    Growth rates of contract-reared versus home-reared replacement dairy heifers

    peer-reviewedSuccessful heifer rearing is dependent on achieving optimal average daily gain (ADG) targets to calve for the first time at 24 months. Whilst dairy farmers internationally have traditionally managed their heifers on-farm to achieve these targets, recent dairy herd expansion within Europe has resulted in increased demand for labour-saving heifer-rearing strategies, such as off-farm contractrearing. However, loss of direct influence on the day-to-day management decisions affecting the ability of heifers to grow adequately to achieve this age of first calving may represent a potential barrier to uptake. Hence, the aim of this longitudinal observational study was to compare the growth rates of contract- vs home-reared heifers. Approximately 6 500 heifers from 120 commercial Irish dairy farms were enrolled in a 3-year study. For 65 of these farms, heifers were reared at a contract-rearing facility. For the remaining 55 farms, heifers were reared on their home farm. Over the course of 20 months from birth until precalving, heifers were examined and weighed at four farm visits. The relationship between several independent variables (farm type, herd size, heifer breed, economic breeding index (EBI) and health events) and ADG at different time points was investigated and analysed utilising linear mixed models. Overall ADG for heifers throughout the rearing period was 0.71 kg/day. There was a significant association between farm type and ADG for all five linear ADG models; home-reared heifers grew, on average, 0.025, 0.039, 0.11 and 0.059 kg/day more than contract-reared heifers between visit 1 and visit 4 (overall ADG), visit 1 and visit 2, visit 2 and visit 3 and visit 1 and visit 3, respectively. The occurrence of diarrhoea during farm visit 2 (median age 8.5 months) was associated with a significant reduction in ADG between visit 3 and visit 4. Calf-hood disease (diarrhoea, respiratory disease or navel ill) was not associated with the growth rate during any of the subsequent visit periods. While home-reared heifers had greater ADG during four of the five periods studied, median heifer ADG in both cohorts exceeded the minimum published target weight gains at each developmental stage required for heifers to reach puberty, conceive at 15 months and calve for the first time at 24 months. Importantly, there was wide variation both within enterprises and between farms. It was concluded that while the absolute difference in daily growth rates of home- and contract-reared heifers was minimal, when considered in the context of the entire heifer-rearing period, these growth rate differences have the potential to impact the future reproductive and milk production performance of heifers

    Assessing the long-term impact of urease and nitrification inhibitor use on microbial community composition, diversity and function in grassland soil

    peer-reviewedReductions in ammonia (NH3) and nitrous oxide (N2O) emissions from agricultural systems are critical for achievement of sustainability targets that underpin international efforts on climate and biodiversity. Urease inhibitors (UI) such as N-(n-butyl) thiophosphoric triamide (NBPT) and nitrification inhibitors (NI) such as dicyandiamide (DCD) slow down microbial and chemical N transformation rates in soil, resulting in decreased environmental N losses. To date there has been minimal assessment of the long-term non-target impacts of UI and NI on soil microbial communities and biological function in grasslands. Utilising a temperate grassland field experiment where fertilisers (with or without inhibitors) were repeatedly applied over a five year period, we assessed the impact of individual or combined inhibitor use on microbial community composition, abundance and function via a combination of functional assays, quantitative polymerase chain reaction (qPCR) assays and amplicon sequence analysis. We also investigated the effect of N inhibitor use on the N functional community, and whether the form of applied N fertiliser (i.e. calcium ammonium nitrate (CAN) or Urea) affected microbial community composition and function. Treatments included a Control (no N); CAN; Urea; Urea + NBPT (UI); Urea + DCD (NI); and Urea + DCD + NBPT (NI & UI). There was no impact of either UI or NI use on non-target microbial community composition or abundance. Function and the abundance of N cycling communities were mainly unaffected by fertilisation or the use of inhibitors. The observed effect of NI was primarily on the nitrification process. There was a significant reduction in nitrification potential associated with the use of NI, and in the case of the Urea + DCD treatment a reduction in COMAMMOX nitrifier abundance, and an increase in potential N mineralisation and N2O emissions. Finally, there was a significant impact of fertilisation and fertiliser type (i.e. CAN or Urea) on the fungal community structure but no impact on bacterial community structure. These results provide a knowledge base that will inform policy regarding the utilisation of N inhibitors as a mitigation measure for reducing gaseous N losses in grasslands

    Bio-economic modelling of sheep meat production systems with varying flock litter size using field data

    peer-reviewedSheep meat producers derive the majority of income from sales of weaned lambs, determined by flock conception rates, litter size, and lamb survival. Field data from commercial flocks can inform sensitivity analyses of the effect of litter size on flock productivity, feed demand, and gross margin. This study adapted an established bio-economic model of a flock of breeding ewes informed by statistical relationships (from linear models) between flock litter size (lambs born per ewe lambing) and production factors (such as flock barren rate, litter birth type and lamb birth weight) identified using 156 145 animal records from the Irish national sheep breeding database. Sensitivity analyses were undertaken to investigate the effects of flock litter size on flock production, feed demand, and gross margin. Results showed that as flock litter size increased, the proportion of lambs born as multiples increased, with 14 % of lambs born as singles when flock litter size was 2.2 lambs born per ewe lambing. Flock gross margin increased from €2 205 to €7 730 as litter size increased from 1.0 to 2.0 lambs born per ewe lambing. As litter size increased from 1.0 to 2.2 lambs born per ewe lambing, flock gross margin increased linearly by, on average, €52 per 0.01 increase in litter size. At a litter size of > 2.2 lambs born per ewe lambing, flock gross margin increased on average €12 per 0.01 increase in litter size. At a litter size of 2.2 lambs born per ewe lambing, flock efficiency (at 65.0 kg of lamb weaned per ewe presented for breeding), weaning rate (at 1.5 lambs weaned per ewe presented for breeding; not including excess lambs from large litters sold within a week after birth and thus not weaned on-farm), and gross margin (at €8 500) began to plateau. The results indicate lower marginal returns in gross margin at very high flock litter size, due to the lower value of additional lambs born as triplets and quadruplets compared with single- and twin-born lambs. However, the diminishing economic returns occurred at higher flock litter size than are currently biologically achieved in most flocks. Quantification from this analysis demonstrates how the value of increasing the number of lambs born changes at very high flock litter size, which can inform the priorities and performance benchmarking for international sheep meat production industries

    The impact of pasture and non-pasture diets on the sensory and volatile properties of whole milk powder

    peer-reviewedThis study evaluated the impact of three distinct diets; perennial ryegrass (GRS), perennial ryegrass/white clover (CLV) and total mixed ration (TMR), on the sensory properties and volatile profile of whole milk powder (WMP). The samples were evaluated using a hedonic sensory acceptance test (n = 99 consumers) and by optimised descriptive profiling (ODP) using trained assessors (n = 33). Volatile profiling was achieved by gas chromatography mass spectrometry using three different extraction techniques; headspace solid phase micro-extraction, thermal desorption and high capacity sorptive extraction. Significant differences were evident in both sensory perception and the volatile profiles of the WMP based on the diet, with WMP from GRS and CLV more similar than WMP from TMR. Consumers scored WMP from CLV diets highest for overall acceptability, flavour and quality, and WMP from TMR diets highest for cooked flavour and aftertaste. ODP analysis found that WMP from TMR diets had greater caramelised flavour, sweet aroma and sweet taste, and that WMP from GRS diets had greater cooked aroma and cooked flavour, with WMP derived from CLV diets having greater scores for liking of colour and creamy aroma. Sixty four VOCs were identified, twenty six were found to vary significantly based on diet and seventeen of these were derived from fatty acids; lactones, alcohols, aldehydes, ketones and esters. The abundance of δ-decalactone and δ-dodecalactone was very high in WMP derived from CLV and GRS diets as was γ-dodecalactone derived from a TMR diet. These lactones appeared to influence sweet, creamy, and caramelised attributes in the resultant WMP samples. The differences in these VOC derived from lipids due to diet are probably further exacerbated by the thermal treatments used in WMP manufacture

    Investigation of breakage behavior and its effects on spray-dried agglomerated whey protein-lactose powders: Effect of protein and lactose contents

    peer-reviewedParticle breakage of dairy powders occurs easily during many processes, reducing the powder functionality. The characteristics of particles and the applied stress from processing conditions on the particles are 2 main factors that can be manipulated to reduce breakage. In this study, we explored the effect of whey protein and lactose contents on dynamic breakage in agglomerated whey protein-lactose powders to provide useful information, in terms of particle characteristics, for controlling unwanted dairy powder breakage. A series of model agglomerates with different whey protein:lactose ratios were produced under the same spray-drying conditions, through a pilot plant trial. We evaluated physical characteristics, composition, and structure of samples; analyzed dynamic breakage under different mechanical stresses; and investigated the rehydration and water adsorption properties of model powders before and after breakage. The particle size and irregularity of agglomerates with more lactose was significantly higher than of samples that contained more protein. This resulted in higher particle breakage during dynamic breakage for samples with more lactose. The breakage of agglomerates was affected by the moisture content of powders and fatigue, where particle breakage happens when mechanical loads, lower than the strength of particles, occur multiple times. Breakage changed the morphology and surface composition of particles and decreased particle size. It also decreased the dispersibility of powders and increased the wetting time of wettable samples but decreased the wetting time of powders with poor wettability. Breakage accelerated time-dependent crystallization and decreased the crystallization temperature but did not affect the glass transition temperature of samples. Thus, under the same drying conditions, composition of powders significantly affected breakage, mainly by altering the physical properties of their particles, which resulted in deteriorated functionality

    Improving the efficacy of herbicides for the control of Rumex obtusifolius L. in Irish grassland using aspects of Integrated Weed Management

    Doctoral thesisDocks are a troublesome weed of conventional grassland and can reduce grass dry matter (DM) production by up to 50%. Conventional farmers largely rely on herbicides to manage docks but often report disappointing results. With imminent EU legislation likely to restrict future herbicide use, there is an impetus to develop new dock management strategies. Three dock management experiments were conducted on a conventionally managed grassland farm (52°21N, 7°18W). Experiment I compared herbicides licensed for use in new leys (NLH) with herbicides licensed for use in established grassland (EGH). Experiment II examined the role of soil test Potassium (K) concentrations (Morgan’s solution; Na acetate + acetic acid, pH 4.8; STK) on competitiveness of docks in grassland. Experiment III combined data from the two field studies to develop control thresholds of seedling docks in new leys. Dock numbers were measured over seven years (2010–2015) and herbage production over five years (2012–2016). There was a 3.4-fold increase (due to clonal propagation) and strong correlation (R2=0.9; P-0.776; P1.0 docks m-2 in new leys

    Modelling transmission of Mycobacterium avium subspecies paratuberculosis between Irish dairy cattle herds

    peer-reviewedBovine paratuberculosis is an endemic disease caused by Mycobacterium avium subspecies paratuberculosis (Map). Map is mainly transmitted between herds through movement of infected but undetected animals. Our objective was to investigate the effect of observed herd characteristics on Map spread on a national scale in Ireland. Herd characteristics included herd size, number of breeding bulls introduced, number of animals purchased and sold, and number of herds the focal herd purchases from and sells to. We used these characteristics to classify herds in accordance with their probability of becoming infected and of spreading infection to other herds. A stochastic individual-based model was used to represent herd demography and Map infection dynamics of each dairy cattle herd in Ireland. Data on herd size and composition, as well as birth, death, and culling events were used to characterize herd demography. Herds were connected with each other through observed animal trade movements. Data consisted of 13 353 herds, with 4 494 768 dairy female animals, and 72 991 breeding bulls. We showed that the probability of an infected animal being introduced into the herd increases both with an increasing number of animals that enter a herd via trade and number of herds from which animals are sourced. Herds that both buy and sell a lot of animals pose the highest infection risk to other herds and could therefore play an important role in Map spread between herds

    Effect of mechanical premilking stimulation on milking duration in late lactation

    peer-reviewedThis study documents the effect of mechanical prestimulation on the milking duration of pasture-based cows in late lactation to better harness increased capacity of automation in the milk harvesting process. Premilking stimulation, provided via manual or mechanical means, has been shown to promote the milk letdown reflex and assist in achieving quick, comfortable, and complete milk removal from the udder. The literature is lacking knowledge on the effect of mechanical premilking stimulation on milking duration, especially in late lactation and in pasture-based systems, and many pasture-based farms do not practice a full premilking routine because of a lack of labor availability. The current study addresses this gap in knowledge. In this study, we tested 2 treatments: (1) the No Stim treatment used normal farm milking settings with no premilking preparation and (2) the Stim treatment used 60 s of mechanical premilking stimulation, with a rate of 120 cycles per minute and a pulsator ratio of 30:70 on cluster attachment. Once the 60 s of stimulation had elapsed, normal milking settings resumed for the remainder of the milking. Sixty cows were enrolled in the study, which ran for 20 d. The effect of treatment on a.m. milking duration was significant, a.m. milking duration for Stim was 12 s shorter than that of No Stim. The effect of treatment on p.m. milk duration was not significant. Treatment had no effect on a.m./p.m. milk yields, average milk flowrates or peak milk flowrates. Significant differences emerged between treatments on a.m. and p.m. dead time (time from cluster attachment to reach a milk flowrate of 0.2 kg/min). The a.m. and p.m. dead times were 6 s shorter for Stim compared with No Stim. The time taken to achieve peak milk flowrate (time to peak) at morning milking was 7 s shorter for Stim compared with No Stim, and treatment yielded no significant effects on time to peak at p.m. milkings. Treatment also had no significant effect on log10 somatic cell count. Although the percentage of congested teat-ends and teat-barrels was numerically lower for Stim versus No Stim, no statistical differences were detected across these measures. Based on the results of the study, we found merit in applying 60 s of mechanical pre-stimulation at a.m. milking from a milking duration perspective. However, the strategy was not as successful for the p.m. milking. Analysis of the milk flowrate profiles recorded during the study suggest potential utility in employing different machine settings for various milkings based on anticipated yield and level of udder fill.Science Foundation Irelan

    Angiotensin-I-Converting Enzyme Inhibitory Activity of Protein Hydrolysates Generated from the Macroalga Laminaria digitata (Hudson) JV Lamouroux 1813

    Seaweeds have a long history of use as both food and medicine, especially in Asian cultures. Moreover, there is growing interest in the use of seaweed ingredients and bioactive compounds in pharmaceutical and nutraceutical products. One ailment that seaweed bioactive compounds may impact is hypertension caused by the enzyme Angiotensin Converting Enzyme 1 (ACE-1; EC 3.4.15.1), found within the Renin-Angiotensin Aldosterone System (RAAS), which causes vasoconstriction of blood vessels, including veins and arteries. The aim of this paper is to generate bioactive peptide containing protein hydrolysates from the brown seaweed Laminaria digitata (Hudson) JV Lamouroux 1813. Proteins were extracted from this seaweed by disrupting the seaweed cell wall using a combination of carbohydrases and proteolytic enzymes. Bioactive peptide containing permeates were generated from L. digitata protein hydrolysates, and both hydrolysates and permeates were screened for their ability to inhibit the enzyme ACE-1. The protein content of the permeate fractions was found to be 23.87% compared to the untreated seaweed, which contained 15.08% protein using LECO analysis. Hydrolysis and filtration resulted in a “white” protein powder, and the protein content of this powder increased by 9% compared to the whole seaweed. The total amino acid (TAA) content of the L. digitata protein permeate was 53.65 g/100 g of the sample, and contains over 32% essential amino acids (EAA). Furthermore, the L. digitata permeate was found to inhibit the ACE-1 enzyme by 75% when compared to the commercial drug Captopril© when assayed at a concentration of 1 mg/mL. The inhibition of ACE-1 (the IC50 value) of 590 g/mL for the L. digitata permeate compares well with Captopril©, which had 100% inhibition of ACE-1, with an IC50 value of 500 g/mL. This study indicates that there is potential to develop protein powders with ACE-1 inhibitory bioactivities from the brown seaweed L. digitata using enzymatic hydrolysis as a cell disruption and protein extraction/hydrolysate generation procedure
