    Do Digestive Bacteria Suffer from Oxidative Stress: A Study on Ruminal Bacteria

    International audienceBackground: Heating oils and oilseeds results in oxidized fatty acids with potential antimicrobial properties that could affect digestive microbiota, and therefore health. Objectives:The objectives of this study were to explore the effect of lipid oxidation products on digestive bacteria using rumen content culture.Methods: A long-term in vitro study was conducted to check the effects of one hydroperoxide, 13OOH cis-9,trans-11-C18:2 (13HPOD), and two aldehydes, hexanal and trans-2,trans-4-decadienal (T2T4D), on rumen bacteria and superoxide dismutase (SOD) activity. Rumen fluid was incubated with one of the three oxidation products for 54 h or 102 h. Controls without oxidation products were also incubated. Results:The bacterial community was highly affected by T2T4D, in particular the Bacteroidetes/Firmicutes ratio and the relative abundance of the Bacteroidales S24-7 group family, leading to an alteration of fermentation responsible for a higher final pH than those in control cultures, and a diminution of SOD activity. Other oxidative products did not affect SOD activity, as well as hexanal did not modify the bacterial community compared to that of the control. Conclusion:The modification of the ruminal bacterial community by T2T4D represents a potential risk for the health of its host. Oxidative stress needs to be investigated in digestive ecosystems.</div

    Technical, societal and ethical feasibility of epigenetic editing

    Epigenome editing is a set of molecular approaches that aims at modifying one or several epigenetics marks in one or a few target genomic regions. It can be used to decipher the causal regulatory mechanisms governing gene expression, to validate putative distal regulatory elements, and to modify gene expression levels. The long term repression or activation of a target gene of interest without altering the DNA sequence of a cell constitutes one of the most promising applications of epigenome editing, including in farm animals. Here we present the main molecular tools currently used to perform epigenome editing in vitro or in vivo: the guidance systems (including CRISPR based), the epigenetic effector domains in use, and the delivery method available in vitro or in vivo. A list of studies reporting in vivo epigenome editing in animals (mostly in mice) is proposed, highlighting methodological choices that have been made by their authors. For example, some approaches require an alteration of the host DNA sequence (i.e. using lentiviral infection), while others let the host genome intact. Some approaches have effects across the whole organism, and even putative inter-generational effects, while other approaches target a single somatic organ. Finally, we list potential applications in farm animal species, ranging from research to commercial solutions. We discuss the implications of the different methodological options, including in terms of economic feasibility and public acceptability

    L'ingestion, un critère pertinent de détection du stress thermique chez les chèvres en élevage

    National audienceDes distributeurs automatiques d’aliments (concentré, fourrage et eau) dédiés aux petits ruminants ont été développés puis déployés à l’UE P3R (Unité Expérimentale Pôle de Phénotypage des Petits Ruminants) pour répondre à des besoins scientifiques au sein du département Génétique Animale d’INRAE. Ces distributeurs permettent d’étudier de nouveaux caractères d’intérêt à l’échelle individuelle tels que l’efficacité et le comportement alimentaires. Associés une interface web, ils facilitent également le suivi de l’ingestion et de la croissance des animaux par les utilisateurs et renforcent ainsi leur surveillance. Les données sont facilement accessibles sous la forme de tableaux et de graphes, à différentes échelles (par visite, jour, distributeur, animal, lot, etc.) pour le suivi quotidien de l’élevage et du protocole expérimental. Des alertes paramétrables sont également disponibles pour avertir d’un disfonctionnement du matériel et offrent des perspectives en termes de diagnostic précoce de problèmes sanitaires et/ou cliniques sur un animal.Dans le cadre du projet européen SMARTER, 41 chèvres de race Alpine ont été conduites dans la halle de phénotypage de l’UE P3R au cours de l’année 2023, en deux lots. Chaque lot était équipé d’un distributeur de concentré, de trois distributeurs de fourrage et d’un distributeur d’eau permettant également de peser l’animal. A partir des données d’ingestion et de comportement alimentaire collectées au cours de la lactation, plusieurs variables journalières, disponibles sur l’interface web, ont été calculées pour chaque aliment telles que le nombre moyen de visites par jour, la consommation moyenne à la visite, la consommation totale journalière, la durée totale des visites par jour et la vitesse d’ingestion moyenne journalière. Des relevés de température et d’humidité ont également été réalisés par des capteurs de type LoRa© toutes les 10 minutes de façon à calculer un indicateur de confort thermique moyen journalier (THI) selon la formule de Kendall &amp; Webster. En mai 2023, en milieu de lactation, le THI a dépassé le seuil de stress léger et modéré (&gt; 70) à cause de températures élevées ce qui s’est traduit par une augmentation des consommations journalières d’eau (+ 3,2 L en moyenne). A posteriori, des analyses statistiques ont permis d’estimer une corrélation égale à 0,86 (P &lt; 0,0001) sur cette période entre la consommation journalière d’eau et le THI moyen journalier, indépendamment du niveau de besoin lié à la production laitière. Ce résultat indique que la consommation d’eau, quand elle est mesurée quotidiennement, peut témoigner de perturbations environnementales tel que le stress thermique

    International audienceWe have previously demonstrated that protein-O-mannosylation (POM), a widespread post-translational glycosyl modification of proteins, is a key virulence factor of Mycobacterium tuberculosis (Mtb), the world’s deadliest infectious agent. Here, we report a detailed analysis of the structure-function relationship of MtPMT, the enzyme that catalyzes POM in Mtb. Using mutagenesis and in cellulo monitoring of POM activity, we demonstrate that, despite notable structural differences, MtPMT shares functional homologies with yeasts’ PMTs in the mechanism of the sugar transfer from lipidic donors. Furthermore, we provide evidence that the selectivity for proline-rich target glycosylation sites that differentiates MtPMT from its eukaryotic homologues, relies on a WW-like domain, which preferentially interacts with proline-rich acceptor substrate analogues. This first identification of a functional WW-like domain in a prokaryotic protein raises questions about its potential evolutionary linkage with eukaryotic WW modules and provides new insights into PMT’s acceptor-substrate recognition mechanism paving the way for the development selective inhibitors of MtPMT with potential therapeutic application against tuberculosis

    Comparaison de potentiels biomarqueurs de vieillissement entre des chiens jeunes et séniors

    Aging in dogs is associated with progressive changes in physiological parameters, which depend on age and size. The study focuses on 106 healthy large-breed dogs, either young adult (2-5 years) or seniors (&gt;8 years), all fed a standardized diet, and aims to evaluate age-related effects through clinical, biochemical, hematological and oxidative stress parameters. Fourteen parameters showed significant differences between the two groups: heart rate, MCS, and coat score were higher in senior dogs. Albumin, the albumin/ globulin ratio, ASAT, phosphorus, and white blood cells (lymphocytes, monocytes, basophils) were lower in seniors dogs. Globulins, triglycerides and platelets were higher. These findings suggest that such parameters could serve as relevant biomarkers during geriatric consultations.Le vieillissement du chien s’accompagne de modifications sur les paramètres physiologiques, variables selon la taille et l’âge du chien. Ce travail porte sur 106 chiens sains de grande et moyenne taille, jeunes adultes (2-5 ans) ou séniors (&gt; 8 ans), nourris avec une alimentation standardisée, et vise à évaluer les effets du vieillissement en s’intéressant à des paramètres cliniques, biochimiques, hématologiques et liés au stress oxydant. 14 paramètres montrent des différences significatives entre les chiens jeunes et séniors : la fréquence cardiaque, le MCS et le score du pelage sont plus haut chez les chiens séniors par rapport aux jeunes. L’albumine, le rapport albumine/globuline, les ASAT, le phosphore, les eucocytes (lymphocytes, monocytes et basophiles) sont plus bas chez les chiens séniors tandis que les globulines, les triglycérides et les plaquettes sont plus élevés. Ces paramètres pourraient constituer des biomarqueurs intéressants à évaluer en consultation gériatrique

    Choc anaphylactique chez les bovins‎ : étiologie et mécanismes physiopathologiques

    Between 2020 and 2023, the prevalence of anaphylactic manifestations in cattle increased abruptly across multiple European countries, with France being particularly affected. The unusually high frequency of these reactions within groups of animals managed in cohorts, combined with the severity of the clinical signs observed, prompted the initiation of epidemiological investigations. However, these studies did not succeed in identifying a common underlying cause. The involvement of povidone - a compound present in several veterinary pharmaceutical for-mulations - was considered, although no definitive causal relationship could be established. The objective of this study is to review the known mechanisms underlying anaphylactic reactions in order to assess the potential role of povidone in the emergence of these atypical hypersensitivity events. Several lines of evidence suggest that pov-idone may contribute to the development of allergic responses in cattle, thereby offering a plausible explanation for this phenomenon.La prévalence des manifestations anaphylactiques a soudainement augmenté dans l’espèce bovine entre 2020 et 2023 simultanément dans plusieurs pays d’Europe, et plus particulièrement en France. La fréquence anormalement élevée au sein de groupes d’animaux élevés en lots et la sévérité des manifestations cliniques ont motivé la réalisation d’enquêtes épidémiologiques qui n’ont pu conclure sur l’existence d’une cause commune sous-jacente de ce phénomène. Le rôle de la povidone, qui est présente dans plusieurs spécialités médicamenteuses, a été envisagé sans qu’un lien formel puisse être établi. L’objet de ce travail est de recenser les mécanismes associés à l’apparition d’une réaction anaphylactique afin d’investiguer l’implication possible de cette molécule dans l’apparition de ces réactions anormales. Plusieurs observations sont en faveur de la participation de la povidone au développement d’une réaction chez les bovins comme élément explicatif de ce phénomène allergique

    Mechanisms underlying alterations of the gut microbiota by exercise and their role in shaping ecological resilience

    International audienceThe gut microbiota (GM) is a dynamic ecosystem intricately linked to human health, including metabolic, immune, endocrine, and gastrointestinal functions. Exercise is recognized as a significant modifier of this microbial ecosystem, yet the complexities of this relationship are underexplored. Here, we delve into the multifaceted interactions between structured physical activity and the GM, emphasizing the role of exercise-induced stressors in shaping microbial composition and function. Unique to our review, we discuss the acute effects of different forms of exercise-induced stress on the GM and explore how these responses may influence long-term adaptability, stability, and resilience. Furthermore, we address critical junctures in microbial dynamics leading to shifts between different stable states. Finally, we explore the implications of host-controlled factors such as diet, exercise training, and nutritional supplementation in modulating the microbial community in the gut to optimize athletic performance. We conclude that while the potential to harness the synergistic effects of exercise-induced stressors, dietary interventions, and microbial adaptations appears promising, current evidence remains preliminary, highlighting the need for additional targeted research to guide future strategies that manipulate the GM for optimal health and athletic performance
