    Treatment Effect of Budesonide in 5xFAD mouse model of Alzheimer´s disease

    This project investigated the therapeutic potential of Budesonide, a glucocorticoid commonly used for asthma therapy, in the treatment of the 5xFAD mouse model of Alzheimer’s disease. The findings indicate that inhalation of Budesonide may influence several aspects of AD pathology. Key results showed that inhalation treatment with Budesonide prevented cognitive decline, impairment of learning, and memory performance in behavioral tests. Additionally, Budesonide treatment was associated with a reduction in brain atrophy without affecting plaque load and amyloid levels. This was paralleled by reduced neuron and synapse loss, and lower blood levels of the neurodegeneration marker NfL. We identified anti-inflammatory changes in microglia of Budesonide treated 5xFAD animals, together with a lower percentage of C1q opsonized synapses as one potential mechanism of Budesonide treatment. Moreover, we showed that inhaled Budesonide reached the brain regions affected by AD pathology, confirming effective drug delivery. Further exploration into the molecular mechanisms of Budesonide’s effects is essential. Understanding these pathways could lead to optimized treatment protocols that maximize therapeutic benefits while minimizing side effects.Dieses Projekt untersuchte das therapeutische Potenzial von Budesonid, einem Glukokortikoid, das üblicherweise zur Behandlung von Asthma eingesetzt wird, in der Therapie des 5xFAD-Mausmodells der Alzheimer-Krankheit. Die Ergebnisse deuten darauf hin, dass die Inhalation von Budesonid verschiedene Aspekte der AD-Pathologie beeinflussen könnte. Wichtige Ergebnisse zeigten, dass eine Inhalationsbehandlung mit Budesonid kognitiven Abbau, Beeinträchtigungen des Lernens und der Gedächtnisleistung in Verhaltenstests verhinderte. Darüber hinaus war die Budesonid-Behandlung mit einer Reduktion der Gehirnatrophie verbunden, ohne jedoch die Plaquelast oder Amyloidspiegel zu beeinflussen. Parallel dazu wurden ein verringerter Neuronen- und Synapsenverlust sowie niedrigere Blutwerte des Neurodegenerationsmarkers NfL beobachtet. Zudem identifizierten wir entzündungshemmende Veränderungen in der Mikroglia von Budesonid-behandelten 5xFAD-Tieren sowie einen geringeren Anteil an C1q-opsonisierten Synapsen, was als ein möglicher Wirkmechanismus von Budesonid angesehen werden kann. Darüber hinaus konnten wir nachweisen, dass inhaliertes Budesonid die von der AD-Pathologie betroffenen Gehirnregionen erreichte, was eine effektive Wirkstoffverabreichung bestätigt

    Investigating the molecular mechanisms involved in the developmental assembly of the mushroom body calyx in <em>Drosophila melanogaster</em>

    How a circuit functions can be investigated by examining the organisation of the circuit to be able to decipher the connectivity logic and the logic of information flow within that circuit. Hence, to understand how the sparse coding of the Kenyon cells (KCs) is wired into the circuit, the organisational logic must be resolved. To that effect, a major question emerges - how do KCs find their synaptic partners during development? This thesis aims to answer that question by taking advantage of available transcriptomic datasets (Alyagor et al. 2018; Li et al. 2020; Li et al. 2017; Xie et al. 2021) and focusing on the possible role of cell surface molecules in circuit assembly by means of RNAi-mediated knockdown in the PNs and KC. The effect of the knockdown was analysed by measuring number and distribution of PN boutons, calyx organisation and synapse formation. Based on this primary RNAi screen, candidate molecules that displayed circuit defects in the calyx were isolated and examined for the pattern of expression, the mutant phenotype and its cell autonomy in order to elucidate the role the molecules play in the development of the calyx. In addition, a method for in vivo imaging of the pupa was developed to be able to image developmental milestones in the context of normal development as well as mutants. With this work, we hope to shed light in understanding how the non-stereotypical circuit of the calyx is assembled during development

    Dispersive analyses of three-body decays and scattering reactions

    More than half a century after the discovery of quantum chromodynamics as the quantum-field-theoretical description of the strong interaction, its non-perturbative nature in the low-energy regime still poses a unique challenge. Particularly, the fundamental properties and scattering of light hadrons need to be studied precisely to unravel the structure of the strong interaction. In this thesis, we consider manifold applications of this kind using dispersive techniques that are based on the S-matrix principles of unitarity and analyticity. We perform a study of rescattering effects in 3π final states, which are often described in terms of two-body resonances and a non-interacting spectator particle. Using Khuri–Treiman dispersion relations, we include crossed-channel effects and estimate the rescattering effects beyond the simplest isobar model for a selected set of quantum numbers. This allows for an estimate when rescattering effects become important and more complicated analysis techniques are needed to extract meaningful physical information from experiments. However, Khuri–Treiman equations are limited to low energies as they are built from truncated partial-wave expansions. Therefore, a new parameterization is presented that can fulfill all theoretical expectations and connects the essential physics of hadron scattering both near threshold and in asymptotic limits. In this construction, dynamical information is entirely contained in Regge trajectories that generalize resonance poles in the complex-energy plane to poles in the angular-momentum plane. While the construction and first results are successfully presented in this thesis, this formalism allows for many extensions that can be explored in the future. Properties of kaons are investigated using the kaon electromagnetic form factor and the Primakoff reactions γK→Kπ and γK→γK. These are constructed obeying the constraints of analyticity, which enables the extrapolation to unphysical energies or the pole positions on the second Riemann sheet. Using the kaon form factor allows for a calculation of the electromagnetic charge radius from a fit to experimental data in timelike and spacelike regions, while the Primakoff reactions can be used in the future to extract the chiral anomaly, radiative K*(892) couplings, and kaon polarizabilities. Additionally, there are several different applications for these results, e.g., the anomalous magnetic moment of the muon and corrections to Dashen's theorem

    Untersuchung zur intraoperativen Volumen- und Kreislauftherapie bei Patientinnen mit Ovarialkarzinom

    Hintergrund: Bei Patientinnen mit Ovarialkarzinom besteht während Tumordebulkingoperationen ein ausgeprägter intraoperativer Volumenmangel. Das Ziel dieser Arbeit war es, Zusammenhänge zwischen der Volumeneinfuhr- und -bilanz, sowie dem postoperativen Outcome zu untersuchen. Dabei wurden intraoperative Faktoren detailliert analysiert, um deren Auswirkungen postoperativ, sowie auf das Langzeitüberleben der Patientinnen zu ermitteln. Methoden: Im Rahmen einer retrospektiven Analyse wurden 73 Patientinnen untersucht, die zwischen 2012 und 2015 in der Universitätsklinik Bonn operiert wurden. Es gab keine festen Vorgaben für das Infusionsregime. Neben der Volumenein- und -ausfuhr wurde die Volumenbilanz erfasst und in Bezug auf die Operationsdauer, zusätzliche abdominalchirurgische Eingriffe, die Beatmungsdauer sowie postoperative Komplikationen analysiert. Zudem wurde der zeitliche Verlauf des Blut- und Plasmavolumens während der Operation untersucht. Abschließend wurde der Einfluss der Volumenbilanz auf das Langzeitüberleben bewertet. Ergebnisse: Es zeigten sich signifikante Korrelationen zwischen der Volumeneinfuhr (Mittelwert 11,2 l) und Volumenbilanz (Mittelwert 9,2 l) zur Operationsdauer und einzelnen abdominalchirurgischen Resektionen. Zudem war eine erhöhte Flüssigkeitszufuhr mit einer verlängerten postoperativen Beatmungszeit und einer höheren Rate an postoperativen Komplikationen wie Pleuraergüssen (18,25 %) und Anastomoseninsuffizienzen (5,1 %) assoziiert. Blut- und Plasmavolumina stiegen zum Operationsende nur moderat an. In Zusammenschau mit anderen Studien könnte die Entwicklung eines Kapillarlecks eine wahrscheinliche Ursache für die beobachteten Komplikationen darstellen. Auf das Langzeitüberleben nach 5 Jahren konnte kein signifikanter Einfluss der Volumenbilanz eruiert werden. Zusammenfassung: In dieser Studie wurde ein deutlicher Zusammenhang zwischen der Volumeneinfuhr und der Volumenbilanz auf das Outcome der Patientinnen gezeigt. Weitere prospektive Studien sind erforderlich, um optimale Flüssigkeitsregime für diese Patientinnengruppe zu entwickeln

    Aphantasie: Fakt oder Fiktion? : Experimentell-neurowissenschaftliche Untersuchungen zur Charakterisierung von Personen ohne sensorisches Vorstellungsvermögen

    Aphantasie beschreibt die Abwesenheit oder merkliche Reduktion des sensorischen Vorstellungsvermögens und ist somit in ihrer Extremform ein einzigartiges natürliches Knock-Out-Modell zur Untersuchung der Einflüsse des sensorischen Vorstellungsvermögens auf unterschiedliche neurokognitive, -affektive und -sensorische Funktionen. Im Rahmen der vorliegenden Dissertation wurden 11 Studien durchgeführt, um durch experimentelle und zum Teil neurowissenschaftliche Untersuchungen das Phänomen der Aphantasie zu charakterisieren, Aphantasisten von Nicht-Aphantasisten abzugrenzen und ein neurobehaviorales Profil von Personen mit Aphantasie zu erstellen. In den Studien 1 und 2 wurden Auswirkungen auf die Performanz in visuellen Suchaufgaben, in Studie 3 Auswirkungen auf das verbale und visuelle Kurz- und Langzeitgedächtnis und in den Studien 4 und 5 Auswirkungen auf die Gesichtserkennung in Abhängigkeit der Aufgabenschwierigkeit sowie bei verbaler Interferenz untersucht. Studie 6 beschäftigte sich mit den neuronalen Grundlagen autobiographischer Gedächtnisdefizite bei Aphantasie und Studie 7 untersuchte, inwiefern auch empathische Prozesse durch das Vorstellungsvermögen beeinflusst werden. Die Studien 8 bis 11 tragen zur Einordnung von Aphantasie hinsichtlich Krankheitswertigkeit, Beeinträchtigungen im Alltag und Beeinträchtigung nicht-visueller Vorstellungsmodalitäten bei. Die Ergebnisse der Studien zeigen, dass das visuelle Vorstellungsvermögen sowohl als Top-Down-Strategie bei der visuellen Suche als auch als Gedächtnisstrategie genutzt werden kann. Defizite durch die Abwesenheit des sensorischen Vorstellungsvermögens werden vor allem bei erhöhter Aufgabenschwierigkeit deutlich, nicht aber wenn feature-basierte Prozesse gefordert werden, das heißt Prozesse, die sich nur auf einzelne Merkmale eines komplexen visuellen Reizes beziehen (z. B. die Nase in einem Gesicht). Die Ergebnisse zu den neuronalen Grundlagen von autobiographischen Gedächtnisdefiziten bei Aphantasie deuten auf eine Beteiligung des Hippocampus und des visuellen Kortex beim Abruf episodischer Gedächtnisinhalte hin. Unsere Befunde konnten zeigen, dass bei Personen mit Aphantasie diese Netzwerke im Vergleich zu Nicht-Betroffenen dahingehend verändert sind, dass der Hippocampus hypo- und der visuelle Kortex hyperaktiviert ist. Eine Erklärung für autobiographische Gedächtnisdefizite bei Aphantasie könnte somit sein, dass potenzielle Vorstellungssignale im visuellen Kortex aufgrund der Hyperaktivierung untergehen. Dennoch zeigen sich im Alltag von Personen mit Aphantasie kaum Beeinträchtigungen (z. B. in der Schule), obwohl eine Teilstichprobe von 34.7 % von Leidensdruck berichtet. Dieser resultiert jedoch nicht zwangsläufig aus der Aphantasie, sondern kann auch durch Drittvariablen verursacht werden. Weitere Ergebnisse deuten darauf hin, dass auch nicht-visuelle Vorstellungsmodalitäten bei Aphantasie betroffen sind und in Zukunft stärker untersucht werden sollten. Insgesamt trägt die vorliegende Arbeit zur Erweiterung des neurobehavioralen Profils von Aphantasie bei, aus dem zukünftige Fragestellungen abgeleitet werden können. So bleibt zum gegenwärtigen Zeitpunkt beispielsweise noch offen, inwiefern sich Aphantasie auf die Wirksamkeit imaginativer Therapieverfahren auswirkt und welche neurochemischen Prozesse dem Vorstellungsvermögen zugrunde liegen. Es werden zwei Stage 1 Registered Reports vorgestellt, die diese Fragestellungen beantworten sollen. Ziel ist unter anderem die experimentelle Manipulation des sensorischen Vor-stellungsvermögens, welche schlussendlich auch Personen mit Aphantasie Einblicke in das sensorische Vorstellungsvermögen ermöglichen könnte.Aphantasia: Fact or fiction? Experimental-neuroscientific studies on the characterization of persons without sensory mental imagery Aphantasia describes the absence or marked reduction of sensory mental imagery and is thus, in its extreme form, a unique natural knock-out model for investigating the influences of sensory mental imagery on different neurocognitive, -affective and -sensory functions. In the present dissertation, 11 studies were conducted to characterize the phenomenon of aphantasia through experimental and partly neuroscientific investigations, to differentiate aphantasics from non-aphantasics, and to create a neurobehavioral profile of individuals with aphantasia. Studies 1 and 2 investigated effects on performance in visual search tasks, Study 3 investigated effects on verbal and visual short- and long-term memory, and Studies 4 and 5 investigated effects on face recognition as a function of task difficulty and verbal interference. Study 6 dealt with the neural basis of autobiographical memory deficits in aphantasia and Study 7 investigated the extent to which empathic processes are also influenced by the ability to create mental imagery. Studies 8 to 11 contribute to the classification of aphantasia in terms of pathological significance, impairment in daily life and impairment of non-visual mental imagery modalities. The results of the studies show that visual imagery can be used both as a top-down strategy in visual search and as a memory strategy. Deficits due to the absence of sensory imagery are particularly evident when the task is more difficult, but not when feature-based processes are required, i.e. processes that only relate to individual features of a complex visual stimulus (e.g., the nose in a face). The results on the neural basis of autobiographical memory deficits in aphantasia indicate an involvement of the hippocampus and the visual cortex in the retrieval of episodic memory content. Our findings showed that these networks are altered in individuals with aphantasia compared to non-aphantasic individuals in that the hippocampus is hypo-activated and the visual cortex is hyperactivated. One explanation for autobiographical memory deficits in aphantasia could therefore be that potential imagery signals are lost in the visual cortex due to a bad signal-to-noise ratio. Nevertheless, there are hardly any impairments in the everyday lives of people with aphantasia (e.g., at school), although a sub-sample of 34.7 % reported suffering. However, this does not necessarily result from aphantasia, but can also be caused by third-party variables. Further results indicate that non-visual mental imagery modalities are also affected in aphantasia and should be investigated more closely in the future. Overall, the present dissertation contributes to the expansion of the neurobehavioral profile of aphantasia, from which future questions can be derived. For example, the extent to which aphantasia affects the effectiveness of mental imagery-based therapy methods and which neurochemical processes underlie the ability to visualize remain open questions at the present time. Two Stage 1 Registered Reports will be presented that aim to answer these questions. One of the aims is the experimental manipulation of sensory mental imagery, which could ultimately give people with aphantasia insights into the process of sensory imagination

    The effects of fat amount and fatty acid composition on postprandial metabolic events in older adults with a risk phenotype for cardiometabolic diseases

    Due to frequent food intake, most humans spend the majority of the day in the postprandial state, associated with alterations in blood concentrations of triglycerides (TGs), glucose, and insulin. The meal composition is known to influence the postprandial response. According to the current knowledge, an excessive metabolic response increases the risk of cardiovascular diseases (CVDs). Strategies to mitigate the metabolic response could contribute to reducing the risk of CVDs. The core of this thesis was a randomized controlled crossover trial of the acute metabolic and vascular effects of four isoenergetic, isonitrogenous meals enriched with 25 or 50 g of canola oil (rich in unsaturated fatty acids [FAs]) or coconut oil (rich in saturated FAs [SFAs]) in 29 participants at increased risk for cardiometabolic diseases. In the fasting state and during a postprandial period of 6 h, blood samples for the analysis of parameters of lipid and glucose metabolism as well as parameters of pro-inflammatory and pro-oxidative processes were collected. In addition, serum concentrations of individual FAs were measured and parameters of arterial stiffness were determined. The main findings were that canola oil induced a stronger TG and interleukin-6 response than coconut oil. Serum concentrations of lauric and myristic acid were higher after coconut oil, while serum concentrations of oleic, linoleic, and alpha-linolenic acid were higher after canola oil. Compared to 25 g test oil, 50 g test oil elicited a stronger TG response but lower glucose and insulin responses. The parameters of arterial stiffness decreased postprandially, independent of fat amount and FA composition. The intervention study was complemented by two systematic reviews of the effects of fat amount and FA composition on lipemia and vascular function. The first review revealed that in individuals with increased CVD risk, SFA-rich meals often induce a stronger TG response than meals rich in unsaturated FAs, whereas the TG response in metabolically healthy individuals is not influenced by the FA composition. The analysis confirmed the influence of fat amount on lipemia. The second review showed that the FA composition largely has no influence on the postprandial vascular function. Data demonstrate that the fat amount and FA composition of a meal are relevant determinants of acute metabolic processes.Aufgrund häufiger Nahrungsaufnahme verbringen die meisten Menschen einen Großteil des Tages im postprandialen Zustand, einhergehend mit Veränderungen der Blutkonzentrationen von Triglyzeriden (TG), Glukose und Insulin. Die Mahlzeitenzusammensetzung beeinflusst die postprandiale Reaktion. Nach derzeitigem Kenntnisstand erhöht eine übersteigerte Stoffwechselantwort das Risiko für Herz-Kreislauf-Erkrankungen (HKEs). Strategien zur Abmilderung der Stoffwechselreaktion könnten zur Verringerung des HKE-Risikos beitragen. Kern dieser Arbeit war eine randomisierte kontrollierte Crossover-Studie zu den akuten metabolischen und vaskulären Effekten von vier isoenergetischen, isonitrogenen Mahlzeiten, angereichert mit 25 oder 50 g Rapsöl (reich an ungesättigten Fettsäuren [FS]) oder Kokosöl (reich an gesättigten FS [SAFAs]), bei 29 Teilnehmern mit erhöhtem Risiko für kardiometabolische Erkrankungen. Nüchtern sowie über 6 Stunden postprandial wurden Blutproben für die Analyse von Parametern des Lipid- und Glukosestoffwechsels sowie von Parametern proinflammatorischer und prooxidativer Prozesse entnommen. Zudem wurden die Serumkonzentrationen einzelner FS gemessen und Parameter der arteriellen Gefäßsteifigkeit bestimmt. Die zentralen Ergebnisse waren, dass Rapsöl eine stärkere TG- und Interleukin-6-Reaktion auslöste als Kokosöl. Die Serumkonzentrationen an Laurin- und Myristinsäure waren nach Kokosöl höher, während die Serumkonzentrationen an Öl-, Linol-, und Alpha-Linolensäure nach Rapsöl höher waren. Im Vergleich zu 25 g Testöl lösten 50 g Testöl eine stärkere TG-Antwort, aber schwächere Glukose- und Insulinantworten aus. Die Parameter der arteriellen Gefäßsteifigkeit sanken postprandial, unabhängig von Fettart und Fettmenge. Die Interventionsstudie wurde durch zwei Übersichtsarbeiten zu den Effekten von Fettmenge und FS-Zusammensetzung auf die Lipämie und Gefäßfunktion ergänzt. Die erste Analyse ergab, dass bei Personen mit erhöhtem HKE-Risiko SAFA-reiche Mahlzeiten im Vergleich zu Mahlzeiten reich an ungesättigten FS oft eine stärkere TG-Antwort auslösen, während die FS-Zusammensetzung die TG-Reaktion bei Stoffwechselgesunden nicht beeinflusst. Die Analyse bestätigte den Einfluss der Fettmenge auf die Lipämie. Die zweite Analyse zeigte, dass die FS-Zusammensetzung meist keinen Einfluss auf die postprandiale Gefäßfunktion hat. Die Daten zeigen, dass die Fettmenge und FS-Zusammensetzung einer Mahlzeit relevante Determinanten akuter Stoffwechselprozesse sind

    Charge collection studies of MAPS in a 65 nm CMOS imaging process

    Monolithic Active Pixel Sensors (MAPS) are candidates to serve as vertex and tracking detectors in the future lepton colliders. This is due to the many advantages they provide compared to hybrid sensors, such as a reduced material budget and lower power consumption. The TANGERINE project aims for the development of the next generation of monolithic silicon pixel sensors produced in the 65 nm imaging process and investigates their possible use as vertex detectors in future lepton colliders. To investigate the 65 nm imaging process, simulations are implemented to reproduce the sensor response and thus save time and resources during the development phase. Technology Computer-Aided Design (TCAD) simulations permit the production of complex electric field profiles by the use of generic doping concentrations, a solution to the restricted access that commercial foundries implement in their fabrication process. A combination of Monte Carlo (MC) simulations using electrostatic TCAD fields was introduced to address the intensive computational requirements of using transient TCAD simulations. This allows the utilization of realistic electric fields combined with a high particle rate, resulting in simulations with high statistics. Furthermore, to include the effects that the electronics have on the signal produced by the silicon detector, Simulation Program with Integrated Circuit Emphasis (SPICE) is integrated into the simulation chain. The sensors used for the validation of the simulation chain are the ALICE Analogue Pixel Test Structure (APTS) are MAPS produced in a 65 nm CMOS imaging process designed at CERN for the CERN EP R&D program and are part of the studies for the ALICE ITS3 upgrade. In particular, the Analogue Pixel Test Structure Source Follower (APTS-SF) allows for the study of the different layouts and their effect on the signal due to several source follower stages used a simple electronic readout. This work showcases the results of a simulation approach combining TCAD, MC, and SPICE, a comparison of charge studies of these simulations with MAPS using a Fe-55 source for calibration and measurements taken at the DESY-II test beam facility using a 4 GeV electron beam. A discussion of the highlights and discrepancies between simulation and experimental data, and the possible reasons for these disagreements, is presented

    The rapid growth of biomedical literature makes it increasingly difficult to identify and organize meaningful knowledge. This project addresses the problem by focusing on relation extraction (RE), i.e., detecting and classifying semantic relationships between biomedical entities within scientific abstracts. Our objective is to evaluate and compare multiple paradigms for scientific relation extraction on the BioRED dataset, a manually annotated benchmark of PubMed abstracts with diverse entities and relation types. Specifically, we investigate three complementary approaches: a classification-based model using BioBERT, a question answering formulation QA4RE, and lastly, generative models SciFive and REBEL. The central research question guiding our study is: Which modeling paradigm offers the most effective and generalizable solution for biomedical relation extraction under the constraints of the BioRED dataset?</em
