    Metabolomic approach of the antiprotozoal activity of medicinal Piper species used in Peruvian Amazon

    International audienceEthnopharmacological relevance : In the Peruvian Amazon as in the tropical countries of South America, the use of medicinal Piper species (cordoncillos) is common practice, particularly against symptoms of infection by protozoal parasites. However, there is few documented information about the practical aspects of their use and few scientific validation. The starting point of this work was a set of interviews of people living in six rural communities from the Peruvian Amazon (Alto Amazonas Province) about their uses of plants from Piper genus: one community of Amerindian native people (Shawi community) and five communities of mestizos. Infections caused by parasitic protozoa take a huge toll on public health in the Amazonian communities, who partly fight it using traditional remedies. Validation of these traditional practices contributes to public health care efficiency and may help to identify new antiprotozoal compounds.Aims of study : To record and validate the use of medicinal Piper species by rural people of Alto Amazonas Province (Peru) and annotate active compounds using a correlation study and a data mining approach.Materials and methods : Rural communities were interviewed about traditional medication against parasite infections with medicinal Piper species. Ethnopharmacological surveys were undertaken in five mestizo villages, namely: Nueva Arica, Shucushuyacu, Parinari, Lagunas and Esperanza, and one Shawi community (Balsapuerto village). All communities belong to the Alto Amazonas Province (Loreto region, Peru). Seventeen Piper species were collected according to their traditional use for the treatment of parasitic diseases, 35 extracts (leaves or leaves and stems) were tested in vitro on P. falciparum (3D7 chloroquine-sensitive strain and W2 chloroquine-resistant strain), Leishmania donovani LV9 strain and Trypanosoma brucei gambiense. Assessments were performed on HUVEC cells and RAW 264.7 macrophages. The annotation of active compounds was realized by metabolomic analysis and molecular networking approach.Results : Nine extracts were active (IC50 ≤ 10 μg/mL) on 3D7 P. falciparum and only one on W2 P. falciparum, six on L. donovani (axenic and intramacrophagic amastigotes) and seven on Trypanosoma brucei gambiense. Only one extract was active on all three parasites (P. lineatum). After metabolomic analyses and annotation of compounds active on Leishmania, P. strigosum and P. pseudoarboreum were considered as potential sources of leishmanicidal compounds.Conclusions : This ethnopharmacological study and the associated in vitro bioassays corroborated the relevance of use of Piper species in the Amazonian traditional medicine, especially in Peru. A series of Piper species with few previously available phytochemical data have good antiprotozoal activity and could be a starting point for subsequent promising work. Metabolomic approach appears to be a smart, quick but still limited methodology to identify compounds with high probability of biological activity

    Water-Induced Damage in Microporous Carbonate Rock by Low-Pressure Injection Test

    International audienceIn the present work, we investigate the mechanical weakening and deformation induced by water on a microporous carbonate rock, the Obourg Chalk from Mons Basin (Belgium), through conventional triaxial tests and injection tests. The injection tests were conducted by waterflooding critically loaded rock samples, initially in dry condition, in a way to minimize the variations in the effective pressures. Furthermore, the samples were instrumented with P-wave piezoelectric transducers to provide active ultrasonic monitoring while injecting. The results show a significant reduction in the mechanical strength of this chalk. Analysis of the mechanical tests and the associated deformation allows us to describe the mechanical behavior as a function of the confining pressure, which draws a brittle-ductile transition spanning from low to high confining pressure. The injection tests, moreover, revealed that the amount of water injected before triggering mechanical instability decreases exponentially with respect to the applied differential stress on the rock sample. The data, therefore, suggest that the failure might be controlled by a mechanical coupling between the water-invaded zone and the dry one. Since water-weakening plays an important role in several fields like oil industry, through secondary and tertiary recovery of hydrocarbons, Enhanced Geothermal Systems (EGS), as well as in the mechanical stability of underground cavities, the outcome of this work is of primary importance in mitigating any kind of problems related to these operations

    An Urban Labor Market with Frictional Housing Markets: Theory and an Application to the Paris Urban Area

    International audienceWe build a tractable model of frictional labor markets and segmented housing markets to study welfare effects of regulations, including spatial misallocation and deviation from competitive pricing of rents. The model is summarized by a labor demand curve depending on rents and wages, a wage curve reflecting labor market tightness and rents, and finally a rent curve reflecting employment. In this economy, the rent gradient in the flexible rent sector is higher than in a purely competitive housing market. This leads to spatial misallocation due to some employees commuting too much and some non-employed living inefficiently close to jobs. In turn, reducing generalized commuting costs reduces the rent gradient in the flexible rent sector and the cost of spatial misallocation of workers. The reduction in market rents is maximal when labor markets are less frictional and housing markets are more frictional, and welfare gains are larger when both are more efficient

    Dynamical effects of the radiative stellar feedback on the H I-to-H2 transition

    International audienceContext. The atomic-to-molecular hydrogen (H/H2) transition has been extensively studied as it controls the fraction of gas in a molecular state in an interstellar cloud. This fraction is linked to star-formation by the Schmidt–Kennicutt law. While theoretical estimates of the column density of the H I layer have been proposed for static photodissociation regions (PDRs), Herschel and well-resolved Atacama Large Millimeter Array observations have revealed dynamical effects in star forming regions, caused by the process of photoevaporation.Aims. We extend the analytic study of the H/H2 transition to include the effects of the propagation of the ionization front, in particular in the presence of photoevaporation at the walls of blister H II regions, and we find its consequences on the total atomic hydrogen column density at the surface of clouds in the presence of an ultraviolet field, and on the properties of the H/H2 transition.Methods. We solved semi-analytically the differential equation giving the H2 column density profile by taking into account H2 formation on grains, H2 photodissociation, and the ionization front propagation dynamics modeled as advection of the gas through the ionization front.Results. Taking this advection into account reduces the width of the atomic region compared to static models. The atomic region may disappear if the ionization front velocity exceeds a certain value, leading the H/H2 transition and the ionization front to merge. For both dissociated and merged configurations, we provide analytical expressions to determine the total H I column density. Our results take the metallicity into account. Finally, we compared our results to observations of PDRs illuminated by O-stars, for which we conclude that the dynamical effects are strong, especially for low-excitation PDRs

    European first-year university students accept evolution but lack substantial knowledge about it: a standardized European cross-country assessment

    International audienceBackground: Investigations of evolution knowledge and acceptance and their relation are central to evolution education research. Ambiguous results in this field of study demonstrate a variety of measuring issues, for instance differently theorized constructs, or a lack of standardized methods, especially for crosscountry comparisons. In particular, meaningful comparisons across European countries, with their varying cultural backgrounds and education systems, are rare, often include only few countries, and lack standardization. To address these deficits, we conducted a standardized European survey, on 9200 first-year university students in 26 European countries utilizing a validated, comprehensive questionnaire, the "Evolution Education Questionnaire", to assess evolution acceptance and knowledge, as well as influencing factors on evolution acceptance.Results: We found that, despite European countries’ different cultural backgrounds and education systems, European first‑year university students generally accept evolution. At the same time, they lack substantial knowledge about it, even if they are enrolled in a biology‑related study program. Additionally, we developed a multilevel‑model that determines religious faith as the main influencing factor in accepting evolution. According to our model, knowledge about evolution and interest in biological topics also increase acceptance of evolution, but to a much lesser extent than religious faith. The effect of age and sex, as well as the country’s affiliation, students’ denomination, and whether or not a student is enrolled in a biology‑related university program, is negligible.Conclusions: Our findings indicate that, despite all their differences, most of the European education systems for upper secondary education lead to acceptance of evolution at least in university students. It appears that, at least in this sample, the differences in knowledge between countries reflect neither the extent to which school curricula cover evolutionary biology nor the percentage of biology‑related students in the country samples. Future studies should investigate the role of different European school curricula, identify particularly problematic or underrepresented evolutionary concepts in biology education, and analyze the role of religious faith when teaching evolution
