    Retrospective study of tumors from cattle slaughtered in Lombardy (Italy): preliminary evaluation on the establishment of a bovine cancer registry

    Forty‑one tumors were detected in a population of 1,649,003 cattle slaughtered in 4 abattoirs in Lombardy over a 5‑year period, for an overall prevalence of 2.5 tumors per 100,000 cattle. Tumors were classified according to the WHO histological classification of tumors of domestic animals. Alimentary and hemopoietic systems were commonly affected with 9 cases each. Other affected sites were the respiratory (n = 3), urinary (n = 2), endocrine (n = 2), musculoskeletal (n = 2), nervous (n = 1), and cardiovascular (n = 1) systems. The peritoneum was affected by 6 cases, while the primary location of 3 tumors of the connective tissues and 3 metastatic carcinomas was unidentified. Liver tumors and mesotheliomas, for which environmental risk factors are well‑known in humans, were common, as well as tumors typically encountered in pediatric human patients (tumors of mesenchymal tissues, pulmonary blastomas and nephroblastomas). These findings suggest the useful role of bovines as sentinel and model for human carcinogenesis. Our study indicates that the establishment of a bovine cancer registry in Lombardy is feasible considering its potential contribution to understanding the role of environmental risk factors in the genesis of tumors in animals and humans

    Allelic polymorphisms of the BoLA-DRB3 gene and resistance to brucellosis in Kazakh cattle

    The Authors investigated polymorphism of the bovine BoLA‑DRB3 gene in connection with resistance or susceptibility to brucellosis of two Kazakh meat breeds, Auliekol and Kazakh Whiteheaded breeds, using PCR‑RFLP. In Auliekol cattle (n = 158), 22 alleles were detected in the brucellosis group, and 24 alleles were shown in the healthy group. BoLA-DRB3 alleles *3, *4, *19, *21 were more common in healthy animals, while Brucella‑positive cattle were more frequently carriers of alleles *7, *10, *18. In Kazakh Whiteheaded cattle (n = 146), 21 alleles were detected in infected and 23 alleles in healthy cattle. Alleles *3, *8, *21 significantly predominate in healthy cattle, while alleles *7, *11, *16 are typical for animals with brucellosis. This study identified BoLA-DRB3 alleles associated with genetic resistance (*3 and *21) and susceptibility (*7) to brucellosis; remarkably, resistance alleles are shared by two important meat breeds of Kazakhstan

    The role of molecular-genetic techniques in BVDV eradication in Lower Austria

    A voluntary bovine viral diarrhoea virus (BVDV) control programme, which later became a compulsory eradication programme, based on the Swedish model was introduced in Lower Austria in 1997. The persistently infected animals were detected by Ag-ELISA and all samples were re-tested by the improved single-tube RT-PCR, employing panpestivirus primers targeting the 5’-UTR of the virus genome. In 2010, the BVDV eradication programme, which became compulsory from 2004, reached the final stage with only five remaining infected herds in which BVDV was difficult to eradicate. To resolve the problem in those herds, a molecular epidemiology approach was used. No differences in the spectrum of BVDV-1 subgenotypes at the beginning and at the final stage of eradication programme were found. The genetic study revealed the importance of human risk factor when finishing an eradication programme. Molecular epidemiology was also used to analyse BVDV isolates associated with re-introductions to BVDV-free herds

    Effects of isoflavone supplementation on endometrial thickness, endometrial hyperplasia, and cancer in ovariectomized cats

    Ovariectomy is identified as a standard treatment in different European countries. Isoflavones, as nonsteroidal compounds in plants, are common constituents of soy and soy products. Some available cat diets contain different concentrations of soy products. This study aimed to examine the effects of isoflavone supplementation on endometrial hyperplasia and endometrial thickness in ovariectomized cats. Fifteen neutered adult cats were divided into control, estradiol, and isoflavone groups (five cats per group). Subcutaneous injection of estradiol (0.5 μg) in sesame oil (100 μL) was done for 30 days in estradiol-treated cats. Isoflavone-treated cats ingested a single oral tablet of soy extract for 30 days, while the controls received subcutaneous injections of the vehicle and oral placebo for 30 days. Histopathological findings of hematoxylin and eosin-stained sections revealed a significant difference between the estradiol group and other groups in terms of hyperplastic epithelium and simple hyperplasia. Thickness of myometrium was greater in the estradiol group compared to the isoflavone and control groups. Higher concentrations of estrogen can affect the endometrium and myometrium, while 30-day ingestion of isoflavone didn't have any uterine effect.

    Transition of dominant canine parvovirus genotype from 2b to 2c in Vietnamese dogs

    Canine parvovirus (CPV) is one of the most important pathogens causing enteritis in dogs. Although there have been a few reports of CPV in Vietnam, recent information on CPV infection in domestic dogs in Vietnam is limited. Faecal samples collected from 30 diarrheic and 50 healthy dogs were examined by PCR for detection of CPV DNA. The prevalence of CPV in diarrheic dogs (43.3%, 13/30) was significantly higher than in healthy dogs (4.0%, 2/50), indicating that CPV was a major cause of diarrhoea in domestic dogs. Genotyping of 15 CPV strains showed that both CPV‑2a and CPV‑2c were circulating and that CPV‑2c was a dominant CPV variant in Vietnam. Virus isolation was performed from faecal samples using A72/cSLAM cells, and nine CPV strains were successfully isolated. The dominant genotype spreading among Vietnamese dogs has changed from CPV‑2b to CPV‑2c

    Peste des petits ruminants (PPR) is a vaccine-preventable transboundary animal disease of goats and sheep majorly, and is regarded as a major constraint to small ruminant production especially in developing countries like Nigeria. Despite different strategies that have been employed to control PPR in Nigeria, cases of the disease are still reported in PPR-vaccinated and unvaccinated small ruminant farms. In this study, molecular detection of field PPR virus (PPRV) strains was carried out to determine the presence of PPRV. A total of 135 samples (45 oculo-nasal swabs and 90 tissue samples) were purposively collected between August and October 2020 from goats and sheep at the Akinyele live small ruminant market and at Akinyele and Amosun abattoirs in Ibadan, Oyo State, Nigeria. Using reverse transcriptasepolymerase chain reaction with primers targeting the partial N-gene of PPRV, 10 out of the 135 (7.4%) field samples yielded positive results. The results of this study reveal that PPRV currently circulates in Ibadan. These findings underscore the need for continuous PPR surveillance, more extensive characterization of circulating PPRV strains and the importance of consistent use of quality vaccines in the country to achieve more effective preventive and control strategies against the disease

    Kisspeptina-10 e ormone inibitore delle gonadotropine nella gravidanza della bovina: Kisspeptina-10 e GnIH nella gravidanza della bovina

    Recently, two different molecules have been discovered to play an important role in reproduction: kisspeptin (Kp) and gonadotropin inhibiting hormone (GnIH). The aim of this study was to establish the trend of kisspeptin 10 (Kp‑10) and GnIH concentrations, during all phases of pregnancy in cattle, in order to understand their possible role in the physiology of pregnancy. To examine the correlation between these hormones and steroid hormones, cortisol and oestradiol 17β (E2) were also analyzed. Eighty pregnant cows were enrolled and the pregnancy was divided into 8 periods of 30 days each (from 30‑60 days to 240‑270 days). Blood samples were collected from all cows, once only for cow. Kp‑10, GnIH, cortisol and E2 were measured in sera. After an initial plateau, Kp‑10 concentrations increased at 90‑120 days and then decreased until 180‑210 days, undergoing a further increase until 240‑270 days. GnIH concentrations decreased until 90‑120 days, then increased until the end of gestation. These trends were opposing until 180‑210 days, whereat concentrations of both increased until the end of gestation. Cortisol concentrations were homogenous at all times, except at the final period, in which they were higher. E2 showed two peaks, at 90‑120 days and 240‑270 days. The trends in Kp‑10 and GnIH concentrations suggest that these two hormones might act to maintain the delicate endocrine equilibrium of pregnancy.Lo scopo di questo studio è stato quello di stabilire le concentrazioni di kisspeptina 10 (Kp-10) e dell’ormone inibitore delle gonadotropine (GnIH), durante tutte le fasi della gravidanza, nella bovina. Sono state prese in considerazione 80 bovine gravide e suddivise in 8 gruppi, in base all’epoca gestazionale (da 30-60 giorni a 240-270 giorni). Sono stati raccolti campioni di sangue da tutte le bovine, una volta sola per soggetto. Sono stati, quindi, misurati nel siero Kp-10, GnIH, cortisolo ed E2. Le concentrazioni di Kp-10 e GnIH hanno mostrato un trend opposto nelle diverse fasi di gravidanza, tranne nel periodo finale, in cui si è rilevato un simultaneo aumento. Le concentrazioni di cortisolo sono risultate sempre omogenee, tranne nel periodo finale, in cui sono aumentate. E2 ha mostrato due picchi, a 90-120 giorni e 240-270 giorni. Le curve di concentrazione di Kp-10 e GnIH suggeriscono che questi due ormoni potrebbero agire in sinergia per mantenere il delicato equilibrio endocrino della gravidanza

    Amelioration of bleomycin induced pulmonary fibrosis by administration of Salvianolic acid B in mice

    Pulmonary fibrosis is the end‑stage manifestation of wide range of respiratory diseases and during pulmonary fibrosis, pulmonary inflammation and epithelial‑mesenchymal transition (EMT) play important roles. Salvianolic acid B (SAB) from the herb Salviae miltiorrhiza has been reported to possess an excellent anti‑inflammatory, antifibrotic and antioxidant activity. The present study aimed to investigate the ameliorative effect of SAB on bleomycin induced pulmonary fibrosis in mice. Adult albino mice were divided as SHAM/control group (saline alone), BLM group (bleomycin @ 1mg/kg intratracheally once) and SAB groups (BLM challenged once and SAB administration in three dosages @ 5, 10 and 15 mg/kg intraperitoneally daily for 30 days). Lungs wet/dry ratio and protein concentration in bronchoalveolar lavage fluid, MPO activity, oxidative stress markers, hydroxyproline assay, levels of inflammatory cytokines (TNF‑α, IL‑6 and TGF‑β1), NF‑κB activity, histopathology, immunostaining (E‑cadherin, vimentin and alpha ‑smooth muscle actin) and ultrastructural changes were studied. SAB showed anti‑inflammatory and anti‑fibrotic effects through inhibition of inflammatory cell infiltration, alveolar structure disruption, and collagen deposition and the expression of several fibrogenic cytokines. SAB also up‑regulate E‑cadherin and down‑regulated vimentin and alpha‑smooth muscle actin expression. In conclusion, Salvianolic acid B is effective in alleviating the BLM induced lung fibrosis through suppression of oxidative stress, inflammation, histological, ultrastructural changes and EMT

    Detection of Testudinid alphaherpesvirus, Chlamydia spp., Mycoplasma spp., and Salmonella spp. in free‑ranging and rescued Italian Testudo hermanni hermanni

    Testudo hermanni is included as near‑threatened in the Red List of the International Union for Conservation of Nature, while T. hermanni hermanni is considered endangered in the Italian Red List. Appropriate management of smuggled or seized wild individuals is recommended before their reintroduction into the wild. Accordingly, a health monitoring study was carried out. During 2014‑2016, 133 oral swabs and 121 cloacal swabs were collected from a total of approximately 180 free‑ranging and rescued T. hermanni hermanni from eight different Italian regions to investigate the presence of DNA of Testudinid alphaherpesvirus (TeAHV), Chlamydia spp. and Mycoplasma spp. in the oral cavity, and Salmonella spp. isolates in the cloaca. Mycoplasma spp. was detected in 52 out of 87 (59.77%) of rescued and in 1 out of 46 free‑ranging (2.17%) individuals; 33 out of 53 (62.26%) Mycoplasma spp. positive samples were typed as M. agassizii by PCR. Salmonella spp. was isolated from 45 out of 121 (37.19%) cloacal swabs, typed into 14 serovars, and characterized for complete antimicrobial susceptibility. A significantly different distribution of Salmonella spp. isolates was found in 2016 in comparison with 2014 and 2015, without any difference between free‑ranging and rescued tortoises. All the tested tortoises were negative for TeAHV and Chlamydia spp. These results are considered a baseline information critical to monitor the dynamics of these microorganisms in free‑ranging and rescued populations of T. h. hermanni, and to correctly approach the management of rescued animals and possible relocation programs

    Malassezia overgrowth in dogs in northern Italy: frequency, body distribution, clinical signs and effects of pharmacologic treatments

    The present study describes Malassezia populations in clinically healthy dogs (HD) and dogs with Malassezia overgrowth (MO), and evaluates the correlation with clinical signs and previous treatments. Thirteen clinically HD and 84 dogs with MO were enrolled. Clinical history and previous treatments were recorded. After a complete physical and dermatological examination, Canine Atopic Dermatitis Extent and Severity Index_03 scores were calculated. Samples for cytology and mycological cultures were obtained from four body regions and from skin lesions. Malassezia overgrowth was diagnosed by cytology. A global score (GS) for quantitative evaluation of the population of Malassezia was calculated. In dogs with MO, the highest frequency of yeast detection was found in skin lesions (82%, P < 0.001). Sum of GS (GSs) obtained from dogs with MO (68, 0‑621) was significantly higher compared to those of HD (3, 0‑48, P < 0.001). GSs in dogs previously treated with antibiotics (312.5, 30‑975) was significantly higher compared to those of dogs that not have received antibiotics (80, 0‑975, P = 0.015). No difference was found between dogs treated and those not treated with steroids
