    Dislessia e biblioteche

Engineering of a reporter mouse to study in vivo the role of ΔHER2 splice variant in breast cancer progression

    No full text
One-third of patients with breast cancer overexpress the ERBB2 receptor tyrosine kinase, which is associated not only with a more aggressive phenotype but also reduced responsiveness to hormonal therapies. (Ursini-Siegel J., Schade Babette et al. 2007) (54). Overexpression of human epidermal growth factor-2 (HER2) oncogene in human breast carcinomas has been associated with a more aggressive course of disease (Menard S., Pupa S.M et al. 2003) (16). The reason for this association is still unclear, although it has been suggested to rest in increased proliferation, vessel formation, and/or invasiveness. Alternatively, prognosis may not be directly related to the presence of the oncoprotein on the cell membrane, but instead to the breast carcinoma subset identified by HER2 overexpression and characterized by a peculiar gene expression profile. HER2 currently represents one of the most appropriate targets for specific therapy. Indeed, trastuzumab, a monoclonal antibody, is therapeutically active in HER2-positive breast carcinomas. However, a consistent number of HER2-positive tumors is not responsive to HER2-driven therapy, indicating the need for a better understanding the action mechanism of a new biological drug in vivo.\ud Recent data (Menard S., Pupa S.M et al. 2003) (16) have shown that HER2 positive breast carcinomas represent a particularly aggressive tumor subset with increased proliferation and metastatic potential.\ud However, HER2/neu overexpression is relevant but not sufficient to induce transformation, as clearly established in mice transgenically expressing the rat proto-oncogene HER2/neu. In these transgenic mice, tumors arose only when the oncoprotein carried mutations, in particular (Castiglioni F., Tagliabue E. et al. 2006) (1) small deletions in the extracellular domain that promote HER2/neu transforming activity of this protein through formation of intermolecular disulfide bonds. Interestingly, an alternative splice form of the human homologous HER2 gene containing an in-frame deletion in the same region that is mutated in neu protoncogene transgenic mice, has been detected in human breast carcinomas. This splice variant encodes a receptor lacking exon 19, which immediately precedes the transmembrane domain.\ud Over the past two decades, many ERBB2 mouse models for breast cancer have conclusively shown that this receptor has a causal role in breast cancer development (Ursini-Siegel J., Schade Babette et al. 2007) (54). These mouse models have also enabled the mechanisms controlling tumor growth, angiogenesis, metastasis, dormancy and recurrence in ERBB2-positive breast cancer to be elucidated. In addition, a mouse model has recently been described that accurately recapitulates many of the hallmarks associated with the early stages of the human disease.Mouse models have made an important contribution to our understanding of breast cancer progression and metastasis. Novel, ‘humanized’ mouse models are at present being developed and will be pivotal to pinpoint the cardinal (Jonkers Jos and Derksen P.W.B 2007)(53) events that govern intrinsic cues of tumor cells and microenvironmental interactions that propel metastatic disease. \ud This research program proposes to generate a new mouse transgenic GEM (genetically engineered mice) that express HER2 splicing oncogene under control MMTV specific promoter and enhancer elements, to examine the ability of altered HER-2 receptors to transform mammary epithelium in vivo. To monitor the transgene expression and eventually HER2 driven tumorigenesis in living mice, our transgenic mouse has been generated using a bicistronic vector with luciferase reporter gene under the control of MMTV promoter. The transgenic mouse model we generated will be a useful tool for the study of the dynamics of human HER2 splice variant transcriptional activation in vivo and ΔHER2 involvement in mammary tumorigenesis.\u

ARCHITETTURA INSTABILE. SPIAGGIA E COSTRUZIONE DI SPAZIO COLLETTIVO \ud [UNSTABLE ARCHITECTURE. BEACH AND CONSTRUCTION OF COLLECTIVE SPACE]

EVALUATION OF RESPIRATORY FUNCTION IN STANDARDBRED HORSE AND CORRELATION TO ATLETIC PERFORMANCE

Moreover, clinical and subclinical respiratory diseases have a very high incidence among foals and yearling. To assess the influence on performance of respiratory diseases arising during stabling, 24 standardbred horses have been investigated from birth to the age of 18 months. Clinical examination has been performed monthly to detect symptoms of respiratory diseases. Samples of venous and arterial blood were collected for blood-gas analysis, starting when the horses were six months old and successively every six months. Samples of arterial and venous blood it was also collected before and after 15 minuts of slow trot at the age of 18 months. Alveolar arterial oxygen gradient [(A-a)DO2] and arteriovenous gradient in the oxygen content [(a-v)CO2] were also obtained. The followed performance of each horse was recorded. There was a correlation between (A-a)DO2 and the assessment of respiratory disease but no one of the other values were associated with good or poor performances. Probably, blood gas analysis should be used in association with other diagnostic procedures to assess its real usefulness in evaluation of the future performance ability.\u

    Quantum Mechanics as Quantum Information?

    No full text
This thesis analyzes the epistemological implications and the role of basic concepts like information, physical law and matter in characterizing the physical reality. The\ud key point will be a quantum description of this physical reality. Once fixed at ontic level two basic elements, namely the laws of physics and the matter, we argue that\ud the underlying physical reality emerges from the interconnection between these two elements. We consider any physical process, including measurement, modeled by\ud unitary evolution. In this context, we deduce quantum randomness as a consequence of inclusion of the observer into the quantum system. The global picture of the\ud universe is in a sense deterministic, but from our own local perspective (as part of the system) we perceive quantum mechanical randomness. Then, the notion\ud of "information" turns out to be a derivative concept. Thus, quantum mechanics cannot be simply intended as quantum information theory.\u

I VETTORI DI LEISHMANIOSI IN ITALIA: ABITUDINI ALIMENATARI E PROSPETTIVE DI PREVENZIONE E LOTTA

    No full text

Nuclear Spectroscopy near the proton drip-line in the Lanthanide region:the 122La nucleus\ud

    No full text
One of the most exciting subjects in contemporary nuclear physics is the study of nuclei at the limits of stability with respect to particle emission. Recently, there has been an intensive experimental activity in measuring proton decay and a large variety of proton emitters were observed in the region of nuclei with 50 < Z < 82. Very recently, the proton radioactivity from 117La and 121Pr has been identified and the decay rate deviates significantly from\ud calculations assuming spherical configurations, thus indicating the onset of large deformations in the drip line nuclei below Z = 69. Predictions by the microscopicmacroscopicmass model and by relativistic mean field calculations support these experimental results and suggest the position of the proton drip line. The lifetimes\ud of the proton-decaying isomers are extremely sensitive to the orbital angular momentum ` of the emitted proton and can vary over several orders of magnitude when changing the angular momentum of the occupied orbital. The position of\ud certain orbitals at the Fermi surface depends strongly on the beta2 deformation. It is therefore very important to determine the quadrupole deformation of the nuclei\ud at and beyond the proton drip line, in order to locate the orbitals close to the Fermi surface and to estimate the lifetime of the isomers, which is essential when\ud looking for new proton emitters. The deformation of the nuclei at the proton drip line in the A 130 mass region can be experimentally tested by comparing the lifetimes of proton emitters with theoretical predictions and by investigating the level structure of particle bound nuclei close to the drip line.\ud For Z = 57 the proton drip line is predicted to correspond to the isotope 118La. The lightest La nucleus for which spectroscopic information has been published is 124La . This thesis bescribes an experiment realised to\ud identify for the first time excited states of the doubly-odd nucleus 122 57 La65.\ud Another aspect that make particularly interesting a study of these nuclei is a well-established phenomenon in two-quasiparticle rotational bands in odd-odd nuclei: the signature inversion. Despite the observation of signature\ud inversion in many different nuclei, and numerous theoretical interpretations, a satisfactory explanation of the phenomenon has yet to be found. We want verify\ud if the experimental level scheme of 122La is in agreement with the systematics of the other nuclei in this mass region.\ud The common feature relating the studies of these neutron deficient nuclei is that they are populated with extremely low cross section and require special techniques to be identified. The standard efficient way to achieve this is the use of arrays of high-purity germanium detectors coupled to specific ancillary devices for their selection.\ud Our experiment has been performed at the Legnaro National Laboratory (LNL) where heavy-ion fusion-evaporation reaction 40Ca(@ 200 MeV)+92Mo was performed to establish the scheme of 122La by using the GASP -ray spectrometer\ud in conjunction with the ancillary charged-particle ISIS detector and the Neutron Ring. This experimental set-up led to numerous achievements in the A 130 mass region.\ud Chapter 1 introduces to the physics of the nuclei close and beyond to the proton drip-line.\ud In Chapter 2 we will briey see the various theoretical models created to describe several types of nuclear excitations and discuss in more detail the cranking\ud model.\ud Chapter 3 describes the experimental techniques of -ray spectroscopy and the setup used for the present measurement.\ud In Chapter 4 the steps followed in the sorting of the raw data to produce particle-gated matrices are described, together with the techniques used to select the different reaction channels and to clean the spectra.\ud Chapter 5 shows the systematic features of the rotational bands of the A 130 proton rich odd-odd nuclei.\ud Chapter 6 will be devoted to the construction of the decay scheme and to the spins assignments, performed through the analysis of gated matrices, transition intensities, DCO, and the comparison with the systematics of the La isotopes.\ud Finally, in Chapter 7, the results are discussed in the light of the deduced experimental quantities like single-particle spin alignment, dynamic moments of\ud inertia, signature splitting, and B(M1)=B(E2) branching ratios to assign the Nilsson orbitals to the found bands.\u
