    The Metagenomic Analysis of the Gut Microbiome of the South American Locust (Schistocerca cancellata)

    abstract: Locusts are generalist herbivores meaning that they are able to consume a variety of plants. Because of their broad diet, and ability to respond rapidly to a favorable environment with giant swarms of voracious insects, they are dangerous pests. Their potential impacts on humans increase dramatically when individuals switch from their solitarious phase to their gregarious phase where they congregate and begin marching and eventually swarming together. These swarms, often billions strong, can consume the vegetation of enormous swaths of land and can travel hundreds of kilometers in a single day producing a complex threat to food security. To better understand the biology of these important pests we explored the gut microbiome of the South American locust (Schistocerca cancellata). We hypothesized generally that the gut microbiome in this species would be critically important as has been shown in many other species. We extracted and homogenized entire guts from male S. cancellata, and then extracted gut microbiome genomic DNA. Genomic DNA was then confirmed on a gel. The initial extractions were of poor quality for sequencing, but subsequent extractions performed by collaborators during troubleshooting at Southern Illinois University Edwardsville proved more useful and were used for PCR. This resulted in the detections of the following bacterial genera in the gut of S. cancellata: Enterobacter, Enterococcus, Serratia, Pseudomonas, Actinobacter, and Weisella. With this data, we are able to speculate about the physiological roles that they hold within the locust gut generating hypotheses for further testing. Understanding the microbial composition of this species’ gut may help us better understand the locust in general in an effort to more sustainably manage them. (abstract

    Performing Arts Programming for Disadvantaged Youth: An Analysis of Effective Marketing Strategies

    abstract: This thesis research aims to define, identify, and promote community theatre as a “third space� for disadvantaged youth. A third space is defined by the Oxford dictionary as “...the in-between, or hybrid, spaces, where the first and second spaces work together to generate a new third space. First and second spaces are two different, and possibly conflicting, spatial groupings where people interact physically and socially: such as home (everyday knowledge) and school (academic knowledge)� (Oxford Dictionary, 2021). For disadvantaged youth, the creation of a third space in the theatre can give them a safe environment away from issues they may have at home or at school, it can further their learning about themselves and others, and it can also help those youth feel a sense of belonging to a community larger than themselves. Because of these benefits, it is clear that performing arts programs can offer a great impact on disadvantaged youth; however, many theatre companies struggle to market their programming to said communities. This may be in part, due to low marketing budgets, no specificity in labor resources dedicated to youth programming, or ineffective marketing strategies and tactics. This research aims to provide tangible recommendations for youth programmers to better involve their target audience. (abstract

    The Effects of Acoustic Stress on the Behavior of Drosophila melanogaster

    abstract: Veterans are approximately 30% more likely than non-veterans to suffer from severe hearing impairment. Tinnitus, or ringing in the ears, which is increasingly common among military service men and women, has been linked to significant cognitive and psychological impairment and can be worsened by the same sounds that trigger post-traumatic stress disorder (PTSD). In fact, tinnitus and PTSD often present as comorbidities, and recent studies suggest these two disorders may share a common neurological pathway. Additional studies are required to better understand the connection between hearing loss and impaired cognitive function such as that observed in with PTSD. Here, we use the fruit fly, Drosophila melanogaster, to explore the relationship between hearing loss and cognitive function. Negative geotaxis climbing assays and courtship behavior analysis were used to examine neurobehavioral changes induced by prolonged, intense auditory stimulation. Preliminary results suggest that exposure to loud noise for an extended period of time significantly affected Drosophila behavior, with males being more sensitive than females. Based on our results, there appears to be a potential connection between noise exposure and behavior, further suggesting that Drosophila could be an effective model to study the link between hearing loss and PTSD.     Developing a Cooperative Food Business for Food-Insecure University Students

    abstract: Food insecurity among university students in the U.S. is a pressing sustainability problem due to its prevalence, complex socio-economic drivers, and adverse effects. A national survey from the Hope Center for College, Community, and Justice found that 45% of university students (n=86,000) had experienced some form of food insecurity in the past 30 days (Goldrick-Rab et al., 2019). Students at Arizona State University (ASU) are similarly impacted by this sustainability problem—a recent study found that approximately 35% of ASU students have experienced food insecurity (Bruening et al., 2016). Food insecurity has a variety of detrimental effects on university students’ physical health, psychological well-being, and academic achievement (El Zien et al., 2019; Payne-Sturges et al., 2018; Meza et al., 2019), and these resulting issues have complex inter-regional, intrageneration, and intergenerational implications. To mitigate food insecurity among university students, the project proposes the development of a sustainable, student-run food cooperative business at Arizona State University (ASU). Food cooperative businesses have long been utilized by communities to advance food access, economic self-determination, and food justice (DePasquale, Sarang, & Vena, 2017), so the project aims to lay the foundation for the establishment of such an enterprise at ASU. Through the development of an enterprise start-up plan and the execution of preliminary coalition-building efforts, the project seeks to demonstrate the plausibility of this solution while empowering stakeholders with the strategies needed to enact it

    abstract: The use of drone warfare in Pakistan has long been a contentious topic. Drones have had a number of adverse effects on the people of Waziristan. The focus of this thesis will be on the effects drone warfare have had on the mental and psychological health of the victims and children of Waziristan. Scholarly articles, research studies, and reports were researched in order to discuss the background of drone strikes in Waziristan, the legality of drone warfare, the data around drone strike casualties in Waziristan, and the effects of drone strikes on the psychological and mental health of children and victims in Waziristan. After discussion of these topics, several suggestions for reparations for the people of Waziristan are addressed in the conclusion. (abstract

    Qualitative Determination of Exopolymeric Substances in Particle-Associated Bacteria from the Sargasso Sea

    abstract: The biological carbon pump acts as part of the global carbon cycle through the photosynthetic fixation of inorganic carbon into dissolved and particulate organic carbon by phytoplankton. Previously, the biological carbon pump was attributed to large aggregates and zooplankton fecal pellets since their size and density results in faster sinking rates, efficiently exporting organic carbon to deeper depths in the ocean. However, recent studies have indicated that small cells, known as picoplankton, contribute significantly to the formation of sinking particles. The presence of exopolymeric substances (EPS), among them sticky transparent exopolymeric particles (TEP) and proteinaceous coomassie stainable particles (CSP), serve as influential factors of export flux and aggregation. The presence of heterotrophic bacteria can also affect aggregation and sinking velocity, as seen in previous studies, and is likely attributed to their EPS and TEP production. The staining and visualization of TEP and CSP allow for the qualitative determination of these types of EPS from bacteria isolated from sinking particles collected with particle interceptor traps at various depths in the Sargasso Sea. I study the presence of TEP and CSP in particle-associated bacteria. Cultures of picocyanobacteria, consisting of xenic Synechococcus and axenic Prochlorococcus, were used to establish positive and negative controls for stained isolate analysis. Marinobacter adhaerens served as a tertiary control for an axenic culture that stains positive for TEP. I chose six isolates of bacteria isolated from sinking particles to be stained and visualized to test for the secretion of TEP and CSP. Four of the isolates stained positive for both TEP and CSP, including Pseudoalteromonas sp., Erythrobacter sp., and Marinobacter sp., while one isolate, Micrococcus sp., stained positive only for TEP, and the last isolate, another Marinobacter sp., stained positive for only CSP. These results are important in understanding the role of plankton organisms in the formation of sinking particles. (abstract
