    Design, synthesis, and structure–activity relationship studies of 6 H -benzo[ b ]indeno[1,2- d ]thiophen-6-one derivatives as DYRK1A/CLK1/CLK4/haspin inhibitors

    International audienceA series of sulfur-containing tetracycles was designed and evaluated for their ability to inhibit protein kinase DYRK1A, a target known to have several potential therapeutic applications including cancers, Down syndrome or Alzheimer's disease. Our medicinal chemistry strategy relied on the design of new compounds using ring contraction/isosteric replacement and constrained analogy of known DYRK1A inhibitors, thus resulting in their DYRK1A inhibitory activity enhancement. Whereas a good inhibitory effect of targeted DYRK1A protein was observed for 5-hydroxy compounds 4i–k (IC50 = 35–116 nM) and the 5-methoxy derivative 4e (IC50 = 52 nM), a fairly good selectivity towards its known DYRK1B off-target was observed for 4k. In addition, the most active compound 4k, having an ATP-competitive mechanism of action, proved to be also a potent inhibitor of CLK1/CLK4 (IC50 = 20 and 26 nM) and, to a lesser extent, of haspin (IC50 = 76 nM) kinases. In silico docking studies within the DYRK1A, CLK1/CLK4 and haspin ATP binding sites were carried out to understand the interactions of our tetracyclic derivatives 4 with these targets. Antiproliferative activities on U87/U373 glioblastoma cell lines of the most potent compound 4k showed a moderate effect (IC50 values between 33 and 46 μM). Microsomal stabilities of the designed compounds 4a–m were also investigated, showing great disparities, depending on benzo[b]thiophene ring 5-substitution

    Intervenir par le sensible: Un enjeu pour l’ergonomie

    International audienceErgonomics has always paid attention to the body(s) in work activity: demands and exposure to multiple risks; gestures, postures, movements; subjective dimension. . . . So many variables through which we can grasp the complexity and singularity of human activity, and on which we can try to act to transform work situations. However, ergonomists still often approach the body through the visible, operational manifestations of work activity and its effects. Much work in the human and social sciences today calls for a sensitive approach to the world and human functioning as a lever for transforming the relationship between people and their environment. These approaches challenge the way we intervene in ergonomics. In this article, we present an experiment carried out as part of a training programme on the occupational risks faced by musicians, using an approach based on the body in movement and sensitivity (inspired by dance and Qi Gong). We discuss the challenges for ergonomics of integrating knowledge and methodologies related to the sensitive body in order to develop our fields of intervention and ensure that work systems evolve in a more ecological and respectful way towards living beings.L’ergonomie a toujours porté une attention au(x) corps dans l’activité de travail : sollicitations et exposition à des dangers multiples ; gestuelles, postures, déplacements ; dimension subjective… Autant de variables à travers lesquelles on peut saisir la complexité et la singularité de l’activité humaine, et sur lesquelles on peut chercher à agir pour transformer les situations de travail. Cependant, les ergonomes approchent encore souvent le corps au travers des manifestations visibles, opératoires, de l’activité de travail et de ses effets. De nombreux travaux en sciences humaines et sociales revendiquent aujourd’hui une approche sensible du monde et du fonctionnement humain, comme un levier pour transformer le rapport des humains à leur milieu. Ces approches interrogent nos manières d’intervenir en ergonomie. Dans cet article, nous présentons une expérimentation réalisée dans un cadre de formation sur les risques professionnels des musiciens, mettant en œuvre une démarche basée sur le corps en mouvement et le sensible (inspirée de la danse et du qi gong). Nous discutons des enjeux pour l’ergonomie d’intégrer les connaissances et les méthodologies qui portent sur le corps sensible, pour développer nos espaces d’intervention et faire évoluer les systèmes de travail dans une visée plus écologique et respectueuse du vivant

    Computation and stability analysis of periodic orbits using finite differences, Fourier or Chebyshev spectral expansions in time

    International audienceWe analyse and compare several algorithms to compute numerically periodic solutions of high-dimensional dynamical systems and investigate their Floquet stability without building the monodromy matrix. The solution and its perturbation are discretised in time either using finite differences, Fourier-Galerkin or Chebyshev expansions. The resulting nonlinear set of equations describing the periodic orbit is solved using a Newton-Raphson algorithm. The linearised equations determining the stability lead to a generalised eigenvalue problem. Unlike the Fourier-Galerkin method, the use of Chebyshev polynomials or finite differences has the advantage that the relevant Floquet exponents are directly given without the well known issue of having to sort out the eigenvalues. The speed of convergence of these three methods is illustrated with examples from the Lorenz system, the Langford system and a two-dimensional thermal convection flow inside a differentially heated cavity. This last example demonstrates the potential of the newly proposed Chebyshev expansion for large-scale problems arising from the discretisation of the incompressible Navier-Stokes equations

    Évaluation de l’endurance électrique d'isolant solide sous tension carrée

    International audienceMedium-frequency transformers (MFTs) are subjected to distinct electrical stresses compared to conventional 50 Hz transformers. It is particularly due to the integration of wide bandgap semiconductor devices such as silicon carbide (SiC), which significantly enhances the efficiency of power electronic converters. However, the MFTs are then submitted to medium frequency square voltage with high dv/dt, which can induce a nonuniform distribution on its turns. In this context, Nomex is considered as a candidate for inter-turn insulation in MFTs. To assess inter-turn stress endurance, virgin Nomex samples were subjected to an electric field of 3.6 kV/mm thanks to a 20 kHz square voltage for 904 hours under ambient temperature. The condition of the insulation was monitored using frequency-domain dielectric spectroscopy and partial discharge (PD) measurements. Experimental results show that, in the absence of PD activity, no significant variation was observed in permittivity, dielectric loss factor, PD inception voltage, or PD extinction voltage, indicating stable dielectric properties and no ageing effects under the applied stress conditions.Les transformateurs à moyenne fréquence (TMF) sont soumis à des contraintes électriques différentes de celles des transformateurs conventionnels fonctionnant à 50 Hz. Cette différence s’explique notamment par l’intégration de dispositifs à semi-conducteurs à large bande interdite, tels que le carbure de silicium (SiC), qui améliorent significativement l’efficacité des convertisseurs d’électronique de puissance. Cependant, les TMF sont exposés à une tension carrée de moyenne fréquence présentant un fort dv/dt, susceptible d’entraîner une répartition non uniforme de la tension entre les spires. Dans ce contexte, le Nomex est considéré comme un matériau candidat pour l’isolation inter-spires des TMF. Afin d’évaluer l’endurance du Nomex face aux contraintes électriques inter-spires, des échantillons vierges ont été soumis à un champ électrique de 3,6 kV/mm, appliqué sous une tension carrée de 20 kHz pendant 904 heures à température ambiante. L’état des isolants a été évalué à l’aide de la spectroscopie diélectrique dans le domaine fréquentiel et de mesures de décharges partielles (DP). Les résultats expérimentaux montrent qu’en l’absence d’activité de DP, aucune variation significative n’est observée sur la permittivité, le facteur de pertes diélectriques, ni sur les tensions d’apparition et d’extinction des DP. Ces observations indiquent une stabilité des propriétés diélectriques et l’absence d’effet de vieillissement sous les conditions de contraintes appliquées

    Gene and transposable element expression in response to stress in temperate and tropical populations of Drosophila

    International audienceThe study of stress response in natural populations is crucial for understanding species local adaptation and evolution. In Drosophila, significant genetic diversity across populations from different geographical origins has been observed, emphasizing the influence of local environments.Results: In this study, we explored the impact of starvation and cold stress on the phenotypic and transcriptomic response of two natural populations of D. melanogaster and D. simulans from temperate and tropical regions. Additionally, we investigated the behavior and influence of transposable elements (TEs) in these types of stress, combining RNA-seq and ChIP-seq experiments, with high-quality long-read genome assemblies of all the strains. Our findings in D. melanogaster revealed that the transcriptomic response to stress is similar across geographical origins, whereas in D. simulans there is more variability. Notably, neither starvation nor cold induced a general activation of TEs in D. melanogaster or D. simulans, at least in the tissue and strains analysed in this study. Finally, we found three polymorphic TEs producing TE-chimeric transcripts associated with changes in nearby gene expression levels after stress.Conclusions: Overall, this study highlights the complexity of stress-TE interactions and their potential impact on adaptation. Understanding these dynamics contributes to the broader knowledge of how genetic and environmental factors interact to modulate gene expression, shaping an organism’s ability to adapt to varying conditions

    Fatphobia and physical activity: from the co-construction of stigma to the potential for change

    International audienceThis article contributes to the research into the conditions and forms of incorporation of gender norms relating to body size and shape (Smith et al., 2015), as people age. The specific aim is to understand these norms in relation to the dispositions that individuals have built up through the successive sites of socialisation in which they have been involved in French society, which may be contradictory or very diverse (Mauger, 2023). In this article, we will examine the role of adapted physical activity upon medical prescription (APAP) for individuals with chronic illnesses or other long-term medical conditions. Our analysis draws on comparative work conducted in five regions across mainland France. APAP is a range of programmes designed for individuals with moderate functional limitations who are not currently engaged in physical activity. In all of the programmes, a particular focus is placed on 'bridging programmes' (e.g. 'ateliers passerelles'), which are designed to provide a transitional period at the end of which participants are expected to continue practising in a mainstream context. These programmes are generally funded by decentralised state services.They exhibit considerable heterogeneity with regard to both duration and the methods of exercise employed. APAP occupies a singular position in France: although it is in the process of being integrated into the medical field, it remains relatively unknown or unrecognised by general practitioners (GPs) who are expected to prescribe it. It was initially limited to individuals with the long-term medical and administrative status of Affection de Longue Durée (ALD), entitling them to reimbursement of long-term medication. In 2023, it was expanded to encompass a broad array of chronic diseases. However, no social security coverage has been provided, which reflects the incomplete inclusion of this practice in the legitimate therapeutic system. This field of activity is directly relevant to the theme of this issue 1 , as it lies at the intersection of two distinct fields-medicine and sports-in which body weight is subject to control, measurement, and standardisation. In the medical field, the medicalisation of overweight and obesity (Jutel, 2006; Lupton, 2018; Rail, 2012) due to the pathologies objectively associated with them or likely to be so (Murray, 2008) contributes to the classification of 'healthy weight' as a health standard. A number of studies have explored the multifaceted consequences of the social and moral stigma of overweight and obesity as a result of health standards (Carof, 2017; Lang et al., 2008). As pioneer in France, Baszanger (1986) showed that GPs' professional expertise intersects with patterns associated with their social background. In various forms, class contempt persists during medical consultations with individuals classified as overweight or obese (Carof, 2015). Meanwhile, the field of sport is structured by standards of performance, mostly combined with those of youth, thinness, and muscular bodies. Several studies have explored the effects of injunctions to be thin, in almost all sports and at all ages (cf., in particular, Byrne & McLean, 2002). At the crossroads between medicine and sport, APAP, a product of recent health policies, has been developed in France in conjunction with policies related to nutrition (Boubal, 2018), namely successive 'national nutrition and health policy' plans and their focus on 'preventing sedentary lifestyles' and promoting physical activity (Clément, Genolini, 2010). The communication and slogans used appear to be aimed particularly at 'fat bodies', which are gradually becoming associated with unhealthy lifestyles (Vigarello, 2013), while the construction of obesity as a social problem gains strength (Grossetête, 2015), in France as in other Western countries (Stern, 1997; Rail, 2012; Wright, Harwood, 2012). 1 We would like to thank the reviewers for their careful reading and suggestions, which have greatly contributed to improve our analysis. L'APAP occupe une place singulière en France : bien qu'elle soit en cours d'intégration dans le domaine médical, elle reste relativement méconnue ou non reconnue par les médecins généralistes qui sont censés la prescrire. Elle était initialement réservée aux personnes bénéficiant du statut médical et administratif d'Affection de Longue Durée (ALD), qui leur donne droit au remboursement des médicaments à long terme. En 2023, elle a été étendue à un large éventail de maladies chroniques. Cependant, aucune couverture sociale n'a été prévue, ce qui reflète l'intégration incomplète de cette pratique dans le système thérapeutique légitime. Ce domaine d'activité est directement lié au thème de ce numéro 1 , car il se situe à l'intersection de deux domaines distincts, la médecine et le sport, dans lesquels le poids corporel est soumis à un contrôle, à une mesure et à une normalisation. Plusieurs études ont exploré les conséquences multiples de la stigmatisation sociale et morale du surpoids et de l'obésité résultant des normes de santé (Carof, 2017 ; Lang et al., 2008). Pionnier en France, Baszanger (1986) a montré que l'expertise professionnelle des médecins généralistes recoupe des schémas associés à leur milieu social. Sous diverses formes, le mépris de classe persiste lors des consultations médicales avec des personnes classées comme en surpoids ou obèses (Carof, 2015). Parallèlement, le domaine du sport est structuré par des normes de performance, souvent associées à celles de la jeunesse, de la minceur et de la musculature. Plusieurs études ont exploré les effets des injonctions à la minceur, dans presque tous les sports et à tous les âges (cf. notamment Byrne & McLean, 2002). À la croisée de la médecine et du sport, l'APAP, produit des politiques de santé récentes, s'est développée en France en lien avec les politiques liées à la nutrition (Boubal, 2018), à savoir les plans successifs de « politique nationale de nutrition et de santé » et leur accent sur la « prévention de la sédentarité » et la promotion de l'activité physique (Clément, Genolini, 2010).
