RAC2 gain-of-function variants causing inborn error of immunity drive NLRP3 inflammasome activation
No full textInternational audienceA growing number of patients presenting severe combined immunodeficiencies attributed to monoallelic RAC2 variants have been identified. The expression of the RHO GTPase RAC2 is restricted to the hematopoietic lineage. RAC2 variants have been described to cause immunodeficiencies associated with high frequency of infection, leukopenia, and autoinflammatory features. Here, we show that specific RAC2 activating mutations induce the NLRP3 inflammasome activation leading to the secretion of IL-1β and IL-18 from macrophages. This activation depends on the activation state of the RAC2 variant and is mediated by the downstream kinase PAK1. Inhibiting the RAC2–PAK1–NLRP3 inflammasome pathway might be considered as a potential treatment for these patients

Spot Spine, a freely available ImageJ plugin for 3D detection and morphological analysis of dendritic spines
No full textInternational audienceBackground Dendritic spines are tiny protrusions found along the dendrites of neurons, and their number is a measure of the density of synaptic connections. Altered density and morphology is observed in several pathologies, and spine formation as well as morphological changes correlate with learning and memory. The detection of spines in microscopy images and the analysis of their morphology is therefore a prerequisite for many studies. We have developed a new open-source, freely available, plugin for ImageJ/FIJI, called Spot Spine, that allows detection and morphological measurements of spines in three dimensional images. Method Local maxima are detected in spine heads, and the intensity distribution around the local maximum is computed to perform the segmentation of each spine head. Spine necks are then traced from the spine head to the dendrite. Several parameters can be set to optimize detection and segmentation, and manual correction gives further control over the result of the process. Results The plugin allows the analysis of images of dendrites obtained with various labeling and imaging methods. Quantitative measurements are retrieved including spine head volume and surface, and neck length. Conclusion The plugin and instructions for use are available at https://imagej.net/plugins/spot-spine

El vocabulario de cierta ecología contemporánea utiliza un léxico centrado en los recursos, lo que es justamente parte del problema. La mitología y la literatura nos proponen, por el contrario, embarcarnos en una palabra muy antigua, arca, que comporta entrelazamientos entre distintas lenguas y transporta innumerables historias y sueños. Esta contribución estudiará, pues, los desafíos de las metáforas que nos orientan hacia el arca viva de la naturaleza, el arca rígida de la historia natural y de sus clasificaciones en celdas y, por supuesto, el arca-refugio. Esta bien podría ser, más que una nave, el lenguaje creador o una biblioteca.« Système-Terre », « conservation des espèces », « réserves naturelles », « gestion des ressources biologiques », « biobanques », « environnement », « anthropocène »… Un certain vocabulaire de l’écologie contemporaine fait décidément tourner le monde autour de l’humain moderne, sans remettre en question la structure d’un rapport au monde conçu comme un donné extérieur utile, dont il faudrait assurer la préservation. Alors même que la catastrophe a déjà commencé, que peuvent nous apporter aujourd’hui les récits les plus anciens du déluge et de l’arche – de "L’Épopée de Gilgamesh" à la Bible ? De quelles façons sont-ils revitalisés dans la littérature des XXe-XXIe siècles ? Nous verrons qu’il s’agit, peut-être, de procéder à une remise en question zoopoétique de notre rapport au langage
