Surface Characterization and in situ Protein Adsorption Studies on Carbene-Modified Polymers
Polystyrene thin films were functionalized using a facile two-step chemical protocol involving carbene insertion followed by azo-coupling, permitting the introduction of a range of chemical functional groups, including aniline, hexyl, amine, carboxyl, phenyl, phosphonate diester, and ethylene glycol. X-ray photoelectron spectroscopy (XPS) confirmed the success of the two-step chemical modification with a grafting density of at least 1/10th of the typical loading density (1014–1015) of a self-assembled monolayer (SAM). In situ, real-time quartz crystal microbalance with dissipation (QCM-D) studies show that the dynamics of binding of bovine serum albumin (BSA) are different at each modified surface. Mass, viscoelastic, and kinetic data were analyzed, and compared to cheminformatic descriptors (i.e., c log P, polar surface area) typically used for drug discovery. Results show that functionalities may either resist or adsorb BSA, and uniquely influence its adsorption dynamics. It is concluded that carbene-based surface modification can usefully influence BSA binding dynamics in a manner consistent with, and more robust than, traditional systems based on SAM chemistry
Nano-in-Micro Self-Reporting Hydrogel Constructs.
Highly reproducible Nano-in-Micro constructs are fabricated to provide a well-defined and self-reporting biomimetic environment for hepatocytes. Based on a protein/hydrogel formulation with controlled shape, size and composition, the constructs enable efficient nutrient exchange and provide an adhesive 3D framework to cells. Co-encapsulation of hepatocytes and ratiometric optical nanosensors with pH sensitivity in the physiological range allows continuous monitoring of the microenvironment. The lobule-sized microbeads are fabricated using an automated droplet generator, Sphyga (Spherical Hydrogel Generator) combining alginate, collagen, decellularized hepatic tissue, pH-nanosensors and hepatocytes. The pH inside the Nano-in-Micro constructs is monitored during culture, while assaying media for hepatic function and vitality markers. Although the local pH changes by several units during bead fabrication, when encapsulated cells are most likely to undergo stress, it is stable and buffered by cell culture media thereafter. Albumin secretion and urea production are significantly higher in the microbeads compared with controls, indicating that the encapsulated Nano-in-Micro environment is conducive to enhanced hepatic function

Policies to Support Inclusive Innovation
Innovation can help fight inequality and social exclusion. But, at present, there are too many barriers to this form of “inclusive innovation”. New government policies are essential to reduce these barriers and new measures are needed to drive inclusive innovation forward. These must encourage formal innovation systems to focus on the poor; help low-income actors to adapt, diffuse and use innovations; and work to address structural roadblocks. In addition, new policy worldviews, processes and structures are required. This paper outlines a systemic approach that explains the rationale, objectives, goals, instruments and governance which will help governments support the expansion of inclusive innovation
Burden of serious fungal infections in the Czech Republic.
We have estimated the number of serious fungal infections in the Czech Republic. All published epidemiology papers reporting Czech fungal infection rates were identified. Where no data existed, we used specific populations at risk and fungal infection frequencies in those populations. Population statistics were obtained from the 2011 Census data, prevalence and incidence data for at-risk conditions were obtained from publicly accessible healthcare statistics and relevant surveys. We estimate that 152 840 Czech women suffer with recurrent vaginal thrush. Allergic bronchopulmonary aspergillosis is likely in 4739 adults and 6581 more have severe asthma with fungal sensitisation. Hypersensitivity pneumonitits secondary to fungi is estimated in 1050 cases and 365 people may have chronic pulmonary aspergillosis. Oesophageal candidiasis is estimated in 210 HIV-positive people. There are 12 cases of Pneumocystis pneumonia in HIV population and 60 more cases in non-HIV population. There are an estimated 526 cases of candidaemia, 79 cases of Candida peritonitis and 297 cases of invasive aspergillosis a year. About 176 000 (1.67%) Czech people suffer from severe fungal infections each year, predominantly from recurrent vaginitis and allergic respiratory conditions. Substantial uncertainty surrounds these estimates except for invasive aspergillosis in haematology and candidaemia in critical care
