Apport de la métabolomique à l’étude de la phase postprandiale
International audienceLa métabolomique a ouvert de nouvelles perspectives dans les études des relations nutrition/santé : elle permet actuellement d’avoir une idée des changements métaboliques à l’échelle de l’organisme entier, et ainsi d’intégrer la complexité des effets métaboliques de l’alimentation. L’objectif du présent projet était d’explorer l’effet de différentes matières premières protéiques laitières sur la réponse postprandiale induite par un repas riche en graisse. L’intervention a consisté en 3 journées d’exploration postprandiale sur 6 heures, selon un dispositif croisé, en ordre randomisé, chez 11 hommes volontaires sains en surpoids. Les volontaires ont consommé à jeun un des trois repas-tests apportant 1200 kcal, composé de 70% de lipides, 15% de glucides et 15% de protéines laitières (caséines, protéines solubles du lactosérum, ou protéines solubles enrichies en α-lactalbumine). Des prélèvements de sang ont été réalisés avant le repas-test, puis en cinétique à 30 min, 60 min, 90 min, 2h, 3h, 4h et 6h après pour le dosage des triglycérides et des apolipoprotéines B-48 plasmatiques. Des prélèvements d’urines ont été réalisés avant le repas- test, puis toutes les deux heures. Ces échantillons d’urines ont été randomisés puis analysés par une approche métabolomique non ciblée: les urines diluées ont été injectées sur un couplage UPLC/QToF, équipé d’une source électrospray. Les données brutes ont tout d’abord été extraites par XCMS avant analyses statistiques. Toutes les données collectées ont été traitées avec des modèles mixtes, avec ajustement de Bonferroni-Hochberg pour les données de métabolomique, et des tests a posteriori de Bonferroni. De plus, pour les données métabolomiques, des analyses multivariées sur tous les ions et sur des sous-groupes d’ions ont ensuite été effectuées tous temps confondus ou par temps de cinétique. Enfin, des corrélations entre données cliniques et données métabolomiques ont été réalisées. Au niveau clinique, les aires sous courbes des cinétiques plasmatiques des triglycérides ou des apolipoprotéines B-48 sont diminuées après consommation du repas hyperlipidique avec des caséines, par rapport aux deux autres repas-tests, ce qui suggère une modification de l’absorption des lipides. Au niveau métabolomique, 139 ions sont significativement modulés en fonction des protéines testées, avec une capacité équivalente à discriminer les 3 types de repas-tests, mais notablement plus d’ions discriminant le repas riche en protéines du lactosérum des deux autres. 36 de ces 139 ions sont caractéristiques de l’interaction temps-type de protéine, dont 20 ions davantage discriminants dans les modèles multivariés (VIP>1). Par ailleurs, tous repas-tests confondus, les corrélations entre les données métabolomiques et cliniques sont significatives pour la moitié de ces ions caractéristiques de l’interaction temps-protéine, dont 8 ions avec VIP>1. Le nombre d’ions corrélés aux données cliniques est différent entre les repas-tests si nous les considérons séparément. En conclusion, les 3 types de protéines consommées dans des repas-tests hyperlipidiques induisent des réponses postprandiales cliniques et métaboliques différentes. La métabolomique non ciblée effectuée sur des prélèvements urinaires a permis d’apporter des informations complémentaires aux mesures cliniques : l’identification des métabolites caractéristiques de l’interaction temps-protéines permettra de confirmer que ces protéines ont, entre autres, un effet différent en lien avec le métabolisme postprandial des lipide
AGRAF: un modèle d'échanges de matières premières agricoles et de biocarburants au sein de l'Union Européenne
National audienceThe search for energetic independence, for boosting local economies and for reducing polluting emissions, has supported bioenergy’s development and biofuels in particular. Mandatory biofuels incorporation rates have been recently established in several countries, including the European Union, in order to promote the use of renewable energetic resources in spite of several criticisms toward biofuels relating to their impacts on food security, the environment and the conflicts associated with resource allocations. To study existing concerns on the growth, the profitability, the limiting factors biofuels, as well as the impacts of current policies in force, a highly parameterized operational modelling tool (AGRAF) dedicated to the European area has been developed. Based on the coupling of three economic models (AROPAj, OURSEurope and GIRAF), each one representing a key sector of the biofuel production chain (respectively agriculture, refining and biofuel manufacturers), his approach allows to assess different scenarios based on different technical, economic, environmental and political criteria. Furthermore, it helps highlighting the biofuel production potential growth (quantity, nature,location, by products and polluting emissions), related costs (investments, transportation, raw material) and exchanges between the agents. AGRAF incorporates biomass and biofuels importations from outside Europe and takes into account both first and second generation biofuels (involved in the European biofuel policy scheme for 2030). These features help identifying the levers and opportunities for reaching European Union bioenergy’s targets over a medium-long term.Malgré leurs impacts négatifs sur la sécurité alimentaire, sur l’utilisation des terres agricoles et de la ressource en eau, les biocarburants ont récemment bénéficié de nombreuses initiatives de développement à travers le monde, afin de réduire les émissions de gaz à effet de serre et pour atteindre l’indépendance énergétique. Dans le but d’étudier les impacts des politiques en vigueur ainsi que les contre parties négatives associées à la croissance de ce secteur, un modèle (AGRAF) entièrement paramétré représentant le secteur européen a été développé. Sa conception repose sur le couplage de trois modèles économiques représentants la production agricole (AROPAj), la transformation (GIRAF) et le raffinage (OURSEurope), chacun étant une entité clé dans leur chaîne de production. Cette approche permet d’étudier différents scénarios selon des critères techniques, économiques, environnementaux et politiques. De plus, il met en lumière le potentiel de croissance du secteur des biocarburants de première et de seconde génération, les coûts associés ainsi que les échanges entre les opérateurs, en Europe et à travers le monde. Ainsi, les caractéristiques du modèle AGRAF permettent d’identifier les leviers et les opportunités qui permettraient d’atteindre les objectifs d’incorporation fixés par l’Union Européenne à moyen terme
Organisation des filières de ruminants : Quelles évolutions ? Quelles alternatives pour les éleveurs ?
National audienceThe durability of French ruminant livestock systems is currently uncertain, in particular in terms of economic viability. This depends especially on the evolution of the downstream sectors’ organization. This synthesis provides an overview on the processes occurring and identifies the stakes for breeders. It refers to a literature review and to some case studies on transformations in the bovine and ovine meat sectors and bovine, ovine and caprine milk sectors. At first, we looked at the main movements regarding the demand of dairy and meatbased products. Secondly, we examined the important reorganizations of these sectors under the influence of the markets’ liberalization: concentration, contractualization. Thirdly, we noted the stakes for breeders and how they may be taken care of: cooperatives, demarcation, shortening of circuits and transformation of breeding activities.Cette synthèse a pour objectif de faire une analyse critique des grandes évolutions de l’organisation des filières bovins et ovins viande ainsi que bovins, ovins et caprins lait pour ensuite identifier et discuter des possibilités d’actions qu’ont les éleveurs. Elle s’appuie sur une recherche bibliographique ainsi que sur des cas d’études concernant les transformations de ces filières. Dans un premier temps, nous dégageons les principaux mouvements en matière de demande des produits laitiers et carnés. Dans un second temps, nous examinons les recompositions très fortes au sein des filières sous l’effet de la libéralisation des marchés : concentration, contractualisation. Puis, la troisième partie fait état des possibilités d’action qu’ont les éleveurs face à ces recompositions : coopération, démarcation, raccourcissement des circuits et transformations des activités d’élevage
Modelling anaerobic digestion and compost of organic residues: towards organic matter fate prediction in soils
Communication oraleInternational audienceIn a context of environmental biorefinery, organic residues such as sludge, biowaste, manure are no more considered as wastes but as potential resources for some targeted services. These services encompass the energy production through the methane produced by their anaerobic digestion, the cropped soils fertilisers and amendments production from digestates, and other value-added compounds production (e.g. organic acids and hydrogen obtained during fermentation). There are currently only few studies focused on combined optimization of energetic and agronomic values of organic wastes. One way to optimize both objectives is through process modelling. However, input variables from all existing models are different and there is a need to find relevant and shared input variables to model the fate of organic residues during their treatments. Recently, a new promising methodology of organic matter characterization has been successfully used to predict anaerobic and aerobic biodegradability and bioaccessibility of a broad range of organic residues (Jimenez et al., 2015). This methodology is based on the combination of chemical fractionation simulating organic matter accessibility and of a 3D fluorescence spectroscopy highlighting fraction’s complexity. The fractions obtained were used as models input variables. Existing models (Batstone et al., 2002, Zhang et al., 2012, Garnier et al., 2003) have been modified to take into account these new variables. In order to calibrate and validate the models, a lab scale reactor of anaerobic digestion was operated to treat wastewater sludge. Biogas, methane proportion, pH and input and output digestate quality were monitored. After 60 days of steady state operation, the digestate was then centrifuged to separate the liquid from the solid part. The solid part was then added with green waste into a lab scale compost reactor. Temperature, CO2 production, moisture and compost quality were monitored. Finally, the final compost was used as substrate for cropped soil incubation. First results showed that, the new anaerobic digestion model was successfully able to predict biogas and digestate quality when model and experimental data were compared. The final quality of the solid part of the simulated digestate was then introduced with the green waste fractionation to model composting. The modified compost model also fitted the quality of compost during the different steps of the composting. Concerning soil incubation modelling, experimental results are still ongoing but will be part of the oral presentation. Overall, the obtained results constitute a strong basis for the development of a decision support system to optimize the design and operation of organic wastes treatment plants

Independent components analysis: theory, applications and difficulties
Independent Components Analysis (ICA) is a blind source separation method that has been developed to extract the underlying source signals from a set of observed signals where they are mixed in unknown proportions. This is made possible by making the assumption that the “pure” sources are by definition totally unrelated (“independent”) with non-gaussian intensity distributions, whereas their mixtures have more gaussian distributions due to the Central Limit Theorem. ICA can thus be used not just to analyse 2D datasets (e.g. IR spectra) but also multiway datasets (e.g. 3D Excitation-Emission fluorescence spectra) after unfolding. Like other methods based on latent variables, a careful investigation has to be carried out to determine which components are significant and which are not. Therefore, it is important to dispose of valid procedures to decide on the optimal number of independent components (ICs) to extract in the final ICA model. The objective of this presentation is to introduce ICA through the study of several real cases, and to show how it performs compared to other more classical multivariate methods such as Principal Components Analysis (PCA). In this way, the relative advantages and disadvantages of ICA will be pointed highlighted
Kinetic study of furan and furfural generation during baking of cake models
International audienceThis study describes the kinetics of furan and furfural generation in a cake model, for the first time. These process-induced compounds impact safety and sensory aspects of baked products. Understanding their generation with regards to process dynamics will serve food quality design. However, the complexity of real products makes this task challenging. This work provides a novel approach to understand and model chemical reactivity by implementing an inert cake model (starch, water and cellulose), specifically designed for mimicking a sponge cake structure. The addition of reaction precursors (glucose and leucine) to follow Maillard and caramelization reactions, resulted in browning and generated considerable levels of furanic compounds (up to 17.61 ng/g for furan and 38.99 mu g/g for furfural, dry basis). Multiresponse data modeling resulted in a kinetic model which adequately describes experimental concentrations and makes it possible to estimate the degradation of precursors and the behavior of two hypothetic intermediates
Whole-genome resequencing of honeybee drones to detect genomic selection in a population managed for royal jelly
International audienceFour main evolutionary lineages of A. mellifera have been described including eastern Europe (C) and western and northern Europe (M). Many apiculturists prefer bees from the C lineage due to their docility and high productivity. In France, the routine importation of bees from the C lineage has resulted in the widespread admixture of bees from the M lineage. The haplodiploid nature of the honeybee Apis mellifera, and its small genome size, permits affordable and extensive genomics studies. As a pilot study of a larger project to characterise French honeybee populations, we sequenced 60 drones sampled from two commercial populations managed for the production of honey and royal jelly. Results indicate a C lineage origin, whilst mitochondrial analysis suggests two drones originated from the O lineage. Analysis of heterozygous SNPs identified potential copy number variants near to genes encoding odorant binding proteins and several cytochrome P450 genes. Signatures of selection were detected using the hapFLK haplotype-based method, revealing several regions under putative selection for royal jelly production. The framework developed during this study will be applied to a broader sampling regime, allowing the genetic diversity of French honeybees to be characterised in detail
