Facile preparation of water soluble curcuminoids extracted from turmeric (Curcuma longa L.) powder by using steviol glucosides
AbstractCurcuminoids from rhizomes of Curcuma longa possess various biological activities. However, low aqueous solubility and consequent poor bioavailability of curcuminoids are major limitations to their use. In this study, curcuminoids extracted from turmeric powder using stevioside (Ste), rebaudioside A (RebA), or steviol glucosides (SG) were solubilized in water. The optimum extraction condition by Ste, RebA, or SG resulted in 11.3, 9.7, or 6.7mg/ml water soluble curcuminoids. Curcuminoids solubilized in water showed 80% stability at pH from 6.0 to 10.0 after 1week of storage at 25°C. The particle sizes of curcuminoids prepared with Ste, RebA, and SG were 110.8, 95.7, and 32.7nm, respectively. The water soluble turmeric extracts prepared with Ste, RebA, and SG showed the 2,2-diphenyl-1-picrylhydrazyl radical scavenging (SC50) activities of 127.6, 105.4, and 109.8μg/ml, and the inhibition activities (IC50) against NS2B-NS3pro from dengue virus type IV of 14.1, 24.0 and 15.3μg/ml, respectively
Diversity of retrievable heterotrophic bacteria in Kongsfjorden, an Arctic fjord
AbstractThe diversity and abundance of retrievable pelagic heterotrophic bacteria in Kongsfjorden, an Arctic fjord, was studied during the summer of 2011 (June, August, and September). Retrievable bacterial load ranged from 103 to 107CFUL−1 in June, while it was 104–106CFUL−1 in August and September. Based on 16S rRNA gene sequence similarities, a higher number of phylotypes was observed during August (22 phylotypes) compared to that during June (6 phylotypes) and September (12 phylotypes). The groups were classified into four phyla: Firmicutes, Actinobacteria, Proteobacteria, and Bacteroidetes. Bacteroidetes was represented only by a single member Leewenhoekiella aequorea during the three months and was dominant (40%) in June. However, this dominance changed in August to a well-known phytopathogenic species Rhodococcus fascians (32%), which could be a result of decrease in the phytoplankton biomass following the secondary bloom. It is the first report of Halomonas titanicae isolation from the Arctic waters. It showed an increase in its abundance with the intrusion of Atlantic water into Kongsfjorden. Increased abundance of Psychrobacter species in the late summer months coincided with the presence of cooler waters. Thus, the composition and function of heterotrophic bacterial community was fundamentally different in different months. This could be linked to the changes in the water masses and/or phytoplankton bloom dynamics occurring in Arctic summer
Impact of rapid diagnostic testing for chlamydia and gonorrhea on appropriate antimicrobial utilization in the emergency department
AbstractProlonged turnaround time of Neisseria gonorrhoeae (NG) and Chlamydia trachomatis (CT) test results may delay time to notification and treatment of test-positive patients and result in unnecessary antimicrobial use in test-negative patients. This quasi‐experimental study evaluated the impact of NG/CT rapid diagnostic testing (RDT) in an urban emergency department (ED) on treatment appropriateness, time to notification, and cost. Patients tested in December 2013–January 2014 (traditional group, n=200) were compared with those in December 2014–January 2015 (RDT group, n=200). There was a significant increase in treatment appropriateness in the RDT group, 72.5% versus 60% (P=0.008) and time to results notification was significantly faster (median 17.4 versus 51.5hours, P=0.010). Availability of test result prior to discharge was associated with increased treatment appropriateness (odds ratio, 22.65 [95% confidence interval, 2.86–179.68]). The RDT would save approximately $37,000 annually. These results support the use of NG/CT RDT to expand antimicrobial stewardship efforts within the ED
Negative Staining for COL4A5 Correlates With Worse Prognosis and More Severe Ultrastructural Alterations in Males With Alport Syndrome
IntroductionAlport syndrome (AS) is a genetic disorder characterized by progressive hematuric nephropathy with or without sensorineural hearing loss and ocular lesions. Previous studies on AS included mostly children.MethodsTo determine the prognostic value of loss of staining for collagen type IV alpha 5 (COL4A5) and its relationship with the ultrastructural glomerular basement membrane alterations, we performed direct immunofluorescence using a mixture of fluorescein isothiocyanate-conjugated and Texas-red conjugated antibodies against COL4A5 and COL4A2, respectively, on renal biopsies of 25 males with AS (including 16 who were diagnosed in adulthood).ResultsAll patients showed normal positive staining of glomerular basement membranes and tubular basement membranes for COL4A2. Of the 25 patients, 10 (40%) patients showed loss of staining for COL4A5 (including 89% of children and 13% of adults) and the remaining 15 (60%) had intact staining for COL4A5. Compared with patients with intact staining for COL4A5, those with loss of staining had more prominent ultrastructural glomerular basement membrane alterations and were younger at the time of biopsy. By Kaplan-Meier survival analysis and Cox regression analysis, loss of staining for COL4A5 predicted earlier progression to overt proteinuria and stage 2 chronic kidney disease or worse. By multivariate Cox regression analysis, loss of staining for COL4A5 was an independent predictor of the development of overt proteinuria and stage 2 chronic kidney disease or worse.DiscussionThus, the COL4A5 expression pattern has an important prognostic value and it correlates with the severity of ultrastructural glomerular basement membrane alterations in males with AS. Loss of COL4A5 staining is uncommon in patients with AS diagnosed in their adulthood
Evaluation of recombinant antigens in combination and single formula for diagnosis of feline toxoplasmosis
AbstractCats are the only definitive hosts of Toxoplasma gondii and constitute an essential source of infection to all warm blooded animals and humans. Diagnosis of T. gondii infection in cats is fundamental for proper management and control of infection in humans and animals. In the current study, we have evaluated the diagnostic performance of tachyzoite lysate antigen (TLA) and different T. gondii recombinant antigens including surface antigen 2 (SAG2), dense granule proteins 2, 6, 7, 15 (GRA2, GRA6, GRA7, GRA15) and microneme 10 protein (MIC10) in immunoglobulin G enzyme linked-immunosorbent assay (IgG ELISA) using cat serum samples, with reference to latex agglutination test (LAT). Remarkably, TLA showed better performance than other recombinant antigens in IgG ELISAs as compared to LAT, with concordance and Kappa values of 94.27% and 0.93, respectively. Furthermore, to improve the reactivity of the recombinant antigens, we have developed IgG ELISAs using different combinations with these recombinant antigens. Strikingly, a combination of SAG2 and GRAs has relatively similar performance as TLA evidenced by concordance and Kappa values of 94.27% and 0.81, respectively. The developed ELISA with a combination of recombinant antigens can be used as a promising diagnostic tool for routine testing of T. gondii infection and mass screening in cats. The major advantages of this assay are the high sensitivity and specificity, lower cost, safer production and easiness of standardization in various laboratories worldwide
On mechanism of antioxidant effect of fullerenols
AbstractFullerenols are nanosized water-soluble polyhydroxylated derivatives of fullerenes, specific allotropic form of carbon, bioactive compounds and perspective pharmaceutical agents. Antioxidant activity of fullerenols was studied in model solutions of organic and inorganic toxicants of oxidative type – 1,4-benzoquinone and potassium ferricyanide. Two fullerenol preparations were tested: С60О2–4(ОН)20–24 and mixture of two types of fullerenols С60О2–4(ОН)20–24+С70О2–4(ОН)20–24. Bacteria-based and enzyme-based bioluminescent assays were used to evaluate a decrease in cellular and biochemical toxicities, respectively. Additionally, the enzyme-based assay was used for the direct monitoring of efficiency of the oxidative enzymatic processes. The bacteria-based and enzyme-based assays showed similar peculiarities of the detoxification processes: (1) ultralow concentrations of fullerenols were active (ca 10–17–10−4 and 10–17–10−5g/L, respectively), (2) no monotonic dependence of detoxification efficiency on fullerenol concentrations was observed, and (3) detoxification of organic oxidizer solutions was more effective than that of the inorganic oxidizer. The antioxidant effect of highly diluted fullerenol solutions on bacterial cells was attributed to hormesis phenomenon; the detoxification was concerned with stimulation of adaptive cellular response under low-dose exposures. Sequence analysis of 16S ribosomal RNA was carried out; it did not reveal mutations in bacterial DNA. The suggestion was made that hydrophobic membrane-dependent processes are involved to the detoxifying mechanism. Catalytic activity of fullerenol (10−8g/L) in NADH-dependent enzymatic reactions was demonstrated and supposed to contribute to adaptive bacterial response
Developing an online cooperative police patrol routing strategy
AbstractA cooperative routing strategy for daily operations is necessary to maintain the effects of hotspot policing and to reduce crime and disorder. Existing robot patrol routing strategies are not suitable, as they omit the peculiarities and challenges of daily police patrol including minimising the average time lag between two consecutive visits to hotspots, as well as coordinating multiple patrollers and imparting unpredictability to patrol routes. In this research, we propose a set of guidelines for patrol routing strategies to meet the challenges of police patrol. Following these guidelines, we develop an innovative heuristic-based and Bayesian-inspired real-time strategy for cooperative routing police patrols. Using two real-world cases and a benchmark patrol strategy, an online agent-based simulation has been implemented to testify the efficiency, flexibility, scalability, unpredictability, and robustness of the proposed strategy and the usability of the proposed guidelines
An approach to prospective consequential life cycle assessment and net energy analysis of distributed electricity generation
AbstractIncreasing distributed renewable electricity generation is one of a number of technology pathways available to policy makers to meet environmental and other sustainability goals. Determining the efficacy of such a pathway for a national electricity system implies evaluating whole system change in future scenarios. Life cycle assessment (LCA) and net energy analysis (NEA) are two methodologies suitable for prospective and consequential analysis of energy performance and associated impacts. This paper discusses the benefits and limitations of prospective and consequential LCA and NEA analysis of distributed generation. It concludes that a combined LCA and NEA approach is a valuable tool for decision makers if a number of recommendations are addressed. Static and dynamic temporal allocation are both needed for a fair comparison of distributed renewables with thermal power stations to account for their different impact profiles over time. The trade-offs between comprehensiveness and uncertainty in consequential analysis should be acknowledged, with system boundary expansion and system simulation models limited to those clearly justified by the research goal. The results of this approach are explorative, rather than for accounting purposes; this interpretive remit, and the assumptions in scenarios and system models on which results are contingent, must be clear to end users
Spatial processes and politics of renewable energy transition: Land, zones and frictions in South Africa
AbstractThis paper seeks to make a contribution to on-going debates about how to conceptualise the spatial processes of renewable energy transition. It makes a case for understanding renewable energy transitions as simultaneously spatial and political processes, constitutive of new territories and configuring development pathways. Drawing on a case study of South Africa's Renewable Energy Independent Power Procurement Programme (REI4P), the paper explores the ways in which energy transitions are intrinsically bound up with both the materiality and the historical and contemporary politics of land. It then examines the relationship between energy transitions and territory to conceptualise the ways in which transitions take on an experimental shape in the form of 'zones'. The paper argues that these zones are new territories deploying forms of spatial and political-administrative exceptionality, which allow political and economic actors to exercise authority and commercial power. Two types of zone emerging from South Africa's energy transition exemplify these processes: legally-defined zones for the development of solar and wind energy and zones of socioeconomic development required by REI4P. The paper explores the spatial and political consequences of these strategies and suggests that these may not necessarily translate into conflict and confrontation, but instead produce uneasy co-existences of different political, social and spatial projects and interests, with potential to create new polities
