Investigating the Role of Genetic Interactions in Breast Cancer Risk
Objectives: Small effect gene mutations like single nucleotide polymorphisms (SNPs) are emerging as an important factor in defining breast cancer risk. Considering the role of epistasis in cancer development, we hypothesize that the interaction between multiple SNPs can modify breast cancer risk through a pathway involving DNA modulation.
Methods: With data from a case-control study completed in 2010, 45 SNPs from 8 candidate genes involved in the proposed pathway, which included genes involved in apoptosis and DNA repair, were examined for their main effects and higher-order geneinteractions (i.e. epistatic interactions) to determine their potential impact on breast cancer risk.
Results: Three SNPs were identified to have associations with breast cancer. Upon initial investigation, 2,113 models of epistatic interactions were observed to have significant association with modified breast cancer risk; however, after adjusting for the false discovery rate, none of the models met our threshold for significant interactions.
Conclusions: Three SNPs linked to different genes associated with apoptosis and DNA repair were associated with increased or decreased breast cancer. This particular candidate gene approach and proposed pathway of interaction did not reveal a significant modification in breast cancer interactions after FDR adjustment. Further investigation with more SNPs or different genes may be necessary to better understand the role that DNA modulating genes have on breast cancer
