    The genome sequence of the Dark Arches Apamea monoglypha (Hufnagel, 1766)

    We present a genome assembly from an individual male Apamea monoglypha (the Dark Arches, Arthropoda; Insecta; Lepidoptera; Noctuidae). The genome sequence is 576 megabases in span. Most of the assembly is scaffolded into 31 chromosomal pseudomolecules, including the assembled Z sex chromosome. The mitochondrial genome has also been assembled and is 16.5 kilobases in length. Gene annotation of this assembly on Ensembl has identified 17,963 protein coding genes

    Too much is too much: Influence of former stress levels on food craving and weight gain during the COVID-19 period

    The COVID-19 pandemic and associated social restrictions had an extensive effect on peoples' lives. Increased rates of weight gain were widely reported, as were declines in the general populations' mental health, including increases in perceived stress. This study investigated whether higher perceived levels of stress during the pandemic were associated with greater levels of weight gain, and whether poor prior levels of mental health were a factor in higher levels of both stress and weight gain during the pandemic. Underlying changes in eating behaviours and dietary consumption were also investigated. During January-February 2021, UK adults (n = 179) completed a self-report online questionnaire to measure perceived levels of stress and changes (current versus pre-COVID-19 restrictions) in weight, eating behaviours, dietary consumption, and physical activity. Participants also reported on how COVID-19 had impacted their lives and their level of mental health prior to the pandemic. Participants with higher levels of stress were significantly more likely to report weight gain and twice as likely to report increased food cravings and comfort food consumption (OR = 2.3 and 1.9-2.5, respectively). Participants reporting an increase in food cravings were 6-11 times more likely to snack and to have increased consumption of high sugar or processed foods (OR = 6.3, 11.2 and 6.3, respectively). Females reported a far greater number of COVID-19 enforced lifestyle changes and both being female and having poor mental health prior to the pandemic were significant predictors of higher stress and weight gain during the pandemic. Although COVID-19 and the pandemic restrictions were unprecedented, this study suggests that understanding and addressing the disparity of higher perceived stress in females and individuals' previous levels of mental health, as well as the key role of food cravings, is key for successfully addressing the continuing societal issue of weight gain and obesity.     Microplastics in European sea salts – An example of exposure through consumer choice and of interstudy methodological discrepancies

    Microplastics are contaminants of emerging concern, not least due to their global presence in marine surface waters. Unsurprisingly, microplastics have been reported in salts harvested from numerous locations. We extracted microplastics from 13 European sea salts through 30% H O digestion and filtration over 5-µm filters. Filters were visually inspected at magnifications to x100. A subsample of potential microplastics was subjected to Raman spectroscopy. Particle mass was estimated, and human dose exposure calculated. After blank corrections, median concentrations were 466 ± 152 microplastics kg ranging from 74 to 1155 items kg . Traditionally harvested salts contained fewer microplastics than most industrially harvested ones (t-test, p < 0.01). Approximately 14 µg of microplastics (< 12 particles) may be absorbed by the human body annually, of which a quarter may derive from a consumer choosing sea salt. We reviewed existing studies, showing that targeting different particle sizes and incomplete filtrations hinder interstudy comparison, indicating the importance of method harmonisation for future studies. Excess salt consumption is detrimental to human health; the hazardousness of ingesting microplastics on the other hand has yet to be shown. A portion of microplastics may enter sea salts through production processes rather than source materials. [Abstract copyright: Copyright © 2023 The Authors. Published by Elsevier Inc.     Wastewater-based epidemiology for comprehensive community health diagnostics in a national surveillance study: Mining biochemical markers in wastewater

    This manuscript showcases results from a large scale and comprehensive wastewater-based epidemiology (WBE) study focussed on multi-biomarker suite analysis of both chemical and biological determinants in 10 cities and towns across England equating to a population of ∼7 million people. Multi-biomarker suite analysis, describing city metabolism, can provide a holistic understanding to encompass all of human, and human-derived, activities of a city in a single model: from lifestyle choices (e.g. caffeine intake, nicotine) through to health status (e.g. prevalence of pathogenic organisms, usage of pharmaceuticals as proxy for non-communicable disease, NCD, conditions or infectious disease status), and exposure to harmful chemicals due to environmental and industrial sources (e.g. pesticide intake via contaminated food and industrial exposure). Population normalised daily loads (PNDLs) of many chemical markers were found, to a large extent, driven by the size of population contributing to wastewater (especially NCDs). However, there are several exceptions providing insights into chemical intake that can inform either disease status in various communities or unintentional exposure to hazardous chemicals: e.g. very high PNDLs of ibuprofen in Hull resulting from its direct disposal (confirmed by ibuprofen/2-hydroxyibuprofen ratios) and bisphenol A (BPA) in Hull, Lancaster and Portsmouth likely related to industrial discharge. An importance for tracking endogenous health markers such as 4-hydroxy-2-nonenal-mercapturic acid (HNE-MA, an oxidative stress marker) as a generic marker of health status in communities was observed due to increased levels of HNE-MA seen at Barnoldswick wastewater treatment plant that coincided with higher-than-average paracetamol usage and SARS-CoV-2 prevalence in this community. PNDLs of virus markers were found to be highly variable. Being very prevalent in communities nationwide during sampling, SARS-CoV-2 presence in wastewater was to a large extent community driven. The same applies to the fecal marker virus, crAssphage, which is very prevalent in urban communities. In contrast, norovirus and enterovirus showed much higher variability in prevalence across all sites investigated, with clear cases of localized outbreaks in some cities while maintaining low prevalence in other locations. In conclusion, this study clearly demonstrates the potential for WBE to provide an integrated assessment of community health which can help target and validate policy interventions aimed at improving public health and wellbeing

    Bungarus fasciatus venom from eastern and north-east India: venom variation and immune cross-reactivity with Indian polyvalent antivenoms

    Bungarus fasciatus is one of the medically important elapid snakes of South and South-eastern Asia and is responsible for several snakebite incidents some of which were fatal. In this study, the venom compositional variation of Bungarus fasciatus from three different geographical locations in eastern and north-east India: two adjacent villages of Hooghly (West Bengal), Aizawl (Mizoram) and Guwahati (Assam) are reported. In vitro biochemical assays exhibit variation in phospholipase A2 activity, fibrinogenolytic activity, caseinolytic activity and anti-coagulation activity. The immunoreactivity of three Indian polyvalent antivenoms against the venoms revealed incomplete recognition. Bharat Serums antivenom demonstrated that at a venom–antivenom ratio of 1:16, the antivenom exhibited different immunocapturing abilities for all the venom samples. The percentage of non-retained fractions was highest for Guwahati (60.00%) and lowest for Hooghly 1 (18.91%). The study demonstrates intra-population (or inter-individual) variation of B. fasciatus venom from two nearby locations of Hooghly (West Bengal), intra-specific variation of B. fasciatus from three geographical locations and also inter-specific venom variation with B. caeruleus from Tamil Nadu. Thus, the venom variation leads to partial immune cross-reactivity by Indian polyvalent antivenoms. Inclusion of non-recognized venom proteins in the immunization mixture during antivenom production would help to improve the efficacy of the antivenom. Further study of the neutralizing ability of Indian polyvalent antivenoms against medically important snakes from different geographical regions would help to understand the effectiveness of the antivenoms and would invariably assist in the designing and development of safe and effective antivenoms

    An Experiment on Animal Re-Identification from Video

    In the face of the global concern about climate change and endangered ecosystems, monitoring individual animals is of paramount importance. Computer vision methods for animal recognition and re-identification from video or image collections are a modern alternative to more traditional but intrusive methods such as tagging or branding. While there are many studies reporting results on various animal re-identification databases, there is a notable lack of comparative studies between different classification methods. In this paper we offer a comparison of 25 classification methods including linear, non-linear and ensemble models, as well as deep learning networks. Since the animal databases are vastly different in characteristics and difficulty, we propose an experimental protocol that can be applied to a chosen data collections. We use a publicly available database of five video clips, each containing multiple identities (9 to 27), where the animals are typically present as a group in each video frame. Our experiment involves five data representations: colour, shape, texture, and two feature spaces extracted by deep learning. In our experiments, simpler models (linear classifiers) and just colour feature space gave the best classification accuracy, demonstrating the importance of running a comparative study before resorting to complex, time-consuming, and potentially less robust methods

    Modernising operational risk management in financial institutions via data-driven causal factors analysis: A pre-registered study

    In an effort to contribute a quantitative, objective and real-time tool to proactively and precisely manage the factors underlying and exacerbating operational risks, this pre-registered study executes the empirical methodology approved in the associated pre-registered report (Cornwell et al., 2023). The application of the Bayesian network-based approach to an Australian insurance company shows that integrating a financial institution's loss and operational data in this way can effectively model the probability of an operational loss event within its interconnected operational risk environment. Further insights and efficiencies are gained by modelling multiple operational loss events together, rather than in isolation. A novel two-module framework derived specifically for causal factors analysis from the resulting operational risk model helps to highlight the relative importance of causal factors, their collective effects and critical thresholds requiring proactivity. These insights derived from the framework are expected to be strategically valuable in helping an organisation design intentional and targeted controls for and monitoring of operational risks. Given existing knowledge of the improvements quantitative risk management tools make to risk management effectiveness and subsequently firm value, the enhanced risk management and the operational efficiencies this tool seeks to afford should ultimately contribute to driving financial performance and firm value
