Alpha-terpineol esters: occurrence of the reaction by enzymatic route – Experimental analysis and molecular docking
This thesis chapter explores the enzymatic synthesis of ester derivatives of α-terpineol, a tertiary alcohol of high relevance in aroma chemistry. The study involved experimental screening with lipase variants and computational modeling to evaluate the reaction feasibility. Results revealed that under the tested conditions, enzymatic esterification of α-terpineol was unsuccessful, emphasizing the structural limitations imposed by its molecular conformation. Complementary chemical assays confirmed that esterification is viable via acid catalysis, reinforcing the hypothesis that steric hindrance prevents efficient biocatalysis in this case. These findings contribute to a better mechanistic understanding of enzyme-substrate interactions involving tertiary alcohols and inform future strategies for the biosynthesis of aroma-related esters

Optimized multiplex immunofluorescence for the characterization of tumor immune microenvironment in neoplastic paraffin-preserved tissues
The study of neoplastic cells enabled the discovery of important tumor-related biomarkers which resulted in new forms of early diagnosis, therapeutic options, and prognostic markers. Thus, immunofluorescence (IF), a high throughput imaging technology, represents a valuable method that enables the virtual characterization and localization of diverse cell types and targets, preserving tissue architecture and spatial surroundings. IF staining and analysis of formalin-fixed paraffin-embedded (FFPE) tissues are considered a challenge due to several difficulties, such as tissue autofluorescence, non-specific antibody binding, and image acquisition and quality. This study aimed to develop a multiplex-fluorescence staining technique with high-contrast and high-quality multiple-color images to enrich the investigation of important biomarkers. We present a robust optimized multiple-immunofluorescence procedure that reduced sample autofluorescence, enabled the use of simultaneous antibodies on the same sample, and showed super-resolution imaging through precise antigen localization. We illustrated the utility of this IF powerful method in FFPE neoplastic appendix, lymph node and bone marrow biopsies, and a 3D-coculture system, in which cells are enabled to grow and interact with their surroundings in all 3D dimensions. This optimized multiple-immunofluorescence method represents a powerful tool for better understanding the complexity of tumor cells, characterizing cell populations and spatial localization, revealing predictive and prognostic biomarkers, and identifying immunologic phenotypes in a single and limited sample. This IF represents a valuable method for tumor microenvironment profiling that could contribute to the study of cellular crosstalk and the niche, and to the identification of predictive biomarkers for neoplasms

Dataset on the enzymatic hydrolysis of rice bran protein concentrate using alcalase and flavourzyme
Below data refer to the enzymatic hydrolysis of rice bran protein concentrate (RBPC) using the commercial enzymes Alcalase and Flavourzyme. Hydrolysis assays were performed at three degrees of hydrolysis (3%, 6% and 12%), and all experiments were carried out in triplicate.
The dataset includes raw titration data obtained during pH-stat hydrolysis, using 1 M NaOH to maintain the reaction medium at the optimal pH for each enzyme. The recorded NaOH volumes correspond to the amount required to keep the reaction medium at the enzyme’s optimal pH and were used to calculate the degree of hydrolysis.
Two separate spreadsheets are provided, one for Alcalase and one for Flavourzyme. Each spreadsheet also includes information on the preparation conditions of the protein solution (solids content and heating parameters), the optimal temperature for enzyme activity, the enzyme inactivation temperature, and the corresponding hydrolysis kinetic data
