Contrôle de la géométrie et de l'anisotropie par la combinaison des effets de coordination stérique et anionique dans les complexes Co II avec ligands N 6 -tripodaux : l'impact de la taille du ligand sur le temps de relaxation magnétique
International audienceFour mononuclear CoII complexes of formula [Co(L)(SCN)2(CH3OH)0.5(H2O)0.5]·1.5H2O·0.75CH3OH (1), [Co(L1)Cl2]·H2O·2CH3CN (2), [Co(L1)(SCN)2]·1.5H2O·CH3OH (3) and [Co(L1)]ClO4·2CH3OH (4) were prepared from the N6-tripodal Schiff base ligands (S)P[N(Me)N[double bond, length as m-dash]C(H)2-Q]3 (L) and (S)P[N(Me)N[double bond, length as m-dash]C(H)1-ISOQ]3 (L1), where Q and ISOQ represent quinolyl and isoquinolyl moieties, respectively. In 1, the L ligand does not coordinate to the CoII ion in a tripodal manner but using a new N,N,S tridentate mode, which is due to the fact that the N6-tripodal coordination promotes a strong steric hindrance between the quinolyl moieties. However, L1 can coordinate to the CoII ions either in a tripodal manner using CoII salts with poorly coordinating anions to give 4 or in a bisbidentate fashion using CoII salt-containing medium to strongly coordinating anions to afford 2 and 3. In the case of L1, there is no steric hindrance between ISOQ moieties after coordination to the CoII ion. The CoII ion exhibits a distorted octahedral geometry for compounds 1–3, with the anions in cis positions for the former and in trans positions for the two latter compounds. Compound 4 shows an intermediate geometry between an octahedral and trigonal prism but closer to the latter one. DC magnetic properties, HFEPR and FIRMS measurements and ab initio calculations demonstrate that distorted octahedral complexes 1–3 exhibit easy-plane magnetic anisotropy (D > 0), whereas compound 4 shows large easy-axis magnetic anisotropy (D Quatre complexes mononucléaires de Co II de formule [[Co(L)(SCN) 2 (H 2 O) 0,5 (CH 3 OH) 0,5 ]•1,5H 2 O•0,75CH 3 OH (1), [Co(L1)Cl 2 ]•H 2 O•2CH 3 CN (2), [Co(L1)(SCN) 2 ]• 1,5H 2 O•CH 3 OH (3) et [Co(L1)]ClO 4 •2CH 3 OH (4) ont été préparés à partir des ligands de bases de Schiff N 6 -tripodes (S)P[N(Me)N=C(H)2-Q] 3 (L) et (S)P[N(Me)N=C(H)1-ISOQ] 3 (L1), où Q etISOQ représentent respectivement les fractions quinolyle et isoquinolyle. Dans le cas 1, le ligand L ne se coordonne pas à l'ion Co II de manière tripodale mais utilise un nouveau mode tridenté N, N, S, ce qui est dû au fait que la coordination N 6 -tripodale favorise un fort encombrement stérique entre les groupements quinolyle. Cependant, L1 peut coordonner les ions Co II soit de manière tripodale en utilisant des sels de Co II avec des anions faiblement coordonnants pour donner 4, soit de manière bisbidentate en utilisant des sels de Co II contenant des anions moyennement à fortement coordonnants pour donner 2 et 3. Dans le cas de L1, il n'y a pas d'encombrement stérique entre les groupements ISO après coordination à l'ion Co II. L'ion Co II présente une géométrie octaédrique déformée pour les composés 1 à 3, avec les anions en position cis pour le premier et en position trans pour les deux derniers. Le composé 4 présente une géométrie intermédiaire entre le prisme octaédrique et le prisme trigonal, mais plus proche de ce dernier.Les propriétés magnétiques DC, les mesures HFEPR et FIRMS et les calculs ab initio démontrent que les complexes octaédriques déformés 1-3 présentent une anisotropie magnétique dans le plan facile (D > 0), tandis que le composé 4 présente une grande anisotropie magnétique dans le plan facile (D < 0). L'analyse comparative des données magnétostructurales souligne le rôle important joué non seulement par la géométrie de coordination mais aussi par les effets électroniques dans la détermination de l'anisotropie des ions Co II. Les composés 2-3 présentent une relaxation lente de l'aimantation induite par le champ. Malgré sa grande anisotropie magnétique dans le plan facile, le composé 4 ne présente pas de relaxation lente (SMR) significative au-dessus de 2 K sous un champ magnétique appliqué nul, mais sa dilution magnétique avec Zn II déclenche une SMR à champ nul. Enfin, il convient de noter que les composés 2 à 4 présentent des temps de relaxation plus courts que les complexes analogues avec le ligand tripode portant dans ses bras des fractions pyridine au lieu d'isoquinoléine, ce qui est très probablement dû à l'augmentation de la taille moléculaire dans le premier.</div
Apport de l'IRM pour le diagnostic précoce des syndromes parkinsoniens chez une population de patients en situation d'incertitude diagnostique
International audienceBackground: International clinical criteria are the reference for the diagnosis of degenerative parkinsonism in clinical research, but they may lack sensitivity and specificity in the early stages. Objectives: To determine whether magnetic resonance imaging (MRI) analysis, through visual reading or machine‐learning approaches, improves diagnostic accuracy compared with clinical diagnosis at an early stage in patients referred for suspected degenerative parkinsonism. Methods: Patients with initial diagnostic uncertainty between Parkinson's disease (PD), progressive supranuclear palsy (PSP), and multisystem atrophy (MSA), with brain MRI performed at the initial visit (V1) and available 2‐year follow‐up (V2), were included. We evaluated the accuracy of the diagnosis established based on: (1) the international clinical diagnostic criteria for PD, PSP, and MSA at V1 (“Clin1”); (2) MRI visual reading blinded to the clinical diagnosis (“MRI”); (3) both MRI visual reading and clinical criteria at V1 (“MRI and Clin1”), and (4) a machine‐learning algorithm (“Algorithm”). The gold standard diagnosis was established by expert consensus after a 2‐year follow‐up. Results: We recruited 113 patients (53 with PD, 31 with PSP, and 29 with MSA). Considering the whole population, compared with clinical criteria at the initial visit (“Clin1”: balanced accuracy, 66.2%), MRI visual reading showed a diagnostic gain of 14.3% (“MRI”: 80.5%; P = 0.01), increasing to 19.2% when combined with the clinical diagnosis at the initial visit (“MRI and Clin1”: 85.4%; P < 0.0001). The algorithm achieved a diagnostic gain of 9.9% (“Algorithm”: 76.1%; P = 0.08). Conclusion: Our study shows the contribution of MRI analysis, whether by visual reading or machine‐learning methods, for the early differentiation of parkinsonism.Contexte : Les critères cliniques internationaux sont la référence pour le diagnostic des syndromes parkinsoniens dégénératifs, mais ils peuvent manquer de sensibilité et de spécificité aux stades précoces. Objectifs : Déterminer si l'analyse par imagerie par résonance magnétique (IRM), par lecture visuelle ou par approche d'apprentissage automatique, améliore la précision diagnostique par rapport au diagnostic clinique à un stade précoce chez les patients orientés pour un parkinsonisme dégénératif suspecté. Méthodes : La population incluait des patients avec une suspicion de maladie de Parkinson (MP), paralysie supranucléaire progressive (PSP) ou atrophie multisystématisée (AMS), ayant réalisé une IRM cérébrale lors de la visite initiale (V1) et ayant bénéficié d'un suivi à 2 ans (V2). Nous avons évalué la précision du diagnostic établi en fonction : (1) des critères diagnostiques cliniques internationaux pour la MP, la PSP et l'AMS lors de la V1 (« Clin1 ») ; (2) de l'analyse radiologique de l'IRM en aveugle du diagnostic clinique (« IRM ») ; (3) de la combinaison de l'analyse radiologique de l'IRM et des critères cliniques lors de la V1 (« IRM et Clin1 ») ; et (4) d'un algorithme d'apprentissage automatique (« Algorithme »). Le diagnostic de référence a été établi par consensus d'experts après un suivi de 2 ans. Résultats : 113 patients ont été recrutés, incluant 53 avec MP, 31 avec PSP et 29 avec AMS). En considérant l'ensemble de la population, comparé aux critères cliniques lors de la visite initiale (« Clin1 » : précision: 66,2 %), la lecture visuelle de l'IRM a montré un gain diagnostique de 14,3 % (« IRM » : 80,5 % ; P = 0,01), augmentant à 19,2 % lorsqu'elle est combinée avec le diagnostic clinique lors de la visite initiale (« IRM et Clin1 » : 85,4 % ; P < 0,0001). Control over the spatial correlation of perforations in silica thin films as a function of solution conditions

Deformation of drops at low Reynolds number impact
International audienceDrop impact experiments are performed with very viscous fluids to propose a description of the drop deformation at low Reynolds number. We focus on a specific case where dimensionless parameters other than the Reynolds number play no role, which means that only kinetic energy and viscous dissipation determine the final deformation. The same situation in the case of a Reynolds number larger than ten has been clarified years ago. The maximum diameter of the spread drop is well described by a 1/5 power law of the Reynolds number only. Here the deformation of the drop, defined as the contact diameter rescaled by the drop size, is also a power law of the Reynolds number. From experimental data and scaling arguments, the exponent of the power law is shown to be 1/3
Medical hypnosis mitigates laboratory dyspnoea in healthy humans: a randomised, controlled experimental trial
International audienceQuestion Dyspnoea persisting despite treatments of underlying causes requires symptomatic approaches. Medical hypnosis could provide relief without the untoward effects of pharmacological approaches. We addressed this question through experimentally induced dyspnoea in healthy humans (inspiratory threshold loading –excessive inspiratory effort– and carbon dioxide stimulation –air hunger–) Material and Methods 20 volunteers (10 women, 21–40) were studied on 4 separate days. The order of the visits was randomised in two steps, firstly “inspiratory threshold loading first” versus “carbon dioxide first” group (n=10 in each group), secondly “medical hypnosis first” versus “visual distraction first” subgroup (n=5 in each subgroup). Each visit comprised three 5-minutes periods (reference, intervention, washout) during which participants used visual analog scales to rate dyspnoea's sensory and affective dimensions and after which they completed the Multidimensional Dyspnea Profile. Results Medical hypnosis reduced both dimensions of dyspnoea significantly more than visual distraction (inspiratory threshold loading: sensory reduction after 5 min 34% of full VAS versus 8% –p=0.0042–; affective reduction 17.6% versus 2.4% –p=0.044–; carbon dioxide; sensory reduction after 5 min 36.9% versus 3% –p=0.0015–; affective reduction 29.1% versus 8.7% –p=0.0023–). The Multidimensional Dyspnea Profile showed more marked sensory effects during inspiratory threshold loading and more marked affective effects during carbon dioxide stimulation. Answer to the question Medical hypnosis was more effective than visual distraction at attenuating the sensory and affective dimensions of experimentally induced dyspnoea. This provides a strong rationale for clinical studies of hypnosis in persistent dyspnoea patients
Auto-assemblage dynamique de bactéries dans des cristaux liquides
Despite the importance of bacterial dynamics in anisotropic environments, such as intestinal mucus, many essential questions about their transport remain unresolved. Water-soluble liquid crystals have proven to be a model system for studying the influence of anisotropy on the swimming and space exploration behavior of microswimmers. Recent studies have shown that bacteria are forced to follow the nematic director, inhibiting their intrinsic run-and-tumble explorationdynamics.Before studying the complex dynamics of bacteria in liquid crystals, we focus on the swimming mechanisms of bacteria in viscous Newtonian fluids. Using 3D Lagrangian tracking and a direct visualization of flagella, we simultaneously measure the swimming velocity, and flagella and body rotation frequency of E. coli bacteria. These simultaneous measurements allow the validation of a minimal model of swimming bacteria, with the measurement of torque generated by a flagellabundle. Varying the viscosity of the Newtonian fluid induces a variation in swimming velocity; from this variation, we can extract flagellar motor characteristics. The visualization of flagella and the body simultaneously also enables the study of wobbling, the off-axis rotation of the body. We measure directly the angle between the body and the flagellar bundle and study its impact on velocity. These measurements are the first to characterize these dynamics under free-swimmingconditions.Liquid crystals enable new types of collective behavior, such as the spontaneous formation of self-propelled trains of bacteria, which we investigate here. Bacteria enforce a planar anchoring of the liquid crystals at their surface, creating a distortion that results in an elastic attraction between bacteria. This attraction competes with a hydrodynamic repulsion due to the pusher flow field generated by bacteria swimming. Experiments at different liquid crystal concentrations, thusat different elastic strengths, always show train formation demonstrating the dominance of elastic attraction over the hydrodynamic repulsion.Our study focuses on understanding the aggregation dynamics and velocities of these trains, revealing that velocity increases with chain length. The aggregation follows a new kind of dynamics, limited by the velocity of the bacteria. We propose a model and simulate this aggregation, studying the evolution of the chain length distrution and of the chain velocity for different lengths over time. Data of the swimming speed as a function of chain length are completed by direct observation of the flagella using the 3D Lagrangian tracking device. These observations reveal the formation of asuper-bundle, where flagella from all bacteria in the train bundle together enabling train propulsion.Finally, we investigate a new system in which bacteria are confined in liquid crystal droplets. The liquid crystal is constrained to form topological defects at the poles in such droplets. We show that bacteria are attracted by these defects and swim following the director at the droplet surface.These findings give insights into bacteria transport dynamics in complex anisotropic environments. It can serve as the basis for further studies of aggregation of self-propelled colloids, and the study of living liquid crystal in complex and curved geometries. This work also paves the way for applications in the control of active matter and cargo transport, for example in drug deleveries.Malgré l'importance de la dynamique bactérienne dans les environnements anisotropes, tels que le mucus intestinal, de nombreuses questions essentielles sur ce transport restent sans réponse. Les cristaux liquides solubles dans l'eau se sont révélés être un système modèle pour étudier l'influence de l'anisotropie sur le comportement de nage et d'exploration spatiale des micro-nageurs. Des études récentes ont montré que les bactéries sont forcées de suivre le directeur nématique, ce qui inhibe leur dynamique intrinsèque d'exploration en run-and-tumble.Avant d'étudier la dynamique complexe des bactéries dans les cristaux liquides, nous nous concentrons sur les mécanismes de nage des bactéries dans les fluides visqueux newtoniens. En utilisant un tracking lagrangien 3D et une visualisation directe des flagelles, nous mesurons, pour des bactéries E. coli simultanément la vitesse de nage, et la fréquence des flagelles et du corps. Ces mesures simultanées permettent de valider un modèle minimal de la nage des bactéries, avec la mesure du couple généré par un faisceau de flagelles. La variation de la viscosité du fluide newtonien induit une variation de la vitesse de nage ; de cette variation, nous pouvons extraire les caractéristiques du moteur flagellaire. La visualisation simultanée des flagelles et du corps permet également d'étudier le wobbling, la rotation désaxée du corps. Nous mesurons directement l'angle entre le corps et le faisceau flagellaire et étudions son impact sur la vitesse. Ces mesures sont les premières à caractériser cette dynamique dans des conditions de nage libre.Les cristaux liquides permettent de nouveaux types de comportements collectifs, tels que la formation spontanée de trains de bactéries autopropulsés, que nous étudions ici. Les bactéries imposent un ancrage planaire des cristaux liquides à leur surface, créant une distorsion qui entraîne une attraction élastique entre les bactéries. Cette attraction est en compétition avec une répulsion hydrodynamique due à l'écoulement de type pusher généré par la nage des bactéries. Dans des expériences à différentes concentrations de cristaux liquides, donc à différentes forces élastiques, on observe toujours la formation de trains démontrant la dominance de l'attraction élastique sur la répulsion hydrodynamique.Notre étude se concentre sur la compréhension de la dynamique d'agrégation et des vitesses de ces trains, révélant que la vitesse augmente avec la longueur de la chaîne. L'agrégation suit un nouveau type de dynamique limitée par la vitesse des bactéries. Nous proposons un modèle et simulons cette agrégation, en étudiant l'évolution de la distribution de la longueur de la chaîne et celle de la vitesse pour les différentes longueurs de chaîne au fil du temps. Les données sur la vitesse de nage en fonction de la longueur de la chaîne sont complétées par l'observation directe des flagelles à l'aide du dispositif lagrangien de tracking 3D. Ces observations révèlent la formation d'un super-faisceau, dans lequel les flagelles de toutes les bactéries du train s'agrègent, ce qui permet la propulsion de la chaine.Enfin, nous étudions un nouveau système dans lequel les bactéries sont confinées dans des gouttes de cristal liquide. Le cristal liquide est contraint de former des défauts topologiques aux pôles de ces gouttes. Nous montrons que les bactéries sont attirées par ces pôles et nagent en suivant le directeur à la surface des gouttelettes.Ces résultats donnent une idée de la dynamique du transport des bactéries dans des environnements complexes. Ils ouvrent de nouvelles études sur la nouvelle dynamique d'agrégation limitée par la nage. Et ils proposent aussi l'étude d'un nouveau système avec les gouttes de cristal liquide vivant.Ces travaux ouvrent également la voie à des applications dans le contrôle de la matière active et le transport de cargo, par exemple dans le domaine de la santé pour une libération ciblée de médicament
Réactions de couplage croisé séquentiel asymétrique avec des réactifs prochiraux C(sp3)-gem-bimétalliques à base de zinc
This thesis presents the development of asymmetric sequential cross-coupling reactions using prochiral C(sp3)-gem-dizincio reagents. While enantioselective consecutive cross-coupling reactions have been explored for various gem-bimetallics, a significant gap exists in the literature for 1,1-dizincio reagents. The motivation behind this work is the establishment of novel enantioselective sequential cross-coupling reactions employing these reagents to synthesize complex, optically active molecules in a modular fashion. The first part introduces a highly enantioselective palladium-catalyzed sequenced arylation/acylation of prochiral 1,1-bis(iodozincio)alkanes achieving α-disubstituted ketones bearing enolizable tertiary stereocenters in excellent yields and enantiomeric ratios. In the second part, a significant advancement was attempted using silyl-substituted gem-dizincio reagents, potentially enabling three consecutive reactions in a single pot.Cette thèse présente le développement de réactions asymétriques de couplages croisés séquentiels à partir des réactifs prochiraux C(sp3)-gem-dizinc. Alors que la réalisation de deux couplages croisés consécutifs en version énantiosélective a été explorée pour divers composés gem-bimétalliques, dans la littérature, un manque significatif de travaux existe pour les réactifs 1,1-dizinc. Ce travail consiste ainsi en l'établissement de nouvelles réactions asymétriques de couplages croisés séquentiels en utilisant ces réactifs pour synthétiser des molécules complexes et optiquement actives de manière modulaire. La première partie présente une séquence arylation/acetylation pallado-catalysée hautement énantiosélective de 1,1-bis(iodozincio)alcanes prochiraux, conduisent à des cétones α-disubstituées portant des centres stéréogenes tertiaires énolisables avec d’excellents rendements et rapports énantiomériques. Dans la deuxième partie, une avancée significative a été tentée en utilisant des réactifs gem-dizinc silyl-substitués, permettant potentiellement trois réactions consécutives en une seule étape

Single ion mobility monitoring (SIM2) stitching method for high-throughput and high ion mobility resolution chiral analysis
International audienceAbstract Chiral analysis is of high interest in many fields such as chemistry, pharmaceuticals and metabolomics. Mass spectrometry and ion mobility spectrometry are useful analytical tools, although they cannot be used as stand-alone methods. Here, we propose an efficient strategy for the enantiomer characterization of amino acids (AAs) using non-covalent copper complexes. A single ion mobility monitoring (SIM 2 ) method was applied on a TIMS-ToF mass spectrometer to maximize the detection and mobility separation of isomers. Almost all of the 19 pairs of proteinogenic AA enantiomers could be separated with at least one combination with the chiral references L-Phe and L-Pro. Furthermore, we extended the targeted SIM 2 method by stitching several mobility ranges, in order to be able to analyze complex mixtures in a single acquisition while maintaining high mobility resolution. Most of the enantiomeric pairs of AAs separated with the SIM 2 method were also detected with this approach. The SIM 2 stitching method thus opens the way to a more comprehensive chiral analysis with TIMS-ToF instruments. Graphical Abstrac
Rotaxane-catalyzed aerobic oxidation of primary alcohols
International audienceNitroxide radicals are widely utilized as catalysts for the oxidation of primary alcohols. Here, the aerobic catalytic oxidation cycle of nitroxide radicals has been implemented within a mechanically interlocked rotaxane architecture consisting of a paramagnetic crown ether, which is confined by a molecular axle containing a dialkylammonium station and a 1,2,3-triazole unit. The rotaxane is engineered to exploit the oxidation of a primary alcohol: the primary catalyst is the wheel, a nitroxide radical capable of altering its oxidation state during the catalytic cycle, while the co-oxidant is the Cerium(IV)/O 2 couple. The synthesis of the proposed rotaxane, along with its characterization using EPR, HRMS, voltammetry and NMR data, is reported in the paper. The aerobic catalytic oxidation cycle was further investigated using EPR, NMR and GC-MS analyses. This study can aid in the design of autonomously driven molecular machines that exploit the aerobic catalytic oxidation of nitroxide radicals.</div
