Wachstum und Optimierung von Ga(N,As,P,Sb)/(B,Ga)(As,P) Heterostrukturen für Laser Anwendungen auf Si(001)
During the past years the field of photonic integrated circuits has gained increased attention by the scientific and industrial community. The integration of photonic devices into the existing CMOS technology (complementary metal-oxide-semiconductor), i.e. Si chip manufacturing, offers a lot of room for improvement concerning data transmission towards high bandwidth density, but also in the field of gas sensing or biomedical applications as affordable and wearable, small chip-sized devices. Especially in the field of data transmission, efficient and reliable laser light sources are required in the near infrared spectral range. Due to the lack of efficient Si-based light sources, the integration of a laser on CMOS compatible Si(001) 300 mm wafers, which are standardized in silicon industry, remains a large challenge up to date. This work aims to realize lasers grown lattice matched on exactly oriented Si(001) substrates via metalorganic vapour phase epitaxy (MOVPE). Therefore, the optimization of Ga(N,As,P,Sb)/(B,Ga)(As,P) heterostructures is pursued for the goal of room temperature lasing. This includes the MOVPE growth of the heterostructures themselves, their characterization via high resolution X-ray diffractometry (HR-XRD), atomic force microscopy (AFM), electrochemical capacitance-voltage profiling (ECV) and photoluminescence spectroscopy (PL). Broad area laser structures are processed for electrically pumped lateral carrier injection geometries as required for on-chip devices. Laser structures are characterized via electroluminescence spectroscopy (EL). The lattice matched bulk layers of realized electroluminescence devices consist of the indirect band gap semiconductor (B,Ga)(As,P). In investigated devices (B,Ga)(As,P) layers lead to an increased onset voltage as well as differential resistance compared to well established materials such as GaAs in established devices. In this work the (B,Ga)(As,P) material system is extensively investigated in terms of growth conditions and doping in order to optimize the layers for laser applications. A significant reduction of the onset voltage from 4.5(1.5) V down to 1.6(1) V is achieved. This improvement is associated with large reductions of energy and heat dissipation of the devices which makes them electrically competitive to industrially established laser devices on other substrates. The optically active Ga(N,As,P,Sb) quantum well (QW) layers are grown under various MOVPE conditions and improved towards smooth interfaces with the surrounding barriers and stronger luminescence. Extended defect, that form mainly during the overgrowth of modulated upper QW interfaces, become visible in AFM topographies due to the formation of 3 dimensional platelets on the heterostructures surfaces. At 575°C growth temperature Ga(N,As,Sb) was optimized in which Sb is assumed to mainly act as a surfactant. Test structures that contain about 9.5% N in the QW are found to result in low defect formation during overgrowth compared to Sb-free structures while also resulting in high optical output powers in PL- as well as EL-structures. At about 11% N the test structures surface roughness, i.e. of overgrown QWs, could be reduced down to about 0.25 nm RMS roughness in AFM topography scans and the platelet formation was completely suppressed. The optical output power of structures containing 11% N and more reduces compared to those containing 9.5% N despite the smoother interfaces which is likely caused by non-radiative recombination from N related states like N-chains or N-cluster configurations in the QWs. Unsymmetrical di-methylhydrazine (UDMHy), a standard MOVPE precursor for nitrogen incorporation, as well as the novel N-As precursor ditertiarybutylaminoarsane (DTBAA) are compared in detail for the growth of Ga(N,As,P) QW layers. Nearly comparable optical output power of electroluminescence structures is realized using DTBAA as N-precursor showing its great potential due to the high N-incorporation efficiency. Furthermore, fundamental research is provided concerning the growth of Ga(N,P) and Ga(N,As,P) layers grown lattice matched to Si(001), of Ga(P,Sb) QW layers and their investigation towards a potential direct band gap, and of high Sb-containing Ga(P,Sb) quantum dots (QDs) on the same substrates.In den vergangenen Jahren hat das Feld der integrierten Optik (photonic integrated circuits) in der wissenschaftlichen und industriellen Gemeinschaft zunehmend an Aufmerksamkeit gewonnen. Die Integration optischer Bauelemente in die CMOS Technologie (complementary metal-oxide-semiconductor), d.h. Si-Chipfertigung, bietet sehr viel Raum für Fortschritt bezüglich größerer Bandbreiten-Dichte in der Datenübertragung, aber auch bezüglich Gas-Sensorik oder für biomedizinische Anwendungen als günstige und tragbare, Chip-große Geräte. Insbesondere im Bereich der Datenübertragung werden dabei effiziente und zuverlässige Laser-Lichtquellen benötigt, deren Emission im nahinfraroten Spektralbereich liegt. Aufgrund des Mangels einer effizienten Si-basierten Lichtquelle ist die Integration eines Lasers auf CMOS kompatiblen Si(001) 300 mm Wafern, welche in der Si-Industrie standardisiert sind, bis heute eine große Herausforderung.. Diese Arbeit zielt auf diese Realisierung eines Lasers ab, der per Metalloraganischer Gasphasenepitaxie (MOVPE) gitterangepasst auf exakt orientierten Si(001) Substraten abgeschieden wird. Für dieses Ziel werden im Zuge der Arbeit Ga(N,As,P,Sb)/(B,Ga)(As,P) Heterostrukturen mit dem Ziel, Lasing bei Raumtemperatur zu erreichen, optimiert. Dies umfasst neben dem Wachstum per MOVPE selbst auch deren Charakterisierung mittels hochauflösender Röntgenbeugung (HR-XRD), Rasterkraftmikroskopie (AFM), elektrochemischen Kapazitäts- Spannungsmessungen (ECV) und Photolumineszenzspektroskopie (PL). Da für on-Chip integrierte Bauteile eine laterale Strominjektion gefordert ist, werden Breitstreifenlaser in dieser entsprechenden Geometrie prozessiert. Die Laserstrukturen werden dann via Elektrolumineszenzspektroskopie (EL) analysiert. Die gitterangepassten Volumenschichten der gewachsenen Elektrolumineszenzstrukturen basieren auf dem indirekten Halbleiter (B,Ga)(As,P). Bei Untersuchungen von entsprechnenden Strukturen zeigt sich, dass, verglichen mit z.B. GaAs Schichten in etablierten Lasern, die (B,Ga)(As,P) Schichten zu erhöhter Einsatzspannung sowie erhöhtem differentiellen Widerstand führt. In dieser Arbeit wird das (B,Ga)(As,P) Materialsystem eingehend bezüglich unterschiedlicher Wachstumsbedingungen und Dotierungen untersucht, um die Schichten für Laseranwendungen zu optimieren. Eine deutliche Verbesserung der Einsatzspannung von 4.5(1.5) V auf 1.6(1) V wird dabei erreicht, was mit einer erheblichen Reduktion von Energie- und Wärmeverlusten einhergeht. Damit sind diese Bauelemente elektrisch bereits mit industriell etablierten Lasern auf anderen Substraten konkurrenzfähig. Die optisch aktiven Quantentopf (QW v. engl. quantum well) Ga(N,As,P,Sb) Schichten werden unter diversen MOVPE Bedingungen gewachsen und hinsichtlich glatter Grenzflächen mit den umgebenden Barrieren und verstärkter Lumineszenz optimiert. Ausgedehnte Defekte, die sich hauptsächlich durch das Überwachsen der modulierten oberen QW Grenzfläche ausbilden, können sich bis zur Probenoberfläche ausdehnen und in AFM Topographien als 3 dimensionale Platelets auf den Oberflächen der Heterostrukturen sichtbar werden. Ga(N,As,Sb) wird bei einer Wachstumstemperatur von 575°C optimiert, bei der angenommen wird, dass Sb hauptsächlich als Surfactant wirkt. In entsprechenden Teststrukturen, die ca. 9.5% N im QW enthalten, wird eine verringerte Defektbildung im Vergleich zu Sb-freien Strukturen erzielt, wobei sie gleichzeitig in hohen optischen Ausgangsleistungen sowohl in PL- als auch EL-Strukturen resultieren. Bei N-Gehältern von ca. 11% können die Oberflächen der Teststrukturen, also an überwachsenen QWs, gemessen als RMS Rauigkeit der AFM Topographien, auf 0.25 nm Rauigkeit reduziert werden. Die Plateletbildung wird dabei komplett unterdrück. Die optischen Ausgangsleistungen von Strukturen, die 11% N oder mehr enthalten, reduziert sich allerdings gegenüber Strukturen, die 9.5% N enthalten, was vermutlich auf nicht-strahlende Rekombination von N-korrelierten Zuständen durch beispielsweise N-Ketten oder N-Clusterkonfigurationen im QW zurückzuführen ist. Anhand des Wachstums von Ga(N,As,P) QW Schichten werden der standard MOVPE Präkursor für N-Einbau unsymmetrisches Dimethylhydrazin (UDMHy) und der neuartige N-As Prükursor Ditärtiärbutylaminoarsan (DTBAA) detailliert miteinander verglichen. Mit beiden werden in Elektrolumineszenzstrukturen fast gleiche optische Ausgangsleistungen erreicht, was das große Potential des DTBAA als N-Präkursors, auch wegen seiner hohen N-Einbaueffizienz, aufzeigt. Desweiteren wird in der Arbeit grundlegende Forschung zum Wachstum von gitterangepassten Ga(N,P) und Ga(N,As,P) Schichten auf Si(001) Substrat, zu Ga(P,Sb) QW Schichten und deren Untersuchung einer möglichen direkten Bandlücke und zu Ga(P,Sb) Quantenpunkten (QD v. engl. quantum dot) mit hohen Sb-Gehältern auf den gleichen Substraten präsentiert
Skewness-Adjusted Bootstrap Confidence Intervals and Confidence Bands for Impulse Response Functions
This article investigates the construction of skewness-adjusted con�dence intervals and joint confidence bands for impulse response functions from vector autoregressive models. Three different implementations of the skewness adjustment are investigated. The methods are based on a bootstrap algorithm that adjusts mean and skewness of the bootstrap distribution of the autoregressive coeffcients before the impulse response functions are computed. Using extensive Monte Carlo simulations, the methods are shown to improve the coverage accuracy in small and medium sized samples and for unit root processes for both known and unknown lag orders
Cryptocurrencies and Gold – Similarities and Differences
This article investigates similarities and differences between gold and four
cryptocurrencies (Bitcoin, Ethereum, Bitcoin Cash and Litecoin). To do so,
we estimate a system-GARCH-in-mean with respect to four determinants for
the period starting 7/18/2014 at earliest until 7/12/2021. We find that, first,
liquidity premia are less important. Second, volatility premia exist in either
gold and cryptocurrencies. Third, the response of cryptocurrencies to ex-
change rate changes is more pronounced than for gold at least if developing
countries are included. Fourth, gold exhibits a safe haven status, while cryptocurrencies do not. So those cannot be seen as a store of value but rather as speculative assets
Boon and Bane: How Perfectionism Shapes Employee Well-Being Through Health-Impairing and Motivational Processes
Es scheint wenig verwunderlich, dass Perfektionismus in einer zunehmend kompetitiven Welt ansteigt (Curran & Hill, 2019). Diese multidimensionale Persönlichkeitsdisposition betrifft besonders den Lebensbereich der Arbeit und kann für Beschäftigte eine Quelle von erhöhtem Stress und vermindertem Wohlbefinden darstellen (Ocampo et al., 2020; Stoeber & Stoeber, 2009). Ziel der vorliegenden Dissertation war es, die ambivalente Rolle von Perfektionismus in diesem spezifischen Kontext zu verstehen, wobei ein Schwerpunkt darauf lag, die unterschiedlichen Zusammenhänge der Dimensionen “Self-Oriented Perfectionism“ (SOP; äußerst hohe Ansprüche, die Personen für sich selbst haben) und „Socially Prescribed Perfectionism“ (SPP; der Eindruck, dass andere Perfektion von einem selbst erwarten) mit dem Wohlbefinden von Beschäftigten zu erklären. Konkret wurde das „Job Demands-Resources“-Modell (JD-R, Bakker & Demerouti, 2007; Demerouti et al., 2001; Schaufeli & Bakker, 2004) als orientierender Rahmen genutzt, um relevante Arbeitsanforderungen und Ressourcen zu identifizieren, auf die Perfektionismus sich auswirken könnte (Forschungsfrage 1). Als weiteres Ziel dieser Arbeit sollten gesundheitsschädliche und motivationale Prozesse des Modells betrachtet werden, um aufzuzeigen, über welche Mechanismen Perfektionismus mit dem Wohlbefinden von Beschäftigten zusammenhängt, sowie auf diesen Prozessen basierende Interventionsansätze (Forschungsfrage 2).
Die vier Manuskripte, welche die Grundlage dieser Dissertation bilden, bezogen sich jeweils auf eine oder mehrere dieser Forschungsfragen. Jedes der Manuskripte hatte dabei einen eigenen Fokus und behandelte bislang unbeantwortete Fragen bisheriger Forschung. Manuskript 1 beleuchtete die Frage, wie perfektionistische Mitarbeitende von potentiellen Teammitgliedern gesehen werden und in welchem Ausmaß sie an ihrem Arbeitsplatz eine soziale Einbindung erleben. In Manuskript 2 untersuchten wir, ob Zeitdruck als Arbeitsanforderung in Kombination mit dem mentalen Abschalten von der Arbeit in der Freizeit (“Detachment“) als Erholungserfahrung erklärt, warum manche Perfektionisten vulnerabel für beeinträchtigten Schlaf sind. Manuskript 3 fokussierte die Frage, ob die drei Grundbedürfnisse nach Autonomie, Kompetenz und Zugehörigkeit eine Schnittstelle zu einerseits vermindertem aber auch gesteigertem Wohlbefinden von Beschäftigten darstellen können. Zuletzt untersuchten wir in Manuskript 4, ob bereits eine kurze App-basierte Achtsamkeitsintervention perfektionistische Kognitionen reduzieren sowie Detachment und die Schlafqualität verbessern kann.
In Manuskript 1 bauten wir auf dem „Perfectionism Social Disconnection“- Modell (Hewitt et al., 2006) und der darin enthaltenen Annahme auf, dass Perfektionismus zu interpersonellen Schwierigkeiten beitragen kann und untersuchten, inwiefern die verschiedenen Dimensionen unterschiedlich mit sozialen Arbeitsanforderungen und Ressourcen zusammenhängen. Studie 1 bezog sich dabei auf die Perspektive der Teammitglieder (experimentell; N = 184) und zeigte, dass Beschäftigte es vorziehen mit einem nicht perfektionistischen Teammitglied zu arbeiten, außer sie haben selbst hohe Ansprüche an ihr Umfeld. In Studie 2 (querschnittlich, N = 279), welche die Perspektive von perfektionistischen Beschäftigen selbst fokussierte, war SPP die einzige Dimension, die ein konsistentes Muster von geringer sozialer Eingebundenheit zeigte, das soziale Ausgrenzung und zwischenmenschliche Konflikte als soziale Arbeitsanforderungen und ein Fehlen von sozialer Unterstützung als Arbeitsressource umfasste.
In Manuskript 2 verknüpften wir die Perfektionismus-Literatur mit dem Stressor-Detachment Modell (Sonnentag & Fritz, 2015) und nahmen an, dass Unterschiede in der täglichen Generierung von Stress, operationalisiert als täglicher Zeitdruck, und im Detachment relevante gesundheitsschädliche Prozesse sein können, die erklären, warum üblicherweise SPP, nicht aber SOP mit schlechter Schlafqualität einhergeht. Die Ergebnisse der Tagebuchstudie (N = 70) unterstützten Zeitdruck, eine aufgabenbezogene Arbeitsanforderung, und anschließendes vermindertes Detachment als seriellen Prozess zwischen SPP und beeinträchtigtem Schlaf. Die angenommene entgegengesetzte serielle Mediation für SOP durch geringeren Zeitdruck, sowie Detachment als separater Mechanismus wurden nicht bestätigt, aber es zeigte sich ein positiver direkter Effekt von SOP auf Schlafqualität.
In Manuskript 3 setzten wir an der Self-Determination Theory und der Erfüllung der Bedürfnisse nach Autonomie, Kompetenz und Zugehörigkeit an (Deci & Ryan, 2000) und untersuchten die drei Bedürfnisse als gesundheitsschädliche und motivationale Pfade zwischen den Dimensionen von Perfektionismus und dem Arbeitsengagement, der Arbeitszufriedenheit und der Erschöpfung von Beschäftigten. Die Befunde einer Online-Studie über zwei Messzeitpunkte im Abstand von drei Monaten (N = 328 zu MZP1 und N = 138 zu MZP2) betonen die spezifischen Rollen der drei Bedürfnisse darin, die unterschiedlichen Zusammenhänge von SOP und SPP mit dem Wohlbefinden von Beschäftigten zu erklären. Dabei zeigte sich die Erfüllung des Autonomiebedürfnisses sich als besonders relevant für das Arbeitsengagement.
Schließlich betrachteten wir in Manuskript 4 Perfektionismus und die dazu gehörigen dynamischen Kognitionen als eine Antipode zur persönlichen Ressource der Achtsamkeit. Wir testeten, ob eine App-basierte Achtsamkeitsintervention perfektionistische Kognitionen reduzieren und Detachment und beeinträchtigten Schlaf als wichtige erholungsrelevante Konzepte verbessern kann. Im Vergleich zu Teilnehmenden in der Wartelisten-Kontrollgruppe (n = 45) berichteten jene in der Interventionsgruppe (n = 38) vorübergehend einen Anstieg der Achtsamkeit und eine Verringerung der sorgenbezogenen perfektionistischen Kognitionen. Es wurden keine signifikanten Veränderungen bezüglich der erholungsbezogenen Outcomes festgestellt. Dennoch liefern die Befunde dieser randomisierten kontrollierten Studie einen ersten Nachweis dafür, dass App-basierte Achtsamkeitsinterventionen eine Möglichkeit darstellen, um dynamische Aspekte von Perfektionismus zu reduzieren.
Zusammenfassend betonen die vier Manuskripte den zweischneidigen Einfluss von Perfektionismus auf verschiedene Aspekte des Arbeitslebens und das Wohlbefinden von Beschäftigten. Es kommt dabei auf die spezifische Dimension an, ob Perfektionismus Fluch oder Segen ist. Unseren Ergebnissen zufolge hängt SPP darüber negativ mit dem Wohlbefinden zusammen, dass diese Dimension zu Arbeitsanforderungen beiträgt und den gesundheitsschädlichen Prozess anstößt und Ressourcen und den motivationalen Prozess hemmt. Demgegenüber hängt SOP positiv mit dem Wohlbefinden zusammen, weil diese Dimension, zumindest zu einem gewissen Grad, den gesundheitsschädlichen Prozess hemmt und gleichzeitig den motivationalen Prozess anstößt. Dynamische Aspekte von Perfektionismus können bereits durch niederschwellige Interventionen adressiert werden. Die Herausforderung, Perfektionismus in das JD-R Modell zu integrieren, Ansätze für zukünftige Forschung, um den konzeptionellen Rahmen dieser Dissertation zu stärken und zu erweitern, sowie praktische Implikationen werden diskutiert.It is unsurprising that perfectionism is rising in a world that is increasingly competitive (Curran & Hill, 2019). This multidimensional personality disposition especially affects the domain of work and may place employees at risk for high stress and poor well-being (Ocampo et al., 2020; Stoeber & Stoeber, 2009). The present dissertation aimed to understand the ambivalent role of perfectionism in this specific context with an emphasis on explaining the different associations of self-oriented perfectionism (SOP; exceptionally high demands that people have for themselves) and socially prescribed perfectionism (SPP; the belief that others expect perfection from oneself) with employee well-being. Specifically, the job demands-resources (JD-R, Bakker & Demerouti, 2007; Demerouti et al., 2001; Schaufeli & Bakker, 2004) model was used as a guiding framework to identify relevant job demands and resources (Research Question 1) that perfectionism may shape. Drawing on this model, this work also aimed to capture health-impairing and motivational processes that may explain by which mechanisms perfectionism relates to employee well-being and possible intervention approaches that build on these processes (Research Question 2).
The four manuscripts that are the basis of this dissertation each referred to one or more aspects of these research questions. Each of the manuscripts had a unique focus and related to questions that remained unanswered from previous research. First, Manuscript 1 addressed the question of how perfectionist colleagues are seen by potential team members and to what extent perfectionists experience social disconnection or integration in the workplace. Second, we investigated, in Manuscript 2, whether time pressure as a job demand in combination with the recovery experience of detachment (the mental disconnection from work during leisure time) explains why some perfectionists are vulnerable to impaired sleep. Third, Manuscript 3 focused on whether the three basic needs of autonomy, competence, and relatedness represent a crossroad linking the dimensions of perfectionism to either impaired or enhanced well-being of employees. Lastly, in Manuscript 4, we investigated whether a short app-based mindfulness intervention may reduce perfectionistic cognitions and improve detachment and impaired sleep.
Concretely, in Manuscript 1 we built on the perfectionism social disconnection model (Hewitt et al., 2006) with its proposition that this disposition may contribute to interpersonal difficulties and examined the role of the perfectionism dimensions in differently shaping social demands and resources. Study 1 concerned the colleagues’ perspective (experimental; N = 184) and indicated that employees would favour working with a non-perfectionist colleague unless they have high demands for their colleagues themselves. In Study 2 (cross-sectional, N = 279), which addressed the perfectionists’ perspective, SPP was the only dimension that displayed a consistent pattern of social disconnection, including social exclusion and interpersonal conflicts as social job demands and a lack of social support as a resource.
In Manuscript 2, we linked the perfectionism literature and the stressor-detachment model (Sonnentag & Fritz, 2015) and assumed that differences in daily stress generation, operationalised as daily time pressure, and detachment may be relevant health-impairing processes explaining why SPP but not SOP is typically related to poor sleep quality. Findings from the diary study (N = 70) provided support for time pressure, a task-related job demand, and subsequently reduced detachment as a serial process that underlies SPP and impaired sleep. The assumed opposite serial mediation concerning SOP via decreased time pressure and detachment as a separate mechanism were not confirmed, but a direct effect of SOP indicating restful sleep was revealed.
In Manuscript 3 we drew on self-determination theory and the concept of autonomy, competence, and relatedness satisfaction (Deci & Ryan, 2000) and examined the three needs as health-impairing and motivational pathways relating the dimensions of perfectionism to employees’ work engagement, job satisfaction, and exhaustion. Results from a two-wave online study conducted over 3 months (N = 328 at T1 and N = 138 at T2) emphasize the unique roles of the three needs in explaining the different relationships of SOP and SPP to employee well-being with autonomy satisfaction being especially relevant for work engagement.
Lastly, in Manuscript 4, we considered perfectionism with its more state-like cognitions as an antipode to the personal resource of mindfulness and tested the effectiveness of an app-based mindfulness intervention in reducing perfectionistic cognitions and improving detachment and impaired sleep quality as important concepts related to recovery. As compared to participants in the wait-list control group (n = 45), those in the intervention group (n = 38) reported a temporary increase in mindfulness and a decrease in perfectionistic concern cognitions. No significant changes in the recovery-related outcomes were found. However, findings from the randomised controlled trial provide initial evidence that app-based mindfulness interventions are a possibility to reduce state-like aspects of perfectionism.
To conclude, the four manuscripts highlight the duality of perfectionism in shaping various aspects of employees’ working life and their well-being. Whether perfectionism can be considered as a boon or bane, depends on the specific dimension. According to our findings, SPP negatively relates to well-being in that it contributes to job demands and the health impairment process and inhibits resources and the motivational process. By contrast, SOP positively relates to well-being because this dimension hinders, at least to some extent, the health impairment process while it may also foster the motivational process. State-like aspects of perfectionism may be already addressed by low-threshold interventions. The challenge of integrating perfectionism in the JD-R model, avenues for future research to strengthen and extend the conceptual framework of this dissertation, and practical implications are discussed
Pharmakologische Eingriffe in mitochondriale Mechanismen bei Hemin-Toxizität und Ferroptose
The growing prevalence of neurological disorders, such as Parkinson's disease, Alzheimer's disease, and strokes, represents a growing challenge for our aging society and is one of the leading causes of morbidity and mortality. These diseases are closely linked to oxidative stress, and intensive research over the last decades has established a link to various cell death mechanisms such as apoptosis, oxytosis, ferroptosis, necroptosis, and hemin toxicity. This is based on evidence of different pathophysiological features, including lipid peroxidation, loss of iron homeostasis, and mitochondrial impairment. In order to gain a more comprehensive understanding of the previously unexplained factors and molecular mechanisms that contribute to neurodegenerative diseases and whose interrelationships have not yet been fully explored, this study investigated different aspects of ferroptosis.
In the first section of this study, mitochondrial involvement in ACSL4/LPCAT2-driven ferroptosis was investigated in more detail. For this purpose, stably transfected HEK293T cell lines with overexpression of the enzymes ACSL4 and LPCAT2 or a control line with an empty vector were used to uncover the missing insights. The overexpression of lipid synthesis-related enzymes resulted in increased sensitivity to the ferroptosis inducer RSL3 compared to the control cell line. Elevated mitochondrial damage was observed through increased mitochondrial ROS production, loss of mitochondrial membrane potential, and reduced mitochondrial respiration. These effects were prevented by ferroptosis inhibitors such as deferoxamine, ferrostatin-1, and 5- and 12/15-LOX inhibitors. Inhibition of the overexpressed enzyme ACSL4 by thiazolidinediones also protected against ferroptosis. VDAC1 inhibitor Akos-22 and the mitochondrial ROS scavenger MitoQ were used to investigate mitochondrial involvement in more detail. Both showed high efficacy in the overexpressing cells and provided protective effects at the cytosolic and mitochondrial levels. MitoQ prevented ferroptosis through its antioxidant effect and influenced metabolism. To determine whether metabolic intervention was sufficient, inhibitors of mitochondrial complex I of the respiratory chain and glutamine deprivation were examined. However, these interventions failed to protect HEK293T cells from ferroptosis. The results suggest that mitochondrial ROS production determines ACSL4/LPCAT2-driven ferroptosis while mitochondria-targeted antioxidants protect against it.
In the second section of this research, the link between erastin- and hemin-induced ferroptosis in neuronal HT22 cells was characterized in more detail, focusing on mitochondrial involvement. Differences in the response to FCS deprivation were observed. Furthermore, the pharmacological 12/15-LOX inhibitor PD146176 appeared to protect against erastin-induced ferroptosis but was ineffective against hemin toxicity. In contrast, the 5-LOX inhibitors were effective against both forms of ferroptosis. The protection was also able to prevent mitochondrial ROS production and the loss of mitochondrial membrane potential induced by hemin. MitoQ was utilized for a more detailed investigation of the role of mitochondrial ROS formation, showing protective effects against erastin but not against hemin-mediated ferroptosis. Metabolic intervention by the complex I inhibitor metformin also failed to protect the HT22 cells from hemin toxicity. As a result, hemin was found to induce a form of ferroptosis that is 5-LOX-dependent and associated with mitochondrial damage, but the latter was not causal for oxidative death. Erastin, on the other hand, induces a 5- and 12/15-LOX-dependent form of ferroptosis with major mitochondrial involvement, as metformin and MitoQ application were effective. Thus, the hemin toxicity differs from the classical ferroptosis induced by erastin.
Finally, in the last section of the study, novel selenium compounds were investigated for their efficacy against ferroptosis in HT22 cells. These compounds are modifications of the already-known GPx4-mimicking compound ebselen. The selenium-containing compounds showed a remarkably potent effect against ferroptosis which was 15-20 fold more protective compared to ebselen. These compounds acted at cytosolic and mitochondrial levels by affecting lipid peroxidation and ROS production. Additionally, they exhibited strong antioxidant properties, as glutathione levels were maintained and one of the compounds even increased GPx4 protein expression. The diselenides were also found to be more potent in comparison to the benzisoselenazoles and were able to withstand an increasing ferroptosis stimulus. Furthermore, differences in efficacy against erastin- or RSL3-mediated ferroptosis were identified with lower concentrations being sufficient for erastin-induced damage. Application up to 8 hours after the onset of the cell death stimulus was still sufficient to preserve cell integrity. These promising in vitro results characterize the novel selenium compounds as potent agents against ferroptosis, warranting further studies to gain new insight into the efficacy of the selenium compounds in vivo.Die zunehmende Prävalenz neurologischer Erkrankungen, wie Morbus Parkinson, Morbus Alzheimer und Schlaganfälle, stellt eine wachsende Herausforderung für unsere alternde Gesellschaft dar und zählt zu den führenden Ursachen von Morbidität und Mortalität. Diese Erkrankungen sind eng mit oxidativem Stress verbunden und die intensive Forschung der letzten Jahrzehnte konnte verschiedenen Zelltodmechanismen wie Apoptose, Oxytose, Ferroptose, Nekroptose und die Hemintoxizität damit verknüpfen. Dies basiert auf dem Nachweis unterschiedlicher pathophysiologischer Merkmale, darunter Lipidperoxidation, Verlust der Eisenhomöostase und Beeinträchtigung der Mitochondrien. Um ein umfassenderes Verständnis für bislang ungeklärte Faktoren und molekularen Mechanismen zu gewinnen, die zu neurodegenerativen Erkrankungen beitragen und deren gegenseitigen Verbindungen noch nicht vollständig erforscht sind, wurden in dieser Studie zahlreiche Untersuchungen im Bereich der Ferroptose durchgeführt.
Im ersten Abschnitt dieser Studie wurde die mitochondriale Beteiligung an der ACSL4/LPCAT2-gesteuerten Ferroptose näher untersucht. Hierzu wurden stabil transfizierte HEK293T-Zellen mit einer Überexpression der Enzyme ACSL4 und LPCAT2 oder einem Leervektor als Kontrolle verwendet. Die Überexpression der lipidsyntheserelevanten Enzyme führte zu einer erhöhten Sensitivität gegenüber der Ferroptose-induzierenden Substanz RSL3 im Vergleich zur Kontrollzelllinie. Eine verstärkte mitochondriale Schädigung wurde durch erhöhte mitochondriale ROS- Produktion, Verlust des mitochondrialen Membranpotenzials und reduzierte mitochondriale Respiration festgestellt. Diese Effekte konnten durch Inhibitoren der Ferroptose verhindert werden. Die Hemmung des überexprimierten Enzyms ACSL4 durch Thiazolidindione schützte ebenfalls vor dem Zelltod. Zur genauen Untersuchung der mitochondrialen Beteiligung wurde der VDAC1-Inhibitor Akos-22 und der mitochondriale ROS-Scavenger MitoQ eingesetzt. Beide zeigten eine hohe Wirksamkeit in den überexprimierenden Zellen und protektive Effekte auf zytosolischer und mitochondrialer Ebene. MitoQ wirkte aufgrund seiner antioxidativen Wirkung schützend und beeinflusste den Stoffwechsel. Es wurden Inhibitoren des mitochondrialen Komplexes I der Atmungskette getestet, und es wurde ein Glutamin-Entzug durchgeführt, um festzustellen, ob ein Eingriff in den Stoffwechsel ausreichend war. Diese Maßnahmen konnten jedoch nicht gegen Ferroptose schützen. Die Ergebnisse deuten darauf hin, dass die mitochondriale ROS-Produktion die ACSL4/LPCAT2-gesteuerte Ferroptose bestimmt und Mitochondrien-gerichtete Antioxidantien die Zellen davor schützen können.
Im zweiten Abschnitt dieser Forschungsarbeit wurde die Verbindung zwischen Erastin- und Hemin-induzierter Ferroptose in neuronalen HT22-Zellen näher charakterisiert, wobei der Fokus auf der mitochondrialen Beteiligung lag. Es konnten Unterschiede in der Reaktion auf den FCS-Entzug festgestellt werden. Des Weiteren zeigte sich, dass der pharmakologische 12/15-LOX-Inhibitor gegen Erastin schützen konnte, jedoch gegen die Hemintoxizität wirkungslos war. Im Gegensatz dazu waren die 5-LOX-Inhibitoren gegen beide Formen der Ferroptose wirksam. Die Protektion konnte auch die durch Hemin- induzierte mitochondriale ROS-Produktion und den Verlust des mitochondrialen Membranpotenzials aufhalten. MitoQ wurde erneut verwendet, um die mitochondriale ROS-Produktion genauer zu untersuchen, wobei ein Schutz gegen Erastin, aber nicht gegen die Hemin-Toxizität erreicht wurde. Auch die metabolische Intervention durch den Komplex I Inhibitor Metformin konnte die HT22-Zellen nicht vor der Hemintoxizität schützen. Dadurch kann geschlussfolgert werden, dass Hemin eine Form der Ferroptose induziert, die 5-LOX-abhängig ist und mit mitochondrialen Schäden verbunden ist, welche jedoch nicht ursächlich für den oxidativen Tod sind. Erastin hingegen induziert eine 5- und 12/15-LOX-abhängige Form der Ferroptose mit hauptsächlicher Beteiligung der Mitochondrien, da MitoQ und Metformin wirksam waren. Die Hemintoxizität unterscheidet sich somit von der Erastin-induzierten Ferroptose.
Abschließend wurden in dieser Studie neuartige Selenverbindungen auf ihre Wirksamkeit gegen Ferroptose in HT22-Zellen untersucht. Diese Verbindungen stellen Modifikationen der bereits bekannten GPx4-nachahmenden Verbindung Ebselen dar. Die selenhaltigen Verbindungen zeigten eine bemerkenswert potente Wirkung gegen Ferroptose, die 15–20 Mal niedrigere Konzentrationen für den Schutz benötigten im Vergleich zum Ebselen. Die Selenverbindungen verhinderten die Lipidperoxidation und die ROS-Produktion auf zytosolischer und mitochondrialer Ebene. Zusätzlich wiesen sie starke antioxidative Eigenschaften auf, da die Glutathionspiegel aufrechterhalten werden konnten und eine der Verbindungen sogar die GPx4-Proteinexpression erhöhte. Es konnte auch festgestellt werden, dass die Diselenide wirksamer sind und einem ansteigenden Ferroptosestimulus länger standhalten können im Vergleich zu den Benzisoselenazolen. Es wurden auch Unterschiede in der Wirksamkeit zwischen Erastin- oder RSL3 identifiziert, wobei niedrigere Konzentrationen für die Erastin-induzierte Schädigung ausreichten. Die Anwendung bis zu 8 Stunden nach dem Ferroptosestimulus war immer noch ausreichend, um die Zellintegrität zu bewahren. Diese vielversprechenden in vitro-Ergebnisse charakterisieren die neuen Verbindungen als potente Substanzen gegen Ferroptose, was weitere Untersuchungen rechtfertigt, um Aufschluss über die Wirksamkeit der Selenverbindungen in vivo zu erhalten
The Role of R&D-intensive Clusters for Regional Competitiveness
Modern cluster theory provides reasons for positive external effects that ac-crue from the interaction of spatially proximate firms operating in common and related fields of economic activity. In this paper, we examine the impact of R&D-intensive clus-ters as a key factor of regional competitiveness on productivity and innovation growth. In analogy to the industry-oriented concepts of related and unrelated variety (Frenken, Van Oort, Verburg 2007), we differentiate between effects of cluster specialisation and diversity. The identification of R&D-intensive clusters is based on a hybrid approach of qualitative input-output analysis and spatial scanning (Kosfeld and Titze 2017). Our empirical study is conducted for a panel of German NUTS-3 regions in 2001-2011. To comprehensive account for specialisation and diversity effects of clustering we adopt a spatial econometric approach, which allows us to identify these effects beyond the geographical boundaries of a single region. After controlling for regional characteristics and unobserved heterogeneity, a robust ‘cluster strength’ effect (i.e. specialization) on productivity growth is found within the context of conditional convergence across Ger-man regions. With regard to the underlying mechanisms, we find that the presence of a limited number of R&D-intensive clusters in specific technological fields is most strongly linked to higher levels of regional productivity growth. While we also observe a positive effect of cluster strength on innovation growth once we account for spatial spillovers, no significant effects of ‘cluster diversity’ can be identified. This indicates that some but not all cluster-based regional development strategies are promising pol-icy tools to foster regional growth processes
Gliosis-dependent expression of complement factor H truncated variants attenuates retinal neurodegeneration following ischemic injury
Inherited, age-related, and acute retinal diseases are often exacerbated by an aberrant or excessive activity
of the complement system. Consequently, cells not directly affected by an acute event or genetic variants may
degenerate, resulting in enhanced visual impairment. The therapeutic potential of supplementation of complement
factor H (FH), a key regulator of the complement cascade, is therefore particularly promising in the context of retinal
diseases caused by complement activation. In this study, we engineered adeno-associated viruses (AAVs) containing
sequences of two truncated human FH variants. The expression of these variants was regulated by the glial
fibrillary acidic protein (GFAP) promoter, which is selectively active in gliotic Müller cells. Both FH variants consisted
of FH domains 19-20, which were connected to domains 1–4 and 1–7, respectively, by a polyglycine linker. These
AAVs were intravitreally injected following ischemic injury of C57BL/6J mouse retinas. We observed transgene expression
in gliotic Müller cells and to some extent in astrocytes. The expression correlated directly with damage severity.
Interventions resulted in decreased complement activation, accelerated normalization of microglia activity and morphological
improvements. Reduced levels of C3 transcripts and C3d protein in conjunction with higher transcript
levels of inhibitory regulators like Cfi and Cfh, hinted at attenuated complement activity. This study demonstrates
the great potential of complement regulatory gene addition therapy. With further in vivo testing it could be applied

Investigation of SAMD1 ablation in mice
SAM domain-containing protein 1 (SAMD1) has been implicated in atherosclerosis, as well as in chromatin and transcriptional regulation, suggesting a versatile and complex biological function. However, its role at an organismal level is currently unknown. Here, we generated SAMD1−/− and SAMD1+/− mice to explore the role of SAMD1 during mouse embryogenesis. Homozygous loss of SAMD1 was embryonic lethal, with no living animals seen after embryonic day 18.5. At embryonic day 14.5, organs were degrading and/or incompletely developed, and no functional blood vessels were observed, suggesting failed blood vessel maturation. Sparse red blood cells were scattered and pooled, primarily near the embryo surface. Some embryos had malformed heads and brains at embryonic day 15.5. In vitro, SAMD1 absence impaired neuronal differentiation processes. Heterozygous SAMD1 knockout mice underwent normal embryogenesis and were born alive. Postnatal genotyping showed a reduced ability of these mice to thrive, possibly due to altered steroidogenesis. In summary, the characterization of SAMD1 knockout mice suggests a critical role of SAMD1 during developmental processes in multiple organs and tissues.Gefördert durch den Open-Access-Publikationsfonds der UB Marburg
PACAP regulates VPAC1 expression, inflammatory processes and lipid homeostasis in M1- and M2-macrophages
Background: Pituitary adenylate cyclase-activating polypeptide (PACAP) acts as an
anti-atherogenic neuropeptide and plays an important role in cytoprotective, as
well as inflammatory processes, and cardiovascular regulation. Therefore, the
aim of this study is to investigate the regulatory effects of PACAP and its
receptor VPAC1 in relation to inflammatory processes and lipid homeostasis in
different macrophage (MΦ) subtypes.
Methods: To investigate the role of PACAP deficiency in the pathogenesis of
atherosclerosis under standard chow (SC) or cholesterol-enriched diet (CED) in vivo,
PACAP−/− mice were crossbred with ApoE−/− to generate PACAP−/−/ApoE−/− mice.
Lumen stenosis in the aortic arch and different MΦ-subtypes were analyzed in
atherosclerotic plaques by quantitative immunohistochemistry. Undifferentiated
bone marrow-derived cells (BMDC) from 30-weeks-old ApoE−/− and PACAP−/
−/ApoE−/− mice were isolated, differentiated into BMDM1- and BMDM2-MΦ, and
incubated with oxidized low-density lipoprotein (oxLDL). In addition, PMAdifferentiated
human THP-1 MΦ were further differentiated into M1-/M2-MΦ and
subsequently treated with PACAP38, the VPAC1 agonist [(Ala11,22,28)VIP], the
antagonist (PG 97–269), and/or oxLDL. Uptake/accumulation of oxLDL was analyzed
by oxLDL-DyLightTM488 and BodipyTM 493/503. The mRNA expression was
analyzed by qRT-PCR, protein levels by Western blot, and cytokine release by ELISA.
Results: In vivo, after 30 weeks of SC, PACAP−/−/ApoE−/− mice showed increased
lumen stenosis compared with ApoE−/− mice. In atherosclerotic plaques of
PACAP−/−/ApoE−/− mice under CED, immunoreactive areas of VPAC1, CD86, and
CD163 were increased compared with ApoE−/− mice. In vitro, VPAC1 protein
levels were increased in PACAP−/−/ApoE−/− BMDM compared with ApoE−/−
BMDM, resulting in increased TNF-α mRNA expression in BMDM1-MΦ and
decreased TNF-α release in BMDM2-MΦ. Concerning lipid homeostasis, PACAP
deficiency decreased the area of lipid droplets in BMDM1-/M2-MΦ with
concomitant increasing adipose differentiation-related protein level. In THP-1
M1-/M2-MΦ, the VPAC1 antagonist increased the uptake of oxLDL, whereas the
VPAC1 agonist decreased the oxLDL-induced intracellular triglyceride content.
Conclusion: Our data suggest that PACAP via VPAC1 signaling plays an important
regulatory role in inflammatory processes in atherosclerotic plaques and in lipid
homeostasis in different MΦ-subtypes, thereby affecting foam cell formation.
Therefore, VPAC1 agonists or PACAP may represent a new class of antiatherogenic

Quorum sensing- and contact-dependent inhibition-based population control in synthetic microbial communities
The utilization of microbial communities instead of single strain populations in biotechnological processes has been under investigation over the last decades. While some natural communities have already for example been employed for the production of fermented food the establishment of synthetic microbial communities for the production of valuable compounds or bioremediation is still under investigation. One of the most important aspects that has to be considered to maintain microbial communities for biotechnological applications stably is to be able to tune growth and growth rate of individual populations to prevent outgrowth and unbalancing of the community. To address this challenge, we aimed to develop a multicellular network in two engineered Escherichia coli populations with which the growth of one population is controlled in a density-dependent manner by the respective other population and vice versa.
In Synthetic Biology standardization and modularization represent a key aspect in order to be able to predict the performance of circuits and networks. For this to be possible all the utilized parts needed to be characterized. To get an understanding of gene expression strength as well as the dynamics over time, we characterized the promoters we implemented in our synthetic network. We expressed representatively a fluorescent protein from the promoters and thus monitored the expression of GFP under different conditions.
To regulate the population growth in our synthetic microbial community we took three different approaches and investigated their characteristics and suitability to be implemented: 1. Antibiotic resistances 2. RNA polymerase and 3. Contact-dependent inhibition.
For the antibiotic resistance, we utilized the chloramphenicol acetyltransferase as a mechanism to regulate population growth. The expression of the corresponding cat gene was placed under the control of a quorum sensing promoter (PluxI or PlasI). While the cognate transcription factor (LuxR or LasR) was expressed in the same cell the cognate communication molecule (acyl-homoserine lactone: AHL) (AHL-C6 or AHL-C12) was produced in cells of the other population. Thus the resistance to chloramphenicol (CAM) should only be conferred by high cell density of the respective other population when enough cognate AHL is produced to induce the gene expression of the resistance cassette. However, our data showed that the cells grew in the presence of CAM, even when no AHL or transcription factor was available. In consecutive rounds of design improvements plasmid backbones, promoters, and also the antibiotic resistance cassette were exchanged for alternative variants or versions but did not result in a system exhibiting the desired performance. This led us to conclude that the implementation of antibiotic resistance cassettes could not achieve the regulation of population growth in bacteria.
The RNA polymerase approach was performed by controlling the expression of the rpoBC operon which encodes the β and β’ subunit of the RNA polymerase. Additionally, we replaced the Las quorum sensing system with the Rpa system (RpaR, RpaI, AHL-pC, and Prpa*). First experiments showed that the cells did not exhibit any differences in growth or growth rate in comparison to the wild-type. Therefore, we improved the design by tagging each component with an LVA tag to enhance protein degradation and thus shortening the protein half-life; all genes were also chromosomally integrated to reduce gene copy number. The results showed that the growth and growth rate were reduced; nevertheless, we did not obtain a quorum sensing-dependent growth switch. To conclude, even though the overall aim could not be achieved so far we got valuable insights into quorum sensing-regulated RNA polymerase expression. Implementation of further improvements is promising, and we identified measures which could be taken next in order to design and build a functional RNA polymerase-based system.
Additionally, we explored the potential of contact-dependent growth inhibition (CDI) to be utilized in a similar system as previously described. The CDI system is a touch-dependent and receptor-mediated toxin delivery system which can inhibit neighboring cells. We characterized two CDI systems from two E. coli strains, EC93 and EC869, under different culture and induction conditions in coculture with either CDI negative cells or two CDI strains. We observed that CDI is very effective in inhibiting growth however the cells carrying the pore-forming protein toxin exhibited a stronger toxin effect than DNase toxin-carrying cells. We can report that induction type and levels, inoculation ratios, and selected toxins influence the dynamics and changes in community composition over time. We also coupled the expression of CDI to quorum sensing resulting in quite controlled and stable cocultures over the course of 26 hours. Valuable characteristics were determined which lay the foundation for developing downstream applications harnessing these contact-dependent inhibition properties in microbial communities. To conclude, CDI is a very interesting and promising tool for growth regulation that has to be further explored and adapted to be employed in a growth regulating application.
To summarize, for all three mechanisms, the antibiotic resistance cassettes, the RNA polymerase, and contact-dependent inhibition we explored their suitability to be employed to regulate population growth in a density-dependent manner. We obtained valuable data exhibiting the difficulties yet potentials of each system and how they can be further improved to obtain the desired systems performance.Die Verwendung mikrobieller Gemeinschaften anstelle von Einzelpopulationen in biotechnologischen Prozessen wurde im letzten Jahrzehnt erforscht. Während einige mikrobielle Gemeinschaften bereits zur Produktion fermentierter Lebensmittel eingesetzt wurden, wird die Etablierung synthetischer mikrobieller Gemeinschaften für die Produktion wertvoller Stoffe oder für die biologische Dekontamination noch untersucht. Ein sehr wichtiger Aspekt, um beständig dauerhaft mikrobielle Gemeinschaften für biotechnologische Anwendungen aufrecht zu erhalten, ist die Fähigkeit, das Wachstum und die Wachstumsrate der individuellen Populationen beeinflussen zu können. So sollen übermäßigem Wachstum und Ungleichgewicht in der Gemeinschaft vorgebeugt werden. Um diese Problematik zu adressieren, haben wir uns zum Ziel gesetzt, ein multizelluläres Netzwerk zwischen zwei gentechnisch veränderten Escherichia coli Populationen zu entwickeln, mit dem das Wachstum der einen Population dichteabhängig von der anderen Population kontrolliert wird und umgekehrt. In der Synthetischen Biologie spielen Standardisierung und Modularisierung eine wichtige Rolle, um Vorhersagen machen zu können, wie sich genetische Schaltkreise und Netzwerke verhalten werden. Damit dies umgesetzt werden kann, müssen alle verwendeten genetischen Bausteine charakterisiert werden. Um einen Einblick in die Genexpressionsstärke sowie die Dynamiken über die Zeit zu bekommen, haben wir das Verhalten der in unserem synthetischen Netzwerk verwendeten Promotoren charakterisiert.
Zur Regulierung des Populationswachstums in unserer synthetischen mikrobiellen Gemeinschaft haben wir drei Ansätze verfolgt und Eigenschaften der Mechanismen untersucht, um ihre Eignung für unser System zu testen: 1. Antibiotikaresistenzen 2. RNA Polymerase und 3. Kontaktabhängige Hemmung. Für die Antibiotikaresistenzen haben wir die Chloramphenicol Acetyltransferase als Wachstumsregulationsmechanismus verwendet. Die Expression des korrespondierenden cat Gens wurde durch einen Quorum sensing Promoter (PluxI oder Plas) kontrolliert. Während der zugehörige Transkriptionsfaktor (LuxR oder LasR) in derselben Zelle exprimiert wurde, wurde das zugehörige Kommunikationsmolekül (Acyl-Homoserin-Lacton: AHL) (AHL-C6 oder AHL-C12) in Zellen der jeweils anderen Population produziert. Demnach sollte die Resistenz gegen Chloramphenicol nur gewährleistet sein, wenn die jeweils andere Population in hoher Zelldichte vorliegt und dementsprechend genügend zugehöriges AHL produziert wird, um die Genexpression der Resistenzkassette zu aktivieren. Jedoch haben unsere Ergebnisse gezeigt, dass die Zellen in chloramphenicolhaltigem Medium wachsen können, auch wenn kein AHL oder Transkriptionsfaktor verfügbar ist. In aufeinanderfolgenden Zyklen wurde das Netzwerkdesign verbessert, es wurden Plasmidvektoren, Promotoren sowie die Antibiotikaresistenz durch Alternativen ersetzt; durch die Änderungen erhielten wir jedoch kein System, das die gewünschten Merkmale aufzeigt. Das führte zu der Erkenntnis, dass die Regulierung des Wachstums einer bakteriellen Population nicht durch die Implementierung einer Antibiotikaresistenzkassette erreicht werden kann.
Der RNA Polymerase Ansatz wurde durch die kontrollierte Expression des rpoBC Operons erreicht, welches für die β und β’ Untereinheit der RNA Polymerase kodiert. Zudem haben wir das Las Quorum sensing System durch das Rpa System ersetzt (RpaR, RpaI, AHL-pC and Prpa*). Unsere Ergebnisse haben gezeigt, dass das Zellwachstum und die Wachstumsrate im Vergleich zu Wildtypzellen keinen Unterschied aufweisen. Daher haben wir unser Design verbessert und jede Komponente mit einem LVA-Tag versehen, um den Proteinabbau zu steigern und somit die Proteinhalbwertszeit zu verkürzen. Außerdem wurden alle Gene chromosomal integriert, um die Anzahl der Genkopien zu reduzieren. Die Ergebnisse haben gezeigt, dass das Wachstum und die Wachstumsrate reduziert werden konnten; jedoch haben wir keinen Quorum sensing abhängigen Wachstumsschalter erhalten. Zusammenfassend kann gesagt werden, dass obwohl das Gesamtziel bisher nicht erreicht werden konnte, wir nützliche Erkenntnisse für die Quorum sensing regulierte RNA Polymerase Expression erhalten haben. Die Implementierung weiterer Verbesserungsansätze ist vielversprechend und wir haben bereits Maßnahmen identifiziert, die umgesetzt werden könnten, um ein funktionierendes RNA Polymerase basiertes Expressionssystem zu entwickeln.
Zusätzlich wurde das Potential der kontaktabhängigen Wachstumshemmung (CDI) für die Nutzung in einem wie oben beschriebenen System untersucht. Das CDI System ist ein berührungsabhängiges und rezeptorvermitteltes Toxinliefersystem, das das Wachstum benachbarter Zellen hemmen kann. Wir haben zwei CDI Systeme von zwei E. coli Stämmen, EC93 und EC869, unter verschiedenen Kultur- und Induktionsbedingungen in Kokultur mit nicht-CDI Zellen oder mit zwei CDI Stämmen charakterisiert. Wir haben beobachtet, dass CDI sehr effektiv das Wachstum hemmen kann, jedoch zeigten Zellen, die ein porenformendes Toxin produzieren, einen stärkeren Toxineffekt aufwiesen als DNase Toxin tragende Zellen. Wir können berichten, dass Induktionsart und -level sowie Inokulationsverhältnisse und ausgewählte Toxine die Dynamik und Veränderungen in der Gemeinschaftszusammensetzung über die Zeit beeinflussen. Zudem haben wir die Expression von CDI an Quorum sensing gekoppelt, was zu relativ kontrollierten und stabilen Kokulturen über einen Zeitraum von 26 Stunden geführt hat. Wertvolle Eigenschaften konnten ermittelt werden, welche die Voraussetzungen schaffen für die Entwicklung weiterführender Anwendungen, die sich diese kontaktabhängigen wachstumshemmenden Eigenschaften in mikrobiellen Gemeinschaften zunutze machen. Abschließend lässt sich sagen, dass CDI ein sehr interessanter und vielversprechender Mechanismus für die Wachstumsregulierung ist, der tiefergehend untersucht und angepasst werden kann, um in einer wachstumsregulierten Anwendung zum Einsatz zu kommen.
Zusammenfassend kann gesagt werden, dass wir alle drei untersuchten Mechanismen, die Antibiotikaresistenzkassette, die RNA Polymerase und die kontaktabhängige Wachstumshemmung auf ihre Eignung zur Wachstumskontrolle von Populationen auf eine dichteabhängige Weise hin untersucht haben. Wir haben nützliche Datensätze erhalten, die die Schwierigkeiten aber auch das Potential jedes Systems aufweisen und wie diese weiter verbessert werden können, um ein System mit der erwünschten Leistung zu erhalten
