    This Master's thesis explores the role of design in promoting sustainable practices through the innovative use of local resources in the Alentejo bioregion of Portugal, focusing on biomaterial experimentation with pine cones, a by-product of the pine nut industry. By integrating elements of Brazilian Modernism, the research contextualises design solutions within the Alentejo bioregion, demonstrating how localised approaches can promote environmentally conscious outcomes and contribute to a more sustainable future. Using methodologies such as Material Driven Design (MDD) and Design Tinkering, the study emphasises the exploration of materials in their natural context. This approach positions design as an agent of change, promoting circularity and ecological awareness. The central question is: "How can I, as a designer, explore a specific territory through contextual and experimental design processes to better understand local resources? The research aims to enhance the functional and aesthetic value of local ecosystems and materials through innovative design practices. A key focus is on bio-material experimentation with pine cones, with the aim of transforming them into functional and aesthetically meaningful materials that contribute to the local economy and support environmental conservation efforts. The research also incorporates Brazilian cultural elements, combining Alentejo vernacular architecture with modernist innovation to create regionally relevant solutions that respect cultural heritage while advancing material innovation and sustainability. By aligning material experimentation with the specific context of the Alentejo, the research promotes sustainable, contextually appropriate designs that are rooted in the local environment.     Die Baubranche ist eine der grössten Industrien weltweit und hat auch immer wieder mit einem negativen Image zu kämpfen. Sei es Dumpinglöhne bei Arbeitnehmenden oder der enorme Ressourcenverbrauch, welcher auf die Branche zurückzuführen ist. Diese Bachelor-Arbeit versucht aus der Sicht eines Prozessgestalters und autodidaktischen Handwerkers kollaborative und partizipative Kultur in der Branche zu verorten. Durch kollektiv gesammelte Inputs und Feedback sowie durch gesammelte Erfahrungen werden Themen wie hierarchische Strukturen oder Entlohnungsstrukturen hinterfragt.     3D selection through hand tracking in XR for point clouds using signed distance fields

    Three-dimensional (3D) visualizations are commonly used in many contexts, e.g., medicine, geography, astronomy, and they are gaining ever more popularity with the recent developments in extended reality (XR). While 3D visualizations can offer unique spatial insights and thus are useful in a variety of ways, interacting with them remains challenging despite being of essential importance in working with 3D visualizations. A specific problem with interacting with 3D visualizations is the lack of an effective and efficient way to select data. Traditional selection methods are primarily designed for, and limited by, 2D interfaces, therefore often do not provide the spatial precision, necessary degrees of freedom and sufficient flexibility necessary for effective 3D selection. Existing approaches for 3D are restricted to simple selection volumes, and do not consider optimization large datasets such as point clouds. Given the above, in this paper, we tackle the challenges associated with the 3D selection problem by designing, developing and user testing a GPU-based selection approach utilizing Signed Distance Fields (SDFs). Our approach allows for computationally efficient, realtime selection of near-arbitrary subsets of point clouds visualized in XR environments.     Beyond the pews. Exploring queer korean protestants’ experiences and ways of co-existing

    Since 2007, the strong anti-queer stance of Korean Protestant churches has hindered the enactment of anti-discrimination laws and erased the presence of queer Protestants both within churches and queer communities. This study looks into the lived experiences of seven queer Protestants in South Korea through individual interviews and a group session, using a browser-based, game-inspired dialogue tool developed by the researcher. Despite differing identities and experiences, all participants recognised the harmful effects of church-based queer antagonism and the need for more inclusive faith communities. Based on these insights, the study proposes the necessity of a community grounded in difference and diversity rather than uniformity, and speculates on new forms of solidarity, primarily through digital spaces.https://hdl.handle.net/20.500.11806/data/NextGen25-rl6bpeu

    Rekrutierung adipöser Personen für Vor-Ort-Studien: Eine Analyse zu Motivationsfaktoren und Partizipationsbarrieren

    Trotz wachsender Bedeutung nutzerzentrierter Entwicklung werden bestimmte Zielgruppen in Usability-Studien zu wenig berücksichtigt. Dazu zählen auch adipöse Personen, deren Perspektive für die Gestaltung neuer medizinischer Geräte und Behandlungsmethoden besonders relevant ist. Die vorliegende Masterarbeit untersucht, welche Faktoren die Teilnahmebereitschaft dieser Zielgruppe an Vor-Ort-Studien erhöhen können. In Kooperation mit dem Medizintechnikunternehmen Ypsomed Holding AG wurde ein Mixed-Methods-Design umgesetzt. Auf eine qualitative Erhebung mittels Interviews und Usability-Tests (N = 10) folgte eine quantitative Online-Befragung (N = 119). Die Ergebnisse zeigen, dass verschiedene Motivationsfaktoren wie persönliches Interesse, der Wunsch, zur Forschung beizutragen oder finanzielle Anreize die Teilnahme fördern. Im Gegensatz dazu wirken emotionale und strukturelle Hürden, insbesondere Schamgefühle, Angst vor Stigmatisierung sowie organisatorische Einschränkungen wie mangelnde Terminflexibilität, hemmend. Die Analyse wird durch theoretische Modelle wie die Self-Determination Theory, das Health Belief Model und Goffmans Stigmatheorie gestützt. Auf Grundlage dieser Erkenntnisse wurden praxisnahe Empfehlungen entwickelt, die darauf abzielen, bestehende Barrieren zu reduzieren und die Motivation zur Teilnahme zu stärken. Die Arbeit liefert Anregungen für die zielgerichtete Gestaltung zukünftiger Studien mit medizinischen Geräten

    Reprogramming the melanoma and immunosuppressive myeloid cells with esomeprazole-loaded PLGA nanoparticles

    Proton pump inhibitors have been explored for potentiating cancer therapies via reverting the tumor acidity and promoting the activation of anti-tumor immune responses. To regulate the intracellular pH of melanoma and immunosuppressive myeloid cells, we developed poly(L-lactide-co-glycolide) nanoparticles loaded with esomeprazole (ESO-NPs). The effect of ESO-NPs on melanoma cells was observed as alkalinization and reduction of melanin content accompanied by a decrease of microphthalmia-associated transcription factor (MITF), poliovirus receptor (PVR), and programmed death ligand 1 (PD-L1) immune checkpoint expression. ESO-NP treatment of melanoma-patient-derived and in vitro-induced myeloid-derived suppressor cells (MDSCs) reduced the expression of immunosuppression-associated molecules PD-L1, CD206, and CD163 on patient-derived myeloid cells while inducing the expression of co-stimulatory molecule CD86 and HLA-DR in the in vitro model. Our findings suggest that reprogramming the intracellular pH of melanoma and immune-suppression-associated myeloid cells with ESO-NPs can modulate the expression of proteins involved in resistance to cancer therapy and immunosuppression, thus potentially improving the response to immunotherapies
