The Effect of Volume of the Soil Raising Chinese Cabbage on the 
Severity of the Soft Rot Disease

Summary 
The present study was carried out to find out the effect of volume of soil raising Chinese cabbage on 
the occurrence and severity of the soft rot disease. The plants (cv. Matsushima-kohai Shin No. 6) were 
raised in the clay pots of No. 10, No.13, No. 15 and No. 18 twice a year (spring sowing and summer sowing) 
in 197 8. The occurrence, the severity of the soft rot disease and the multiplication of soft rot bacteria in 
rhizosphere soil of the plants were examined periodically. The growth of the plants and the severity of the 
disease varied with the size of a pot in which the plants were raised. They were reduced as the pot became 
smaller in size. On the contrary, the multiplication of soft rot bacteria in rhizosphere soil occurred in all 
pots. In addition to the multiplication of soft rot bacteria, the occurrence and sev:erity of the disease were 
found to be also governed by the growing conditions of the plants influenced by volume of the soil in which 
they were raised. 
I . Introduction 
The occurrence and severity of the soft rot disease 
of Chinese cabbage (Brassica campestris L. (pekinensis 
group)), cited by Erwinia carotovora subsp. carotovora were 
confirmed to be largely influenced by the growing stage 
and the sowing date of the plant1-4.5>. Namely, the occurr-
ence of the disease is restricted to the late wrapping 
stage of 45-50 days after sowing">. In summer sowing, 
the severity of the disease depends upon the sowing date 
: the earlier the sowing date is, the heavier the damage 
is1•7•8>. Furthermore, it was observed that the severity 
was mild in the plants grown in pots and was not so se-
rious as occurred in fields. Consequently the severity 
seems to be concerned with volume of the soil raising the 
plant. 
In order to examine the effect of volume of soil on the 
severity of the soft rot disease of Chinese cabbage, the 
plant was raised in the pots with different sizes and the 
severity of the disease was compared among the plants.

I . Materials and Methods 
The present study was carried out on our university 
farm. Four clay pots with different sizes, No. 10, No. 13, 
No.15 and No. 18 were u\>ed. After the pots were filled 
with the soil obtained on the field of our university farm, 
they were hurried near the soil level. The soil of the 
field was clay loam and the soil pH value was 6.20. In 
the field Chinese cabbage was continuously raised. 
Chinese cabbage (cv. Matsushima-kohai Shin No. 6) was 
sown on April 27 (spring sowing) and August 8 (summer 
sowing) in 1978. The plants were suitably thinned out 
and finally one plant per pot was raised. They were fer-
tilized as reported in the previous paper4> . The plants 
raised in field were used as control. Soft rot bacteria in 
rhizosphere soil of the plants were counted by dilution 
plating method with the modified Drigalski's medium•>. 
Disease indexes were calculated at intervals of a week as 
described by Shimizu et at3>. 
][. Results 
1. Spring sowing 
The growth of Chinese cabbage and the severity of the

soft rot disease varied considerably with the size of a pot. 
They declined with a decrease of the size of a pot. The 
vigorus growth and excellent wrapping were obtained in 
No.18 and in control (Table 1). For example, in control, 
Number of countable leaves was 56; and plant weight 
was about 2.0 kg in June 29. Moreover soft rot occurred 
at the beginning of June and developed rapidly from the 
end of June to July (Fig. l). Disease indexes were 90-100 
at the middle of July. In No.18 and No.15, the disease 
occurred as serious as control. However, the growth and 
the wrapping were insufficient in No.10 and No.13. 
Though the disease occurred, the severity was found to 
be slight. Survival rates ( % ) of the plant by the disease

were examined as a preliminary study in 1977 (Fig. 2). 
The results showed that the survival rates were very 
different among the pots. 
As shown in Table 1, soft rot bacteria multiplied ex-
clusively in the rhizosphere soils. at wrapping stage of 
45-50 days after sowing and amounted to the level of 
104-lO'cfu/g of the oven dried soil regradless of the size 
of pots. 
2. Summer sowing 
As spring sowing, the growth of the plant and the 
severity of the disease varied with the size of pots. They 
were reduced as the pot became smaller in size. The 
plants grew vigrously and wrapped in control and in No.

18. For example. Number of countable leaves was about 
64 and plant weight was 3.9 kg in October 11 in control 
(Table 2). The disease occurred at the late wrapping 
stage. Disease indexes were approximately 90-100 at 
October 11 to 17 (Fig. 3). The poor growth and insuffi-

cient wrapping were obtained in No.10 and in No. 13. 
Disease indexes of the plant approximated 32.3 and 4 7.5 
in October 11 respectively. Soft rot bacteria multiplied 
exclusively in rhizosphere soils of the plants in all pots. 
N. Discussion 
It was widely accepted that the severity of the soft rot

of Chinese cabbage was mild when the plant was raised 
in pots. The fact seems to indicate that the volume of soil 
raising the plant is a factor which determines the sever-
ity of the disease. In the present study, Chinese cabbage 
was raised in the pots with different sizes and the effect 
of the volume of soil on the severity of the soft rot dis-
ease was investigated. 
When the plant was raised in the pots of No. 15 and 
No. 18, it grew vigorously and the disease occurred as 
severe as the one grown in field (control). However, in 
the plants raised in the pots of No. 10 and No. 13, the 
growth and the severity were reduced. Therefore they 
were largely concerned with the size of a pot, in other 
words, the volume of soils which the plants were raised. 
On the other hand, soft rot bacteria multiplied in rhi-
zosphere soils of the plants in all pots from the late 
wrapping stage as reported in the previous paper3•6) . 
From the present results, it can be said that the multi-
plication of the soft rot organism (establshment of the 
inoculum)'> does not always lead to the occurrence of the 
disease. 
Multiplication of the organism was found to play an 
imortant role for the occurrence of the disease2•4) . In 
addition to the multiplication2 •4 •6), it was also found that 
the growing conditions of the plants caused by different 
volume of soil influenced the occurrence and the severity 
of the disease. In future, it could be possible to prevent 
the disease by controling effectively the growing condi-
tions of the plant through the changing volume of soil.

ハクサイの生育する土壌の容積が軟腐病の発生に及ぽす影響

摘要
大きさの異なる素焼の植木鉢にハクサイを栽培し，軟腐病の発生に及ぽす土壌の容積の影響を調べた． 1978年4月27
日および8月8日にハクサイ（松島交配新六号）を10号， 13号， 15号および18号の素焼鉢に播種し，その生育，軟腐病
の発病および根圏士壌における軟腐病菌の増殖等を調べ，比較した．この結果，鉢が小さくなるに従って生育が不良と
なり，軟腐病の発病指数も小さくなる傾向がみられた．すなわち， 10号の鉢では株当り約 1kgの重量で軟腐病の発病
程度も軽症であった． しかし， 18号の鉢では対照区のほ場に栽培したハクサイとほぽ同程度に生育した．軟腐病も同様
に発生し， 90~100の発病指数に達した．一方，根圏土壌における軟腐病菌の増殖は鉢の大きさに関係なくおこり， 4番
種後40~50日に全鉢で104~106の菌数（乾土 lgあたり）に達した．これらのことから，ハクサイ軟腐病の発病や被害
程度は根圏士壌における病原菌の増殖に加え，ハクサイが栽培される植木鉢の大きさ，換言すれば土壌の容積に左右さ
れる地上部の生育状態にも依存していることが示された．

Summary 
A typical yellowing and wilting in gyojaninniku (Allium victorialis L. subsp. platyphyllum Hulten) 
grown in the experimental field of Faculty of Agriculture, Yamagata University occurred in June, 1988. 
White and silky mycelia grew luxuriantly on the diseased leaf sheath at the basal stem and produced 
many light brown mustard seed-like sclerotia on the lesions and on the soil surrounding the diseased 
plant. Consequently, the diseased plant wilted and fel down. The inoculation test using the fungus iso-
lated from diseased plant showed the same symptom as the natural occurrence under field condition. 
From the symptom and sign appeared on the diseased plant, characteristics of the colony and scler-
otium and the pathogenicity test of the isolated fungus, the pathogen of the present disease was identi-
fied as Corticium rolfsi. Therefore, the present disease was diagnosed as southern blight of gyOjaninn1-
ku caused by the fungus. 
This is the first report on southern blight of gyOjaninniku (Allium victorialis L. subsp. platyphyllum 
Hulten) in Japan.

緒 言
1988年6月，山形大学農学部学内圃場において生理
学的研究のため試験的に栽培しているギョウジャニン
ニク (Alliumvictoriales L. subsp. platyphyllum Hulten) 
に黄化および萎凋症状を示す原因不明の病害が発生し
た．そこで本病害の診断を行った結果，白絹病菌
(Corticium rolfsii)による白絹病であることが判明し
た．ギョウジャニンニクの白絹病はこれまで本邦では
未記載であるので，その概要を報告する．

材料および方法
ギョウジャニンニクに発生した病害の病微観察
1988年山形大学農学部学内圃場に栽培されているギョ
ウジャニンニクに発生した原因不明の病害について，
病徴および標徴等の観察を行った．
病原菌の分離および培養試験 罹病部の病斑より組
織分離法により病原菌の分離を行い，また標徴部から
も病原菌の分離を行った．すなわち，各々病斑部およ
び標徴部をとり， 70％エタノール，次いで20％次亜塩
素酸ナトリウム溶液で表面殺菌し， PSA(potato sue-
rose agar)培地に置床し， 25℃で培養した．さらに，
分離菌の形態的な特徴および生理的性質を調べる目的
で， 25℃，暗黒下および光照射下で培養した．
分離菌株の病原性の検定接種試験は飯田，渡辺の

方法により以下のように行った151．すなわち， 1989
年10月，直径約 18cmのプラスチック製ポットにギョ
ウジャニンニクの株を移植した同年11月にこれら移
植株の地際部に，あらかじめ PSA培地で培養した分
離菌株の菌叢および菌核を寒天培地ごと静置し，軽く
覆土して接種した．その後，ポットは圃場の木陰に静
置し，発病調査を行った．
結果および考察
ギョウジャニンニクはユリ科ネギ属の多年草で，奈
良県以北の山地や低地の林に生育し，最近山菜として
注目されその活用が有望視されている見本学部の学
内圃場で生理学的研究のため試験的に栽培されていた
が，栽培数年目に黄化，萎凋して，ついには枯死，倒
伏する個体が多く見られた．病徴を観察すると，はじ
め葉身が黄化し， しだいに株全体に進展した．その後
葉鞘の地際部は黒変して壊死し，その部分を中心とし
て土壌の表面に白色の菌糸が認められ，さらに白色か
ら淡褐色のケシ粒状の菌核が多数形成された（図版ー
1). 病勢が進むとやがて萎凋し，最後には株全体が
枯死し，倒伏したこのような罹病植物の葉鞘の病斑
部を採取し，表面殺菌したのち PSA培地上で培養し
たところ，白色で絹糸状の菌糸からなる特徴的な糸状
菌の菌叢が形成された（図版ー 2). さらに培養を続け
ると，菌叢上の，特にペトリ皿の縁部に，白色から淡
褐色の球形の菌核が形成された（図版ー 3). この菌核
形成は，暗黒条件下より光照射下の方が良好で，光照
射下では菌核直径は小さくなる傾向が見られたものの
菌核数は明らかに増加した．これらの分離菌を用いた
接種試験の結果，翌年の高温多湿時である 6月頃に，
接種植物の葉鞘地際部に白色の菌糸が生育し，同時に
微小な菌核が形成された．一方，ギョウジャニンニク
の生育は不良となり，最後には株全体が枯死し，倒伏
した（図版ー 4). このように枯死した茎葉部およびそ
の周辺部にも菌糸がさらに旺盛に生育し，同時に淡褐
色ケシ粒状の茜核が多数形成され，圃場での自然発病
と同一の症状が再現された．
上記のように，病徴および標徴の特徴（図版ー 1)'
分離菌の培養的所見（図版ー 2, 3),菌核の形成およ
び接種試験の結果（図版— 3, 4) などから，本分離
菌は白絹病菌と同定され，本病を白絹病と診断した．
白絹病は多犯性の土壌伝染性病原菌2)で，わが国で
は66科251種の作物を侵害することが知られている見
これらの中でギョウジャニンニクと同じ Allium属の
ネギ，タマネギ等には白絹病の発生が報告されている
3)が，ギョウジャニンニクについては未だ知られてい
ない． しかし，今回の実験によりギョウジャニンニク
にも白絹病が発生することが確認されたので，本病を
ギョウジャニンニク白絹病と呼ぶことを提案したい．
摘 要
1988年6月，山形大学農学部学内圃場で試験的に栽
培されているギョウジャニンニク (AIlium victorialis 
L. subsp. platyphyllum Hulten)に黄化，萎凋を示す病
害が発生した．病徴と標徴の観察を行った結果，葉鞘
の地際部に白色で絹糸状の菌糸が生育し，白色から淡
褐色ケシ粒状の菌核が多数形成された．このため，罹
病植物は黄化，萎凋し，最終的には株全体が枯死し，
倒伏した．罹病植物の病斑から病原菌を分離し，それ
らの培養的性質菌核の形成および病原性の試験の結
果，白絹病菌（Corticiumrolfsii)と同定された．以上
のことから，本病はギョウジャニンニクの白絹病と診
断した．なお，ギョウジャニンニクの白絹病は本邦初
記載の病害である．

(1) 微生物とは
微生物という言葉は，肉眼ではその個体を確認でき
ない極く微小（数μmから100μ m程度）で多様な生
物を総称する用語で，通常，細菌，菌類（変形菌類，
カビ・きのこ・酵母類），単細胞性の藻類，原生動物
（繊毛虫類・鞭毛虫類・アメーバ等の単細胞の動物）
を指す．これらの微生物のうち，カビときのこ以外の
微生物の生存の単位（個体）は基本的に単細胞である．
また，細菌以外の微生物は全て核のある細胞を持って
いる真核生物である．しかし細菌は，その細胞内に核
膜で閉ざされた核がなく，核物質であるDNAが細胞
質中にむき出しになっているため，原核生物と呼ばれ
る．また，ウイルスを微生物の中に含めて考える場合
もあるが，ウイルスは，細胞構造を持たないため単独
では増殖できず，その増殖を細胞性の他の生物に依存
している点で，一般の生物とは異なる特殊な存在であ
る．
(2) 好気性生物と嫌気性生物
ビトを含め我々の知っている多細胞生物（動物・植
物を問わず）は，酸素（空気）がないと生存できない
好気性（酸素呼吸する）生物である．しかし，単細胞
の微生物の中には，酸素がなくても平気で生きていけ
るものがたくさんいて，これらの生物は“嫌気性’'生
物と呼ばれる．嫌気性生物は，酸素があってもなくて
も増殖できるもの（通性嫌気性）と，無酸素環境下で
のみ増殖でき，酸素があると死んでしまうもの（偏性
嫌気性）とに区別できる．清酒の製造等に使われるい
わゆる酵母は，酸素があれば酸素呼吸をするが，無酸
素環境下では糖を発酵してアルコールを生成すること
により増殖する．また，乳酸菌は無酸素環境下で乳酸
発酵を行う通性嫌気性の細菌である．酒類，味噌・醤
油，漬物，さらにヨーグルトやチーズ等の発酵食品は
これらの微生物の性質を利用して製造される．なお，
無酸素環境下でのみ増殖できる偏性嫌気性生物として
これまでに知られているものは細菌がほとんどである
が，そのほかにも原生動物や単細胞性の下等な菌類が
知られている．
微生物は，我々ピトの目では確認できない，ほんの
小さなスペースがあれば十分生存できる．微生物が棲
息できるそのような極く微小で，しかも無酸素の環境
は我々の周囲にいくらでも存在しており，嫌気性微生
物は我々の生活と深い関わりをもっている．
(3) ヒトのからだと微生物
ビトからだの，特に外界と接している場所には，い
ろいろな微生物が常在している．毛穴や頭皮，脇の下
等の皮膚にはたくさんの微生物が棲みつくか，あるい
は付着している．また，口の中は微生物にとっては格
好の棲息環境である．歯垢は食べ物の残りかすを餌に
増殖した細菌と，歯に付着しやすい唾液中の成分等の
有機物でできた塊で，歯垢 1g当たり 2千億個 (2X 
101個）もの細菌細胞がいる (70％が細菌体）とされ
ている．これが虫歯と歯槽膿漏の原因となる．これら
の細菌の中には偏性嫌気性細菌も多数含まれている．
口から取り入れられた食物は，食道，胃，小腸，大
腸と通って，最後に肛門から排泄される．消化管内は
ビトの“からだ”にとっては，食物が通過するいわば
外界である．食物が胃から小腸まで通過する間に， ヒ
トのからだの元来持っている消化酵素等による消化と
吸収が行われ，そして大腸（盲腸，結腸，直腸）では
未消化残涜から水分が吸収される．ヒトを含め，ほと
んど全ての多細胞動物は植物の繊維（セルロース）を
自前の消化力では全く消化できないため，食物繊維は
単なる残澄として大腸まで素通りしてしまう．
ヨーグルトや漬物，あるいは納豆など，生きている
微生物も一緒に食べることになる食品を除くと，我々
が日常摂取する食物は十分な洗浄や加熱調理を経てお
り，ほとんど無菌といっても良い状態なっていると考
えられる．しかし，上述したように口の中には多くの
細菌が棲息しており，また周囲の環境中から様々な微
生物が口の中に入り込み，これらは食物と一緒に消化
管に入って行く．しかし，ヒトの胃の中は胃酸の分泌
によって普通は強い酸性(pH1.0~3.0程度）になっ
ていて，これらの微生物もここで大半が死滅してしま
う．胃は外部からの侵入者に対するバリアーである．
しかし，小腸下部から大腸内のpHはほぽ中性に近く，
微生物が十分棲息できる．そして大腸内には内容物 l

g当たり 10°個から1炉個もの細菌が棲息しており，こ
れらの細菌は腸内細庶と呼ばれている．腸内細菌の主
なものは酸素があると全く増殖できない偏性嫌気性細
苗である．なお，腸内細菌と糞便中の細菌とはほぼ同
じであると考えられている（図 1).
大腸は常に一定温度であり，また食物残澄という餌
が一定間隔で供給され，一方老廃物は排泄されるため，
環境変化の非常に少ない，細菌にとっては非常に恵ま
れた安定した無酸素環境であり，嫌気性細菌が旺盛に
増殖できる．腸内細菌は食物残澄の他，腸から分泌さ
れた粘液物質や剥離細胞，細菌体等を餌として増殖す
る．細菌の中には宿主であるヒトのからだが全く消化
できなかった繊維を消化できるもの（セルロース分解
菌）もたくさんいる．これらの細菌は増殖に伴って主
に酢酸やプロピオン酸，酪酸といった脂肪酸，あるい
はメタンや二酸化炭素といったガスを生成する．この
うちガスは体外にそのまま排出されるが，脂肪酸は宿
主の体内に吸収され，宿主のエネルギー源になる．現
代日本人のように，消化の良い，栄養の豊富な食物を
ふんだんに口にすることのできる場合は，腸内細菌に
よるこのエネルギー供給はあまり重要ではない．しか
し，消化の悪い，繊維の多い食物しか摂取できないよ
うな場合は，腸内細菌による食物繊維等の分解によっ
て生成された発酵生成物が宿主に供給するエネルギー
源としての寄与の程度は，かなり大きいものと考えら
れる．また，腸内細菌はビタミン Bやビタミン Kを
生成する．さらに，腸内細菌は腸内における病原菌の
増殖を抑制し，また宿主の免疫力を高めることにより，
病原菌の感染を防御するという働きもする． しかし，
腸内細菌は腐敗産物や細菌毒素，さらに発癌性物質等
を生成する等，ビトの健康にとって有害な側面もある
ことが指摘されており，その二面性も十分に理解する
必要がある．
母親の胎内は無菌状態である．しかし，生まれ落ち
たとたん，ビトは微生物が周囲に無数に存在する環境
にさらされ， 1日もすればその腸内には様々な細菌が
棲息するようになり， 2日後にはほとんど大人と同じ
くらいの数の細菌が定着する．そして生まれて半年か
ら1年後には腸内細菌の種類組成はほぽ安定し，食物
や体調の変化，また老化等に伴い多少の変化をするも
のの，ヒトは一生を通じてはほぼ一定の種類の腸内細
菌と共に暮らすことになる（図 2).
(4) 反蜀動物のからだと微生物
ヒトは単胃動物であり，口から取り込まれた食物は
食道を通過して直接胃に送られる．しかし，ウシ， ビ
ッジ，ヤギといった反蜀動物は我々が持っている通常
の胃の前に 3個の胃を持っている．反蜀動物は本来草
食動物であり，反勢動物が食べた繊維の多い餌は，通
常の胃（第 4胃）に送られる前に，この 3個の胃を通
過することになる．反蜀動物の第一胃を）レーメン（ウ
シのルーメンの大きさは150~2501) というが，ここ
には 1g当たり 1010個から 101個の嫌気性細菌と 1g 
当たり 105個から1『個の，大きいもので200μ m程度
の嫌気性の原生動物が棲みついている．これらの）レー
メン微生物（特に細菌）の持つ性質の最も大きな特徴
は，動物が食べた草の中の，動物が自力では消化でき
ない繊維を消化できる（セルロース分解能を持つ）点
であり，動物によって摂取された繊維の70~80%は
ルーメン微生物によって消化されることが知られてい
る．）レーメン中で繊維は，草の中のタンパク質等の他
の成分と共に旺盛に消化され，ヒトの腸内と同じよう
に酢酸やプロピオン酸，酪酸といった脂肪酸に変換さ
れる．これらの脂肪酸はルーメン壁から宿主動物の体
内に吸収され，これが反蜀動物の主要なエネルギー源
となる．
原生動物は主にルーメン中で増殖した細菌を捕食し
て増殖する．原生動物は単細胞とはいえ，動物性のタ
ンパク質を持っており，宿主動物が摂取した草の中の
植物タンパク質は，ルーメン中で細菌を経て原生動物
に摂取されることにより，動物性のタンパク質に変換
されることになる．
）レーメン微生物が消化しきれなかった残涜，増殖し
た細菌と原生動物の細胞等は，単胃動物と同様に第 4
胃以下の消化管に送られ，ここでビトの消化管内と同
じような消化吸収を受ける．反焼動物は，自分のから
だの中に微生物の増殖しやすい環境を作ることによっ
て膨大な数の微生物を飼い，この微生物の力で自力で
は全く消化できない草の中の繊維を容易に消化させて
エネルギーを得，さらに細菌や原生動物の体という，
消化の良い栄養の豊富な食物に変化させている．肉，
ミルク，羊毛等，人類が反蜀動物から受けている恩恵
は計り知れないが，その恩恵も元をたどれば微生物に
行き着くのである．
(5) まとめ
ヒトや反蜀動物に限らず，全文の動物の消化管内に
はそれぞれに特有の微生物が棲息している．後生動物

（多細胞動物）はその出現の最初から微生物の存在と
共にあり，その数億年単位の進化の歴史と共に，それ
ぞれの動物に特有の消化管内微生物が発達してきたと
考えられる．動物のからだは個体毎にそれぞれが膨大
な数の腸内細菌を初めとする微生物を棲息させており，
それぞれの個体が，微生物に一つの独立した生態系を
提供している．最近，事ある毎に‘‘抗茜，抗菌”とい
う言葉が使われようになった．しかし，我々は無茜状
態で暮らすことは決してできない．一部の微生物の存
在のもつ病原性等のヒトにとってのリスクを上手に制
御しながら，微生物と仲良く共生し，むしろその大い
なる恩恵を享受するという考えが是非とも必要であろ
ぅ．
