Investigation of the Apoptotic Effects of Cinnamaldehyde, Crocin, and Betanin on the MCF-7 Cell Line
Cinnamaldehyde, Betanin ve Crocin bileşiklerinin her birinin, meme kanseri hücrelerinde apoptotik mekanizmaları farklı moleküler yolaklar üzerinden aktive ettiği ve hücre canlılığını anlamlı düzeyde azalttığı önceki çalışmalarda gösterilmiştir. Bu araştırmada ise Cinnamaldehyde, Betanin ve Crocin'in MCF-7 meme kanseri hücrelerindeki apoptotik etkilerinin tekil ve sinerjik olarak incelenerek MCF-10A sağlıklı meme epitel hücreleri üzerindeki etkilerinin karşılaştırmalı olarak değerlendirilmesi amaçlanmıştır. Araştırmada kemoterapötik etkinliği değerlendirmek amacıyla Dosetaksel referans ajan olarak kullanılmıştır. Cinnamaldehyde, Betanin, Crocin ve Dosetaksel'in farklı konsantrasyonları MCF-7 ve MCF-10A hücrelerine uygulanmış, 24, 48 ve 72 saat için IC50 değerleri MTT yöntemiyle belirlenmiştir. Ardından, hücrelere tekil ve kombine uygulamalar yapılmış ve apoptotik etkilerinin değerlendirilmesi amacıyla Bcl-2, Kalpain-1, P53 ve Sitokrom C düzeyleri ELISA yöntemiyle analiz edilmiştir. MCF-7 hücrelerine uygulanan Cinnamaldehyde+Betanin+Crocin kombinasyonu, Bcl-2 düzeyini 0,61 ng/mL'ye düşürerek en belirgin azalmayı göstermiştir (p<0,001). Bu değer, Dosetaksel grubuna (0,59 ng/mL) oldukça yakındır. Kalpain-1 düzeyi Betanin+Crocin grubunda 5,38 ng/mL ile en yüksek seviyeye ulaşmış ve tüm tekli uygulamalardan anlamlı şekilde farklı bulunmuştur (p<0,001). Aynı kombinasyon MCF-10A hücrelerinde Kalpain-1 düzeyini azaltmıştır (p<0,001). P53 düzeyi Cinnamaldehyde+Betanin+Crocin grubunda en yüksek artışı göstermiştir (p=0,006). Sitokrom C düzeyi ise Cinnamaldehyde+Betanin grubunda en fazla artış göstermiştir (p=0,002; p<0,001). Araştırmanın sonuçları Cinnamaldehyde, Betanin ve Crocin'in sinerjik olarak uygulanmasının, MCF-7 hücrelerinde apoptotik süreci etkin bir biçimde tetikleyerek hücre canlılığını azalttığını ve bu etkinin apoptoza ilişkin farklı moleküler mekanizmalar üzerinden gerçekleştiğini ortaya koymuştur. Bu sinerjik etki, bileşiklerin meme kanserinde terapötik potansiyelini vurgulamakta ve ileri araştırmalar için değerli bir temel sunmaktadır. This study aimed to investigate the apoptotic effects of Cinnamaldehyde, Betanin, and Crocin, both as single agents and in combination, on MCF-7 breast cancer cells, while comparatively evaluating their effects on MCF-10A healthy breast epithelial cells. Docetaxel was employed as a reference agent to assess chemotherapeutic efficacy. Different concentrations of Cinnamaldehyde, Betanin, Crocin, and Docetaxel were administered to MCF-7 and MCF-10A cells, and their IC50 values at 24, 48, and 72 hours were determined using the MTT assay. Subsequently, single and combined treatments were applied to the cells, and the levels of Bcl-2, Calpain-1, P53, and Cytochrome C were analyzed via ELISA to evaluate their apoptotic effects. In MCF-7 cells, the combination of Cinnamaldehyde+Betanin+Crocin resulted in the most pronounced reduction in Bcl-2 levels, decreasing it to 0.61 ng/mL (p<0.001), which was comparable to the Docetaxel group (0.59 ng/mL). Calpain-1 levels reached their highest value in the Betanin+Crocin group at 5.38 ng/mL, showing a statistically significant difference compared to all single-agent treatments (p<0.001). Interestingly, the same combination led to a significant decrease in Calpain-1 levels in MCF-10A cells (p<0.001). The highest increase in P53 levels was observed in the Cinnamaldehyde+Betanin+Crocin group (p=0.006), whereas the Cytochrome C level was most elevated in the Cinnamaldehyde+Betanin group (p=0.002; p<0.001). The findings of the study revealed that the synergistic application of Cinnamaldehyde, Betanin, and Crocin effectively triggered the apoptotic process in MCF-7 cells, resulting in a significant reduction in cell viability through distinct apoptotic molecular mechanisms. This synergistic effect highlights the therapeutic potential of these compounds in breast cancer and provides a valuable foundation for future research. Dijital Bankacılıkta Bireysel Müşterilerin Uzaktan Müşteri Edinimi Yöntemiyle Banka Müşterisi Olma Noktasındaki Belirleyiciler
Teknolojinin gelişimi, bankacılık sektöründe operasyonel verimlilikten müşteri hizmetlerine kadar her alanda büyük bir dönüşüm geçirmesini sağlamıştır. Dijitalleşme adı verilen bu dönüşüm süreci bankacılığın daha erişilebilir, hızlı ve verimli hale gelmesini sağlarken aynı zamanda daha geniş kitlelere finansal hizmetlerin ulaşmasını mümkün kılmıştır. Dijitalleşme süreciyle bankacılık hizmetlerinin sunumu köklü bir şekilde değişmiş ve geleneksel şube tabanlı hizmet anlayışından dijital platformlar üzerinden sağlanan hizmetlere doğru geçişi hızlandırmıştır. Dijital platformlar bankaların daha geniş bir müşteri kitlesine ulaşmasını sağlamakla kalmayıp aynı zamanda maliyet etkinliği ve hız gibi avantajlar da sağlamıştır. Türkiye'de özellikle son 10 yılda bankacılık sektörü oyuncuları gerek rakip bankaların dijital stratejilerinden gerekse de kişiselleştirilmiş bir müşteri deneyimi sunan yıkıcı girişimlerden kaynaklanan rekabete cevap vermesiyle dijital dönüşüm süreci ciddi hız kazanmıştır. Dijitalleşme sürecinin etkisiyle Türkiye'de uzaktan müşteri edinimi önemli bir dönüşüm geçirmiştir. Önceleri dijital hizmet kanalları üzerinden yapılan başvuru sonucunda kurye ile gelen sözleşmeleri imzalayıp bankaya geri göndermek suretiyle müşteri edinimi ya da bankaya ait fiziksel cihaz üzerinden video konferans yöntemiyle yapılan görüşme sonrasında sözleşmenin imzalanmasıyla müşteri edinimi gibi farklı uygulamalar ile tabiri caizse yarı dijital bir şekilde müşteri edinimi mümkün iken 01/05/2021 tarihinde yürürlüğe giren yönetmelik ile bankacılık sektöründe dijital hizmet kanalları aracılığıyla kolay, hızlı ve tam dijital bir şekilde anında müşteri ediniminin önü açılmıştır. Bu çalışmada Türkiye'de aktif olarak faaliyet gösteren bir bankanın veri tabanından 01/08/2023 – 31/07/2024 tarihleri arasında şubeden müşteri edinimi ve uzaktan müşteri edinimi yöntemleriyle banka müşterisi olan bireysel gerçek kişilerin müşteri edinim sürecinde girmiş oldukları bilgilerin anonimleştirilerek gerçek kişi ile ilgili hiçbir ilişkisi kalmadan elde edilen veri seti üzerinde tek değişkenli ve çok değişkenli analizler yapılarak şubeden ve uzaktan müşteri edinimi yöntemleriyle banka müşterisi olmasına katkı sağlayan değişken ve çoklu değişken kombinasyonları araştırılmıştır. Analizi yapılan değişkenlerin anlamlılık düzeyini tespit etmek adına kategorik değişkenler için ki-kare analizi, numerik/sürekli değişkenler için ortalamadan bağımsız t-testi sonuçları yayınlanmıştır. Veri analizleri ve istatistiksel sonuçların ışığında belirlenen hipotezler test edilip detaylı bir şekilde değerlendirilmiştir. Ayrıca bir karar ağacı algoritması olan LightGBM algoritması ile oluşturulan modelin hiperparameteleri optimize edilerek 5 katmanlı çapraz doğrulama modeli uygulanarak modelin çıktıları parametreleriyle beraber ortaya koyulmuştur. Modelin genellikle doğru tahminler yaptığı ve güvenilir bir şekilde kullanılabilir olduğu görülmüştür. Modelin daha sağlıklı ve daha doğru sonuçlar üretmesi adına değişkenlerin önemi belirlenmiştir. Oluşturulan modelin sonuç tahminlemesine en çok etki eden belirleyicilerin yaş, meslek ve öğrenim durumu olduğu, en az etki eden belirleyicilerin ise medeni durum, çalışılan sektör ve cinsiyet olduğu belirlenmiştir.The development of technology has enabled a significant transformation in the banking sector, affecting all areas from operational efficiency to customer services. This transformation process, known as digitalization, has made banking more accessible, faster, and efficient, while also making it possible for financial services to reach a broader audience. with the digitalization process, the delivery of banking services has fundamentally changed, accelerating the shift from traditional branch-based service models to services provided through digital platforms. Digital platforms not only help banks reach a wider customer base, but also provide advantages such as cost efficiency and speed. In Turkey, especially in the last decade, the digital transformation process has gained significant momentum as banking sector players have responded to competition arising from both digital strategies of rival banks and disruptive startups offering personalized customer experiences. with the impact of the digitalization process, remote customer acquisition in Turkey has undergone a significant transformation. Previously, customer acquisition was carried out in a semi-digital manner through various practices, such as signing contracts sent via courier following an application made through digital service channels and returning them to the bank, or signing the contract after a video conference conducted via a physical device belonging to the bank. However, with the regulation that came into effect on May 1, 2021, the banking sector has been enabled to facilitate instant, easy, fast, and fully digital customer acquisition through digital service channels. In this study, univariate and multivariate analyses were conducted on a dataset obtained from the database of a bank actively operating in Turkey, covering the period between August 1, 2023, and July 31, 2024. The dataset was derived by anonymizing the information entered during the customer acquisition process by individual real persons who became bank customers through branch customer acquisition methods and remote customer acquisition methods, ensuring no connection to the actual individuals. The study investigated the variables and combinations of multiple variables contributing to becoming a bank customer through branch and remote acquisition methods. To determine the significance level of the analyzed variables, chi-square analysis was performed for categorical variables, and independent t-test results were published for numerical/continuous variables. Based on the data analysis and statistical results, the hypotheses were tested and evaluated in detail. Additionally, a model created using the LightGBM algorithm, a decision tree algorithm, was optimized by tuning hyperparameters, and a 5-fold cross-validation model was applied. The model's outputs were presented with their parameters. It was found that the model generally made accurate predictions and could be used reliably. To produce more accurate and robust results, the importance of the variables was determined. Investigation of the Effects of the Combination of Silibinin and Boric Acid on Apoptosis in the MCF-7 Breast Cancer Cell Line
Bu çalışmada, silibinin ve borik asit kombinasyonunun meme kanseri hücre hattı MCF-7 üzerindeki apoptotik etkileri değerlendirilmiş ve bu etkinin seçiciliği sağlıklı meme epiteli hücre hattı MCF-10A'da incelenmiştir. Hücre canlılığı MTT testi ile değerlendirilerek maddelerin IC₅₀ dozları belirlenmiştir. Apoptozla ilişkili biyobelirteçler olan MMP-2, Kaspaz-8 ve Kaspaz-9 protein düzeyleri ELISA yöntemiyle; NF-κB, p21, MMP-9 ve Ki-67 gen ekspresyon düzeyleri qPCR analizi ile ölçülmüştür. Elde edilen bulgular, MCF-7 hücre hattında silibinin uygulamasının MMP-2 düzeylerinde istatistiksel olarak anlamlı bir düşüşe yol açtığını göstermiştir (p<0,01). Borik asit, kombinasyon ve dosetaksel gruplarında bu azalmanın daha yüksek anlamlılık düzeyinde olduğu saptanmıştır (p<0,001). Tüm tedavi gruplarında Kaspaz-8 ve Kaspaz-9 konsantrasyonları istatistiksel olarak anlamlı düzeyde artmıştır (p<0,001). MCF-10A hücre hattında ise MMP-2, Kaspaz-8 ve Kaspaz-9 düzeyleri tüm tedavi gruplarında anlamlı artış göstermiştir (p<0,001). MCF-7 hücre hattında NF-κB ekspresyonu sadece silibinin grubunda baskılanmıştır (p<0,001). p21 ekspresyonu tüm tedavi gruplarında kontrol grubuna kıyasla artış gösterirken MMP-9 ve Ki-67 ekspresyon düzeyleri azalmıştır (p<0,001). Sonuçlar, silibinin ve borik asidin hem tek başlarına hem de kombine olarak uygulanmasının MCF-7 hücrelerinde apoptozu artırdığını, hücre proliferasyonunu ve invazyon kapasitesini baskıladığını göstermiştir. Bu veriler, silibinin ve borik asit bileşiklerinin kombinasyon halinde uygulanmasının meme kanseri tedavisinde tamamlayıcı ve geleceğe yönelik umut vadeden bir yaklaşım olabileceğini düşündürmektedir.In this study, we evaluated the apoptotic effects of a combination of silibinin and boric acid on the breast cancer cell line MCF-7 and the selectivity of this effect was investigated in the healthy breast epithelial cell line MCF-10A. Cell viability was assessed by MTT assay and the IC₅₀ doses of the substances were determined. Apoptosis-related biomarkers, MMP-2, caspase-8, and caspase-9 protein levels were measured by ELISA, while NF-κB, p21, MMP-9, and Ki-67 gene expression levels were measured by qPCR analysis. The findings showed that silibinin application in the MCF-7 cell line caused a statistically significant decrease in MMP-2 levels (p<0.01). This decrease was found to be higher in the boric acid, combination and docetaxel groups (p<0.001). Caspase-8 and caspase-9 concentrations increased statistically significantly in all treatment groups (p<0.001). In the MCF-10A cell line, MMP-2, caspase-8 and caspase-9 levels increased significantly in all treatment groups (p<0.001). In the MCF-7 cell line, NF-κB expression was suppressed only in the silibinin group (p<0.001). While p21 expression increased in all treatment groups compared to the control group, MMP-9 and Ki-67 expression levels decreased (p<0.001). The results showed that silibinin and boric acid, both alone and in combination, increased apoptosis and suppressed cell proliferation and invasiveness in MCF-7 cells. These data suggest that the combined application of silibinin and boric acid compounds may be a complementary and promising approach for the treatment of breast cancer
The Relationship between Emotional Expression Skills and Psychosocial Care Competencies Among Nurses in Turkey: A Cross-Sectional Study
BackgroundPsychosocial care is a core component of nursing practice aimed at meeting the holistic needs of patients. In this context, nurses' emotional expression skills play a critical role in delivering effective psychosocial care.AimThe aim of this study was to examine the relationship between nurses' psychosocial care competencies and their emotional expression skills, and to explore the subdimensions of this relationship.MethodThis descriptive and correlational cross-sectional study was conducted with 227 nurses working at a training and research hospital in Istanbul. Data were collected using a Socio-Demographic Information Form, the Psychosocial Care Competency Self-Assessment Scale (PCCSS), and the Emotional Expression Skills Scale (EESS). Data were collected between October and December 2023. Analyses were performed using SPSS 22.0, with Pearson correlation and multiple regression techniques.ResultsThe findings revealed statistically significant positive correlations between nurses' psychosocial care competencies and their emotional expression skills. Specifically, the proximity and positive emotion expression subdimensions showed moderate correlations with psychosocial care subscales. However, multiple regression analysis did not identify any of the psychosocial care subdimensions as significant predictors of emotional expression skills. This suggests the possible influence of environmental or personal mediators such as institutional support, burnout, or job satisfaction.ConclusionEnhancing nurses' emotional expression skills may contribute to the development of their psychosocial care competencies. It is recommended to integrate emotional intelligence-based content into nursing education curricula and implement emotional awareness programs in clinical practice. A Proposal for an Elderly Care Home Associated with a Vocational Health School
Günümüzde gelişmekte olan bir dünyada, yaşlı vatandaşlar ekstra bakım ve ilgiye çok ihtiyaç duymaktadır. Yine de, bir noktada, aile üyeleri, yoğun programlar, gerekli ekipman ve malzemeler dahil olmak üzere çeşitli nedenlerle yaşlılarına gereken ilgiyi gösteremezler; Bu nedenle, bir bakım evi, yaşlılar için sürdürülebilir bir yaşam ortamı sağlayabilecek bir seçenek haline gelir. Still, at a point, family members cannot provide proper attention to their older ones for various reasons, including busy schedules, the skills, required equipment, and materials; hence, a care home becomes an option that can provide a sustainable living environment for the older people. This research describes socialization as a major problem faced by the elderly in care homes, which causes isolation and progresses into depression hence, it proposes a paradigm shift in design that goes beyond the boundaries of a hospital or an isolation center. The architectural solutions propose creating a safe space where the elderly make new connections and rediscover themselves by integrating a vocational School that provides adaptive Spaces that identify with their needs, this is one of the factors considered very important as it is necessary to understand that the care of ethe lderly people can be a bit challenging because they become very sensitive as they age. Investigating the Effect of the Atomic Ratio of ClO2 Gas on the Disinfection Process of the Influenza Virus Using Molecular Dynamics Simulation
Influenza virus transmission remains a critical public health concern, necessitating effective disinfection strategies to control outbreaks. However, the molecular mechanisms by which varying atomic ratios of chlorine dioxide (ClO2) gas affect viral destabilization and inactivation are not fully understood. To address this knowledge gap, this study used molecular dynamics simulations using the LAMMPS software to investigate interactions between ClO2 gas and the influenza virus at different atomic ratios. Increasing the ClO2concentration from 15 % to 50 % significantly raised virus-gas interaction energy from 25,377.83 kcal/mol to 83,430.95 kcal/mol and virus-virus interaction energy from 523,570.84 kcal/mol to 558,130.12 kcal/mol. Concurrently, mean square displacement decreased, indicating reduced viral atom mobility, and the radius of gyration contracted from 68.55 & Aring; to 65.58 & Aring;, reflecting structural collapse. These molecular-level findings demonstrate that higher ClO2 atomic ratios strengthened the interactions that led to viral destabilization and accelerated structural breakdown, providing quantitative insights to optimize ClO2dosing protocols for effective disinfection in healthcare and public environments. Thermal and Stability Outcomes of Different Osteotomy Techniques and Implant Macrogeometries in Type IV Bone: An in Vitro Study
Achieving reliable implant stability in type IV (D4) bone remains a clinical challenge due to its thin cortical plates and low trabecular density, which increase the risk of early failure. Novel osteotomy techniques such as osseodensification and the OsseoShaper have been proposed, yet their effects when combined with different implant macrogeometries are not fully understood. This in vitro study evaluated forty osteotomies in standardized polyurethane foam blocks simulating D4 bone density. Three site preparation protocols-conventional osteotomy, osseodensification, and OsseoShaper-were combined with two implant macrogeometries (parallel-walled conical and tri-oval tapered designs). Insertion torque (IT) was measured using a calibrated digital torque meter, and implant stability was assessed by resonance frequency analysis (ISQ). Intraoperative thermal changes were monitored with infrared thermography under constant irrigation. Statistically significant differences were observed among groups (p < 0.05). Osseodensification with parallel-walled implants achieved the highest stability, whereas osseodensification with tri-oval implants showed the lowest. Although osseodensification produced the greatest thermal increase, all values remained below the 47 degrees C osteonecrosis threshold. Within the study's limitations, both the osteotomy technique and implant macrogeometry significantly affected stability and thermal outcomes, with osseodensification plus parallel-walled implants providing the most predictable performance in D4 bone.Science Citation Index Expande

Pauls, Alexander/0000-0002-8117-5376; Fehérkuti, Anna/0000-0002-5043-2958; Zucchetta, Alberto/0000-0003-0380-1172;A search is performed for dark matter (DM) produced in association with a single top quark or a pair of top quarks using the data collected with the CMS detector at the LHC from proton-proton collisions at a center-of-mass energy of 13TeV, corresponding to 138 fb(-1) of integrated luminosity. An excess of events with a large imbalance of transverse momentum is searched for across 0, 1 and 2 lepton final states. Novel multivariate techniques are used to take advantage of the differences in kinematic properties between the two DM production mechanisms. No significant deviations with respect to the standard model predictions are observed. The results are interpreted considering a simplified model in which the mediator is either a scalar or pseudoscalar particle and couples to top quarks and to DM fermions. Axion-like particles that are coupled to top quarks and DM fermions are also considered. Expected exclusion limits of 410 and 380 GeV for scalar and pseudoscalar mediator masses, respectively, are set at the 95% confidence level. A DM particle mass of 1 GeV is assumed, with mediator couplings to fermions and DM particles set to unity. A small signal-like excess is observed in data, with the largest local significance observed to be 1.9 standard deviations for the 150 GeV pseudoscalar mediator hypothesis. Because of this excess, mediator masses are only excluded below 310 (320) GeV for the scalar (pseudoscalar) mediator. Türkiye'de Pole Spor Branşı ile İlgilenen Sporcuların Öz Güven ve Öz Saygı Düzeylerinin İncelenmesi
Çalışma Türkiye'deki tüm pole sporcularının öz güven ve benlik saygısı düzeylerinin incelenmesini hedeflemiştir. Bu amaçla araştırmacı tarafından hazırlanan Katılımcı Onam ve Kişisel Bilgi Formu, Rosenberg tarafından oluşturulmuş Benlik Saygısı Ölçeği ve Ahmet Akın tarafından oluşturulmuş Öz Güven Ölçeği, Türkiye'deki ulaşılabilen tüm pole sporcularına online olarak uygulanmıştır. Yapılan web araştırması sonucu tüm spor okullarının, ulaşılabilen tüm sporcularına ulaşıldığı varsayılmıştır. Elde edilen veriler, SPSS programı ile incelenmiş ve veri toplanan 126 sporcunun 122'sinin öz güven düzeyi, ölçekte belirtilen sınırın üzerinde yani yüksek düzeyde bulunmuştur. 4'ünün ise öz güven düzeyi ölçekte belirtilen sınırın altında yani düşük düzeyde bulunduğu görülmüştür. Bu 4 sporcunun da kişisel bilgi formuna bakıldığında pole spor yapma süresinin, en kısa işaretlenebilecek seçenek olan 1-4 yıl olarak işaretledikleri verisine ulaşılmış ve bu durum da katılımcıların pole spora yeni başlamış olabileceklerini düşündürmüştür. 126 sporcunun 126'sının da Rosenberg Benlik Saygısı Ölçeği'nin Benlik Saygısı alt boyutunun ölçekte belirtilen sınırda veya üzerinde bulunmuştur ve tüm katılımcıların benlik saygısının orta düzeyde veya yüksek düzeyde olduğu bilgisine ulaşılmıştır. Tüm bunların sonucunda pole spora katılım sağlayanların öz güven ve benlik saygısı seviyelerinin yüksek olduğu ve pole spor yapma süresi uzadıkça öz güven ve benlik saygısı seviyelerinin yükseldiği düşünülmektedir. Pole sporun benlik saygısı ve öz güven gelişimi için bir araç olarak kullanılabileceği düşünülmekte ve önerilmektedir.The study aimed to investigate the self-confidence and self-esteem levels of all pole athletes in Turkey. For this purpose, the Researcher's Participant Consent and Personal Information Form, Rosenberg's Self-Esteem Scale, and Ahmet Akın's Self-Confidence Scale were administered online to all accessible pole athletes in Turkey. Due to the absence of an official pole sports federation in Turkey, official figures regarding the number of athletes could not be obtained. Based on a web survey, it is assumed that all sports schools reached all accessible athletes. The collected data were analysed using the SPSS program, revealing that out of the 126 surveyed athletes, 122 had a high level of self-confidence above the specified threshold on the scale. 4 athletes had a low level of self-confidence below the specified threshold on the scale. Upon reviewing the personal information forms of these 4 athletes, it was noted that their duration of pole sports practice was marked as the shortest selectable option of 1-4 years, indicating that these participants might have recently started pole sports. All 126 athletes also scored at or above the threshold of the Self-Esteem subscale of the Rosenberg Self-Esteem Scale, indicating that all participants had moderate or high levels of self-esteem. As a result, it is observed that participants engaging in pole sports have high levels of self-confidence and self-esteem, and as the duration of pole sports practice increases, self-confidence and self-esteem levels rise. It is suggested that pole sports can be used as a tool for the development of self-esteem and self-confidence
Engineering Vascularized Brain Tumor Organoids: Bridging the Gap Between Models and Reality
Traditional two-dimensional cultures and patient-derived xenografts fail to fully mimic the complexity of the tumor microenvironment, limiting their utility in drug discovery and personalized medicine. Recent breakthroughs in three-dimensional tumor modeling have led to the development of brain tumor organoids, patient-derived organoids, and bioengineered tumor-on-chip systems that offer more physiologically relevant platforms for studying glioblastoma biology and therapeutic response. One of the key advancements in these models is the incorporation of vascular networks to mimic the neurovascular unit and the blood-brain barrier (BBB). Various strategies such as co-culturing with endothelial cells, bio-printing vascularized scaffolds, and utilizing microfluidic platforms have been explored to enhance vascularization within glioblastoma organoids. These models have demonstrated improved nutrient and oxygen exchange, reduced hypoxia, and better maintenance of tumor heterogeneity. However, challenges remain in achieving fully functional capillary networks, BBB integrity, and immune cell integration. This review provides a comprehensive analysis of the latest advancements in brain tumor organoid research, focusing on vascularization strategies, their impact on tumor modeling, and their potential applications in drug screening and personalized therapy. We discussed the strengths and limitations of glioblastoma models, highlighted advanced bioengineering techniques for enhancing organoid complexity, and explored future directions for clinically relevant tumor organoids.Science Citation Index Expande
