    Molecular mechanisms affected by boron deficiency in root and shoot meristems of plants

    Boron deficiency is an abiotic stress that negatively impacts plant growth and yield worldwide. Boron deficiency primarily affects the development of plant meristems— stem cells critical for all post-embryonic tissue growth. The essential role of boron in meristem development was first established in 1923. It remains unclear whether boron directly integrates into meristem molecular signalling pathways. In addition to its stabilizing function in the primary cell wall, growing evidence suggests roles for boron in various molecular processes including phytohormone cascades. These indications enhance a mechanistic understanding of why boron is crucial for proper meristem development. In this review we compile and discuss molecular pathways influenced by boron availability in Arabidopsis (Arabidopsis thaliana), maize (Zea mays), rice (Oryza sativa), and oilseed rape (Brassica napus) with a focus on the auxin-, ethylene-, and cytokinin-mediated hormone cascades. We particularly compare and contrast phenotypic and molecular adaptations of shoot and root meristems to boron deficiency and pinpoint tissue-specific differences

    Satellite remote sensing-based crop cover classification over Europe : accuracy of different methodological approaches

    Crop maps play an important role in a variety of applications, from calculating crop areas and forecasting food production quantities to the analysis of agri-environmental interactions, highlighting the necessity of timely and accurate information on agricultural land use. The availability of remote sensing data has permitted numerous crop classification studies, which have investigated a variety of methods to improve classification performance, such as the selection of remote sensing sources, classification algorithms, and preprocessing methods. This paper compares these approaches with respect to classification accuracy in a European context. The study also investigates aspects such as classification level, study area division, and class granularity. The review shows that optical products provide more information for crop identification than radar products, however, combining optical data with radar backscatter increases accuracy. Classification accuracy benefits from specific features such as red-edge and spectral indices for optical products and Haralick textures for radar. Compared to traditional machine learning and distance-based classification methods, deep learning algorithms have been shown to achieve superior performance. Nevertheless, random forest's comparative accuracy at relatively low computational cost makes it a viable alternative for largescale applications. Finally, preprocessing methods and data on topography, climate, and crop growth patterns appear to improve accuracy

    The mathematical properties of the radiative transfer equation

    The kinetic equation describing the interaction of matter with electromagnetic waves is the radiative transfer equation. This thesis has as aim the development of a comprehensive mathematical theory for this equation. In particular, it collects several results about different problems which study the behavior of the temperature distribution in a body where the heat is transferred mainly by radiation. This work is a cumulative thesis which collects five articles written by the author with several collaborators during her PhD studies. Chapter 1 gives a detailed introduction of the radiative transfer equation describing its derivation, its main mathematical features and the available mathematical literature. Chapter 2 deals with the existence theory of the stationary radiative transfer equation in the case in which the absorption and the scattering coefficients depend on the temperature and it is based on the article in Appendix A. Chapter 3 is based on the article in Appendix B and it treats the diffusion approximation of the radiative transfer equation, which is studied via matched asymptotic expansions, introducing the concepts of equilibrium and non-equilibrium diffusion approximations. Chapter 4 studies rigorously the diffusion approximation of the stationary radiative transfer equation when the radiation interacts with matter only through emission and absorption. This chapter is based on the article in Appendix C. Chapter 5 develops the well-posedness theory of a free boundary problem for the melting of ice in the case in which the heat is transferred by conduction in the liquid and by conduction and radiation in the solid. This chapter is based on the article in Appendix D. Chapter 6 is based on the article in Appendix E and it continues the study of the free boundary problem introduced in Chapter 5 and it deals with the existence theory of traveling wave solutions. Finally, Chapter 7 summarizes the main results of this thesis and presents several future research directions. The articles upon which this thesis is based can be found in the appendices

    Novel Mathematical Techniques for Scattering Amplitudes

    In this thesis, we discuss mathematical methods to compute families of Feynman integrals. Central to our approach are two mathematical structures: the geometries underlying integral families, and their associated twisted (co-)homology groups. We use the latter to derive general relations among integrals and understand the so-called canonical form of a basis of master integrals better. Additionally, we present the analytic computation of several integral families: specific Lauricella integrals associated with hyperelliptic curves, the unequal-mass kite integral family, and two-dimensional (fishnet) integrals. We begin by reviewing the physical and mathematical background. Starting with a brief summary of the role of Feynman integrals in perturbative quantum field theory, we introduce key tools such as their parametric representations, their cuts, and the method of differential equations, which relies on finding a so-called canonical basis of an integral family. Then we introduce the mathematical background: We discuss the geometries appearing in the integral families studied in this thesis – including hyperelliptic curves and Calabi–Yau varieties – and review the twisted (co-)homology groups, with an emphasis on the computation of intersection and period matrices. We conclude the first part by discussing how a matrix of cuts of Feynman integrals can be interpreted as a period matrix of twisted (co-)homology groups. This interpretation provides a transition into one of the central results of this thesis: We explore what can be learned from the so-called twisted Riemann bilinear relations for the intersection and period matrices of twisted (co-)homology groups associated to a Feynman integral family. These relations not only explain known and provide new relations, but also enable us to gain a deeper understanding of the canonical basis. Then we use these insights to construct canonical differential equations for Lauricella integral families associated to genus-one and genus-two hyperelliptic curves which serve as models for hyperelliptic maximal cuts. Next we present the analytic computation of the unequal-mass kite integral family, which is related to two distinct elliptic curves. A key step is the parametrisation of the five kinematic variables on two elliptic tori, enabling the solution in terms of iterated integrals on these tori. Then we study single-valued Feynman integrals in two spacetime dimensions. We show how these integrals naturally are bilinear combinations – effectively, double copies – of twisted periods. This construction is equivalent to the one of the Kähler potential of a Calabi–Yau geometry, which we exploit to compute the conformal fishnet integrals directly from the data of their associated Calabi-Yau varieties. The techniques developed here provide a conceptual bridge between the abstract mathematics of (twisted) periods and the analytic computation of Feynman integrals, e.g., for scattering amplitudes

    Lysosomale Proteine in der Biomarkerforschung : Systematische Analyse klinischer Relevanz und proteomischer Nachweisstrategien

    Lysosomale Proteine werden zunehmend als potenzielle Biomarker für zahlreiche Erkrankungen diskutiert, ihre zuverlässige Detektion in klinischen Proben stellt jedoch weiterhin eine analytische Herausforderung dar. In dieser systematischen Literaturrecherche wurden 1.619 Studien ausgewertet, die genomische, transkriptomische oder proteomische Analysen humaner Proben durchführten. Insgesamt konnten 250 lysosomale Gene, Transkripte oder Proteine identifiziert werden, die in 245 unterschiedlichen Krankheitsentitäten als potenzielle Biomarker beschrieben wurden. Die Analyse zeigte, dass insbesondere luminale und lysosomal assoziierte Proteine häufig detektiert wurden, während membranständige lysosomale Proteine deutlich unterrepräsentiert waren. Ungerichtete proteomische Ansätze lieferten zwar eine breite Proteomabdeckung, zeigten jedoch eine unzureichende Sensitivität für lysosomale Proteine. Gezielte massenspektrometrische Verfahren (PRM, SRM, MRM) ermöglichten hingegen eine sensitive und reproduzierbare Quantifizierung lysosomaler Proteine auch in komplexen klinischen Proben und stellen damit die derzeit praktikabelste Strategie für deren Etablierung als Biomarker dar

    Relevanz hinweisgebender Verfahren und Einfluss präanalytischer Variablen auf die Befundinterpretation forensisch-toxikologischer Fragestellungen

    Die forensisch-toxikologische Beurteilung basiert auf der objektiven und validen Erfassung von Substanzen in verschiedenen Matrizes. Die Erhebung von Messwerten gliedert sich dabei in die präanalytische und analytische Phase. Insbesondere während der präanalytischen Phase können u. a. physikalisch-chemische Faktoren (z. B. in vitro-Hämolyse) sowie der Zeitraum zwischen einem Ereignis (z. B. eine Straftat) und der Probenasservierung die Integrität der Substanzkonzentration und damit die Befundinterpretation erheblich beeinflussen. Informationen zu einer möglichen Substanzaufnahme sowie der Einsatz hinweisgebender Verfahren ermöglichen die gezielte Anwendung nachfolgender Analysen. Ziel der in dieser Dissertation zusammengefassten Publikationen war es, die Zuverlässigkeit immunchemischer Drogenvortests in Blut, Mundflüssigkeit und Urin zu bewerten. Zudem wurde der Einfluss präanalytischer Variablen auf die Identifizierung und Konzentration von Analyten im Kontext verschiedener forensisch-toxikologischer Fragestellungen untersucht. Im Rahmen polizeilicher Verkehrskontrollen wurden verschiedene Drogenvortests hinsichtlich ihrer Zuverlässigkeit und Praktikabilität evaluiert. Die Ergebnisse der Studie ergaben, dass die überprüften Drogenvortests insgesamt eine zufriedenstellende Leistung erbrachten, wobei Verbesserungspotenzial, insbesondere bei der Reduzierung falsch-positiver Befunde, besteht. Weiterhin wurde deutlich, dass Polizeibeamte zufrieden und sicher im Umgang mit Speicheltests sind, jedoch Urintests bevorzugten. Die Ergebnisse legen zudem nahe, dass bei einer möglichen Erhöhung des THC-Grenzwertes im Serum eine Anpassung der Cut-off-Werte der Vortests für den weiteren Einsatz im Straßenverkehr erwogen werden sollte. Ein weiterer Schwerpunkt der Arbeit lag in der Untersuchung von Methamphetamin-Enantiomeren in zwei Blutproben. Der eingesetzte Immunoassay zeigte eine geringere Sensitivität gegenüber dem (R)-Enantiomer, was zu falsch-negativen Ergebnissen führen konnte. Derartige Testcharakteristika sind zu beachten, weshalb eine Bestätigung durch chromatographisch-massenspektrometrische Verfahren erforderlich ist. Zusätzlich wurde der Einfluss von Natriumfluorid als Stabilisator in Blutentnahmesystemen auf die Konzentration verschiedener Arzneimittelwirkstoffe im Blutserum untersucht. Es zeigte sich, dass eine NaF-induzierte Hämolyse und analytische Matrixeffekte zu abweichenden Konzentrationsbestimmungen führen können, was bei der Befundinterpretation zu berücksichtigen ist. Abschließend wurde das Potenzial des GHB-Fettsäureesters 4-Palmitoyloxybutyrat (GHB-Pal) als Biomarker zur Verlängerung des Nachweisfensters von GHB im Vollblut geprüft. Die Ergebnisse zeigten jedoch, dass GHB-Pal hierfür nicht geeignet ist. Insgesamt verdeutlichen die Arbeiten die Bedeutung eines umfassenden Verständnisses der eingesetzten Methoden und präanalytischen Einflussfaktoren, um forensisch-toxikologische Befunde valide und gerichtsfest bewerten zu können

    Microbiological Profiles of Patients with Acute Periprosthetic Joint Infection Undergoing Debridement, Antibiotics, Irrigation and Implant Retention (DAIR)

    Background: Periprosthetic joint infection (PJI) is one of the most serious complications following total joint arthroplasty. The debridement, antibiotics, irrigation, and implant retention (DAIR) procedure is commonly employed to treat acute, early-stage infections, but its success is highly variable, influenced by factors such as pathogen virulence and antibiotic susceptibility profiles. This study aimed to evaluate the impact of pathogens responsible for these infections on the outcome of DAIR. Methods: This retrospective, single-center study analyzed the microbiological profiles of 116 patients (66 hips and 50 knees) treated for acute periprosthetic joint infections (PJIs) with DAIR between 2018 and 2022. Acute PJI was defined as a duration of symptom less than three weeks, according to the criteria established by the Tsukayama and Izakovicova classification. Preoperative joint aspirations, intraoperatively collected tissue samples, and sonication of the exchanged mobile parts were analyzed for each case. We differentiated between monomicrobial PJI, polymicrobial PJI (defined as the identification of more than one microorganism from preoperative joint fluid aspiration or intraoperative samples), and difficult-to-treat (DTT) pathogens. Results: In this cohort, the following pathogen profiles were identified: culturenegative cases accounted for 11.1% of infections, while 64.2% were attributed to Grampositive bacteria, 19.8% to Gram-negative bacteria, and 4.9% to fungal pathogens. Among the identified microorganisms, coagulase-negative staphylococci (CNS) were the most frequently detected, exhibiting a notable oxacillin resistance rate of 52.9% and rifampicin resistance rate of 28.7%. Additionally, no significant difference in revision-free implant survival was found between patients with DTT pathogens and/or polymicrobial PJI and those without such infections. Conclusions: This study highlights that pathogens in prosthetic joint infections (PJIs) do not solely determine outcomes, as patient-specific factors (comorbidities, implant type) may also play a key role. Regional variations in pathogens and antibiotic resistance patterns should guide empirical therapy.     Nahrungsmittelunverträglichkeiten bei Hashimoto-Thyreoiditis

    Die Hashimoto-Thyreoiditis (HT) ist eine chronisch-entzündliche Autoimmunerkrankung der Schilddrüse, die vor allem Frauen betrifft. Auch bei euthyreoter Stoffwechsellage berichten viele Patientinnen und Patienten über anhaltende Beschwerden wie Gewichtszunahme, Erschöpfung, metabolische Dysregulationen und Mikronährstoffmängel. Ziel: Ziel dieser Arbeit war es, potenzielle Zusammenhänge zwischen Hashimoto-Thyreoiditis, Nahrungsmittelunverträglichkeiten (NU) sowie ausgewählten serologischen und anthropometrischen Parametern – insbesondere Schilddrüsenantikörpern, Vitamin-D-Spiegeln und dem Body-Mass-Index (BMI) – zu untersuchen. Methodik: Untersucht wurden 133 Patientinnen und Patienten mit gesicherter Hashimoto-Thyreoiditis. Die Datenerhebung erfolgte anhand standardisierter Fragebögen und laborchemischer Parameter. Analysiert wurden die Prävalenz von NU, geschlechtsspezifische Unterschiede sowie mögliche Korrelationen zwischen NU, Antikörpertitern (TPO- und Tg-Antikörpern), Vitamin-D-Status und BMI. Ergebnisse: Erhöhte Prävalenz von NU: 41 % der HT-Patientinnen und -Patienten berichteten über bestehende oder vermutete Nahrungsmittelunverträglichkeiten – ein hochsignifikant erhöhter Anteil im Vergleich zur europäischen Normalbevölkerung (16 %, p Kein Einfluss von NU: Zwischen dem Vorliegen von NU und Geschlecht (p = 0,238), TPO-Antikörpern (p = 0,29), Tg-Antikörpern (p = 0,32) sowie Vitamin-D-Status (p = 0,908) bestand kein signifikanter Zusammenhang. Vitamin-D-Mangel unabhängig von NU: Ein Vitamin-D-Mangel lag bei 65 % aller HT-Patientinnen und -Patienten vor – deutlich häufiger als in der europäischen Allgemeinbevölkerung (15 %; Projekt ODIN; Rabenberg & Mensink 2016; p Assoziation zwischen BMI und Vitamin D: Patientinnen und Patienten mit einem BMI ≥ 25 kg/m² wiesen signifikant niedrigere Vitamin-D-Spiegel auf als Normalgewichtige (p = 0,001; Cohen's d = 0,56). Schlussfolgerung: Die Ergebnisse zeigen eine erhöhte Prävalenz von Nahrungsmittelunverträglichkeiten und einen häufigen Vitamin-D-Mangel bei Hashimoto-Thyreoiditis. Während Nahrungsmittelunverträglichkeiten keinen Einfluss auf Geschlecht, Antikörpertiter oder Vitamin-D-Status hatten, war der Vitamin-D-Mangel unabhängig von NU und signifikant mit einem höheren BMI assoziiert. Diese Befunde unterstreichen die Bedeutung einer individualisierten ernährungs- und präventionsmedizinischen Betreuung von HT-Patientinnen und -Patienten, die Mikronährstoffstatus und Gewichtsregulation berücksichtigt. Weitere Studien sind erforderlich, um die pathophysiologischen Zusammenhänge und therapeutischen Optionen im Kontext von HT, Ernährung und Mikronährstoffmetabolismus zu vertiefen.Food Intolerances in Hashimoto's Thyroiditis Background: Hashimoto's thyroiditis (HT) is a chronic autoimmune inflammation of the thyroid gland that predominantly affects women. Even in a euthyroid state, many patients report persistent symptoms such as weight gain, fatigue, metabolic dysregulation, and micronutrient deficiencies. Objective: The aim of this study was to investigate potential associations between Hashimoto's thyroiditis, food intolerances (FI), and selected serological and anthropometric parameters, in particular thyroid autoantibodies, vitamin D levels, and body mass index (BMI). Methods: A total of 133 patients with confirmed Hashimoto's thyroiditis were examined. Data were collected using standardized questionnaires and laboratory analyses. The prevalence of FI, sex-specific differences, and possible correlations between FI, antibody titers (TPO and Tg antibodies), vitamin D status, and BMI were analyzed. Results: Increased prevalence of FI: 41% of HT patients reported existing or suspected food intolerances, a highly significant increase compared to the European general population (16%, p No effect of FI: No significant associations were found between FI and sex (p = 0.238), TPO antibodies (p = 0.29), Tg antibodies (p = 0.32), or vitamin D status (p = 0.908). High prevalence of vitamin D deficiency independent of FI: Vitamin D deficiency was present in 65% of all HT patients, significantly higher than in the European general population (15%; Project ODIN; Rabenberg & Mensink 2016; p Association between BMI and vitamin D: Patients with a BMI ≥ 25 kg/m² showed significantly lower vitamin D levels compared to normal-weight individuals (p = 0.001; Cohen's d = 0.56). Conclusion: The findings demonstrate an increased prevalence of food intolerances and a high rate of vitamin D deficiency in patients with Hashimoto's thyroiditis. While FI showed no influence on sex, antibody titers, or vitamin D status, vitamin D deficiency occurred independently of FI and was significantly associated with higher BMI. These results highlight the importance of individualized nutritional and preventive medical care for HT patients, including assessment of micronutrient status and weight management. Further studies are needed to elucidate the underlying pathophysiological mechanisms and to develop targeted nutritional and therapeutic strategies in the context of HT, nutrition, and micronutrient metabolism
