    Proteomic Characterization and Cytotoxic Potential of Proteins from \u3cem\u3eCuscuta\u3c/em\u3e (\u3cem\u3eCuscuta epithymum\u3c/em\u3e (L.) Crude Herbal Product Against MCF-7 Human Breast Cancer Cell Line

    Background The burden of breast cancer, the second leading cause of death worldwide, is increasing at an alarming rate. Cuscuta, used in traditional medicine for different ailments, including cancer, is known for containing phytochemicals that exhibit anticancer activity; however, the bioactivities of proteins from this plant remain unexplored. This study aimed to screen the cytotoxic potential of proteins from the crude herbal product of Cuscuta epithymum(L.) (CE) harvested from the host plants Alhagi maurorum and Medicago sativa. Methods The proteins from CE were extracted using a salting-out method, followed by fractionation with a gel filtration chromatography column. Gel-free shotgun proteomics was subsequently performed for protein characterization. The viability assay using MTT was applied to deduce the cytotoxic potential of proteins against MCF-7 breast cancer cells, with further exploration of the effect of treatment on the expression of the apoptotic mediator BCL2-associated X protein (BAX) and B-cell lymphoma protein 2 (BCL-2) proteins, using western blotting to strengthen the findings from the in vitro viability assay. Results The crude proteins (CP) of CE were separated into four protein peaks (P1, P2, P3, and P4) by gel filtration chromatography. The evaluation of potency showed a dose-dependent decline in the MCF-7 cell line after CP, P1, P2, and P3 treatment with the respective IC50 values of 33.8, 43.1, 34.5, and 28.6 µg/ml. The percent viability of the cells decreased significantly upon treatment with 50 µg/ml CP, P1, P2, and P3 (P \u3c 0.001). Western-blot analysis revealed upregulation of proapoptotic protein BAX in the cells treated with CP, P3 (P \u3c 0.01), and P2 (P \u3c 0.05); however, the antiapoptotic protein, BCL-2 was downregulated in the cells treated with CP and P3 (P \u3c 0.01), but no significant change was detected in P2 treated cells. The observed cytotoxic effects of proteins in the CP, P1, P2, and P3 from the in vitro viability assay and western blot depicted the bioactivity potential of CE proteins. The database search revealed the identities of functionally important proteins, including nonspecific lipid transfer protein, superoxide dismutase, carboxypeptidase, RNase H domain containing protein, and polyribonucleotide nucleotidyltransferase, which have been previously reported from other plants to exhibit anticancer activity. Conclusion This study indicated the cytotoxic activity of Cuscuta proteins against breast cancer MCF-7 cells and will be utilized for future investigations on the mechanistic effect of active proteins. The survey of CE proteins provided substantial data to encourage further exploration of biological activities exhibited by proteins in Cuscuta

    This text presents a collection of mathematical exercises with the aim of guiding readers to study topics in statistical physics, equilibrium thermodynamics, information theory, and their various connections. It explores essential tools from linear algebra, elementary functional analysis, and probability theory in detail and demonstrates their applications in topics such as entropy, machine learning, error-correcting codes, and quantum channels. The theory of communication and signal theory are also in the background, and many exercises have been chosen from the theory of wavelets and machine learning. Exercises are selected from a number of different domains, both theoretical and more applied. Notes and other remarks provide motivation for the exercises, and hints and full solutions are given for many.     Co-Designing Situated Displays for Family Co-Regulation with ADHD Children

    Family informatics often uses shared data dashboards to promote awareness of each other’s health-related behaviors. However, these interfaces often stop short of providing families with needed guidance around how to improve family functioning and health behaviors. We consider the needs of family co-regulation with ADHD children to understand how in-home displays can support family well-being. We conducted three co-design sessions with each of eight families with ADHD children who had used a smartwatch for self-tracking. Results indicate that situated displays could nudge families to jointly use their data for learning and skill-building. Accommodating individual needs and preferences when family members are alone is also important, particularly to support parents exploring their co-regulation role, and assisting children with data interpretation and guidance on self and co-regulation. We discuss opportunities for displays to nurture multiple intents of use, such as joint or independent use, while potentially connecting with external expertise

    Bikeability Disparities in Orange County, California: Intersection of Place and Demographics

    Active transportation modes such as walking and biking are gaining popularity for their extensive health and environmental benefits, yet scholars know little about how place-based accessibility varies by area sociodemographic composition. This study is among the first to examine sociodemographic disparities (by both race and socioeconomic status) in bikeability while allowing for heterogeneity in disparities. Consideration of bikeability disparities is particularly critical within the framework of urban planning concepts that promote equitable accessibility and reduced dependency on automobiles, such as the 15-minute city. Geographically Weighted Regressions examined associations between census tract-level bikeability (using an index that combines five components), socioeconomic status, and percentage non-White residents (controlling for age of structures in tracts). Findings showed that the strength and directionality of associations between bikeability and race/socioeconomic status varied throughout the county, providing targeted information on where greater concentrations of low socioeconomic status and non-White residents were associated with lower bikeability

    LGBTQIA+ Pride Month: A Digital Library Display

    June is a time of reflection, celebration, and solidarity as we recognize LGBTQIA+ Pride Month. This annual observance commemorates the 1969 Stonewall Uprising in Manhattan, a landmark event that ignited the Gay Liberation Movement and fundamentally shaped the fight for LGBTQIA+ rights in the United States. Initially observed as Gay Pride Day on the last Sunday of June, this day of remembrance soon expanded into a month-long celebration, now attracting millions of participants across the globe. In addition to parades, concerts, and workshops, June is also a time for memorials honoring those lost to hate crimes and HIV/AIDS. In this digital display, we highlight significant individuals and provide eBooks on LGBTQIA+ health issues. There is also a list of online resources that provide more information, data, and statistics. LGBTQIA+ stands for Lesbian, Gay, Bisexual, Transgender, Queer and/or Questioning, Intersex, Asexual, and more. Although variations of the acronym are common, this is the most inclusive and is used throughout the display. This display is part of the Leatherby Libraries\u27 Diversity, Equity and Inclusion initiatives in alignment with the Chapman University Strategic Plan for Diversity & Inclusion, fostering a diverse and inclusive campus climate

    Cross-Market Effects of Consolidation: Evidence from Banking

    The U.S. banking sector had nearly 70% fewer banks in 2022 relative to 1989, primarily because of mergers. We develop a methodology to estimate cross-market spillover effects of bank mergers and test whether the operations of incumbents facing consolidating competitors in one market are affected in other markets. We find that nonmerging banks within a market that are one standard deviation more exposed to mergers in other markets increase deposits by 2.1% relative to their less exposed competitors. Our methodology may be applied elsewhere to assess the aggregate impacts of industry consolidation and illustrates challenges with product-based or geographic market definitions

    Ideology and Economic Change

    This paper revisits the old theses of the contrasting paths to modernization between Japan and China. It develops a new analytical framework regarding the role of ideology and ideological change—Meiji Japan’s decisive turn towards the West pitted against Qing China’s lethargic response to Western imperialism—as the key driver behind these contrasting paths. Our framework and historical narrative highlight the contrast between Tokugawa Japan’s feudal, decentralized political regime and Qing China’s centralized bureaucratic system as a key determinant driving the differential patterns of ideological realignment. We argue that the 1894-95 Japanese naval victory over China could not be justified under the prevailing Imperial Chinese ideology and thus served as the catalyst for China’s subsequent ideological transformation, which occurred via borrowing Japan’s successful Meiji reforms of both institutions and ideology. Our analytical framework, developed from a comparative historical narrative, sheds new insights on the importance of ideology and ideological change for our understanding of political and economic change

    Non-alcoholic Fatty Liver Disease: Role of PNPLA3 and Its Association with Environmental Chemicals

    Globally, non-alcoholic fatty liver disease (NAFLD) is on the rise with 30-32%, 27-33%, 35-48%, 36%, 9-20%, and 36-38% prevalence in Asia, Europe, North America, South America, Africa, and Australia, respectively. Approximately, 5-10% of NAFLD proceeds to hepatitis called non-alcoholic steatohepatitis (NASH). NASH often progresses to liver fibrosis, cirrhosis, and hepatocellular carcinoma (HCC). Precise mechanism(s) for the development of HCC is not fully understood in NAFLD and NASH patients. Higher insulin levels in type 2 diabetes (T2D) can result in lipolysis of adipose tissue activating lipid synthesizing enzymes such as fatty acid synthase and stearoyl-CoA desaturase-1, resulting in lipid accumulation in liver. Higher levels of glucose in T2D patients activate carbohydrate response element binding protein and insulin which increases the level of active sterol regulatory element binding protein. These lipogenic transcription factors activate patatin-like phospholipase domain-containing protein-3 (PNPLA3) from their response elements in the promoter. Endocrine disrupting chemicals (EDCs) and persistent organic pollutants (POPs) are implicated in T2D development and NAFLD. The emerging association between POPs, including insecticides, fungicides, herbicides, and diabetes has been noted. However, their connection with NAFLD remains less evident. Here, we reviewed association of POPs, especially EDCs, and role of PNPLA3 in the development of NAFLD and NASH.
