    À la carte. Ein spielerischer Ansatz für gemeinsames Essen

    «À la Carte» ist ein Kartenset, das durch Verringern des Drucks rund um das Ritual des gemeinsamen Essens dieses unterstützen und neu beleben will. Statt Planung und Entscheidung zur Belastung werden zu lassen, bietet es einfache Hilfestellungen für alltägliche Situationen. Das Set besteht aus 15 Karten, davon zehn als individuell beschreibbare Blankokarten. Auf der Rückseite hilft ein Navigationssystem, das auf Gefühlen wie low energy oder low time basiert, bei der Orientierung. So können nach einem anstrengenden Tag die eigenen Gefühle evaluiert und entsprechend eine Karte für das gemeinsame Essen ausgewählt werden. Die Blankokarten haben die Funktion, dass man sich auch eigene Strategien für andere Situationen erstellen kann und diese auf der Rückseite in eigene Kategorien einordnen kann.     Wir leben in einem System, welches durch das Prinzip des Wettkampfs und des stetigen Wachstums fortlaufend Stärkere und Schnellere gewinnen lässt. Dabei lernen wir prinzipiell gegeneinander zu kämpfen. Effizienz und Kampfgeist stehen ganz oben und das endlose Vergleichen schwächt unsere Tätigkeit. Unter solchen Bedingungen fällt es dem Menschen äusserst schwer, sich selbst und andere tatsächlich wahrzunehmen, geschweige denn zu lieben. Denn wir versuchen ständig, äusseren Bedingungen gerecht zu werden, während wir uns selbst und Andere immer wieder aufs Neue hintergehen oder missachten.     Mechanistic investigation into the phase separation behavior of soluplus in the presence of biorelevant media

    More than a decade since its introduction, the polymeric excipient Soluplus continues to receive considerable attention for its application in the development of amorphous solid dispersions (ASDs) and its utility as a solubilizer for drugs exhibiting solubility limited absorption. While it is well-recognized that Soluplus forms micelles, the impact of its lower critical solution temperature of approximately 40 °C remains an underexplored aspect. This study investigated the phase behavior of Soluplus in fasted-state simulated intestinal fluid (FaSSIF-V1). It was demonstrated that Soluplus forms a dispersed polymer-rich coacervate phase, which coexists with Soluplus micelles at 37 °C. This behavior was confirmed by cloud point measurements, visually discernible phases after centrifugation, as well as multi-angle dynamic light scattering (MADLS) measurements, and quantitative 1H-nuclear magnetic resonance (NMR) spectroscopy of Soluplus concentrations in the supernatant pre- and post-centrifugation. The practical relevance of these findings was contextualized by solvent shift experiments and dissolution testing of spray-dried ASD. The results demonstrated that the poorly water-soluble drug RO6897779 resided in a polymer-rich coacervate phase and was spun down during centrifugation, which resulted in an amorphous pellet exhibiting the characteristics of a viscous liquid. The entrapment of the drug within the polymer-rich phase was further analyzed by temperature- and time-dependent MADLS experiments. The findings of this study are of particular relevance for a mechanistic understanding, relevant to comprehending in vitro-in vivo relationships of Soluplus-based ASDs. Low sampled drug concentrations in FaSSIF-V1 at 37 °C may originate not only from limited drug release and precipitation but also from the formation of a drug-containing, polymer-rich Soluplus phase. Therefore, a liquid–liquid phase separation occurring from Soluplus-based formulations in a biorelevant medium can be excipient-driven, which is different from the common perception that phase separation in the solution state is triggered primarily by high drug concentrations exceeding their amorphous solubility

    Harnessing generative AI for project management certification: developing high-quality question banks with prompt engineering

    The integration of generative AI into education offers transformative opportunities to streamline the creation of learning materials, particularly for certification preparation. This paper focuses on the development of a question bank tailored for the IPMA Level D Project Management certification using ChatGPT. The core of this research lies in prompt engineering, detailing how prompts were designed to extract relevant, high-quality questions from a 383-page study book comprising 29 chapters. Prompts were crafted to generate open-ended and single-choice questions with corresponding answers, including detailed explanations and references to specific page numbers in the source material. A key aspect of this study is ensuring that the output adheres to a specific structural format compatible with Learning Management Systems (LMS) like Moodle. Questions were generated in XML format to streamline their direct integration into Moodle. This requires prompts to be carefully engineered, not only to yield content of the appropriate complexity but also to conform to the technical specifications for seamless import. This structural requirement plays a pivotal role in assessing the efficiency of using generative AI, as deviations from the required format can significantly increase post-processing time. Prompt engineering methodology included iterative refinement to balance detail, difficulty, and style while meeting structural demands. Key insights are provided on how initial prompts were structured and adjusted based on output quality, addressing challenges such as ambiguous phrasing, misaligned references, overly simplistic questions, or format inconsistencies. The quality of the AI-generated outputs is currently under expert review, with ongoing analyses focusing on: - The proportion of questions requiring no or minor adjustments to content or format. - Cases needing substantial rewriting for clarity, alignment, or adherence to the required structure. - The number of questions deemed unsuitable for inclusion due to technical or content-related issues. A time-based comparison is also underway to evaluate the efficiency of the generative AI approach. This involves measuring the time required for reviewing and refining AI-generated questions, including adjustments to ensure Moodle compatibility, against an estimate of the manual effort required for crafting and structuring comparable questions from scratch. Preliminary findings suggest significant time savings, especially in meeting structural requirements for LMS integration. While this paper emphasizes the technical aspects of prompt engineering, quality assurance, and structural considerations, an outlook for future studies is provided. This includes evaluating participants' feedback on the usability and effectiveness of the question bank as a preparation tool, alongside their performance in the certification process. Metrics such as participant satisfaction, perceived usefulness, and examination outcomes will be assessed to offer deeper insights into the impact of generative AI tools in educational contexts. The findings so far highlight the importance of expert review in ensuring both the quality and structural integrity of AI-generated content, underscoring the potential of generative AI to reduce workload while maintaining robust standards and technical compatibility for certification materials.https://iated.org/archive/inted202

    Die Markthalle besteht aus einer Bäckerei, einem Bistro sowie einem breiten Food- und Non-Food-Sortiment. Viele Produkte stammen aus den stiftungsinternen Werkbetrieben und verbinden hohe Qualität mit regionaler Verankerung. Die Produktion legt grossen Wert auf Inklusion und sozialen Mehrwert. Obwohl die Produkte viele Vorteile bieten, fällt es der Markthalle schwer, diese passend zu kommunizieren. Hinzu kommt, dass das Einzugsgebiet durch die ländliche Lage der Markthalle begrenzt ist. Aus diesen Gründen müssen möglichst viele der potenziellen Kundschaft zukünftig angesprochen werden

    Für Menschen mit eingeschränkter Handbeweglichkeit ist es oft eine Herausforderung, Hilfsmittel zu finden, die dem persönlichen Stil entsprechen. OSA ist eine Kollektion von Mobilitätshilfen bestehend aus orthopädischen Schienen und Werkzeugen. Weil sie wie modische Schmuckstücke wirken, helfen sie, das Stigma zu durchbrechen, das Mobilitätshilfen oft anhaftet. Die Objekte wirken weniger wie medizinische Hilfsmittel, sondern unterstützen die Selbstentfaltung der Träger:innen und mindern das Gefühl, über ihre Erkrankung definiert zu sein. Die Kollektion umfasst Fingerschienen zur Verbesserung von Stabilität, Griffigkeit und Schmerzlinderung sowie Armbänder mit Griffpads zum Flaschenöffnen und Haken zum Tragen schwerer Taschen.     Self‐boosting programmable release of multiple therapeutic agents by activatable heterodimeric prodrug‐enzyme assembly for antitumor therapy

    Endogenous stimuli-responsive prodrugs, due to their disease lesion specificity and reduced systemic toxicity, have been widely explored for antitumor therapy. However, reactive oxygen species (ROS) as classical endogenous stimuli in the tumor microenvironment (TME) are not enough to achieve the expected drug release. Herein, a ROS-activatable heterodimeric prodrug-loaded enzyme assembly is developed for self-boosting programmable release of multiple therapeutic agents. The heterodimeric prodrug NBS-TK-PTX (namely NTP) is composed of 5-(ethylamino)-9-diethylaminobenzo[a]phenothiazinium chloride analog (NBS), paclitaxel (PTX) and ROS-responsive thioketal (TK) linker, which shows a strong binding affinity with glucose oxidase (GOx), thus obtaining NTP@GOx assembly. Notably, the enzymatic activity of GOx in NTP@GOx is inhibited by NTP. The programmable release is achieved by following steps: i) NTP@GOx is partially dissociated in acidic TME, thus releasing a small segment of NTP and GOx. Thereupon, the enzymatic activity of GOx is recovered; ii) GOx-triggered pH reduction further facilitates the dissociation of NTP@GOx, thus accelerating a large amount of NTP and GOx release; iii) The TK linker of prodrug NTP is cleaved by hydrogen peroxide generated by GOx catalysis, thus expediting the release of NBS for Type-I photodynamic therapy and PTX for chemotherapy, respectively. The NTP@GOx shows great potential for multimodal synergistic cancer therapy

    Where should speed-pedelecs ride? Perspectives from riders versus other road users

    This study investigates the integration of speed-pedelecs (S-pedelecs) into the existing mobility system, focusing on the perspectives of various road users across Austria, Germany, and Switzerland. S-pedelecs, capable of pedal-assisted speeds up to 45 km/h, present an alternative to motorized transport but challenge existing infrastructure due to their unique speed and performance characteristics. Using a survey approach that included an image-based stated preference and acceptance survey, the study reveals strong acceptance for sharing cycling facilities among S-pedelec riders and other road users, regardless of country or road type. S-pedelec riders prefer using cycling facilities, with mixed traffic being more acceptable under conditions like lower speed limits (30 km/h) and low vehicular traffic. Results from this study fill a gap in the existing research and can inform policy makers and planners on the next generation of mobility infrastructure that is accepted, safe and inclusive for Spedelec riders as well as for other road users.https://www.heart2025.vtk.ed.tum.d
