Repeated fasting events sensitize enhancers, transcription factor activity and gene expression to support augmented ketogenesis
Abstract Mammals withstand frequent and prolonged fasting periods due to hepatic production of ketone bodies. Because the fasting response is transcriptionally-regulated, we asked whether enhancer dynamics impose a transcriptional program during recurrent fasting and whether this generates effects distinct from a single fasting bout. We found that mice undergoing alternate-day fasting (ADF) respond profoundly differently to a following fasting bout compared to mice first experiencing fasting. Hundreds of genes enabling ketogenesis are ‘sensitized’ (induced more strongly by fasting following ADF). Liver enhancers regulating these genes are also sensitized and harbor increased binding of PPARα, the main ketogenic transcription factor. ADF leads to augmented ketogenesis compared to a single fasting bout in wild-type, but not hepatocyte-specific PPARα-deficient mice. Thus, we found that past fasting events are ‘remembered’ in hepatocytes, sensitizing their enhancers to the next fasting bout and augment ketogenesis. Endothelial force sensing signals to parenchymal cells to regulate bile and plasma lipids
International audienceHow cardiovascular activity interacts with lipid homeostasis is incompletely understood. We postulated a role for blood flow acting at endothelium in lipid regulatory organs. Transcriptome analysis was performed on livers from mice engineered for deletion of the flow-sensing PIEZO1 channel in endothelium. This revealed unique up-regulation of Cyp7a1 , which encodes the rate-limiting enzyme for bile synthesis from cholesterol in hepatocytes. Consistent with this effect were increased gallbladder and plasma bile acids and lowered hepatic and plasma cholesterol. Elevated portal fluid flow acting via endothelial PIEZO1 and genetically enhanced PIEZO1 conversely suppressed Cyp7a1 . Activation of hepatic endothelial PIEZO1 channels promoted phosphorylation of nitric oxide synthase 3, and portal flow-mediated suppression of Cyp7a1 depended on nitric oxide synthesis, suggesting endothelium-to-hepatocyte coupling via nitric oxide. PIEZO1 variants in people were associated with hepatobiliary disease and dyslipidemia. The data suggest an endothelial force sensing mechanism that controls lipid regulation in parenchymal cells to modulate whole-body lipid homeostasis
Propagation of distinct CWD prion strains during peripheral and intracerebral challenges of gene-targeted mice
International audienceSince prion diseases result from infection and neurodegeneration of the central nervous system (CNS), experimental characterizations of prion strain properties customarily rely on the outcomes of intracerebral challenges. However, natural transmission of certain prions, including those causing chronic wasting disease (CWD) in elk and deer, depends on propagation in peripheral host compartments prior to CNS infection. Using gene-targeted GtE and GtQ mice, which accurately control cellular elk or deer PrP expression, we assessed the impact that peripheral or intracerebral exposures play on CWD prion strain propagation and resulting CNS abnormalities. Whereas oral and intraperitoneal transmissions produced identical neuropathological outcomes in GtE and GtQ mice and preserved the naturally convergent conformations of elk and deer CWD prions, intracerebral transmissions generated CNS prion strains with divergent biochemical properties in GtE and GtQ mice that were changed compared to their native counterparts. While CWD replication kinetics remained constant during iterative peripheral transmissions and brain titers reflected those found in native hosts, serial intracerebral transmissions produced 10-fold higher prion titers and accelerated incubation times. Our demonstration that peripherally and intracerebrally challenged Gt mice develop dissimilar CNS diseases which result from the propagation of distinct CWD prion strains points to the involvement of tissue-specific cofactors during strain selection in different host compartments. Since peripheral transmissions preserved the natural features of elk and deer prions, whereas intracerebral propagation produced divergent strains, our findings illustrate the importance of experimental characterizations using hosts that not only abrogate species barriers but also accurately recapitulate natural transmission routes of native strains
Recherche de critères décisionnels pour la gestion médicale de la pyélonéphrite chez la vache : intérêt de l’urémie
Pyelonephritis is a bacterial infection of the kidney secondary to ascending contamination of the urinary tract. It is the most common urinary tract infection in cattle, and occurs exclusively in post-partum cows. The aim of this study was to identify criteria to aid therapeutic decision-making in the case of pyelonephritis. Clinical and paraclinical data collected on 104 cases referred to the ENVT bovine clinic confirm that the disease appears within 3 months of parturition, with marked hematuria, renal abnormalities detected by palpation or ultrasound, and increased uremia and creatininemia. For a uremia value of over 40 mmol/L, the risk of therapeutic failure is two times higher, which is the best predictor of survival in this context. This study needs to be completed to make it possible to use this indicator in first-line cases.La pyélonéphrite est une infection bactérienne rénale secondaire à une contamination ascendante du tractus urinaire. C’est la plus fréquente des affections urinaires chez les bovins et elle apparaît exclusivement chez la vache dans la période post-partum. Cette étude vise à identifier des critères d’aide à la décision thérapeutique lors de pyélonéphrite. Les données cliniques et paracliniques collectées sur 104 cas référés à la clinique bovine de l’ENVT confirme que la maladie s’exprime dans les 3 mois suivant la parturition, avec une hématurie marquée, des anomalies rénales détectées par palpation ou à l’échographie, ainsi qu’une urémie et une créatininémie augmentées. Pour une valeur de l’urémie supérieure à 40 mmol/L, le risque d’échec thérapeutique est deux fois plus élevé, ce qui constitue le meilleur indicateur de la survie dans ce contexte. Cette étude devra être complétée afin de rendre possible l’utilisation de cet indicateur sur des cas en première intention
Molecular responses of chicken embryos to maternal heat stress through DNA methylation and gene expression
Abstract Climate change, with its repercussions on agriculture, is one of the most important adaptation challenges for livestock production. Poultry production is a major source of proteins for human consumption all over the world. With a growing human population, improving poultry’s adaptation to environmental constraints becomes critical. Extensive evidence highlights the influence of environmental variations on epigenetic modifications. The aim of this paper is therefore to explore chickens’ molecular response to maternal heat stress. We employed Reduced Representation Bisulfite Sequencing (RRBS) to generate genome-wide single-base resolution DNA methylation profiling and RNA sequencing (RNA-seq) to profile the transcriptome of the brains of embryos hatched from dams reared under either heat stress (32 °C) or thermoneutrality (22°C). We detected 289 significant differentially methylated CpG sites (DMCs) and one differentially methylated region (DMR) between heat stressed and control groups. These DMCs were associated with 357 genes involved in processes such as cellular response to stimulus, developmental processes and immune function. In addition, we identified 11 genes differentially expressed between the two groups of embryos, and identified ATP9A as a target gene of maternal heat stress on offspring. This study provides a body of fundamental knowledge on adaptive mechanisms concerning heat tolerance in chickens
Genetic determinism of resilience to unrecorded challenges in pigs
Session 85. Genetics of novel health and welfare traitsInternational audienceSelection of animals able to maintain their performance under variable environmental conditions is needed. Pigsin selection farms are raised in a monitored and controlled environment (e.g. temperature). However, pigs couldbe exposed to challenges/disturbances unrecorded and from unknown origin. Therefore, resilience in pigs remainsimportant. The probability of the occurrence of an unrecorded environmental challenge at a given day was estimatedfrom daily feed intake (DFI) data, and the genetic determinism of resilience to such events was evaluated.In total, 186,242 DFI records recorded automatically from 2,517 Large White males were used. The pedigreeinvolved 5,649 individuals. For each day (within each batch), the probability of the occurrence of a challenge wascomputed via a mixture model. As a result, unrecorded events of environmental challenges affecting pigs’ DFIwere identified. Days with a high probability of having a high coefficient of variation (CV) were related to the occurrenceof an environmental challenge. The probabilities (p) of high CV (stressful day) were used as a covariatein a reaction norm model (RNM) as an environmental descriptor, ranging from a non-challenging environment(p=0) to a challenging one (p=1). Variance components were estimated using the average information restrictedmaximum likelihood method. The mean probability of being a day with a high CV was 0.10; thus most of thedays have a low probability of the occurrence of a challenge. The genetic correlation between level and slopewas 0.06±0.13 in RNM. This value suggests that selecting for decreased DFI would have a minimal effect on theanimal’s sensitivity to challenging environmental conditions.Sessio
Conséquences d’une sélection divergente sur des critères de sociabilité sur la communication des émotions chez les ovins
International audienceLes signaux vocaux encodent notamment les émotions. Leur perception par lescongénères jouerait un rôle important dans la coordination et la coopération des groupessociaux, et expliquerait une part de la sélection de ces signaux au cours de l’évolution. Lesovins sont une espèce grégaire, chez laquelle l’héritabilité des caractères sociaux permet unesélection divergente sur la réactivité à l’isolement social. Cette sélection est entreprise depuisune dizaine d’années sur des ovins de race Romane. Nous utilisons deux lignées divergentessur la réactivité à l’isolement social pour tester les conséquences de la sélection sur lacommunication vocale. Nous faisons l’hypothèse que lors d’un isolement social, les agnellesde la lignée fortement sociable sont plus stressées que celle de la lignée moins sociable. Cestress serait quantifiable dans la structure acoustique des bêlements et perçu différemment pardes agnelles plus ou moins sociables. L’analyse spectro-temporelle de 1240 bêlementsenregistrés sur 131 agnelles exposées à un isolement social a révélé des différences dedistribution fréquentielle et de formants entre les deux lignées divergentes. Une expérience deplayback a permis de tester si ces différences acoustiques sont perçues par les agnelles et si uneéventuelle contagion émotionnelle peut s’observer selon les caractéristiques des bêlementset/ou la lignée. Le changement de comportement et les variations temporelles de la températurefaciale en réponse au playback ont été quantifiés. Cette étude ouvre la discussion sur les liensentre sociabilité et communication vocale d’émotions
The genetic architecture of recombination rates is polygenic and differs between the sexes in wild house sparrows ( Passer domesticus )
International audienceMeiotic recombination through chromosomal crossing-over is a fundamental feature of sex and an important driver of genomic diversity. It ensures proper disjunction, allows increased selection responses, and prevents mutation accumulation; however, it is also mutagenic and can break up favourable haplotypes. This cost/benefit dynamic is likely to vary depending on mechanistic and evolutionary contexts, and indeed, recombination rates show huge variation in nature. Identifying the genetic architecture of this variation is key to understanding its causes and consequences. Here, we investigate individual recombination rate variation in wild house sparrows (Passer domesticus). We integrate genomic and pedigree data to identify autosomal crossover counts (ACC) and intra-chromosomal allelic shuffling (r¯intra) in 13,056 gametes transmitted from 2,653 individuals to their offspring. Females had 1.37 times higher ACC, and 1.55 times higher r¯intra than males. ACC and r¯intra = 0.23 and 0.11; r¯intra h2 = 0.12 and 0.14), but cross-sex additive genetic correlations were low (rA = 0.29 and 0.32 for ACC and r¯intra). Conditional bivariate analyses showed that all measures remained heritable after accounting for genetic values in the opposite sex, indicating that sex-specific ACC and r¯intra can evolve somewhat independently. Genome-wide models showed that ACC and r¯intra are polygenic and driven by many small-effect loci, many of which are likely to act in trans as global recombination modifiers. Our findings show that recombination rates of females and males can have different evolutionary potential in wild birds, providing a compelling mechanism for the evolution of sexual dimorphism in recombination
Création de supports pédagogiques destinés à favoriser l’apprentissage de l’échographie abdominale des carnivores domestiques
Ultrasound, commonly used by practicing veterinarians, has become an indispensable diagnostic tool. It requires a sound knowledge of topographical anatomy, as well as a considerable know-how, acquired with practice. As part of a project to improve teaching methods used in the initial training of veterinary students, this work proposes to create a variety of teaching aids designed to help students learn how to perform ultrasound in domestic carnivores. An abdominal organ projection model was created to illustrate the ultrasound accessibility of these organs. Posters of abdominal organs were then displayed in an ultrasound training room. They illustrate the main anatomical points, the examination method, and normal ultrasound images. Finally, an ultrasound-guided cystocentesis workshop was set up to promote the acquisition of manual skills.L’échographie, couramment employée par les vétérinaires praticiens, est devenue un outil de diagnostic indispensable. Elle nécessite une bonne connaissance de l’anatomie topographique mais également un savoir-faire important qui s’acquiert avec la pratique. Dans un contexte d’amélioration des dispositifs pédagogiques mis en œuvre dans la formation initiale des étudiants vétérinaires, ce travail propose de créer différents supports destinés à favoriser l’apprentissage de l’échographie des carnivores domestiques. Un mannequin de projection des organes abdominaux a été créé afin d’illustrer l’accessibilité échographique de ces organes. Puis, des posters relatifs aux organes abdominaux ont été affichés dans une salle d’apprentissage de l’échographie. Ils illustrent les principaux rappels anatomiques, la méthode d’examen ainsi que des images échographiques normales. Enfin, un atelier de cystocentèse échoguidée a été mis en place afin de promouvoir l’acquisition d’une habileté manuelle

Ces substances sont inscrites sur le plan d’action continu communautaire (CoRAP2 ), publié3 sur le site internet de l’Agence européenne des produits chimiques (ECHA) avec une courte description des préoccupations initialement identifiées pour chacune des substances. La proportion de ces deux isomères n'était pas renseignée. En 2016, l'ECHA a envoyé au déclarant une décision relative à la vérification de la conformité au titre du règlement REACH afin de demander une clarification de l'identité de la substance. En décembre 2021, le déclarant principal a mis à jour son dossier d'enregistrement avec de nouvelles informations. L’identité de la substance a été établie comme correspondant à l'isomère 3-BHA.Dans la littérature scientifique, la substance testée est rapportée principalement en tant que BHA, sans information sur le ratio d’isomères.Le BHA avait été sélectionné à l'origine afin de clarifier les préoccupations concernant ses possibles propriétés de toxicité pour la reproduction et de perturbation endocrinienne. La substance présente également des « usages dispersifs ». Les consommateurs et les populations sensibles pourraient être fortement exposés. Au cours de l'évaluation, d'autres préoccupations ont également été identifiées à savoir de possibles propriétés cancérogènes et mutagènes. Le 3-BHA est mis sur le marché à un tonnage compris entre 10 et 100 tonnes selon les informations actuellement disponibles dans le dossier d’enregistrement auprès de l’ECHA.Cette évaluation couvre à la fois le BHA et le 3-BHA
