Discovery of a Novel Chemo-Type for TAAR1 Agonism via Molecular Modeling

    No full text
The search for novel effective TAAR1 ligands continues to draw great attention due to the wide range of pharmacological applications related to TAAR1 targeting. Herein, molecular docking studies of known TAAR1 ligands, characterized by an oxazoline core, have been performed in order to identify novel promising chemo-types for the discovery of more active TAAR1 agonists. In particular, the oxazoline-based compound S18616 has been taken as a reference compound for the computational study, leading to the development of quite flat and conformationally locked ligands. The choice of a ``Y-shape'' conformation was suggested for the design of TAAR1 ligands, interacting with the protein cavity delimited by ASP103 and aromatic residues such as PHE186, PHE195, PHE268, and PHE267. The obtained results allowed us to preliminary in silico screen an in-house series of pyrimidinone-benzimidazoles (1a-10a) as a novel scaffold to target TAAR1. Combined ligand-based (LBCM) and structure based (SBCM) computational methods suggested the biological evaluation of compounds 1a-10a, leading to the identification of derivatives 1a-3a (hTAAR1 EC50 = 526.3-657.4 nM) as promising novel TAAR1 agonists

Weighted Average of Human Motion Sequences for Improving Rehabilitation Assessment

    No full text
International audienceWhile human motion analysis has been widely addressed in recent years, the specific task of rehabilitation motion assessment remains challenging due to the lack of available annotated data. To overcome this challenge, data augmentation can be considered. However, classical augmentation techniques applied to human motion sequences often result in meaningless movements. Moreover, in rehabilitation assessment, labels are often continuous values illustrating the quality of a movement. Hence, associating a continuous label to augmented data is not straightforward. In this work, we propose to address data augmentation using an averaging method, called shapeDBA, adapted to rehabilitation motion sequences represented as multivariate time series. We extend the original proposal by weighting the average, hence allowing us to infer continuous labels associated to augmented motion sequences. We evaluated our proposed method on the Kimore dataset. Experimental results show that our method generates coherent rehabilitation sequences that can be efficiently used to extend a small dataset for rehabilitation assessment

    No full text
International audienceIf the State in France is omnipresent in the conciliation between landscape protection and renewable energy development, local authorities also have an important role to play. They can indeed complicate or, on the contrary, facilitate renewable energy development by expressing their opinion on the quality of the landscape. The more the interest of the landscape is recognised, the more is the damage to the landscape. Avoidance, reduction or compensation measures will then be more significant and costly. In addition, local authorities have several levers for action on renewable energies development projects. They may actually be consulted on these projects, but above all they can guide project holders at an early stage, thanks to their own policies and objectives. The role of local authorities should therefore not be neglected and could even be strengthened as local authorities seize the tools at their disposal.

This thesis analyzes energy quality in distribution networks by identifying and classifying disturbances (overvoltage, outages, harmonics, etc.) through time-frequency methods and machine learning techniques. It aims for real-time detection of signal alterations, introducing time-frequency descriptors, and automatic classification of disturbances. PLL and Hilbert Transform (HT) methods estimate frequency and phase fluctuations, while time-frequency approaches (STFT, Stockwell, OST) detect disturbances precisely. Ten validated descriptors are used by classification algorithms (RF, KNN, SVM) on a simulated network, enabling real-time detection of faults such as interruptions and 3rd and 5th-order harmonics, etc.

A dynamical view of Tijdeman's solution of the chairman assignment problem

    No full text
International audienceIn 1980, R. Tijdeman provided an on-line algorithm that generates sequences over a finite alphabet with minimal discrepancy, that is, such that the occurrence of each letter optimally tracks its frequency. In this article, we define discrete dynamical systems generating these sequences. The dynamical systems are defined as exchanges of polytopal pieces, yielding cut and project schemes, and they code tilings of the line whose sets of vertices form model sets. We prove that these sequences of low discrepancy are natural codings of toral translations with respect to polytopal atoms, and that they generate a minimal and uniquely ergodic subshift with purely discrete spectrum. Finally, we show that the factor complexity of these sequences is of polynomial growth order nd1n^{d-1}, where dd is the cardinality of the alphabet

Récits de transitions, récits en transition

    No full text
International audience“Transition narratives” are a generic term used to describe a range of narratives, whether or not they are part of traditional news coverage, which aim to provide a perspective on climate emergency issues and encourage action. How does this project of mobilizing information, whatever it may be, unfold in the media? What narratives are favoured? This proposal examines this phenomenon from the perspective of information and communication sciences research. From the notions of media narratives and works on the transformations of information writing and journalism, the “Transitions narratives” are seen in mirror of the profound evolutions that have been taking place in the world of information for several decades now.

Engineered Nanocomposite Coatings: From Water-Soluble Polymer to Advanced Hydrophobic Performances

    No full text
International audienceIn this work, a water-soluble (hydrophilic) polymer was used to form a hydrophobic coating on silicon substrates (Si) in a two-step process comprising (i) the transformation of the polymer into an insoluble material and (ii) the structuring of this coating at nanometric and micrometric scales to achieve the desired hydrophobic behavior. Polyvinylpyrrolidone (PVP), a water-soluble commodity polymer, was crosslinked using benzophenone and UV irradiation to produce a water-insoluble PVP coating. The nanometric scale roughness of the coating was achieved by the addition of silica nanoparticles (NPs) in the coating. The micrometric scale roughness was achieved by forming vertical pillars of PVP/NP coating. To prepare these pillars, a perforated polystyrene (PS) template was filled with a PVP/NP suspension. Micrometer scale vertical pillars of PVP/silica NPs were produced by this method, which allowed us to tune the wettability of the surface, by combining the micrometric scale roughness of the pillars to the nanometric scale roughness provided by the nanoparticles at the surface. By adjusting the various experimental parameters, a hydrophobic PVP coating was prepared with a water contact angle of 110°, resulting in an improvement of more than 80% compared to the bare flat film with an equal amount of nanoparticles. This study paves the way for the development of a more simplified experimental approach, relying on a blend of polymers containing PVP and NPs, to form the micro/nano-structured PVP pillars directly after the deposition step and the selective etching of the sacrificial major phase

    No full text
