Study of the validity of a job-exposure matrix for the job strain model factors an update and a study of changes over time
International audienceObjectives - The objectives of the study were to construct a job-exposure matrix (JEM) for psychosocial work factors of the job strain model, to evaluate its validity, and to compare the results over time. Methods - The study was based on national representative data of the French working population with samples of 46,962 employees (2010 SUMER survey) and 24,486 employees (2003 SUMER survey). Psychosocial work factors included the job strain model factors (Job Content Questionnaire): psychological demands, decision latitude, social support, job strain and iso-strain. Job title was defined by three variables: occupation and economic activity coded using standard classifications, and company size. A JEM was constructed using a segmentation method (Classification and Regression Tree-CART) and cross-validation. Results - The best quality JEM was found using occupation and company size for social support. For decision latitude and psychological demands, there was not much difference using occupation and company size with or without economic activity. The validity of the JEM estimates was higher for decision latitude, job strain and iso-strain, and lower for social support and psychological demands. Differential changes over time were observed for psychosocial work factors according to occupation, economic activity and company size. Conclusions - This study demonstrated that company size in addition to occupation may improve the validity of JEMs for psychosocial work factors. These matrices may be time-dependent and may need to be updated over time. More research is needed to assess the validity of JEMs given that these matrices may be able to provide exposure assessments to study a range of health outcomes
Bisphenol S instead of Bisphenol A: Toxicokinetic investigations in the ovine materno-feto-placental unit
International audienceBisphenol S (BPS) is widely used as a substitute for Bisphenol A in consumer products. Despite its potential endocrine-disrupting effects and widespread exposure, toxicokinetic data, particularly during the critical period of pregnancy, are not available for BPS. The objectives of our study were to evaluate the mechanisms determining fetal exposure to BPS and to BPS glucuronide (BPSG) and to compare them with those prevailing for BPA. The disposition of BPS and BPSG was evaluated in the materno-fetal unit of the catheterized pregnant ewe model, following intravenous administrations of BPS and BPSG to mothers and their fetuses. In a second experiment, the rate of BPS accumulation in the fetal compartment was determined under steady-state conditions after repeated intravenous BPS administrations to the mother. In the maternal compartment, BPS was mainly metabolized into BPSG and totally eliminated in urine. Only 0.40% of the maternal dose was transferred to the fetus. However, once in the fetal compartment, 26% of the fetal dose was rapidly eliminated through placental transfer, while 46% of BPS was metabolized into BPSG which remained trapped in the fetal compartment. Thus, the elimination of BPSG from the fetal compartment required its back-conversion into bioactive BPS, leading to an 87% enhancement of the fetal BPS exposure. Our findings demonstrate that, despite the low materno-fetal placental transfer of BPS, this substitute for BPA is able to accumulate in the fetal compartment after repeated maternal exposure, leading to chronic fetal exposure to BPS in a range of concentrations similar to those of BPA

Strongly Participatory Science and Knowledge Justice in an Environmentally Contested Region
International audienceThis article draws insights from a case study examining unanswered health questions of residents in two polluted towns in an industrial region in southern France. A participatory health study, as conducted by the author, is presented as a way to address undone science by providing the residents with relevant data supporting their illness claims. Local residents were included in the health survey process, from the formulation of the questions to the final data analysis. Through this strongly participatory science (SPS) process, the townspeople offered many creative ideas in the final report for how the data could be used to assist in improving their health and environment and policy work is already in evidence, resulting from the study. Drawing from the literature on participatory science and expertise as well as from the initial outcomes of the local health study, I propose that SPS produces a form of knowledge justice. Understanding knowledge and its making as part of a social justice agenda aligns well with environmental justice frames. Through SPS, local residents have a hermeneutical resource to make sense of their embodied lives and augment their claims with strong data supporting actions for improving their health and environment
Effect of DEET-multiple exposures on behavior and life history traits in the malaria mosquito Anopheles gambiae (s.s.)
International audienceBackground: Vector-borne diseases are major public health concerns, and their control is threatened by the spread of insecticide resistance in vector populations. In this context, the use of repellents is an alternative approach to limit vector-host interactions. However, prior exposure to repellents is suspected to affect mosquito behavior at the subsequent exposure, possibly reducing the efficacy of the compound. Despite this, the effect of mosquito experience on repellent efficacy remains poorly documented. In the present study, we tested whether a first blood meal successfully obtained upon a DEET-treated net would affect the success at taking a second blood meal in spite of DEET in the malaria mosquito Anopheles gambiae (s.s.). The impact of DEET on mosquito life history traits after the first and the second exposure was also measured, in order to assess the long-term consequences of multiple exposures to DEET in vector insects.Results: A first blood meal obtained upon a DEET-treated net did not influence the success of An. gambiae females to take a second blood meal in spite of DEET. However, data showed that a prior exposure to DEET negatively affected all life history traits tested in this study related to fecundity and fertility. DEET pre-exposed females displayed a reduction in blood engorgement at the second exposure, as well as a reduction in the number of eggs laid and in the proportion of offspring that reach adult stage. Also, an increase of mosquito activity was observed during the second blood meal in DEET-pre-exposed females. Taken together, these data suggest an overall impact of DEET exposure on mosquito fitness.Conclusions: Our results did not evidence any effect of a prior exposure to DEET on its efficacy during the second exposure. However, data show a negative impact of DEET exposure on mosquito fitness. These results give insights to understand the long-term efficacy of the most used mosquito repellent, and highlight that DEET induces deleterious effects on mosquito fitness in addition to repellency, potentially increasing its efficacy for controlling vector-borne diseases

Alors que certaines recherches se tournent vers des techniques de bio-surveillance, les enquêtes de santé-environnement sont utiles pour dresser un tableau général de la santé d’une communauté (ici, les habitants de deux villes de Fos-sur-Mer et Port-Saint-Louis-du-Rhône) et identifier des points à approfondir dans une recherche ultérieure ou pour de futures interventions de politique publique
Dynamic and differential expression of the gonadal aromatase during the process of sexual differentiation in a novel transgenic cyp19a1a -eGFP zebrafish line
Accepted manuscriptIn zebrafish, there exists a clear need for new tools to study sex differentiation dynamic and its perturbation by endocrine disrupting chemicals. In this context, we developed and characterized a novel transgenic zebrafish line expressing green fluorescent protein (GFP) under the control of the zebrafish cyp19a1a (gonadal aromatase) promoter. In most gonochoristic fish species including zebrafish, cyp19a1a, the enzyme responsible for the synthesis of estrogens, has been shown to play a critical role in the processes of reproduction and sexual differentiation. This novel cyp19a1a-eGFP transgenic line allowed a deeper characterization of expression and localization of cyp19a1a gene in zebrafish gonads both at the adult stage and during development. At the adult stage, GFP expression was higher in ovaries than in testis. We showed a perfect co-expression of GFP and endogenous Cyp19a1a protein in gonads that was mainly localized in the cytoplasm of peri-follicular cells in the ovary and of Leydig and germ cells in the testis. During development, GFP was expressed in all immature gonads of 20 dpf-old zebrafish. Then, GFP expression increased in early differentiated female at 30 and 35 dpf to reach a high GFP intensity in well-differentiated ovaries at 40 dpf. On the contrary, males consistently displayed low GFP expression as compared to female whatever their stage of development, resulting in a clear dimorphic expression between both sexes. Interestingly, fish that undergoes ovary-to-testis transition (35 and 40 dpf) presented GFP levels similar to males or intermediate between females and males. In this transgenic line our results confirm that cyp19a1a is expressed early during development, before the histological differentiation of the gonads, and that the down-regulation of cyp19a1a expression is likely responsible for the testicular differentiation. Moreover, we show that although cyp19a1a expression exhibits a clear dimorphic expression pattern in gonads during sexual differentiation, its expression persists whatever the sex suggesting that estradiol synthesis is important for gonadal development of both sexes. Monitoring the expression of GFP in control and exposed-fish will help determine the sensitivity of this transgenic line to EDCs and to refine mechanistic based-assays for the study of EDCs. In fine, this transgenic zebrafish line will be a useful tool to study physiological processes such as reproduction and sexual differentiation, and their perturbations by EDCs
Occupational exposure to organic solvents and risk of male breast cancer: a European multicenter case-control study
The etiology of male breast cancer (MBC) is largely unknown but a causal role of exposure to organic solvents has been suggested. Previous studies on occupational risk factors of breast cancer were often restricted to women who are frequently exposed to lower levels and at a lower frequency than men. We investigated the association between MBC and occupational exposure to petroleum and oxygenated and chlorinated solvents in a multicenter case-control study of rare cancers in Europe. METHODS: The study included 104 MBC cases and 1901 controls. Detailed lifetime work history was obtained during interviews, together with sociodemographic characteristics, medical history and lifestyle factors. Occupational exposures to solvents were estimated from a job-exposure matrix. Odds ratios (OR) and their 95% confidence intervals (CI) were calculated using unconditional logistic regression models.
