    Putting earthworm conservation on the map: Shortfalls and solutions for developing earthworm conservation

    International audienceEarthworms are keystone organisms that influence both soil function and community assembly of other soil organisms. However, soils are increasingly threatened by global change, so there is an urgent need to consider earthworms in conservation strategies. Earthworm monitoring has been promoted in numerous European and country research programs, and the global interest in earthworm conservation is rising, resulting in a rapid increase in the availability of earthworm data. However, most research focuses on a limited number of local-scale indicators, mainly based on abundance, biomass, and species richness of assemblages along with Bouché's ecological categories. We argue that these metrics are insufficient to effectively address earthworm conservation issues. We suggest four ecological characteristics which may be more informative for the development of conservation plans. Measurement of how much a species is (i) rare or common, (ii) native/nonnative, endemic and invasive, (iii) a specialist or generalist, and (iv) a winner or loser in the Anthropocene are all promising tools to support earthworm diversity conservation. These metrics could also be applied to functional traits, but better definition of these traits is fundamental. Finally, we emphasize the need to broaden spatial scales in earthworm studies by analyzing alpha, beta and gamma components of diversity, as local diversity alone can be misleading

    Comparative genomics and epigenomics of transcriptional regulation

    International audienceTranscriptional regulation in response to diverse physiological cues involves complicated biological processes. Recent initiatives that leverage whole genome sequencing and annotation of regulatory elements significantly contribute to our understanding of transcriptional gene regulation. Advances in the data sets available for comparative genomics and epigenomics can identify evolutionarily constrained regulatory variants and shed light on noncoding elements that influence transcription in different tissues and developmental stages across species. Most epigenomic data, however, are generated from healthy subjects at specific developmental stages. To bridge the genotype-phenotype gap, future research should focus on generating multidimensional epigenomic data under diverse physiological conditions. Farm animal species offer advantages in terms of feasibility, cost, and experimental design for such integrative analyses in comparison to humans. Deep learning modeling and cutting-edge technologies in sequencing and functional screening and validation also provide great promise for better understanding transcriptional regulation in this dynamic field

    Metabolomic analysis of a core collection of Brassica rapa and Brassica oleracea unveils unexpected chemical diversity with potential applications in chemical ecology and breeding

    International audienceSpecialized metabolism plays a crucial role in plant interactions with biotic and abiotic environments, and its characterization in crops is an essential objective for optimizing ecosystem services in breeding programs. However, our knowledge of specialized metabolism in cultivated Brassica species remains insufficient to fully support these breeding strategies. In this study, we conducted a detailed exploration of both constitutive and stress-induced specialized metabolites in response to CuCl₂ in the leaves and roots of 20 Brassica rapa and B. oleracea accessions using UHPLC-HRMS/MS-based untargeted metabolomics. Our analysis revealed a wide range of species-specific chemical signatures and allowed the putative annotation of nearly 200 compounds belonging to various chemical families, including flavonoids, glucosinolates, phenolamides, indoles, amino acids, oxylipins and coumarins. A detailed interpretation of ion fragmentation patterns uncovered both inter- and intra-specific structural variations within these chemical families, providing deeper insights into the phytochemical diversity of B. rapa and B. oleracea species. This study also enabled the identification of previously unreported compounds in these species, including the carotenoid-derivatives blumenols, flavonols decorated with rhamnoside moieties or pentahexosylated flavonols. Finally, we showed that these resources, combined with the high-quality genetic data available for these accessions, allowed the identification of genetic variants of MAM2 genes possibly involved in the control of 4C or 3C/4C-aliphatic glucosinolate chemotypes among B. oleracea accessions. This work thus lays the foundation for future studies on functional genomics of specialised metabolism in Brassica species in a chemical ecology perspective, and aiming to harness metabolic diversity in breeding programs

    Ce numéro traite de résultats de projets lauréats en 2019 l'appel à projet CASDAR (Compte d'Affectation Spéciale Développement Agricole et Rural), "Innovation et Partenariat" et "Recherche Technologique. Ces projets sont financés par le ministère de l'Agriculture et de la Souveraineté Alimentaire.National audienceThe RenouVeau project aimed to explore various innovative veal production methods, focusing on housing and feeding practices to meet new societal expectations. Consumers, animal welfare NGOs, and agriculture students were consulted to gain insight into their vision of veal production in the future. The main expectation from these groups is to provide animals with outdoor access. As a result, different veal production systems that include outdoor access were tested on an experimental farm. Observations showed that calves actively use this new space and move around more compared to actual indoor systems. However, abnormal behaviours are still present, and providing straw bedding significantly increases straw consumption. Carcasses and meat produced in these new systems are more coloured (redder) which creates challenges for product marketability. All project results have been widely disseminated, including a final webinar available at https://idele.fr/renouveau/publications/.Le projet RenouVeau a visé à étudier différents modes de production de veaux de boucherie, innovants du point de vue du logement et des conduites alimentaires appliqués, afin de répondre aux nouvelles attentes sociétales. Des consommateurs, ONG impliquées pour le bien-être animal et étudiants en agriculture ont été interrogés sur leur vision de la production de veau dans le futur. L'attente principale pour ces différents publics est de permettre aux animaux d'avoir un accès à l'extérieur. Différents modes de production de veaux de boucherie avec accès extérieur ont ainsi été testés en ferme expérimentale. Il a été constaté que les veaux occupent ce nouvel espace et se déplacent davantage comparé à un élevage en bâtiment fermé. Les comportements stéréotypés restent néanmoins présents et la présence de litière paillée pour le couchage entraine une consommation importante de paille. Les carcasses et les viandes produites dans ces nouveaux systèmes ressortent plus colorées, générant des problèmes de valorisation de ces produits. L'ensemble des résultats obtenus dans le projet a fait l'objet de nombreuses diffusions, dont la réalisation d'un webinaire 1 final disponible sur https://idele.fr/renouveau/publications/

    The Impact of Fat Graft Reconstructive Surgery on Prognosis in Young Women with Breast Cancer: A Study from the French Insurance Databases

    International audienceBackground: Fat grafting (FG) is widely used in breast reconstructive surgery (RS) after mastectomy or lumpectomy for breast cancer (BC); however, concerns persist about its oncologic safety. This study evaluated the impact of FG reconstruction compared with other techniques on BC survival in young women. Methods: The authors identified patients aged 18 to 45 years treated for nonmetastatic BC in the French National Healthcare System Database between January 1, 2010, and December 31, 2018. Patients undergoing FG were matched with patients undergoing other RS on the basis of BC subtype, chemotherapy, radiotherapy, and RS timing after BC surgery. Analyses were stratified on breast surgery (mastectomy or lumpectomy). Results: A total of 50,178 patients were included: 28,260 with luminal tumors, 7982 with triple-negative BC, 7087 with HER2-positive tumors, and 6849 with undefined tumors. In the mastectomy and lumpectomy group, 7333 and 883 patients, respectively, received FG. In the mastectomy group, after matching, FG was associated with better disease-free survival (DFS) (hazard ratio, 0.76 [95% CI, 0.65 to 0.88]) and overall survival compared with patients with other RS. FG also showed improved DFS (hazard ratio, 0.59 [95% CI, 0.51 to 0.69]) compared with patients without any RS. No significant differences in DFS or overall survival were found in the lumpectomy group. Conclusions: FG appears to improve survival outcomes in young women undergoing mastectomy with or without additional RS. In contrast, no significant impact was observed in the lumpectomy group. These findings suggest that FG can be a safe reconstructive option, particularly after mastectomy. Further research is needed to confirm these results and understand the underlying mechanisms.     La loi Climat et résilience de 2021 a instauré la mise en place d’un affichage environnemental sur les produits alimentaires afin d’informer les consommateurs du coût environnemental de leurs achats. La construction de cet affichage a suscité un important travail méthodologique ouvert aux acteurs. Un bilan intermédiaire a souligné la difficulté à appréhender toutes les dimensions de la biodiversité. C’est dans ce cadre que les ministères en charge de la transition écologique, et de l’agriculture et de l’alimentation, ainsi que l’ADEME, ont sollicité INRAE et l’Ifremer courant 2022 pour mieux documenter ce volet biodiversité, en se focalisant sur les pratiques de production. Afin d’éclairer plus largement les politiques publiques, les pouvoirs publics ont choisi de s’appuyer sur les labels dont les cahiers des charges certifient des pratiques et parce que leur développement les place au cœur de nombreux débats sur les relations entre production et consommation durables. L’étude, intitulée « BiodivLabel », a été menée par un comité pluridisciplinaire d’experts scientifiques issus d’organismes publics de recherche et d’enseignement supérieur
