    Alkannin-Mediated Enhancement of Secondary Metabolite Biosynthesis and Antioxidant Defense in Cadmium-Stressed Vitis vinifera L. cv. Öküzgözü

    Investigating the biochemical defense mechanisms of grapevines against heavy metal contamination is essential for developing sustainable viticulture practices in polluted environments. However, comprehensive knowledge about how bioactive compound supplementation modulates secondary metabolite profiles under cadmium stress remains limited. We investigated phenolic compound accumulation, anthocyanin biosynthesis, and amino acid dynamics in three-year-old Vitis vinifera L. cv. Öküzgözü plants subjected to graduated cadmium concentrations (0, 5, 10, 20 ppm) with and without Alkannin treatment (25 μM), followed by detailed metabolomic profiling. Cadmium concentration and Alkannin application significantly influenced all measured metabolite classes (p < 0.05), with notable interaction effects observed for ferulic acid, quercetin, and myricetin among phenolics, and delphinidin-3-glucoside, cyanidin-3-glucoside, and petunidin-3-glucoside among anthocyanins. Alkannin-supplemented plants consistently demonstrated superior metabolic resilience under low to moderate cadmium stress, maintaining higher phenolic compound concentrations and enhanced anthocyanin accumulation, particularly acylated derivatives such as peonidin-3-glucoside acetyl and malvidin-3-glucoside-p-coumaroyl compared to untreated plants. Recovery of secondary metabolite biosynthesis was most pronounced at moderate stress levels, with Alkannin-treated vines showing better restoration of antioxidant compound levels and enhanced amino acid profiles including asparagine, serine, and arginine. Metabolic profiling revealed distinct clustering patterns, with Alkannin-treated plants forming separate groups characterized by elevated phenolic and anthocyanin concentrations, while severely cadmium-stressed plants showed reduced metabolite accumulation indicative of metabolic disruption. Correlation analysis demonstrated strong positive relationships among phenolic compounds and anthocyanins, confirming coordinated biosynthetic pathway responses. We conclude that Alkannin application significantly enhances grapevine resilience to cadmium toxicity in a concentration-dependent manner through comprehensive protection of secondary metabolite pathways, particularly demonstrating high efficiency under low to moderate Cd stress levels. The differential metabolic responses between treated and untreated plants provide valuable information for bioactive compound-based mitigation strategies, while the quantitative metabolite profiles offer valuable parameters for assessing vine health and fruit quality potential under heavy metal contamination scenarios

    The Impact of Solo and Pair Programming Modes on Problem-solving Skills and Motivation: Students' Reflections on Pair Programming

    This study primarily investigates the effects of solo and pair programming on students’ problem-solving skills and motivation. Additionally, it aims to identify key factors and tensions associated with pair programming based on students’ reflections. An embedded mixed-methods research design was employed. The sample consisted of 42 undergraduate students. Participants in the control group, comprising 19 students, worked individually on computational tasks, while those in the experimental group engaged in the same tasks in pairs. All participants took part in computational thinking activities over seven weeks, structured around game development projects. Data were collected through questionnaires and in-depth interviews. The results indicated no statistically significant differences between the two groups in terms of overall motivation and problem-solving scores. However, the experimental group scored higher in the sub-dimension related to conceptualizing problems through modeling. Student motivation was found to be high across both groups following the intervention, regardless of the programming mode. After analyzing the qualitative data obtained from five students, a total of 122 codes were identified and categorized under six themes: 'team dynamics', 'task and platform characteristics', 'affective domain', 'cognitive strategies and problem-solving', 'pedagogical framework', and 'roles'. The qualitative findings suggest that the pedagogical benefits of pair programming can only be fully realized through the effective management of pairing strategies, peer communication, task difficulty, and collaborative processes

    ÖzetTüm dünyada iş sağlığı ve güvenliğinin önemi her geçen gün artmaktadır. İşletmeler açısındançok önemli bir konu haline gelen iş sağlığı ve güvenliği noktasında çalışanların bilgi düzeyininartması ile birçok iş kazası ve meslek hastalığının önüne geçebilmek mümkün görülmektedir. Buçalışma kapsamında, Hakkâri ilinde bulunan yiyecek-içecek işletmelerinde mutfak bölümündeçalışan kişilerin iş sağlığı ve güvenliğine ilişkin yaşadıkları sorunların incelenmesi amaçlanmıştır.Bu kapsamda, Hakkâri ilindeki yiyecek-içecek işletmelerinde mutfak departmanında görev alanaşçılar, aşçı yardımcıları vb. mutfak çalışanlarıyla yarı-yapılandırılmış görüşmelergerçekleştirilmiş ve kayıt altına alınmıştır. Elde edilen sonuçlar, içerik analizi yöntemiyledeğerlendirilmiştir. Çalışmadan elde edilen bulgulara göre, mutfak bölümü çalışanları iş sağlığıve güvenliği açısından bazı sorunlar yaşamaktadır. Fiziksel, kimyasal, biyolojik, psiko-sosyaltehlikeler ve riskler söz konusu sorunlara örnektir. Çalışma, yiyecek-içecek işletmesi sahiplerine,yöneticilerine ve araştırmacılara yönelik önerilerle son bulmuştur.Anahtar Kelimeler: Mutfak Çalışanları, İş Sağlığı ve Güvenliği, Meslek Hastalığı, Hakkâri.&nbsp;AbstractThe importance of occupational health and safety (OHS) is progressively increasing worldwide.Enhancing the knowledge level of employees in OHS, a critical issue for businesses, is consideredessential to prevent numerous occupational accidents and diseases. Within this study, the aim wasto examine the occupational health and safety problems experienced by kitchen staff in food andbeverage establishments in Hakkâri, Turkey. To this end, semi-structured interviews wereconducted and recorded with kitchen employees, including chefs and assistant chefs, working inthe kitchen departments of food and beverage businesses in Hakkâri. The data obtained wereevaluated using content analysis. The findings revealed that kitchen staff encounter variousoccupational health and safety issues, including physical, chemical, biological, and psycho-socialhazards and risks.
