Longitudinal to transversal conversion of mode-locked states in an empty optical resonator

    No full text
A longitudinal mode-locked state can be converted to a transverse mode-locked state by exploiting the spectral and spatial filtering of an empty optical resonator. Carrier and amplitude modulation sidebands were simultaneously transmitted by the conversion resonator, yielding phase-locked superpositions of up to five transverse modes. Equivalently, an amplitude-modulated beam was converted into a beam that periodically moved across the transverse plane. Precise control over the spatial beam shape during oscillation was gained by independently altering the set of transverse modes and their respective powers, which demonstrated an increased level of control in the generation of transverse mode-locked states

Genetic polymorphisms in the promoter of the FSHB gene and their impact on transcription factor binding

Inhibition of p90 ribosomal S6 kinases disrupts melanoma cell growth and immune evasion

    No full text
Background: The mitogen-activated protein kinase (MAPK) signaling pathway is frequently hyperactivated in malignant melanoma and its inhibition has proved to be an efficient treatment option for cases harboring BRAFV600 mutations (BRAFMut). However, there is still a significant need for effective targeted therapies for patients with other melanoma subgroups characterized by constitutive MAPK activation, such as tumors with NRAS or NF-1 alterations (NRASMut, NF-1LOF), as well as for patients with MAPK pathway inhibitor-resistant BRAFMut melanomas, which commonly exhibit a reactivation of this pathway. p90 ribosomal S6 kinases (RSKs) represent central effectors of MAPK signaling, regulating cell cycle progression and survival. Methods: RSK activity and the functional effects of its inhibition by specific small molecule inhibitors were investigated in established melanoma cell lines and patient-derived short-term cultures from different MAPK pathway-hyperactivated genomic subgroups (NRASMut, BRAFMut, NF-1LOF). Real-time qPCR, immunoblots and flow cytometric cell surface staining were used to explore the molecular changes following RSK inhibition. The effect on melanoma cell growth was evaluated by various two- and three-dimensional in vitro assays as well as with melanoma xenograft mouse models. Co-cultures with gp100- or Melan-A-specific cytotoxic T cells were used to assess immunogenicity of melanoma cells and associated T-cell responses. Results: In line with elevated activity of the MAPK/RSK signaling axis, growth and survival of not only BRAFMut but also NRASMut and NF-1LOF melanoma cells were significantly impaired by RSK inhibitors. Intriguingly, RSK inhibition was particularly effective in three-dimensional growth settings with long-term chronic drug exposure and suppressed tumor cell growth of in vivo melanoma models. Additionally, our study revealed that RSK inhibition simultaneously promoted differentiation and immunogenicity of the tumor cells leading to enhanced T-cell activation and melanoma cell killing. Conclusions: Collectively, RSK inhibitors exhibited both multi-layered anti-tumor efficacy and broad applicability across different genomic melanoma subgroups. RSK inhibition may therefore represent a promising novel therapeutic strategy for malignant melanoma with hyperactivated MAPK signaling

    No full text

When high work engagement is negative for family tasks: mechanisms and boundary conditions

    No full text
Background: Typically, work engagement is positively related to beneficial job outcomes. Earlier studies, however, revealed a "dark side" of work engagement showing negative effects such as more work-family conflict. Using a resource perspective, our study seeks to better understand why and when these negative effects of work engagement occur. Specifically, we test a new model in which the relationship of work engagement with work-family conflict is mediated by organizational citizenship behavior (OCB) and work rumination. Moreover, we argue that employees’ resource-building strategies (i.e., job crafting) and resource levels (i.e., psychological capital) buffer resource depletion due to high work engagement. Methods: We tested our assumptions in a field study that involved data collected on three measurement points with 523 employees from Nigeria. The measures consist of Utrecht Work Engagement Scale, Organizational Citizenship Behavior Scale, Work Rumination Scale, Psychological Capital Scale, Job Crafting Measure, Work-family Conflict Scale, and demographic variables. Structural Equation Modeling (SEM) was used to test the hypotheses. Results and discussion: Results from latent structure equation modelling confirm that work rumination mediates the positive relationship between work engagement and work-family conflict. Additionally, our findings suggest that behavioral engagement (i.e.,OCB) and work rumination mediate the relationship between work engagement and work-family conflict. Moreover, psychological capital mitigated the relationships of work engagement with work rumination, but not job crafting. Our study helps to better understand the "dark side" of work engagement and offers implications on how to mitigate its detrimental relationship with work-family conflict

Zur Rezeption Josquin Desprez' am kurpfälzischen Hof Ottheinrichs: Johannes Mittners Missa Hercules Dux Ferrariae

    No full text

Effect of the Interplay between Polymer–Filler and Filler–Filler Interactions on the Conductivity of a Filled Diblock Copolymer System

    No full text
We investigate the relative roles of the involved interactions and micro-phase morphology in the formation of the conductive filler network in an insulating diblock copolymer (DBC) system. By incorporating the filler immersion energy obtained by means of the phase-field model of the DBC into the Monte Carlo simulation of the filler system, we determined the equilibrium distribution of fillers in the DBC that assumes the lamellar or cylindrical (hexagonal) morphology. Furthermore, we used the resistor network model to calculate the conductivity of the simulated filler system. The obtained results essentially depend on the complicated interplay of the following three factors: (i) Geometry of the DBC micro-phase, in which fillers are preferentially localized; (ii) difference between the affinities of fillers for dissimilar copolymer blocks; (iii) interaction between fillers. The localization of fillers in the cylindrical DBC micro-phase has been found to most effectively promote the conductivity of the composite. The effect of the repulsive and attractive interactions between fillers on the conductivity of the filled DBC has been studied in detail. It is quantitatively demonstrated that this effect has different significance in the cases when the fillers are preferentially localized in the majority and minority micro-phases of the cylindrical DBC morphology
