    Efficient Post-Shrinkage Estimation Strategies in High-Dimensional Cox’s Proportional Hazards Models

    Regularization methods such as LASSO, adaptive LASSO, Elastic-Net, and SCAD are widely employed for variable selection in statistical modeling. However, these methods primarily focus on variables with strong effects while often overlooking weaker signals, potentially leading to biased parameter estimates. To address this limitation, Gao, Ahmed, and Feng (2017) introduced a corrected shrinkage estimator that incorporates both weak and strong signals, though their results were confined to linear models. The applicability of such approaches to survival data remains unclear, despite the prevalence of survival regression involving both strong and weak effects in biomedical research. To bridge this gap, we propose a novel class of post-selection shrinkage estimators tailored to the Cox model framework. We establish the asymptotic properties of the proposed estimators and demonstrate their potential to enhance estimation and prediction accuracy through simulations that explicitly incorporate weak signals. Finally, we validate the practical utility of our approach by applying it to two real-world datasets, showcasing its advantages over existing methods

    Navigating fragmented services: a gender-based violence (GBV) critical feminist analysis of women’s experiences engaging with health and social supports in three Canadian cities

    Background: Gender-based violence (GBV) remains a pervasive public health crisis with devastating impacts on women’s health and well-being. Women experiencing GBV face considerable barriers accessing appropriate and timely health and social services. This study explored women’s experiences with health and social services in three Canadian cities to understand critical challenges and strengths in service provision for women experiencing GBV. Methods: In-depth interviews were conducted with self-identifying women (n = 21) who had accessed health or social care services and with service providers (n = 25) in three Canadian cities between February 2021 and November 2022. Women’s interviews focused on experiences engaging with services including what worked well, the challenges they faced, and their recommendations to enhance service delivery to women experiencing violence. Staff interviews focused on their experiences of providing services within their organization, and the strengths and challenges in providing services to women within their community. Data were analyzed using reflexive thematic analysis with a gender-based violence critical feminist lens. Results: We organized the findings into three interrelated themes. First our results show how the systems within which health and social services are organized, are not designed to meet women’s complex needs, with rigid structures, siloed services, and stigmatizing cultures creating significant barriers. Second, the data illustrate how service providers support and empower women through practices such as providing key information, assisting with administrative tasks, offering material resources, and addressing discrimination through advocacy and accompaniment. Third, our findings demonstrate how building an effective working relationship characterized by trust, non-judgment, and collaboration is crucial for service engagement and women’s overall well-being. Conclusions: Findings illuminate critical public health challenges as women navigate fragmented services across multiple and siloed systems not designed to meet their complex needs. There is an urgent need for systemic change to create more integrated, responsive support systems for women experiencing GBV. This includes addressing underlying structures perpetuating gender inequities and violence. Facilitating safe access to holistic services that consider women’s preferences is crucial. Effective working relationships built on trust, respect, and power-sharing are key to supporting women’s agency and addressing their interconnected needs

    An engineered redox‐switchable streptavidin mutein enables the high‐affinity capture and efficient elution of biotinylated ligands

    Although the high affinity and specificity of the interaction between streptavidin and biotin are widely utilized to isolate biotinylated ligands, their elution over practical timescales requires the use of strongly denaturing conditions that are incompatible with many target biomolecules. To overcome this limitation, we previously engineered a disulfide bond into a critical loop of streptavidin to create the redox‐sensitive M88 mutein, which releases biotinylated ligands ~19,000‐fold faster in the reduced state when compared with the oxidized state. To optimize the speed and efficiency of ligand recovery, we describe how multiple mechanisms like disulfide bond reduction, increasing temperature, elevating pH, and adding organic cosolvents can dramatically increase the rates of dissociation for different types of biotinylated ligands bound to M88 or wild‐type streptavidin coupled to magnetic beads. By combining these mechanisms, we demonstrate how a range of biotinylated biomolecules (peptide, oligonucleotide, and protein) can be efficiently recovered under mild conditions that preserve biological activity. M88 magnetic beads thus provide many advantages over currently available methods using either wild‐type streptavidin or lower affinity biotin‐binding proteins for applications requiring both high affinity capture and efficient release of target biomolecules under non‐denaturing conditions.Natural Sciences and Engineering Research Council of Canada (Award: 05287

    Development and analysis of microbridge resonators for reduced pull-in voltage and preserved resonant frequency

    This study presents a novel design methodology for microbridge resonators aimed at reducing pull-in voltage while maintaining resonant frequency. Previous studies have primarily focused on adjusting resonator plate geometry, modifying anchor conditions, or altering material properties to control pull-in voltage. In contrast, this work introduces the ratio of the bottom electrode length to the microbridge length as a critical and tunable design parameter, which has remained unexplored in previous studies. An analytical model is developed to capture the effects of this ratio, and its predictions are validated through finite element analysis. To demonstrate the concept, two microbridges are designed and fabricated with bottom electrode lengths of 42 and 82 μm, corresponding to 35 % and 68 % of the microbridge length, respectively. All other design parameters, such as plate thickness, material properties and cavity height, are kept constant to enable a fair comparison. Electrical characterizations confirm that increasing the bottom electrode-to-microbridge length ratio effectively lowers the pull-in voltage without degrading resonator performance. Results show a 16 % reduction in pull-in voltage when the bottom electrode length is 68 % of the microbridge length, demonstrating the feasibility and advantages of the proposed methodology over existing techniques

    Changing Emotion with Emotion: The Best Sequence Depends on the Target Concern

    Lingering anger and sadness about a interpersonal interaction is a common problem. However, resolving those feelings may depend on the sequence in which feelings are experienced. Method: Using 167 participants, two experimental groups were identified based on presenting emotional concern: individuals with predominantly lingering anger about an interpersonal interaction (i.e., angry group, n = 70), and individuals with predominantly lingering sadness about an interpersonal interaction (i.e., sad group, n = 97). Participants completed written interventions to facilitate anger and sadness in one of two randomly assigned conditions (i.e., anger-before-sadness condition or sadness-before-anger condition), which differed only by the order in which participants were guided to feel anger and sadness. Results: In the angry group, those guided to feel anger-before-sadness reported a greater decline in the intensity of their presenting anger than those guided to feel sadness-before-anger (d = − 0.56). In contrast, in the sad group, those guided to experience sadness-before-anger reported a greater decrease in lingering sadness than those guided to experience anger-before sadness (d = − 0.26). Conclusions: Strategically ordered sequence of emotion states seems to have a synergistic impact in facilitating change, which has implications for how therapists might best choose to guide client process in psychotherapy
