SOP for analysis of cell wall polysaccharides from yam and plantain
Cell walls in plants are mainly composed of polysaccharides. Among them, pectin, cellulose and hemicellulose are composed of various sugars residues (acidic: galacturonic acid in pectins; or neutral: rhamnose, galactose, arabinose, xylose, glucose in cellulose, hemicellulose and pectin side chains). This fine composition impacts the structure but also the cooking behavior and mechanical properties of cell walls.
This method seeks to determine the cell wall composition (neutral sugars composition and galacturonic acid content) of destarched AIS (Alcohol Insoluble Solid) extracted from raw and cooked yam and banana. It is applicable for AIS extracted from every fruit or vegetable product and containing a low amount of residual starch. For RTB products a destarching step is mandatory before analysis.
First the samples are hydrolyzed in order to obtain sugar residues. Pectin and hemicellulose are degraded into their individual neutral and acidic sugars by a simple acid hydrolysis (hot concentrated sulphuric acid). In contrast, cellulose has to be solubilized under harsher conditions. For this reason, a double hydrolysis is performed. Afterwards, the free neutral sugars are derivatized into volatile alditol acetates before their separation in Gas Chromatography with a Flamme Ionization Detector (GC-FID). They are then quantified thanks to an internal standard of inositol. Whereas free galacturonic acid, transformed to its furfuric derivatives, is quantified by colorimetry after reaction with 3-phenylphenol (MHDP) which produces a fuschia pink complex with a maximum absorption at 520 nm. While long-term organic fertilizer (OF) application tends to decrease copper (Cu) and zinc (Zn) availability in agricultural soils, earthworm bioturbation tends to have the opposite effect. Consequences in terms of Cu and Zn bioavailability to earthworms in bioturbated, OF-amended soils are hence still a matter of debate. Accordingly, we assessed the effect of a decade of agronomically-realistic OF applications on Cu and Zn availability in earthworm-inhabited soils and their consequence on Cu and Zn bioavailability to earthworms. An epi-endogeic species (Dichogaster saliens) was exposed in microcosms to three soils receiving at field scale either no, mineral or organic fertilization for a decade. Dissolved organic matter (DOM) properties (i‧e. concentration, aromaticity, and binding properties towards Cu), pH, and Cu and Zn availability (i‧e. total concentration and free ionic activity) were determined in the solution of earthworm-inhabited soils. Cu and Zn bioavailability was assessed by measuring the net accumulation and concentration of Cu and Zn in earthworms. Despite soil Cu and Zn contamination induced by a decade of OF applications, the organic fertilization-induced increase in pH and DOM properties drove the mitigation of Cu and Zn availability in earthworm-inhabited soils, while bioturbation had little effect. Consistently, Cu and Zn bioavailability did not increase with OF applications. From an ecotoxicological perspective, our results suggest that agronomically-realistic applications of OF for a decade should not constitute a risk for earthworms in terms of bioaccumulation and toxicity of Cu and Zn

This map gives the spatial distribution of the major land use in a cell of 1500m x 1500m=2.25 km2, also called the landscape matrix, over an area of study of landscape dynamics in Madagascar. It consists of an area of 84 square kilometers located at the West of Vavatenina town, in the Analanjirofo Region, on the North-East coast of Madagascar. This map is based on the very high spatial resolution land use / land cover map of the area produced for 2019 (doi:10.18167/DVN1/KDNOV4), processing a majority analysis inside sliding windows of 1500m in dimension. 7 types of land-uses were here identified as landscape matrix: diversified agroforests, introduced timber plantations, low-diversified and diversified woody fallow, bramble-dominated and diversified shrubby fallows, and annual crops or other short vegetation

Gendered boiled cassava product profile in Benin
The WP1 Gendered Food Product Profile for Gari-Eba in Nigeria reflects the final step (step 5) of an interdisciplinary five-step methodology developed to identify demand for quality characteristics among diverse user groups along the food chain (Forsythe et al., 2022). This methodology includes: step 1) interdisciplinary state of knowledge of the product ; step 2) gendered food mapping, which includes participatory research with men and women in rural communities regarding their product preferences and priorities; step 3) participatory processing diagnosis and quality characteristics ; and step 4) consumer studies in rural and urban areas of the product using different RTB varieties

This map gives the spatial distribution of the main agricultural systems and land uses present in an area of study of landscape dynamics in Madagascar. It consists of an area of 84 square kilometers located at the West of Vavatenina town, in the Analanjirofo Region, on the North-East coast of Madagascar. This map is based on Pleiades very high spatial resolution multispectral images (50cm/pixel) acquired in 2019, and field reference data. The nomenclature includes 13 classes, with both agroforestry and annual cropping systems, various fallows discriminated on the basis of their density and diversity, water bodies and anthropized areas. The global precision of this map is 80%
