Proteomic screen of dorsal root ganglion neuron growth states reveals Glypican 1 as an autocrine and paracrine activator of axon growth
Following an injury in the adult central nervous system (CNS), axons fail to regenerate because of repressed intrinsic growth competence and a cellular environment that lacks growth support. Interestingly, injured axons of dorsal root ganglia neurons (DRG) can become growth competent and regenerate under specific conditions. The molecular program underlying their regenerative growth, however, has remained unclear. Thus far, studies of growth and regeneration in DRG axons have focused on candidate- or pathway-specific approaches or employed RNA sequencing for unbiased studies. However, transcriptomes only weakly predict the actual proteome. Especially non-transcriptional mechanisms, such as target degradation via autophagy or the proteasome or secretion of targets, can escape such transcriptomic analyses. Consequently, we set out to investigate the proteome of growing and regenerating DRGs. We used three previously published growth paradigms, in which DRG axons switch between low growth with high branching, to elongating growth with little branching. Proteomic analysis identified 39 proteins that inhabit the intersection of these growth paradigms. An overexpression screen of the identified candidates revealed multiple growth effectors. The largest growth effect was elicited by the cellsurface proteoglycan Glypican1 (Gpc1), which has previously been implicated in developmental axon guidance in both mammals and invertebrates. Overexpression of Gpc1 was sufficient to induce axon growth in cultured neurons, where it can overcome the growth inhibition conferred by chondroitin sulfate proteoglycans (CSPGs). Secretion of Gpc1 to the plasma membrane was necessary to bolster growth. Functional knock out of its heparan sulfate binding domain indicated that Gpc1 modulates cell-surface signalling receptors. We demonstrated that Gpc1 acts both autocrine and paracrine. Gpc1-overexpressing cells confer growth competence to WT neurons via extracellular vesicles which label positive for Gpc1. Further, we show that overexpression of Gpc1 changes the whole- and phospho proteome of the cell, to mirror a number of characteristics we found in the original growth paradigms. Together these results reveal Gpc1 as a promising candidate for growth activation, as it acts extracellularly to activate intracellular growth mechanisms, a challenge on which many other growth activators fail. Prävalenz der Riesenzellarteriitis bei Patienten/-innen mit neudiagnostizierter Polymyalgia rheumatica
Hintergrund: Ein gehäuftes gemeinsames Auftreten von Riesenzellarteriitis (RZA) und Polymyalgia rheumatica (PMR) ist bekannt. Die Prävalenz der RZA bei neudiagnostizierten Patienten/-innen mit PMR wurde bisher noch nicht prospektiv untersucht. Im Rahmen der vorliegenden Arbeit wurde die Prävalenz der RZA bei Patienten/-innen mit neudiagnostizierter PMR im Rahmen eines prospektiven Studiendesigns untersucht.
Methodik: 60 Patienten/-innen mit neudiagnostizierter PMR wurden prospektiv in die Studie eingeschlossen und mittels umfassender Ultraschalluntersuchungen auf das zusätzliche Vorliegen einer RZA hin untersucht. Zudem wurden demographische Daten, Laborparameter und krankheitsspezifische Symptome erhoben. Es wurden nur Patienten/-innen eingeschlossen, die eine Steroid-Einnahmezeit von sieben Tagen nicht überschritten.
Alle Patienten/-innen erhielten Ultraschalluntersuchungen der Schulter- und Hüftgelenke sowie der extrakraniellen Gefäße (A. facialis, A. temporalis, A. carotis, A. vertebralis und A. axillaris). Bei pathologischen Gefäßbefunden, die typisch für eine RZA waren, wurde bei den jeweiligen Patienten/-innen in Zusammenschau mit den klinischen Parametern eine RZA diagnostiziert.
Anhand der gestellten Diagnose (isolierte PMR oder PMR und zusätzlich vorliegende RZA) wurde die Kohorte in zwei Gruppen aufgeteilt (PMR-Gruppe und RZA-Gruppe). Zudem wurde ein Cut-off-Wert für das C-reaktive Protein (CRP) definiert, welcher zwischen Patienten/-innen mit isolierter PMR und zusätzlich bestehender RZA unterschied.
Ergebnisse: Bei 28 Patienten/-innen der Kohorte lag zusätzlich eine RZA vor. Die Prävalenz der RZA in dieser Kohorte von neu diagnostizierten Patienten/-innen mit PMR lag folglich bei 46%. Bei 13 Patienten/-innen (22%) in der PMR-RZA-Gruppe zeigten sich keine RZA-typischen Symptome und die Diagnose konnte nur anhand der Ultraschallbefunde gesichert werden. Ein CRP-Wert von 26,5 mg/l diskriminierte mit einer Sensitivität von 66% und einer Spezifität von 73% zwischen Patienten/-innen mit isolierter PMR und zusätzlich vorliegender RZA
Der Einfluss von Mikroglia auf die Differenzierung cerebellärer Purkinjezellen und deren Protektion durch Engrailed-2
Autismus ist eine Störung sozialer Kommunikation, die in unserer Bevölkerung zunehmende Bedeutung erlangt. Obwohl die Diagnose noch immer auf Basis von Verhaltensauffälligkeiten diagnostiziert wird, gibt es Hinweise, dass der Auslöser in externen Stimuli begründet liegen könnte. So wurden auf neuroanatomischer Ebene bei Mäusen nach maternaler Induktion einer Inflammation in der Schwangerschaft die gleichen Veränderungen der Hirnmasse oder der Zellmorphologie der Purkinjezellen (PC) im Kleinhirn entdeckt wie bei Menschen mit Autism Spectrum Disorder (ASD) postmortem. Daher wurde in dieser Arbeit untersucht, welchen Einfluss eine Inflammation auf murine PC hat, und ob diese Effekte von Mikroglia vermittelt werden. Ebenso sollte gezeigt werden, ob sich diese Entzündungsreaktionen durch das Homöodomänenprotein Engrailed-2 beeinflussen lassen.
Mithilfe von cerebellären Schnittkulturen, die von neugeborenen Mäusen gewonnen wurden, wurden PC über 10 Tage in Kultur gehalten und in dieser Zeit pro- und antiinflammatorischen Substanzen ausgesetzt. Die PC wurden anschließend immunhistochemisch gefärbt, mikroskopiert und deren Morphologie semiautomatisch vermessen. Mit dieser Methode konnte gezeigt werden, dass die Reifung der PC nach Lipopolysaccharid (LPS)-Behandlung am dritten Tag in Kultur weiter vorangeschritten war als in unbehandelten Kontrollzellen. Wurden die Schnittkulturen 3 Tage später (am sechsten Tag in Kultur) mit LPS behandelt, waren die Effekte umgekehrt. LPS verzögerte die PC Differenzierung, die Dendriten waren im Durchschnitt kürzer, es wurden mehr Stammdendriten ausgebildet, die aber weniger Verzweigungspunkte hatten. Dieser Effekt ließ sich durch eine Hemmung der Mikrogliaproliferation mittels PLX3397 und durch die Konversion des proinflammatorischen M1 Typs in den antiinflammatorischen M2 Typ durch IL-4 inhibieren. Gleichzeitig konnte die Blockade des Tumornekrosefaktor alpha Signalweges durch dB-cAMP die LPS-induzierten Effekte verhindern. Zusätzlich konnte gezeigt werden, dass die extrinsische Gabe des Homöoboxproteins Engrailed-2 die inhibierenden Effekte auf PC durch LPS-Applikation zu hemmen vermochte.
Somit kann die neuronale Differenzierung durch LPS beeinflusst werden, was maßgeblich durch Mikroglia vermittelt wird. Zudem konnte extrinsisches Engrailed-2 LPS-induzierte Effekte auf die Dendritogenese von PC unterdrücken
Line Intensity Mapping the Epoch of Reionization with the Fred Young Submillimeter Telescope
The baryonic matter in the universe is primarily hydrogen and helium gas, which existed in a neutral atomic state for approximately 400 thousand years following the Big Bang. With the formation of the first galaxies, their ionizing far-UV radiation initiated the ionization of intergalactic hydrogen, and later of helium. The ionization of hydrogen occurred between approximately 300 to 1000 million years after the Big Bang (redshift 11 to 6), a period referred to as the Epoch of Reionization (EoR). The majority of the early galaxies responsible for the reionization are too faint to be detected individually by current observational methods. However, line intensity mapping (LIM) is a novel technique that complements traditional galaxy surveys by capturing the aggregated emission from galaxies within a particular volume in both angular and frequency space, including those too faint to be detected individually.
This thesis develops models for forecasts of observations with a forthcoming LIM survey at 205 to 440 GHz using the Fred Young Submillimeter Telescope (FYST). Specifically, we predict the power spectrum (PS) of the singly ionized carbon, [CII] 158 μm (1900.5 GHz), fine-structure line emission from EoR galaxies. Our model is based on the IllustrisTNG300 hydrodynamic simulation. We find that a detection of the galaxy PS would be possible at redshift bins z=3.4-3.9, 4.1-4.8, 5.3-6.3, and a detection may be possible at z=6.8-8.3.
However, our predictions did not account for the detrimental, contaminating effect of carbon monoxide (CO) rotational line emission from lower-redshift (foreground) galaxies. These lines overlap in frequency with the higher redshift [CII] emission, and their signal must be separated or suppressed in order to detect the [CII] PS.
The second part of this thesis also models the CO emission and evaluates the effectiveness of a foreground masking technique. Our findings indicate that [CII] emission dominates over CO emission above 300 GHz. However, extensive masking is necessary at lower frequencies to detect [CII]. We anticipate the recovery of the [CII] PS at z=3.4-3.9 and z=4.1-4.8, and likely also at z=5.3-6.3. However, at z=6.8-8.3 this is most challenging, necessitating the development of more effective strategies
Adoption and Economic and Environmental Impact of Laser-assisted Precision Land Leveling in Northwestern India
Globally, agricultural production systems need to become more sustainable to meet future food demand without overexploiting natural resources. Northwestern India, a region with highly intensive agricultural production, is facing a rapid decline in groundwater levels due to excessive water use for irrigation. This dissertation investigates laser-assisted land levelling technology (LLL) that reduces water use for irrigation and has been adopted in northwestern India to manage groundwater depletion. The study examines three interrelated research questions: 1) Why are farmers adopting LLL technology, 2) How did farmers access this technology, and 3) What impact does LLL technology adoption have on groundwater levels? The study uses primary data from 1661 households in 84 villages to determine why and how farmers adopt the technology. Further, combining village-level survey data from 291 villages with monthly observational well data and climate data for 21 years, the study also estimates the effect of LLL adoption on groundwater in northwestern India. LLL is popular and widely adopted in northwestern India, with 93 % of farmers aware of the technology and 84% adopting it in their farms. Analysis based on qualitative interviews and estimation based on regressions and causal machine learning shows that the widespread adoption of LLL is due to positive perceptions about this technology and other co-benefits, such as a marginal increase in yield and preference for levelled fields. The findings highlight the need to recognise farmers' perceptions and co-benefits, integrate them with benefits designed by researchers, and foster feedback loops and knowledge co-creation to promote LLL adoption. The emergence of private service providers, an institutional mechanism for renting technology, facilitates the adoption of LLL on small plots and among small farm owners. Regression analysis shows that the number of service providers is positively associated with adoption rates, particularly on smaller plots and farms. Promoting individual service providers increases the access of small farm owners to LLL technology through flexible on-demand services and competitive rental markets. Furthermore, the analysis reveals that LLL adoption slows down the effect of groundwater decline in northwestern India. Applying a staggered difference-in-difference approach, the study shows that the adoption of LLL at the village level has reduced the decline in groundwater levels by 3.7 meters in the month succeeding the use of technology. However, adopting LLL is not enough to stop or reverse overall groundwater decline. Several technological and policy options that could impact the behaviour of farmers towards saving water for irrigation in agriculture are discussed. This dissertation contributes to understanding the adoption and impact of LLL technology for groundwater sustainability in three ways. First, it provides insights into designing new dissemination strategies based on farmers' experience. Second, it discusses new institutional approaches – individual service providers, for supporting the adoption of technology by smallholder farmers. Third, it examines the extent to which the adoption of technology at the farm level transforms into system-level effects in terms of the actual impact on groundwater savings, reducing the negative externalities of groundwater exploitation for agriculture
Welchen Einfluss hat der Versicherungsstatus auf die Überlebensrate onkologischer Patienten/-innen?
Weltweit bestehen unterschiedliche Versicherungsstrukturen und mehrere Studien konnten bereits soziale Ungleichheiten im Gesundheitssystem aufzeigen. Auch in Deutschland wird zwischen gesetzlicher und privater Krankenversicherung unterschieden. Ziel dieser Studie war es, zu untersuchen, ob der Versicherungsstatus einen Einfluss auf die Überlebensrate onkologischer Patientinnen und Patienten hat.
Zur Beantwortung dieser Fragestellung wurden Daten aus dem Krebsregister des Centrums für Integrierte Onkologie in Bonn herangezogen. In die Analyse einbezogen wurden Patientinnen und Patienten mit den Tumorentitäten Bronchialkarzinom, neuroendokrine Tumoren, hochgradige Gliome, Pankreaskarzinom sowie ZNS-Lymphome.
Es wurden Matching-Paare gebildet, jeweils bestehend aus einem gesetzlich und einem privat versicherten Patienten, um Unterschiede hinsichtlich Therapie, Diagnosezeitpunkt und Überlebenszeit besser vergleichen zu können. Für keine der untersuchten Tumorentitäten konnte ein signifikanter Unterschied in diesen Punkten festgestellt werden.
Dennoch ist bekannt, dass es im deutschen Gesundheitssystem Unterschiede in der Terminvergabe und der Häufigkeit von Verlaufskontrollen in Abhängigkeit vom Versicherungsstatus gibt. Es bleibt daher weiterhin wichtig, dieser Fragestellung nachzugehen, zusätzliche Variablen zu berücksichtigen und die Untersuchung auf weitere Tumorentitäten auszuweiten
Remote sensing of crop parameters using UAV-based multispectral imaging and radiative transfer models
In the face of climate change and a growing global population, it's important to increase the output of agricultural systems and improve crop resilience to challenging environmental conditions. Achieving these objectives requires monitoring crop health in the field, as well as breeding more resilient crop varieties. However, these tasks are labor-intensive and not economically sustainable. Remote sensing tools, such as uncrewed aerial vehicles (UAVs), have demonstrated their potential to successfully measure crop parameters while minimizing the need for human and financial resources.
The mapping of crop parameters has been widely studied in both the satellite and UAV research communities. The UAV community typically uses data-driven and parametric models to predict crop parameters, while satellites rely on physical models called radiative transfer models (RTMs) to retrieve these variables. However a key drawback of using satellites is their low spatial and temporal resolution for applications in precision agriculture.
This thesis explores the use of radiative transfer model PROSAIL to retrieve crop variables with UAVs and multispectral imaging. First, we examine the reflectance calibration workflows of the optical sensor, vital for time-series image analysis. We propose a multi-panel approach for calibrating reflectance of a multispectral sensor, which our analysis shown to perform better than the one-point calibration. Next we address the challenges of retrieving structural and biochemical variables in complex and homogeneous crop canopies. Our findings confirm that the higher spatial resolution provided by UAVs doesn't disrupt the fundamental assumptions of the PROSAIL, which was originally developed for simpler canopies. Finally, we investigate the sensor synergies for crop stress detection. In one study we explore the synergy between terrestrial laser scanner, multispectral imaging, and RTMs to track drought-induced leaf movement in soybean. Reduced Costs for Vehicle Routing and General Mixed-Integer Programs
Mixed-integer linear programming is a powerful tool for modeling combinatorial optimization problems in a standardized way. Sophisticated solvers that calculate optimal solutions for these programs consist of dozens of individual interacting parts. Throughout the last 70 years, a lot of scientific and commercial resources have been invested in developing new components and refining old ones. Due to the diversity of applications, many general-purpose techniques exist. Additionally, when employed in practice, these algorithms often have to solve multiple problems with a similar structure. Consequently, problem-specific extensions are of high interest. In this thesis, we investigate both types of modules, general-purpose mixed-integer linear programming techniques and algorithmic refinements for a specific combinatorial optimization problem, namely, the vehicle routing problem (VRP).
In VRPs, the task is to deliver goods to customers using a fleet of vehicles at minimal travel cost. We introduce two new variants: the VRP with heterogeneous time windows and its dynamic and robust extension. These variants are motivated by a logistics problem from practice. In contrast to their homogeneous pendants, heterogeneous VRPs imply an additional level of decision-making: Which customer gets assigned to which vehicle type? We elaborate that this question is fundamental in our (mixed-)integer linear programming approach. We present efficient adaptations of state-of-the-art techniques to enforce the allocations of these vehicle-assignment variables throughout the solution process. In our extensive computational experiments, we evaluate different techniques, such as branching and reduced cost fixing for these variables in our branch-cut-and-price algorithm. Our results underline their importance for the VRP with heterogeneous time windows. In the dynamic extension, we use robust optimization techniques to introduce a novel delay-resistance objective term. To guarantee optimality, we adapt the dominance check of the labeling algorithm. In a computational study, we evaluate the practical impact of this objective term in our model on a set of real-world instances.
Motivated by the significant impact of enforcing routes in VRP solutions, we developed a general-purpose variable fixing technique. It is based on reduced cost fixing, a famous integer linear programming technique that combines important concepts of linear programming with the integrality condition of the variables and a primal bound U on the optimal objective function value. Our novel reduced cost fix and cut method extends this idea by efficiently increasing the amount of accessed local information via rotation of the objective vector. Its iterative version is an exact method for checking whether some variable values can not be attained in a solution with an objective value better than U. Furthermore, we introduce a new class of cuts, the gap-based optimality cuts. They are designed to cut off not only a given fractional optimum of the linear relaxation but also solutions that do not improve upon U. We show that these cuts imply the classic Dantzig cuts and their strengthened version by Charnes and Cooper. We evaluate the performance of our novel techniques on the MIPLIB 2017 Benchmark set to analyze their effectiveness and efficiency on a heterogeneous class of instances. Our implementation in the open-source solver SCIP shows that the new cuts significantly strengthen the dual bound at the root node relaxation. Additionally, both novel techniques can accelerate the solution process of a sophisticated MILP framework
Resilienz bei koronarer Herzkrankheit: Zusammenhang mit Erkrankungsschwere und Erkrankungsdauer
Hintergrund:
KHK-Patient:innen weisen, wie Personen mit chronischen körperlichen Erkrankungen im Allgemeinen, eine erhöhte Prävalenz von psychischen Belastungssymptomen auf. Resilienz ist ein komplexes Konstrukt und wird im Kontext von Belastungen als schützender Faktor verstanden. Obwohl die meisten Resilienzfragebögen auf einem Eigenschaftskonzept beruhen, existieren Modelle, die Annahmen über die Auswirkungen einer anhaltenden Belastung auf die Resilienz zulassen. Beispielsweise könnte die Resilienz mit zunehmender kardialer Erkrankungsdauer abnehmen ("wear and tear effect") oder durch wiederholte Bewältigungserfahrungen zunehmen ("steeling effect").
Ziel:
Ziel dieser Studie war es, die Beziehungen zwischen Erkrankungsschwere, Erkrankungsdauer und der Resilienz bei KHK-Patient:innen zu untersuchen.
Methoden:
Im Rahmen der multizentrischen TEACH-Studie wurden 457 psychosozial belastete KHK-Patient:innen aus sechs deutschen Universitätskliniken umfassend untersucht. Erfasst wurden soziodemographische Merkmale, krankheitsbezogene Variablen, die psychische Belastung (gemessen durch HADS, PSS-4, HAF-17, D-S) sowie die Resilienz (gemessen durch die RS-13). Eine Netzwerkanalyse wurde durchgeführt, um die Beziehungen zwischen den gemessenen Konstrukten zu visualisieren.
Ergebnisse:
Eine erhöhte körperliche Symptombelastung durch die Herzerkrankung ging mit einer erhöhten psychischen Belastung und einer niedrigeren Resilienz einher.
Entgegen der Hypothese zeigte sich kein moderierender Effekt der Resilienz auf die Beziehung zwischen Schweregrad der KHK und psychischer Belastung. Ebenso bestand keine signifikante Korrelation zwischen der Erkrankungsdauer und der Resilienz. Die Netzwerk- und Clusteranalyse zeigte eine enge Verbindung des Resilienzkonzepts zu Angst und Depression und nur wenige Assoziationen zwischen Resilienz und krankheitsbezogenen Variablen.
Schlussfolgerung:
Ein protektiver moderierender Effekt der Resilienz auf die Beziehung zwischen Erkrankungsschwere und psychischer Belastung konnte anhand der Daten nicht nachgewiesen werden. Ebenso fand sich keine Bestätigung für einen wear and tear oder einen steeling effect auf die Resilienz bei zunehmender Krankheitsdauer. Die enge Korrelation zwischen Resilienz, gemessen anhand eines gängigen, eigenschaftsbasierten Resilienzfragebogens, und Angst und Depression legt eine konzeptionelle Überschneidung nahe und unterstreicht die Problematik eines bisher fehlenden Goldstandards zur zuverlässigen Erfassung des multidimensionalen Resilienzkonstrukts
Variabilität der Plasmaspiegel der direkten oralen Antikoagulanzien Rivaroxaban und Apixaban bei Patienten mit venöser Thromboembolie
Direkte orale Antikoagulanzien (DOAK) stellen neben der Schlaganfallprophylaxe bei Vorhofflimmern die Standardtherapie in der Behandlung und Rezidivprophylaxe venöser Thromboembolien (VTE) dar. In Deutschland sind die Faktor-Xa-Inhibitoren Rivaroxaban, Apixaban und Edoxaban sowie der Thrombininhibitor Dabigatran zugelassen. Im Gegensatz zu Vitamin-K-Antagonisten erfordern DOAK kein routinemäßiges Monitoring, wenngleich die Bestimmung von Plasmaspiegeln in bestimmten klinischen Situationen – etwa vor invasiven Eingriffen oder bei eingeschränkter Eliminationskapazität – von Bedeutung sein kann. Die bislang verfügbaren Referenzbereiche beruhen überwiegend auf Daten aus klinischen Studien mit gesunden Probanden und sind daher nur bedingt auf die VTE-Patientenpopulation übertragbar.
In dieser retrospektiven Kohortenstudie wurden die Daten von 2.252 Patienten unter Erhaltungstherapie mit DOAK nach VTE analysiert. In die finale Auswertung gingen 971 Patienten unter Rivaroxaban und 431 unter Apixaban ein, während Edoxaban- und Dabigatran-Gruppen aufgrund geringer Fallzahlen ausgeschlossen wurden. Die ermittelten 10. – 90. Perzentilbereiche der Rivaroxaban-Spitzen- und Talspiegel lagen enger als die bislang publizierten Erwartungswerte. Die 5. – 95. Perzentilbereiche für Apixaban entsprachen hingegen weitgehend den Literaturangaben.
Darüber hinaus zeigte sich eine schwache, jedoch signifikante Korrelation der Plasmaspiegel mit dem Alter, während das Körpergewicht ausschließlich bei Rivaroxaban in der Dosierung von 20 mg 1×/Tag eine signifikante negative Korrelation aufwies. Ein Zusammenhang mit dem Body-Mass-Index konnte nicht nachgewiesen werden.
Die Ergebnisse belegen die Zuverlässigkeit funktioneller Messmethoden zur Bestimmung von DOAK-Plasmaspiegeln und unterstreichen die Notwendigkeit, populationsbasierte Referenzwerte zu definieren, um die klinische Interpretation in praxisrelevanten Situationen zu optimieren
