    Targeting NgR1/NogoA Signalling Enhances Natural Killer Cell Cytotoxicity and Alters Immune and Oncogenic Pathways in TNBC Cells

    Triple-negative breast cancer (TNBC) is an aggressive subtype with limited therapeutic options, and impairment of natural killer (NK) cell cytotoxicity within the tumour microenvironment remains a major challenge. The present study investigated the role of the NogoA/Nogo receptor 1 (NgR1) axis in modulating NK cell-mediated cytotoxicity against TNBC cells. NK-92 and MDA-MB-231 cells were co-cultured at optimised effector-to-target ratios, followed by treatment with the NgR1 antagonist NEP1-40. Cell viability was assessed by WST-1 assay, apoptosis was evaluated using AO/DAPI staining, gene expression was analysed by RT-PCR, and downstream pathways together with immune infiltration were examined in silico. Blockade of NogoA/NgR1 signalling increased NK-92-mediated cytotoxicity (p < 0.0001). NEP1-40 reduced tumour cell viability from approximately 81% to 50% and significantly increased apoptotic cell death (p < 0.0001). Expression analysis revealed repression of oncogenic and migration-associated genes, including CDK4, MYC, NF-kappa B1, VEGFA, FGF1, N-cadherin and CD155, consistent with enhanced NK cell-mediated cytotoxicity and reduced tumour immune evasion. Functional annotation identified the PI3K/AKT pathway alongside cell cycle-related processes as candidate downstream mechanisms, while correlation analysis showed that most NogoA-regulated genes were significantly negatively associated with NK cell infiltration (p < 0.05), highlighting their role in immune escape. These findings suggest that NogoA regulates both tumour behaviour and immune remodelling in TNBC by enhancing NK cell-mediated cytotoxicity, promoting apoptosis and suppressing immune evasion pathways. Targeting the NgR1/NogoA axis may therefore represent a promising approach to strengthen NK cell-based immunotherapy, offering a novel therapeutic avenue in TNBC

    The primary objective of this study is to identify the factors influencing passengers' preferences for cruise tourism in Türkiye and to evaluate their significance level. The most important criteria were determined using the fuzzy DEMATEL method, and the results were evaluated. The most significant criterion for passengers opting for cruise tourism is travel costs. Accordingly, the criteria of activities and entertainment, ship type and structure, destination, safety, accommodation, and restaurant services have been identified. The criterion of lesser importance is port management. Cruise tourism stakeholders can design upcoming steps according to this research, in which the most important criteria have been identified and the results evaluated. The analysis of these criteria will also contribute to policymakers, practitioners, and entrepreneurs who require novel sources of information. The importance of ranking the criteria influencing the preference for cruise tourism to contribute to sustainable development demonstrated the necessity of identifying the most significant criteria. These criteria are of high significance in the contemporary worldwide cruise tourism industry, which needs to be accentuated.     Synergistic effects of SCC (with CKD) and flexural performance of reinforced concrete beams strengthened with ferro-cement in tension zone

    This paper presents a study investigated ways to enhance the mechanical properties of reinforced concrete beams (RCBs) and reduce their failure under high loading conditions by employing self-compacting concrete (SCC) and ferrocement (FC). A variable replacement level based on Cement Kiln Dust (CKD) was used in the SCC mixture as an eco-friendly alternative material for cement; these levels were 0%, 2.5%, 5%, 7.5%, 10%, and 12.5%. The fresh properties and hardened properties of SCC were analyzed. Further, layers of FC were placed over the RCB samples to understand their effect under flexural load conditions. The results indicated that an increase in CKD content led to reduction in SCC flowability, an increase in viscosity, a reduction in passing ability, and an improvement in segregation resistance. In hardened SCC concrete, increases in CKD content led to reduced dry density, compressive strength, flexural strength, and modulus of elasticity, but increased absorption and porosity. Notwithstanding this fact, samples containing low percentages (2.5% and 5%) displayed increased splitting tensile strengths and impact energy absorption capacity. In RCB samples, the addition of either one or two layers of FC demonstrated substantial improvement in their flexural properties, represented by the first cracking stress, ultimate flexural load (11–15% enhancement), and ductility properties in conditions containing CKD as well as CKD-free conditions. RCBs failed under flexural loads; their first cracks occur at 36–46% of the ultimate loading capacity. The addition of three layers of FC was found to substantially upgrade beam stiffness by 84% for CKD0-3 samples and 27% for CKD12.5-3 samples, while ductility was significantly reduced.     A gentle and colorful touch of epilepsy surgery on a young painter’s art and life: a case report with dramatic paintings

    Purpose: Patients with drug-resistant epilepsy (DRE) may experience significant limitations in their activities of daily living, social interactions, educational processes, and cognitive and artistic abilities. Epilepsy surgery aims primarily to achieve seizure control with drug-resistant epilepsy (DRE), but it may also lead to improvements in quality of life, including cognitive, emotional, and psychosocial recovery. This report presents a 16-year-old patient with DRE who demonstrated a remarkable improvement in artistic and educational abilities following temporal lobe surgery, illustrating the multifaceted benefits of surgical treatment. Methods: An adolescent male with DRE since the age of eight years, characterized by focal seizures of temporal lobe origin, was evaluated with video-EEG, brain MRI, and FDG-PET. The patient underwent right temporal lobectomy after confirming mesial temporal sclerosis. His psychosocial, educational, and artistic progress were followed for two years postoperatively. Results: Following surgery, the patient achieved complete seizure freedom, progressive cognitive improvement, and significant emotional stabilization. His artistic abilities, once limited by the burden of frequent seizures, evolved dramatically, as reflected by increasingly sophisticated and colorful paintings. His educational performance improved, leading to his successful admission to the fine arts program of the university. Conclusion: This case presentation, with its fascinating drawings and paintings, aims to highlight that epilepsy surgery in adolescents with DRE can offer benefits beyond seizure control. Restoration of cognitive and creative capacities can substantially improve quality of life, academic performance, and psychosocial well-being, underscoring the broader impact of surgical intervention in pediatric epilepsy
