs 37,6% in the second-stage group, (p = 0.033).
Conclusions: The present study confirms the effectiveness of the EVAT in the treatment of low anastomotic leaks, in particular when it is used as a first treatment after diagnosis
Untangle sustainable development goal 8 through data visualization and HCI methods
Following the approval of the 2030 Agenda for Sustainable Development in 2015, sustainability became a hotly debated topic. In order to build a better and more sustainable future by 2030, this agenda addressed several global issues, including inequality, climate change, peace, and justice, in the form of 17 Sustainable Development Goals (SDGs), that should be understood and pursued by nations, corporations, institutions, and individuals.
In this thesis, we researched how to exploit and integrate Human-Computer Interaction (HCI) and Data Visualization to promote knowledge and awareness about SDG 8, which wants to encourage lasting, inclusive, and sustainable economic growth, full and productive employment, and decent work for all. In particular, we focused on three targets: green economy, sustainable tourism, employment, decent work for all, and social protection. The primary goal of this research is to determine whether HCI approaches may be used to create and validate interactive data visualization that can serve as helpful decision-making aids for specific groups and raise their knowledge of public-interest issues. To accomplish this goal, we analyzed four case studies. In the first two, we wanted to promote knowledge and awareness about green economy issues: we investigated the Human-Building Interaction inside a Smart Campus and the dematerialization process inside a University. In the third, we focused on smart tourism, investigating the relationship between locals and tourists to create meaningful connections and promote more sustainable tourism. In the fourth, we explored the industry context to highlight sustainability policies inside well-known companies.
This research focuses on the hypothesis that interactive data visualization tools can make communities aware of sustainability aspects related to SDG8 and its targets. The research questions addressed are two: "how to promote awareness about SDG8 and its targets through interactive data visualizations?" and "to what extent are these interactive data visualizations effective?"
Correlations between cardiac magnetic resonance and histologic findings in Anderson Fabry disease
ABSTRACT
Background
Cardiac magnetic resonance (CMR) has been shown as promising diagnostic tool in Anderson-Fabry disease (AFD) cardiomyopathy due to its ability to detect fat deposits through lower native T1 values. However no histological validation has been provided to date.
Objectives
To correlate CMR and histologic findings in different cardiac stages of AFD focusing on T1 mapping. Methods Fifteen AFD patients (49 years [IQR 39-63], 60% females) undergoing CMR (cines, native T1 and T2 mapping, LGE and post-contrast T1 imaging) and endomyocardial biopsy (EMB, n=11) or septal myectomy (n=4), were retrospectively evaluated. Tissue specimens were analyzed with light/electron microscopy and vacuolization amount calculated as percentages of vacuolated myocytes and vacuolated myocyte area (%VMA) through a quantitative histomorphometric color-based analysis.
Results
In patients without increased indexed left ventricular mass (LVMi) at CMR (67%), T1 fell as %VMA increased (r= -0.883; p<0.001), whereas no clear relationship was evident once increased LVMi occurred (r= -0.501; p=0.389). At least 45% of vacuolized myocytes and 10% of VMA were needed for low T1 to occur. %VMA positively correlate with maximal wall thickness (MWT, r=0.860, p<0.0001) and LVMi (r= 0.762; p<0.001). Increased MWT and LVMi were present with at least 45% and 80% of vacuolated myocytes, respectively, and 18% and 22% of VMA.
Conclusions
This study demonstrated an inverse correlation between native T1 and the vacuolization amount in patients without increased LVMi at CMR, providing a histological validation of low native T1 in AFD. Importantly, a significant vacuolization burden was needed before low T1 and left ventricle hypertrophy occurred
Mars delivery service - development of the electro-mechanical systems of the Sample Fetch Rover for the Mars Sample Return Campaign
This thesis describes the development of the Sample Fetch Rover (SFR), studied for Mars Sample Return (MSR), an international campaign carried out in cooperation between the National Aeronautics and Space Administration (NASA) and the European Space Agency (ESA). The focus of this document is the design of the electro-mechanical systems of the rover.
After placing this work into the general context of robotic planetary exploration and summarising the state of the art for what concerns Mars rovers, the architecture of the Mars Sample Return Campaign is presented. A complete overview of the current SFR architecture is provided, touching upon all the main subsystems of the spacecraft. For each area, it is discussed what are the design drivers, the chosen solutions and whether they use heritage technology (in particular from the ExoMars Rover) or new developments. This research focuses on two topics of particular interest, due to their relevance for the mission and the novelty of their design: locomotion and sample acquisition, which are discussed in depth.
The early SFR locomotion concepts are summarised, covering the initial trade-offs and discarded designs for higher traverse performance. Once a consolidated architecture was reached, the locomotion subsystem was developed further, defining the details of the suspension, actuators, deployment mechanisms and wheels. This technology is presented here in detail, including some key analysis and test results that support the design and demonstrate how it responds to the mission requirements.
Another major electro-mechanical system developed as part of this work is the one dedicated to sample tube acquisition. The concept of operations of this machinery was defined to be robust against the unknown conditions that characterise the mission. The design process led to a highly automated robotic system which is described here in its main components: vision system, robotic arm and tube storage
Distributed ledger technologies between anonymity and transparency: AML/CFT regulation of cryptocurrency ecosystems in the EU
The advent of Bitcoin suggested a disintermediated economy in which Internet users can take part directly. The conceptual disruption brought about by this Internet of Money (IoM) mirrors the cross-industry impacts of blockchain and distributed ledger technologies (DLTs). While related instances of non-centralisation thwart regulatory efforts to establish accountability, in the financial domain further challenges arise from the presence in the IoM of two seemingly opposing traits: anonymity and transparency. Indeed, DLTs are often described as architecturally transparent, but the perceived level of anonymity of cryptocurrency transfers fuels fears of illicit exploitation. This is a primary concern for the framework to prevent money laundering and the financing of terrorism and proliferation (AML/CFT/CPF), and a top priority both globally and at the EU level.
Nevertheless, the anonymous and transparent features of the IoM are far from clear-cut, and the same is true for its levels of disintermediation and non-centralisation. Almost fifteen years after the first Bitcoin transaction, the IoM today comprises a diverse set of socio-technical ecosystems. Building on an analysis of their phenomenology, this dissertation shows how there is more to their traits of anonymity and transparency than it may seem, and how these features range across a spectrum of combinations and degrees. In this context, trade-offs can be evaluated by referring to techno-legal benchmarks, established through socio-technical assessments grounded on teleological interpretation.
Against this backdrop, this work provides framework-level recommendations for the EU to respond to the twofold nature of the IoM legitimately and effectively. The methodology cherishes the mutual interaction between regulation and technology when drafting regulation whose compliance can be eased by design. This approach mitigates the risk of overfitting in a fast-changing environment, while acknowledging specificities in compliance with the risk-based approach that sits at the core of the AML/CFT/CPF regime
On the role of AGN feedback in shaping galaxies across cosmic time
Understanding how Active Galactic Nuclei (AGN) shape galaxy evolution is a key challenge of modern astronomy. In the framework where black hole (BH) and galaxy growth are linked, AGN feedback must be tackled both at its “causes” (e.g. AGN-driven winds) and its “effects” (alteration of the gas reservoir in AGN hosts).
The most informative cosmic time is z~1-3, at the peak of AGN activity and galaxy buildup, the so-called cosmic noon.
The aim of this thesis is to provide new insights regarding some key questions that still remain open in this research field: i) What are the properties of AGN-driven sub-pc scale winds at z>1? ii) Are AGN-driven winds effective in influencing the life of galaxies? iii) Do AGN impact directly on star formation (SF) and gas content of their hosts?
I first address AGN feedback as “caught in the act” by studying ultra-fast outflows (UFOs), X-ray AGN-driven winds, in gravitationally lensed quasars. I build the first statistically robust sample of high-z AGN, not preselected based on AGN-driven winds. I derive a first estimate of the high-z UFO detection fraction and measure the UFO duty cycle of a single high-z quasar for the first time.
I also address the “effects” of AGN feedback on the life of host galaxies. If AGN influence galaxy growth, then they will reasonably impact the molecular gas reservoir first, and SF as a consequence.
Through a comparative study of the molecular gas content in cosmic-noon AGN hosts and matched non-active galaxies (i.e., galaxies not hosting an AGN), we find that the host galaxies of more regular AGN (not selected to be the most luminous) are generally similar to non-active galaxies. However, we report on the possibility of a luminosity effect regulating the efficiency by which AGN might impact on galaxy growth
Efficient and low-computational predictive models for spectral sensors
Spectral sensors are a wide class of devices that are extremely useful for detecting essential information of the environment and materials with high degree of selectivity. Recently, they have achieved high degrees of integration and low implementation cost to be suited for fast, small, and non-invasive monitoring systems. However, the useful information is hidden in spectra and it is difficult to decode. So, mathematical algorithms are needed to infer the value of the variables of interest from the acquired data. Between the different families of predictive modeling, Principal Component Analysis and the techniques stemmed from it can provide very good performances, as well as small computational and memory requirements. For these reasons, they allow the implementation of the prediction even in embedded and autonomous devices. In this thesis, I will present 4 practical applications of these algorithms to the prediction of different variables: moisture of soil, moisture of concrete, freshness of anchovies/sardines, and concentration of gasses. In all of these cases, the workflow will be the same. Initially, an acquisition campaign was performed to acquire both spectra and the variables of interest from samples. Then these data are used as input for the creation of the prediction models, to solve both classification and regression problems. From these models, an array of calibration coefficients is derived and used for the implementation of the prediction in an embedded system. The presented results will show that this workflow was successfully applied to very different scientific fields, obtaining autonomous and non-invasive devices able to predict the value of physical parameters of choice from new spectral acquisitions
