Letter to the Editor: Comment on the recent article titled “Diagnostic and economic evaluation of matrix-assisted laser desorption/ionization time-of-flight mass spectrometry with machine learning for screening of Johne's disease from dairy cow serum”
International audienceWe read with great interest the recent article evaluating MALDI-TOF MS combined with machine learning for Johne's disease screening (Sarkar et al., 2026). Although the study is innovative, we believe that both the diagnostic and economic assessments rely on assumptions that substantially undermine the validity of the conclusions.From a methodological standpoint, the diagnostic evaluation suffers from several weaknesses: the limited sample size associated with the risk of overfitting when applying machine learning to complex proteomic profiles, and the lack of clarity on animal selection, which raises concerns about representativeness relative to the intended clinical context. Moreover, the study reports only relative sensitivity (Se) and specificity (Sp) of MALDI-TOF compared with the ELISA, not to the true infection status.Despite this, the economic model assumes absolute Se and Sp of 80% and 83% for MALDI-TOF, and 50% and 99% for the ELISA. These assumptions concerning the MALDI-TOF are unjustified. Given that the MALDI-TOF Se and Sp are only relative to ELISA, its true Se and Sp should-at best-be the product of both: approximately 0.8 × 0.5 = 40% for Se and 0.83 × 0.99 ≈ 82% for Sp. Even this computation presumes that the 27% MALDI-TOF "false positives" are indeed false, and not animals misclassified by the imperfect ELISA reference.</p

Bis(monoacylglycero)phosphate protects murine RAW 264.7 macrophages against 7-Ketocholesterol-induced apoptosis and impaired autophagy
International audienceAtherosclerosis, a leading cause of cardiovascular disease, is driven by the accumulation of oxidized low-density lipoproteins (oxLDL) in arterial walls. 7-Ketocholesterol (7KC), a major oxysterol found in oxLDL and atherosclerotic plaques, triggers multiple cell injuries including loss of lysosomal integrity, oxidative stress, apoptosis, and impaired autophagy in vascular cells. Bis(monoacylglycero)phosphate (BMP), also known as lysobisphosphatidic acid, is a unique phospholipid concentrated in the endolysosomal compartment, known to regulate vesicle dynamics, lysosomal enzyme activities, intracellular cholesterol trafficking and its oxidative metabolism. Using a validated model of BMP enrichment in murine RAW 264.7 macrophages, we investigated whether BMP could exert protective activity against 7KC-induced damage. Our findings revealed that BMP enrichment provides comprehensive protection against 7KC at the cellular level by preserving cell viability, morphology, and neutral lipid balance. Mechanistically, BMP enrichment prevented apoptosis by maintaining mitochondrial integrity and blocking caspase activation. This was demonstrated by normalized BAX/BCL2 ratios, preserved pro-Caspase-3 levels, and reduced PARP cleavage. Remarkably, BMP enrichment also restored autophagic flux, thereby preventing the pathological accumulation of LC3-II and p62 that characterizes autophagy dysfunction. Enhanced colocalization between LC3 and BMP suggests direct functional interactions in the stress response. Gene expression analysis confirmed that BMP enrichment normalized the transcriptional dysregulation of key autophagy regulators, including Sqstm1, Becn1, and Pink1. Taken together, these results suggest that BMP is an endogenous protective factor that counteracts 7KC-induced cellular damage at multiple steps by regulating cell death and autophagy pathways in a coordinated manner
Who are the healthcare professionals involved in interprofessional team meetings in French multidisciplinary primary care centres? A quantitative analysis of eight centres
International audienceBACKGROUND: Interprofessional collaboration is crucial for providing high-quality care to patients with complex conditions in primary care. In France, multidisciplinary primary care centres (MPCC) receive funding if they organise at least 6 interprofessional team meetings (ITM) per year to discuss complex patient situations and collectively define care strategies. It remains unclear how ITM have been implemented in France. This study analyses healthcare professionals’ involvement in ITMs within MPCCs.METHOD: A multicenter retrospective quantitative study in 8 French MPCCs based on the analysis of ITM reports for the period from 2018 to 2019 was conducted. RESULTS: 1733 patients’situations (n = 1733 cases) discussed during ITMs were analysed. The 8 MPCCs were heterogeneous in terms of geographical location, creation date, and size (3 MPCCs with > 20,000 patients followed by more than 60 professionals and 3 MPCCs with < 20 professionals). On average, five healthcare professionals attended each ITM, with huge variations among MPCCs. The nurse-general practitioner (GP) pair was central. At least one GP was present at 89% (n = 1469) and nurses at 38.15% (n = 630) of ITMs. Participation of other professionals was less frequent and varied according to the MPCC. Physiotherapists were present at 7.8% of ITMs, and other healthcare professionals were present at < 6% of ITMs, including pharmacists (3% of ITMs). In some cases, healthcare professionals external to MPCC, particularly those related to mental health, were also involved in ITMs.CONCLUSIONS: Depending on the MPCC, the professionals involved in ITM vary widely, with the nurse-GP pair at the centre. Interpersonal Violence in French Sports Clubs: Who Are the Victims?
International audienceThe purpose of this study was to provide an accurate depiction of interpersonal violence (IV) in French sports. Thus, the aim of this study was to (a) measure the prevalence of IV experienced by French athletes since joining their current club, (b) accurately describe the victim's profile based on several risk factors, (c) and investigate IV according to a specific timeframe. A total of 2,250 athletes completed the French adaptation of the IV in Sport. Our results indicate a high prevalence of IV in the sporting environment, with 59.7% of participants reporting exposure to at least one form of IV and 3.8% reporting exposition to all forms of IV, highlighting the scale of this phenomenon in French sports clubs. Type of sport was the most influential risk factor for experiencing physical violence while the level of competition appeared to increase the experience of psychological violence, neglect and sexual violence. The results point to the relevance of measuring IV in a specific context and during a specific timeframe, even when accounting for the fact that participants who reported IV were significantly more likely to abandon the questionnaire before completing it
La conversion des données est-elle importante ? Évaluation de son impact sur la métabolomique non ciblée LC-MS
International audienceUntargeted LC–MS metabolomics generates highly complex datasets, making chemometric methods essential for exploring and interpreting the data. Although much attention has been given to signal detection, normalization, and statistical modeling, the initial step of data conversion — especially the transformation from profile to centroid mode — is rarely examined, even though it directly influences variance structure and relationships between variables.In this study, biological samples were analyzed in profile mode and then converted into centroided data using three common approaches: a wavelet‑based method, a vendor‑embedded algorithm, and a proprietary converter. Chemometric analyses applied to these converted datasets reveal that each centroiding strategy significantly alters the multivariate structure.Differences were observed in point density, intensity distributions, and the amount of retained information. These discrepancies affected PCA and t‑SNE representations, where samples grouped more according to the conversion method than their biological origin. Supervised models also showed that the discriminant regions of the dataset varied depending on the algorithm used, potentially leading to biased interpretations.Overall, the results demonstrate that data conversion is not a neutral preprocessing step. It strongly shapes the statistical structure of LC–MS data and can introduce hidden variability. Therefore, centroiding choices must be explicitly considered within chemometric validation workflows to improve the robustness and interpretability of metabolomics analyses
AntFlie: Frugal Visual Teach and Repeat on Narrow FoV Micro-Drones
International audienceWe present an insect-inspired visual teach-and-repeat framework demonstrated on Antflie, a 33-gram MAV equipped with an ultra-low-resolution camera (24×24 px) and a narrow 87° field of view (FoV). During a one-shot teach flight along an outbound route, the MAV performs periodic physical scans and uses a local compass based on inertial and optic flow cues to categorize views as left or right relative to the path, storing compact, lateralized visual memories in a Mushroom Body (MB) neural network with a footprint under 4 kB. In the repeat phase, the MAV flies the inbound route by retracing the outbound path, and autonomously lands at its home location using only visual familiarity through direct sensorimotor coupling, rather than map-based reasoning. Offline simulations show that the Route Lateralized (R-Lat) algorithm in Antflie matches the accuracy of a state-of-the-art insect visual compass (V-Comp) while running up to 20× faster and supporting narrow FoVs. Real-world indoor experiments further demonstrate 24 autonomous inbound repeats totaling 110 meters of flight, with a 13-cm median lateral error and a mean landing error of 34 cm. These results highlight the feasibility of frugal, bio-inspired, vision-only navigation for MAVs operating under strict size, weight, power, and cost constraints, inspired by the navigation of Cataglyphis and Melophorus ants
Bilateral parking procedures
We introduce the class of bilateral parking procedures on the integer line. While cars try to park in the nearest available spot to their right in the classical case, we consider more general parking rules that allow cars to use the nearest available spot to their left. We show that for a natural subclass of local procedures, the number of corresponding parking functions of length r is always equal to . The setting can be extended to probabilistic procedures, in which the decision to park left or right is random. We finally describe how bilateral procedures can naturally be encoded by certain labeled binary forests, whose combinatorics shed light on several results from the literature
