Trial by Fire: The Marshall-Bell Kiln Site in Fredericksburg, Virginia
In 2012 and 2013 community members and local professional archaeologists led by Dovetail Cultural Resource Group worked together to salvage a stoneware waster dump in Fredericksburg, Virginia. More than 17,000 artifacts were recovered, representing two successive potters, Hugh R. Marshall and Francis H. Bell. This article brings together the archaeological and documentary evidence to discuss this short-lived pottery operation of the early 1830s. Considered are the physical attributes of the vessel forms and decorations, and the broader aspects of how this pottery operated within the local community and regional markets

Tempering Our Expectations: Drinking, Smoking, and the Economy of a Western Massachusetts Farmstead-Tavern
Between 1800 and 1830, William Sanford and his family operated a tavern in Hawley, a hilltown in western Massachusetts. The establishment was located on the town’s common, adjacent to the community’s Congregational meetinghouse and several other taverns. At the initiative of the local historical preservation group the Sons and Daughters of Hawley, archaeologists, students, teachers, and community members excavated the tavern site between 2011 and 2014. Historical and archaeological research indicates that William Sanford’s homestead functioned not only as a tavern, but also as a farm, store, smithy, and occasionally a court of law. Material evidence of alcohol and tobacco consumption, however, is less pronounced than at heavily-trafficked urban taverns. Research at the Sanford Tavern and other 19th-century public houses indicates that hybrid rural establishments played a variety of social and economic roles within local communities, which is evident in the archaeological record. Our findings show that archaeologists should approach rural farmstead-taverns with a different set of expectations
GlyPro as a self-immolative spacer for the release of payloads from antibody-drug conjugates
Antibody-drug conjugates (ADCs) are an increasingly popular modality for targeted drug delivery in many oncological and immunological applications. The paratope of a monoclonal antibody (mAb) directs the delivery of a conjugated therapeutic payload to antigen expressing cells, resulting in a controlled transport of payload to a desired cell type. Internalization of the ADC followed by lysosomal degradation results in the release of a payload to perform its biochemical function. A chemical linker between the drug and the antibody is responsible for the stability of the conjugate in circulation alongside mediating the release of an unmodified payload under lysosomal conditions. Currently, the lead linker, ValCit-p-aminobenzyl carbamate (PABC), has demonstrated impressive extracellular integrity while maintaining susceptibility to lysosomal proteases, such as Cathepsin B, for controlled intracellular release.1 However, the well-studied ValCitPABC linker system is largely restricted to amine-containing payloads for immuno-modulating ADCs whereas there are few ADC linkage systems that are able to release alcohol containing payloads, regardless of their functional prevalence across a myriad of biologically active small molecules. We propose that upon cathepsin-mediated cleavage of our ADCs, the ValCit release of a GlyPro ester undergoes rapid cyclization to form a diketopiperazine, thereby releasing the alcohol-containing payload. The efficiency of this linker (ValCitGlyPro) was examined using a model system designed to release dexamethasone, a potent glucocorticoid. Kinetic studies demonstrated that our linker system results in rapid GlyPro-dexamethasone release in lysosomes, which undergoes rapid cyclization to release dexamethasone at neutral pH. After conjugation of the linker payload to targeted and untargeted antibodies, several cell assays show that this system is capable of targeted immune suppression of lipopolysaccharide (LPS) stimulated cells. A 3,5 torus knot carved from Olivewood. completed four new wooden torus knot sculptures, two from Purpleheart wood and two from Olivewood. One of the Purpleheart pieces came from a 2 x6 x6 block, and all the others were from 2 x4 x4 blocks. I made two (3,5) torus knots, one from the larger Purpleheart block and one from an Olivewood block.
