Citicoline Triggers Proteome Remodeling and Proteostatic Adaptation: Evidence from Shotgun Proteomics
International audienceBackground/Objectives: Citicoline, also known as CDP-choline, is a nootropic agent currently used in the treatment of glaucoma and is undergoing evaluation as a first-line therapy in a multi-center, international, phase III, randomized clinical trial involving citicoline eyedrops (ClinicalTrials.gov ID: NCT05710198). Numerous clinical and preclinical studies have linked the neuroenhancement and neuroprotective effects of citicoline to its role as a metabolic precursor for structural and functional components of cell membranes (such as phosphatidylcholine and sphingomyelin) and for neurotransmitters (e.g., acetylcholine and dopamine). However, compelling evidence suggests that the molecular mechanisms underlying its cytoprotective activity involve additional as-yet uncharacterized pharmacological actions. Methods: To further elucidate its pharmacology, we investigated the effect of two cytoprotective doses of citicoline (0.1 mM and 1 mM) on the global proteome of neuroblastoma cells using an unbiased shotgun proteomics approach. Results: With over 4000 unique proteins identified and quantified per experimental condition, the proteomics analysis revealed that citicoline, after 6 h of stimulation, induces a profound and robust remodeling of the intracellular proteome compared to untreated cells. Importantly, this effect was observed to significantly diminish by 18 h of stimulation, highlighting its transient nature (data are available via ProteomeXchange with identifier PXD061053). The clustering and rationalization of proteins upregulated by citicoline treatment identified the enrichment of key pathways for mRNA splicing, protein translation, proteostasis balance through the ubiquitin proteasome system (UPS), and mitochondrial metabolism. Conclusions: These proteomics findings introduce previously uncharacterized biological effects of citicoline and foster the working hypothesis that this drug may exert its cytoprotective activity through molecular mechanisms linked to the hormesis principle. These data further support the rationale for its clinical application in neurodegenerative processes and human disorders characterized by proteotoxicity

National audienceCurricula, national assessments, and initial and ongoing teacher training demonstrate a reductive focus on learning, revealing a rather rigid conception of primary school teaching that aims to standardize it. We propose moving beyond this reductive approach so that schools can enable all students to share a common culture by empowering teachers to fully practice their profession.Programmes, évaluations nationales, formation initiale et continue des maîtres manifestent une focalisation réductrice des apprentissages révélatrice d'une conception peu "libérale" des enseignements à l'école primaire, qui vise à les standardiser. Nous proposons de sortir de cette approche réductrice pour que l'école permettre à tous les élèves de partager une culture commune en redonnant aux enseignants la possibilité de faire pleinement leur métier

L'éco-responsabilité en tant que concept dans le cadre des organisations commerciales et éducatives
International audienceThis paper analyses and presents the results of the commercial use of the term eco-responsibility in Spanish (Latin America and Spain) and French in a corpus of websites belonging to business organisations as a starting point for a reflection from a political philosophy perspective on the potential of a coherent eco-responsibility construct that can mobilise organisational change. Following the discursive analysis, a conceptual framework for working on organisational eco-responsibility is proposed, based on reflection on social justice, environmental justice, the notion of responsibility, ecological footprint and planetary boundaries.Esta comunicación analiza y presenta los resultados de la utilización comercial del término eco-responsabilidad en castellano (América Latina y España) y en Francés en un corpus de páginas webs de organizaciones empresariales como punto de partida de una reflexión desde la filosofía política sobre el potencial de un constructo de eco-responsabilidad coherente que puede movilizar el cambio organizacional. Luego del análisis discursivo, se propone un marco conceptual de trabajo sobre la eco-responsabilidad organizacional construida a partir de la reflexión sobre justicia social, justicia ambiental, la noción de responsabilidad, huella ecológica y límites planetarios.Cette communication analyse et présente les résultats de l'utilisation commerciale du terme « éco-responsabilité » en espagnol (Amérique latine et Espagne) et en français dans un corpus de sites web d'organisations entrepreneuriales comme point de départ d'une réflexion philosophique et politique sur le potentiel d'un concept cohérent d'éco-responsabilité susceptible de mobiliser le changement organisationnel. Exploring effects of embodied learning on students’ perception on astronomy and science
International audienceEmbodiment has recently become of interest for educational environments in general and science education in particular. The present study conducted in several classrooms across Europe set out to explore what effects this approach can have on the self-reports on science interest, engagement in activities, social appreciation and knowledge gains. Change in self-report regarding these areas were evaluated in a pre-post design. The learning sequence was found to positively impact students’ perceptions of scientific knowledge gained and increased the self-reported interest during the lessons across all participating countries. The results show that embodied learning has the potential to positively impact science lessons
ALMA millimetre-wavelength imaging of HD 138965: new constraints on the debris dust composition and presence of planetary companions
International audienceHD 138965 is a young A type star and member of the nearby young Argus association. This star is surrounded by a broad, bright debris disc with two temperature components that was spatially resolved at f ar-infrared w avelengths by Herschel . Here, we present Atacama Large Millimiter/Submillimeter Array (ALMA) millimetre-wavelength imaging of the cool outer belt. These reveal its radial extent to be 150 + 10 -7 au with a width ( σ ) of 49 + 7 -6 au ( R/R = 0 . 77), at a moderate inclination of 49 .• 9 + 3 . 3 -3 . 7 . Due to the limited angular resolution, signal-to-noise and inclination we have no constraint on the disc's vertical scale height. We modelled the disc emission with both gravitational and radiation forces acting on the dust grains. As the inner belt has not been spatially resolved, we fixed its radius and width prior to modelling the outer belt. We find astronomical silicate is the best fit for the dust composition. Ho we ver, we could not reject possible scenarios where there are at least 10 per cent water-ice inclusions. Combining the spatially resolved imaging by ALMA with non-detection at optical wavelengths by HST , we obtain a limit on the scattering albedo ω ≤ 0 . 09 for the debris dust in the outer belt. Analysis of the outer belt's architecture in conjunction with simple stirring models places a mass limit of 2.3 ± 0.4 M Jup on a companion interior to the belt ( a ≤ 78 au), a factor of two impro v ement o v er constraints from high contrast imaging.</p
