Wildtype peers rescue social play and 50-kHz ultrasonic vocalization deficits in juvenile female Cacna1c heterozygous rats
Background: Healthy brain development depends on early social practices and experiences. The risk gene CACNA1C is implicated in numerous neuropsychiatric disorders, in which key characteristics include deficits in social functioning and communication. Recently, we reported sex-dependent impairments in social behavior and ultrasonic vocalizations (USV) in juvenile heterozygous Cacna1c+/− (HET) rats. Specifically, HET females displayed increases in rough-and-tumble play that eliminated the typically observed sex difference between male and female rats. Interestingly, female wild-type Cacna1c+/+ (WT) pairs also showed a similar increase in social play when housed with HET females, suggesting their behavior may be influenced by HET cage mates. This indicates that the genetic makeup of the social environment related to Cacna1c can influence social play, yet systematic studies are lacking.
Methods: In the present study, we housed juvenile females in MIXED- or SAME-genotype cages and tested them in a social play paradigm with a same- and opposite-genotype partner.
Results: The results show that the early social environment and the genotype of the play partner influence social play and 50-kHz USV emission. Experience with a WT play partner appears necessary for HET females to show comparable levels of play and 50-kHz USV emission. Same-genotype HET pairs played less and emitted fewer 50-kHz USV than same-genotype WT or opposite-genotype pairs; however, we found that the decrease in social play and 50-kHz USV in HET pairs can be rescued by playing with a WT partner. The effect was particularly prominent when the first play partner was WT, as we found it increased play and 50-kHz USV emission in all subsequent interactions with ensuing partners.
Conclusion: These findings suggest that the genetic makeup related to the social environment and/or social peers influences social play in Cacna1c+/− haploinsufficient rats. Specifically, our results show that WT peers can rescue behavior and communication alterations in Cacna1c female rats. Our findings have important implications because they show that the genetic makeup of the social environment can divulge phenotypic changes in genetic rat models of neuropsychiatric disorders.Gefördert durch den Open-Access-Publikationsfonds der UB Marburg
Above, but close to two percent. Evidence on the ECB’s inflation target using text mining
Due to its official mandate, the European Central Bank (ECB) is assumed to maximize an implied objective function that leads it to pursue inflation with a subordinate focus on supporting the general economic policy of the European Union. This objective is – by its very nature – difficult to quantify. My paper tries to decipher information regarding the ECB’s objective through the use of text mining on all public speeches between 2002 and 2020. The estimation of a sentiment index through a ’bag-of-words’-approach yields the following results. First, the findings of my analysis suggest a concave objective regarding the inflation rate. The implied inflation target is best summarized as an inflation rate of ’above, but close to 2%’. Deviations from this target lead to a reduction in the sentiment of the institutions’ communication. Second, my findings suggest a convex objective towards output growth and a linear objective towards the unemployment rate, with a preference for higher GDP growth and employment independently of the current level. Furthermore, the hierarchical order in the the European Central Bank (ECB)’s mandate does not always appear to be consistent with my findings. Deviations from its primary objective, the inflation rate, appear to be of no greater concern than deviations in its subordinate objective. Third, in periods of heightened uncertainty, there is an additional decrease in the sentiment of speech. Last, over the last two decades, speeches have become more pessimistic, even when controlling for macroeconomic conditions
The development of cryptocurrencies such as Bitcoin after the global financial crises from 2007 onwards has put the financial sector into trouble. Some voices even speak of the possibility of abolishing cash. This article shows, first, whether and to what extent cryptocurrencies can replace cash, second, to what extent they change non-cash payment systems, and third, which obstacles have to be overcome on the way to large acceptance. We shall first scrutinize the financial sector's response to the multifaceted phenomenon of the Distributed Ledger Technology (“blockchain”)
Computed tomography (CT) has become the diagnostic modality of choice for head trauma due to its accuracy, reliability, safety, and its availability. CT scanning of the head is typically used to detect infarction, calcifications, tumors, bone trauma, and hemorrhage.
Motion correction (MC) is of general interest in CT imaging. Motion correction techniques have two categories. The first group requires motion information and consists of motion acquisition and motion compensation processes. The motion acquisition derives the motion information from reference images, surrogate signals, or the data themselves. Motion compensation compensates the motion during a reconstruction process. The second group is based on image-processing techniques and corrects the motion without prior knowledge of the motion. Based on what prior information is available, one can choose the appropriate approach to perform the motion correction.
The study aimed to validate the data-driven 3D iterative motion estimation (ME) and motion compensation algorithm on phantom as well as clinical studies with head movements during computed tomography (CT) scan and to optimize the data-driven 3D iterative algorithm for robust application.
The Hoffman 3D brain phantom provides a quantitative and qualitative study of the three-dimensional effects of scatter and attenuation similar to the human brain. We also performed MC for head movement during the CT part of the scan of the brain PET/CT and examined its significance for final image reconstruction. A series of PET/CT scans of Hoffman's brain phantom filled with fluorodeoxyglucose (18F-FDG) were acquired using mCT Siemens Biograph PET/CT scanner. The phantom was acquired with a variety of movements during the CT part of the acquisition, to simulate patient movements, but the phantom remained stationary during the PET scan. Each motion-corrupted CT scan was reconstructed using fully automated 3D iterative data-driven image reconstruction with motion compensation (MC) to remove motion artifacts and afterward an attenuation map was generated from this MC CT. The PET raw data was reconstructed offline using the JSrecon algorithm with an attenuation map from motion-corrected CT and compared with the PET scan reconstructed with an attenuation map from motion-corrupted CT. The data-driven motion compensation approach was also implemented on patients presenting head movement during the CT part of the brain PET/CT scan. All reconstructed images were independently assessed for qualitative analysis and the scenium analysis was performed for quantitative analysis. The reconstructed PET images of 10 primary brain regions using both nMC-PET and MC-PET were analyzed and the results showed that the SUVmean of all brain regions in nMC-PET was significantly higher than those in MC-PET. The 3D-standard surface projection (3D-SSP) Z score was evaluated on both nMC-PET and MC-PET. The 3D-SSP method compares the data from the individual to a database of healthy controls by defining a large number of points on a spatially normalized brain surface. With the little head motion, motion correction had only a slight impact on the Z score image in qualitative terms.
The data-driven iterative motion compensation approach for head CT significantly increases the quantitative and qualitative accuracy of the PET/CT brain image affected by patient movement. The method could be applied to both stand-alone helical CT scans and the CT component of hybrid imaging systems such as PET/CT and SPECT/CT.Die Computertomographie (CT) hat sich aufgrund ihrer Genauigkeit, Zuverlässigkeit, Sicherheit und Verfügbarkeit zur diagnostischen Methode der Wahl bei Kopftraumata entwickelt.
Effects of Monetary Policy Communication in Emerging Market Economies: Evidence from Malaysia
By conducting a high-frequency event study similar to Gürkaynak et al.
(2005), we find that two factors are needed to adequately capture the effects
ofmonetary policy announcements for a non-inflation targeting emerging
market economy, Malaysia. These factors are the surprise changes in
the policy rate (Overnight Policy Rate, OPR) and the information about
the future path of monetary policy. We find that the path factor has a
strong influence on long-term government bond yields, corporate bond
yields and spreads. Our findings are indicative of the view that monetary
policy communication is mostly about revealing information pertaining to
the central bank’s assessment of the economic outlook, as opposed to an
unconditional binding commitment to follow a specific policy path
Experiences and expectations of family caregivers of people with a migrant background with memory problems or dementia in the GP practice: an interview study
Background: In Germany, there is an increasing number of people with dementia and a migration background, who are often cared for by relatives at home. The needs of migrant family caregivers for primary care physicians are unclear and the experiences and expectations of this population group in Germany are not sufficiently studied.
Research question: How do family caregivers of people with mild cognitive impairment or dementia who have a migration background experience primary care in Germany? What expectations do they have of family physicians?
Methods: 18 family caregivers with a non-German-speaking migrant background were recruited via General practices or support organizations who care for a person with mild cognitive impairment or dementia at home. Semistructured guided interviews (3 face-to-face, 15 by telephone), were conducted. Interviews were recorded, transcribed verbatim, and analyzed thematically using MAXQDA software.
Results: Five main themes were identified. (1) family doctors as coordination centers (2) trust (3) initiative (4) barriers to counseling (5) burden.
Family physicians are seen as people of trust and medical coordination centers who are seen as the first point of contact. There are barriers for family caregivers regarding medical consultation, where family physicians can be supportive. Family caregivers sometimes criticize insufficient information and a lack of recognition of their role as informal caregivers. A supportive and culturally sensitive attitude on the part of family doctors is appreciated. Family caregivers feel obliged to show initiative themselves in order to receive appropriate dementia care. It is often expected that more initiative comes from family doctors. Respondents report high caregiving burdens. Home care is preferred, but also seen as having no alternative, for example because there is a lack of cultural and dementia-specific services.
Discussion: Family doctors play a central role for the surveyed family caregivers. The respondents particularly value trusting care that is proactive and takes into account the barriers and burdens of the family caregivers. They expect information, consideration, helpfulness and holistic care. Culturally sensitive support as well as patient-centered medicine is needed. The wide range of migration backgrounds as well as dementia types in this study may have been limiting, but were equally beneficial due to desired heterogeneity. Further studies of this complex care setting are required
In vitro and in ovo photodynamic efficacy of nebulized curcumin-loaded tetraether lipid liposomes prepared by DC as stable drug delivery system
Lung cancer is one of the most common causes of high mortality worldwide. Current treatment strategies, e.g., surgery, radiotherapy, chemotherapy, and immunotherapy, insufficiently affect the overall outcome. In this study, we used curcumin as a natural photosensitizer in photodynamic therapy and encapsulated it in liposomes consisting of stabilizing tetraether lipids aiming for a pulmonary drug delivery system against lung cancer. The liposomes with either hydrolyzed glycerol-dialkyl-glycerol tetraether (hGDGT) in different ratios or hydrolyzed glycerol-dialkyl-nonitol tetraether (hGDNT) were prepared by dual centrifugation (DC), an innovative method for liposome preparation. The liposomes' physicochemical characteristics before and after nebulization and other nebulization characteristics confirmed their suitability. Morphological characterization using atomic force and transmission electron microscopy showed proper vesicular structures indicative of liposomes. Qualitative and quantitative uptake of the curcumin-loaded liposomes in lung adenocarcinoma (A549) cells was visualized and proven. Phototoxic effects of the liposomes were detected on A549 cells, showing decreased cell viability. The generation of reactive oxygen species required for PDT and disruption of mitochondrial membrane potential were confirmed. Moreover, the chorioallantoic membrane (CAM) model was used to further evaluate biocompatibility and photodynamic efficacy in a 3D cell culture context. Photodynamic efficacy was assessed by PET/CT after nebulization of the liposomes onto the xenografted tumors on the CAM with subsequent irradiation. The physicochemical properties and the efficacy of tetraether lipid liposomes encapsulating curcumin, especially liposomes containing hGDNT, in 2D and 3D cell cultures seem promising for future PDT usage against lung cancer.Gefördert durch den Open-Access-Publikationsfonds der UB Marburg
Bacterial extracellular vesicles repress the vascular protective factor RNase1 in human lung endothelial cells
Background: Sepsis is one of the leading causes of death worldwide and characterized by blood stream infections associated with a dysregulated host response and endothelial cell (EC) dysfunction. Ribonuclease 1 (RNase1) acts as a protective factor of vascular homeostasis and is known to be repressed by massive and persistent inflammation, associated to the development of vascular pathologies. Bacterial extracellular vesicles (bEVs) are released upon infection and may interact with ECs to mediate EC barrier dysfunction. Here, we investigated the impact of bEVs of sepsis-related pathogens on human EC RNase1 regulation.
Methods: bEVs from sepsis-associated bacteria were isolated via ultrafiltration and size exclusion chromatography and used for stimulation of human lung microvascular ECs combined with and without signaling pathway inhibitor treatments.
Results: bEVs from Escherichia coli, Klebsiella pneumoniae and Salmonella enterica serovar Typhimurium significantly reduced RNase1 mRNA and protein expression and activated ECs, while TLR2-inducing bEVs from Streptococcus pneumoniae did not. These effects were mediated via LPS-dependent TLR4 signaling cascades as they could be blocked by Polymyxin B. Additionally, LPS-free ClearColi™ had no impact on RNase1. Further characterization of TLR4 downstream pathways involving NF-кB and p38, as well as JAK1/STAT1 signaling, revealed that RNase1 mRNA regulation is mediated via a p38-dependent mechanism.
Conclusion: Blood stream bEVs from gram-negative, sepsis-associated bacteria reduce the vascular protective factor RNase1, opening new avenues for therapeutical intervention of EC dysfunction via promotion of RNase1 integrity. Benchmark Value Added Chains and Regional Clusters in German R&D Intensive Industries
Although the phase of euphoria seems to be over, policymakers and regional agencies have maintained their interest in cluster policy. Modern cluster theory provides reasons for positive external effects that may accrue from interaction in a group of proximate enterprises operating in common and related fields. While there is some progress in locating clusters, in most cases only limited knowledge on the geographical extent of regional clusters is established. The present paper presents a hybrid approach to cluster identification. While dominant buyer-supplier relations are derived by qualitative input-output analysis (QIOA) from national I-O tables, potential regional clusters are identified by spatial scanning. This procedure is employed to identify clusters of German R&D intensive industries. In a sensitivity analysis, good robustness properties of the hybrid approach are revealed with respect to variations in the quantitative cluster composition
