ENSO and the North American monsoon’s interannual variability
En este trabajo se actualizaron los índices que miden la intensidad de la etapa inicial del Monzón de América del Norte (MAN) para el periodo 1979 - 2019 y se evaluó la influencia de el fenómeno El Niño - Oscilación Sur (ENOS) sobre la etapa inicial y zona de influencia del MAN. Los datos utilizados se obtuvieron de los campos horarios de ERA5, el cual es la quinta generación de reanálisis de ECMWF, así como los campos diarios de CHIRPS un análisis quasi-global de precipitación. Para darle certidumbre estadística a los resultados, a los mapas de correlación entre variables relevantes se les aplicó el método propuesto por Livezey y Chen (1983) con el fin de determinar su significancia espacial. Se ha documentado que durante eventos El Niño (La Niña) la precipitación asociada al MAN es menor (mayor) que el promedio y la teleconexión más fuerte entre estos dos fenómenos ocurre precisamente durante la etapa inicial del MAN. Sin embargo, los resultados de esta tesis muestran que durante las últimas décadas la relación ENOS - MAN se ha debilitado, indicando que la influencia de ENOS sobre la etapa inicial del MAN ha disminuido. Las temperaturas superficiales en las regiones del océano Pacífico ecuatorial asociadas a ENOS han disminuido su correlación con la precipitación en la zona núcleo del MAN, de la misma forma que lo han hecho las alturas geopotenciales a 500 mb (Z500) en la región de la Zona de Convergencia Intertropical. Las anomalías estacionarias en Z500 asociadas a la fase positiva o negativa de ENOS que habían sido identificadas en la costa oeste de Estados Unidos y que presumiblemente podían interferir con la evolución del MAN actuando como un bloqueo al anticiclón de altura del monzón no han estado presentes durante las dos décadas recientes. A pesar de ello, el anticiclón de altura mantiene su trayectoria desde el suroeste en dirección noroeste (noreste) durante años La Niña (El Niño). A escala regional, se encontró que la temperatura superficial del mar del GoC se ha incrementado paulatinamente desde el inicio del periodo de estudio (1979). El calentamiento observado del GoC coincide en la escala temporal con la disminución de la relación entre el contraste térmico océano - continente (CTOC) y la convergencia de flujo de humedad, lo que implica que el CTOC ha disminuido su influencia como modulador del MAN. Además, se identificó una tendencia positiva del gradiente de presión superficial (∆P) entre el sur y el norte del GoC que comenzó en el año..In this work, indices that measure the intensity of the North American Monsoon (NAM) onset were updated to the period 1979 - 2019 and the influence of the El Niño - Southern Oscillation (ENSO) phenomenon on the NAM onset was evaluated. Data used were obtained from ERA5 hourly fields, which is the fifth generation ECMWF reanalysis and CHIRPS, a quasi-global precipitation analysis. To give statistical certainty to the correlation maps that were calculated between relevant variables, the methods proposed by Livezey y Chen (1983) were applied in order to determine their field significance. It has been documented that during El Niño (La Niña) events precipitation associated with NAM is lower (higher) than the average and the strongest teleconnection between these two phenomena occurs during the NAM onset. However, the results presented in this thesis show a decreasing link between the interannual variability of the NAM and ENSO during recent decades. Surface temperatures in the equatorial Pacific associated with ENSO have decreased their correlation with precipitation in the core NAM region, as have the 500 mb geopotential heights (Z500) of the Intertropical Convergence Zone region. Z500 anomalies associated with strong positive and negative ENSO events that had been identified over the western coast of the United States which presumably interfere with the evolution of the NAM by acting as a path obstruction to the upper level monsoon anticyclone have not been present during the last two decades. Despite this, the monsoon anticyclone keeps its displacement from the southwest to the northwest (northeast) during La Niña (El Niño) years. On the regional scale, it was found that the sea surface temperature of the GoC has increased gradually since the beginning of the study period (1979). The observed warming of the GoC coincides in timescale with a decrease in the link between the landsea thermal contrast (LSTC) and moisture flux convergence, which implies that LSTC has decreased its influence as a driver of NAM interannual variability. Furthermore, a positive trend of the surface pressure gradient (∆P) was identified between the south and north of the GoC that began in 2000. This increase coincides with a slight increase in near-surface wind speeds over the GoC. Precipitation analysis as a function of the sign and magnitude of ENSO in the NAM’s influence zone, showed that precipitation is no longer modulated by ENSO in the northwestern Mexico

La computación cuántica nace de la observación de Richard Feynman de la dificultad de simular sistemas cuánticos en la máquina RAM. El interés por este modelo de cómputo llegó con el algoritmo de Shor, donde la factorización de números, un problema en la intersección entre NP y co-NP, y del cual dependen muchos sistemas criptográficos, se pudo hacer en tiempo polinomial. Otra sorpresa fue el algoritmo de Grover, que permite encontrar un objeto en una colección en menos pasos que el tamaño de la colección; contrario incluso a la intuición natural. Con esta motivación, se aborda un problema que también requiere una cantidad lineal de pasos; pero en donde la condición de paro es más compleja que la condición de igualdad. En este trabajo se estudia el problema de jumbled pattern matching (JPM), donde dada una cadena w y un patrón p tales que w, p ∈ ∑ ∗ y |p| ≤ |w|, buscamos los sub-patrones de que estén construidos con los mismos elementos que p; es decir, todos los sub-patrones que sean una permutación de p. El JPM es especialmente útil en problemas de biocomputación relacionados con análisis de patrones, como espectrometría de masa para interpretación de datos, descubrimientos de SNP, alineamiento de secuencias, y aplicaciones de biosecuenciación. Explorando la computación cuántica, se presenta un algoritmo de complejidad O(√N) que genera la solución al problema de jumbled pattern matching casi seguramente; por tanto, acelerando las mejores soluciones clásicas en un factor cuadrático.Quantum computing was born from Richard Feynman’s observation of the difficulty of simulating quantum systems in the RAM machine. The interest in this computational model arose with the Shor’s algorithm, where the integer factorization, a problem in the intersection between NP and co-NP, on which many cryptography systems depend, it can be performed in polynomial time. Another surprise was the Grover’s algorithm, which allows finding an object among a collection in fewer steps than the size of the collection; contrary even to natural intuition. With this motivation we work on a problem that also requires a linear number of steps; but where the halting condition is more complex than the equality condition. In this work we approach the jumbled pattern matching problem, where given a text and a pattern p such that w, p ∈ ∑ ∗ and |p| ≤ |w|, we search for the sub-patterns in built with the same elements of p; that is, all sub-patterns that are a permutation of p. JPM is specially useful in bio computing problems related to pattern analysis, as mass spectrometry for data interpretation, SNP discovery, sequence alignment, and bio sequencing applications. Exploring quantum computing, we present an algorithm whose complexity is O(√ N) that generates the solution for jumbled pattern matching almost certainly; therefore, speeding-up the best classical solutions in a quadratic factor
Effect of nutrient availability on the phototrophic regulation of phytoplankton and bacterial with proteorhodopsins (PRs)
Las proteorodopsinas son proteínas transmembranales que capturan luz para generar energía bioquímica, la cual puede ser utilizada para crecimiento y supervivencia en bacterias marinas. Las mayores concentraciones de proteorodopsina se han observado generalmente en ambientes oligotróficos donde las concentraciones de clorofila-a y nutrientes inorgánicos son bajas. Sin embargo, en un estudio reciente en la zona de surgencia de la cuenca del Sur de California se observó que las máximas concentraciones de proteorodopsina coincidieron con concentraciones de clorofila-a altas durante las proliferaciones algales de primavera. Para comprender mejor la dinámica de comunidades fotótrofas, en este estudio investigamos las relaciones entre la disponibilidad de nutrientes, el crecimiento del fitoplancton y microorganismos con proteorodopsinas, y la estructura de las comunidades microbianas asociadas al desarrollo de proliferaciones algales en un ambiente controlado. Con ese objetivo se realizó un experimento de incubación de mesocosmos durante 20 días con agua de mar colectada en la cuenca del Sur de California, induciendo el crecimiento fitoplanctónico con la adición de nutrientes inorgánicos. Se cuantificó la clorofila-a, proteorodopsina, abundancia bacteriana y se secuenciaron amplicones de las regiones V4-V5 del gen 16S de ARNr. Las máximas concentraciones de clorofila-a se presentaron entre los 8 y 12 días después del inicio del experimento, mientras que los niveles más altos de proteorodopsina y abundancias bacterianas se presentaron entre los 14 y 20 días. Bacterias en los órdenes Flavobacteriales y Rhodobacterales dominaron la estructura de las comunidades bacterianas, sugiriendo la utilización de proteorodopsinas en miembros de estos grupos. Estos datos sugieren que, a diferencia de lo observado en sistemas oligotróficos, en sistemas productivos la asimilación activa de nutrientes y materia orgánica por parte del bacterioplancton está acoplada a la obtención de energía solar mediante proteorodopsinas.Proteorhodopsins are transmembrane proteins that capture light to generate biochemical energy, which can be used for growth and survival in marine bacteria. By quantifying retinal, as a proxy for this photosystem, the highest concentrations of proteorhodopsin have generally been observed in oligotrophic environments where the concentrations of chlorophyll-a and inorganic nutrients are low. However, in a recent study in the upwelling region of the Southern California basin, maximum proteorhodopsin concentrations coincided with those of chlorophyll-a during spring algal bloom seasons. To better understand the dynamics of phototrophic communities, in this study we investigated the relationships between the availability of nutrients, the growth of phytoplankton and microorganisms with proteorhodopsins, and the structure of the microbial communities associated with the development of algal blooms in a controlled environment. With this objective, we carried out a mesocosm incubation experiment for 20 days with seawater collected in the Southern California basin, inducing phytoplankton growth with the addition of inorganic nutrients. We quantified chlorophyll-a, proteorhodopsin and bacterial abundances, and also sequenced and analyzed amplicons of the V4-V5 regions of the 16S rRNA gene. The highest concentrations of chlorophyll-a occurred between 8 and 12 days after the start of the experiment, while the highest levels of proteorhodopsin and bacterial abundances were found between days 14 and 20. Bacteria in the orders Flavobacteriales and Rhodobacterales dominated the structure of bacterial communities, suggesting the use of proteorhodopsins in members of these groups. These data suggest that, unlike what is observed in oligotrophic systems, in productive systems the active assimilation of nutrients and organic matter by bacterioplankton is coupled with obtaining solar energy through proteorhodopsins
Nesting ecology of Lepidochelys olivacea in situ vs. ex situ in the protected natural area in Cabo Pulmo National Park, Baja California Sur, Mexico
El Parque Nacional Cabo Pulmo (PNCP), se encuentra en el límite septentrional de anidación para Lepidochelys olivacea en México, posee características ambientales únicas, poco habituales en comparación con las principales playas de anidación para esta especie. La anidación en tortugas marinas es una etapa crítica para el ciclo de vida, tanto para hembras como crías. El éxito reproductivo en estas especies está limitado a un rango de condiciones físicas y ambientales. No obstante, ha sido complicado conocer la influencia de factores ambientales sobre los parámetros reproductivos (eclosión, emergencia y mortalidad) en este sitio de importancia ecológica. En este trabajo se realizó un análisis espacio temporal de los parámetros reproductivos para la tortuga golfina en incubación ex situ e in situ, basado en 10 años de monitoreo (2009-2019) en cinco playas del PNCP. Se utilizaron modelos lineales generalizados (GLM) para determinar la influencia de las variables ambientales. Se encontró variación entre los años, pero no entre sitios. De acuerdo con los análisis bayesianos, el éxito reproductivo (eclosión y emergencia) en ambos tipos de incubación, no mostro diferencias estadísticas (BF10= 0.424 y BF10= 0.121, respectivamente), en contraste, la mortalidad presentó diferencias en el tipo de incubación (BF10= 21.759) para los años 2010, 2014 y 2018. Se obtuvieron seis GLM interpretables, uno para cada parámetro reproductivo por tipo de incubación, todos los modelos tuvieron un porcentaje de devianza explicada (D2) mayor a 50%. La variable ambiental más importante por su presencia en todos los modelos fue la irradiancia, seguida de las anomalías regionales de temperatura superficial del mar y anomalías regionales de temperatura ambiental, siendo todas estas significativas (P ≤ 0.05). Adicionalmente se analizó la mortalidad embrionaria, parámetro que, en general, no se reporta, pero que puede dar un panorama para entender las tendencias poblacionales en tortugas marinas. Hubo mayor tasa de mortalidad en la tercera fase de desarrollo embrionario siendo esté parámetro igual en ambas condiciones de incubación (BF10 ≤ 3). Con base en los resultados, se proponen recomendaciones para optimizar las estrategias de manejo, con el fin de brindar una mejor protección de las nidadas, ayudando a garantizar la estabilidad de la especie.Cabo Pulmo National Park (PNCP) is part of the olive ridley sea turtle sea turtle (Lepidochelys olivacea) nesting northern limit, and has unique environmental characteristics, different to those of the main nesting beaches for this species. Nesting stage of sea turtles is critical due to the imminent risks to which both females and hatchlings are exposed. Reproductive success in these species is limited to a range of physical and environmental conditions. However, it has been difficult to determine how environmental factors influence the reproductive parameters of the species in this site of great ecological importance. Based on 10 years of monitoring (2009-2019) in five beaches of Cabo Pulmo, a spatial and temporal analysis of the reproductive parameters of olive ridley was carried out in ex situ and in situ incubation sites. Generalized linear models (GLM) were used to determine the influence of environmental variables on reproductive parameters. Variation was found among years, but not among sites. According to the Bayesian analyzes, reproductive success (hatching and emergence) in both types of incubation did not show statistical differences (BF10= 0.424 and BF10= 0.121, respectively). Conversely, the mortality showed differences in the type of incubation (BF10= 21,759) in 2010, 2014, and 2018. Regarding environmental variables and reproductive parameters, six interpretable GLMs were obtained, one for each reproductive parameter by type of incubation, all the models had an explained deviance percentage (D2) greater than 50%. The most important environmental variable was irradiance, followed by regional anomalies of sea surface temperature and regional anomalies of environmental temperature, all of which were significant (P ≤ 0.05). Additionally, embryonic mortality which is a parameter that is seldom reported was analyzed, since can provide an overview of the population trends on sea turtles. We found high mortality rate in the third stage of embryonic development however, this parameter was the same in both incubation conditions (BF10 ≤ 3). Based on these results, some recommendations are proposed to optimize management strategies, and provide better protection of clutches, and help to guarantee the stability of the species
Reliability and Adaptive performance of multi-cloud storage with neural networks
El uso de los servicios en la nube se ha incrementado en los últimos años por lo que usuarios, industrias y gobiernos depositan grandes conjuntos de datos en la infraestructura de los proveedores de servicios (CSP, por sus siglas en inglés). No obstante, algunos servicios como el almacenamiento como servicio involucran severos riesgos de accesibilidad, integridad y privacidad de datos. Una solución a dichos inconvenientes está dada por el uso de múltiples CSPs, evitando así que un único CSP cuente con acceso total a la información confidencial. En este trabajo de tesis, se propone el análisis, diseño e implementación de una red apuntadora para configurar un sistema redundante de números residuales con aproximación de rango (AR-RRNS, por sus siglas en inglés) que permite distribuir la información en CSPs y recuperar la misma solo con de ellos. La red neuronal propuesta busca minimizar la probabilidad de pérdida de información y redundancia, ambos objetivos están en conflicto y es fundamental determinar una adecuada configuración de (, ) donde 2 ≤ ≤ . Asimismo, la red permite seleccionar CSPs específicos y asignarles un segmento del total de la información. La red apuntadora utiliza un modelo codificador decodificador y un sistema de atención entrenados mediante aprendizaje por refuerzo. Esta estructura le permite a la red afrontar cambios en los parámetros de los CSPs como la probabilidad de error. Además, la mayoría de los cálculos se realizan de manera offline. A partir de los resultados, se llevó a cabo un análisis comparativo con diversas versiones de algoritmos genéticos y el algoritmo de ramificación y poda, todos ellos forman parte fundamental del estado del arte en la optimización. El análisis muestra que la red apuntadora es más eficiente en tiempo que los demás algoritmos y genera soluciones similares en calidad al algoritmo genético simple pero la diversidad es menor con respecto a los algoritmos basado en población.The use of cloud services has increased in recent years as users, industries and governments deposit large data sets in the infrastructure of service providers (CSP). However, some services such as storage as a service involve severe data accessibility, integrity and privacy risks. A solution to these drawbacks is given by the use of multiple CSPs, thus preventing a single CSP from having full access to confidential information. In this thesis work, the analysis, design and implementation of a pointing network is proposed to configure a redundant system of residual numbers with range approximation (AR-RRNS) that allows to distribute the information in CSPs and recover the same only with of them. The proposed neural network seeks to minimize the probability of information loss and redundancy, both objectives are in conflict and it is essential to determine an adequate configuration of 2 ≤ ≤ . Likewise, the network allows selecting specific CSPs and assigning them a segment of the total information. The pointer network uses an encoder-decoder model and attention system trained by reinforcement learning. This structure allows the network to face changes in the parameters of the CSPs such as the probability of error. Also, most of the calculations are done offline. Based on the results, a comparative analysis was carried out with different versions of genetic algorithms and the branching and bound algorithm, all of which are a fundamental part of the state of the art in optimization. The analysis shows that the pointing network is more efficient in time than the other algorithms and generates solutions similar in quality to the simple genetic algorithm, but the diversity is lower with respect to population-based algorithms
Bioprospection and characterization of endophytic bacteria from Solanum hindsianum as plant growth promoters in Solanaceae of agricultural interest in Baja California
Las rizobacterias promotoras de crecimiento vegetal (PGPR) ayudan a las plantas a hacer frente a condiciones de estrés mediante distintos mecanismos, como la solubilización de nutrientes, la fijación de nitrógeno y la producción de fito-hormonas. Con el fin de encontrar alternativas a los fertilizantes químicos, se estudió el microbiota asociada a la solanácea endémica de Baja California Solanum hindsianum. Se lograron aislar 31 cepas bacterianas endófitas, las cuales fueron sometidos a una serie de pruebas para caracterizar su capacidad de solubilizar distintos nutrientes, producir ácido indol- 3- acético y biofilms, así como su tolerancia al estrés hídrico y salinidad. A continuación, mediante ensayos in vitro se determinó la variabilidad de los aislados bacterianos para promover la germinación en semillas de distintas solanáceas. Por último, se realizó un ensayo en invernadero inoculando plántulas de tomate, chile, berenjena y rábano con los aislados que habían mostrado mayor potencial. Las medidas morfométricas que se analizaron fue el largo de tallos y raíces, el diámetro del tallo a la altura de la corona, así como el peso seco para determinar el área por masa foliar (LMA) y el área específica de la raíz (RSL). Se encontraron aislados bacterianos pertenecientes al género Pseudomonas y Bacillus, capaces de promover el crecimiento en las distintas solanáceas. Los efectos se observaron principalmente a nivel de las raíces; las plantas de berenjenas fueron las plantas donde los efectos se dieron con mayor intensidad. Se proponen los aislados bacterianos endófitos Pseudomonas sp. EBCAb1, Bacillus sp. EBCAb9 y Pseudomonas sp. EBCAe10 como aislados potenciales para la formulación de biofertilizantes aplicados en zonas de suelos áridos.Plant growth promoting rhizobacteria (PGPR) help plants to cope with stress conditions through different mechanisms, such as nutrient solubilization, nitrogen fixation, and phytohormone production. In order to find alternatives to chemical fertilizers, the microbiota associated with the endemic Baja Californian Solanaceae Solanum hindsianum was studied. 31 endophytic bacterial strains were isolated, which were subjected to a series of tests to characterize their ability to solubilize different nutrients, produce indole3-acetic acid and biofilms, as well as their tolerance to water stress and salinity. Then, through in vitro tests, the variability of the bacterial isolates was determined to promote germination in seeds of different Solanaceae. Finally, a greenhouse assay was carried out, inoculating tomato, chile, eggplant and radish seedlings with the isolates that had shown the greatest potential. The morphometric measurements that were analyzed were the length of stems and roots, the diameter of the stem at the height of the crown, as well as the dry weight to determine the area per leaf mass (AML) and the specific area of the root (RSL). Bacterial isolates belonging to the genus Pseudomonas and Bacillus were found capable of promoting growth in the different Solanaceae. The effects were mainly observed at the root level; the eggplants were the crop where the effects occurred with greater intensity. The endophytic bacterial isolates Pseudomonas sp. EBCAb1, Bacillus sp. EBCAb9 and Pseudomonas sp. EBCAe10 are proposed as potential isolates for the formulation of biofertilizers applied in arid soil areas
Identification and distribution of resting cysts of Gymnodinium catenatum Graham in surface sediments of the northern Gulf of California.
La diversidad, abundancia y distribución geográfica de los quistes de dinoflagelados en sedimentos recientes sirven como bioindicadores de las condiciones ambientales y la productividad de regiones específicas. Este estudio describe ensamblajes de quistes de dinoflagelados de 53 muestras de sedimentos superficiales del norte del golfo de California (nGC) obtenidas en julio de 2016 y febrero de 2017. Se identificaron 47 especies de los órdenes: Gymnodiniales (6), Gonyaulacales (23) y Peridiniales (18). Diez de estas especies son potencialmente productoras de toxinas. La concentración total de quistes en todas las muestras fue de 3481 quistes/g en 2016 y 4223 quistes/g en 2017. La mayor abundancia de Gymnodiniales está relacionada probablemente a las condiciones de temperatura y salinidad del nGC. El aumento en el total de quistes registrados en 2017 se relacionó con un florecimiento algal nocivo (FAN) de Gymnodinium catenatum. Al nGC se han registrado FAN recurrentes de G. catenatum. Sin embargo, no se había investigado la presencia de quistes en reposo y el efecto de la hidrodinámica sobre su distribución en la región. Por esto, se evaluó la abundancia y distribución de quistes en sedimentos superficiales y su relación con la circulación local. Además, se implementó un modelo oceánico numérico para caracterizar el transporte y reubicación de quistes y sedimentos en la región. Los quistes de G. catenatum se distribuyeron heterogéneamente con algunas áreas de alta acumulación (hasta 158 quistes/g y 27 % del total de quistes registrados). El transporte de los quistes parece ser en dirección oeste a este después de la deposición. Por lo tanto, existen áreas extensas de acumulación de quistes que representan probablemente el semillero relacionado con el inicio de los FAN en la región. El nGC es un área de retención de quistes (y sedimentos) que permiten la formación de semilleros que podrían ser importantes para el desarrollo de FAN de G. catenatum. Los resultados proporcionan información clave para comprender la ecología de G. catenatum y, específicamente, para comprender la distribución geográfica y temporal de los FAN al nGC. Además de la identificación y cuantificación por microscopía, se estandarizaron protocolos de extracción de ácidos nucleicos a partir células vegetativas y quistes de G. catenatum en sedimentos, identificando molecularmente dos regiones del DNA ribosomal: LSU D1–D2 e ITS1-5.8S-ITS mediante la reacción en cadena de la polimerasa (PCR)...The diversity, abundance, and geographic distribution of dinoflagellate cysts in recent sediments serve as bioindicators of environmental conditions and productivity in specific regions. This study describes dinoflagellate cyst assemblages from 53 surface sediment samples from the northern Gulf of California (nGC) obtained in July 2016 and February 2017. Forty-seven species from three orders were identified: Gymnodinial (6), Gonyaulacal (23), and Peridinial (18). Ten species are potentially toxin-producers. The total cyst concentration in all samples was 3481 cysts/g in 2016 and 4223 cysts/g in 2017. The higher abundance of Gymnodiniales is probably related to local sea surface temperature and salinity in the nGC. The increase in total cysts in 2017 abundance was related to a harmful algal bloom (HAB) of Gymnodinium catenatum. Recurrent HABs of G. catenatum have been recorded at nGC. However, the presence of resting cysts and the effect of hydrodynamics on their distribution in the region have not been investigated. Therefore, the abundance and distribution of cysts and their relationship with local circulation in surface sediments were evaluated. In addition, a numerical ocean model was implemented to characterize the transport and relocation of cysts and sediments in the region. G. catenatum cysts were heterogeneously distributed with some areas of high accumulation (up to 158 cysts/g and 27 % of the total cysts recorded). Cysts appear to be transported in a west-to-east direction after deposition and accumulated in an extensive area that is probably the seedbed responsible for the initiation of HAB in the region. The nGC is an area of cyst (and sediment) retention that allows for the formation of seedbeds that could be important for the development of HAB of G. catenatum. The results provide key information for understanding the ecology of G. catenatum and, specifically, for understanding the geographic and temporal distribution of HABs to the nGC. Analysis of E2 binding to mitochondrial estrogen receptors in MCF-7 cells
La mitocondria es uno de los organelos de mayor interés en biomedicina debido a la variedad de procesos bioquímicos que se llevan a cabo en ella, por ejemplo, la respiración celular; el metabolismo de la glucosa y de los aminoácidos; y la síntesis de moléculas como el ATP, y hormonas como los estrógenos y los andrógenos. Diversos estudios han demostrado que los estrógenos, en especial el estradiol (E2), tienen efectos neuroprotectores y cardioprotectores. La mayoría de los efectos de los estrógenos son mediados por los receptores a estrógenos (ERs). Existen 3 tipos de ERs: el receptor a estrógeno alfa (ERα), el receptor a estrógeno beta (ERβ) y el receptor a estrógeno acoplado a proteína G (GPER). Tanto el ERα como el ERβ, y sus respectivas isoformas, han sido localizados en diferentes compartimientos celulares: en el núcleo, citosol, membrana celular y mitocondrias. Diversos estudios han sugerido que los efectos benéficos de los estrógenos se dan a nivel mitocondrial. Se han reportado efectos positivos del E2 sobre la función mitocondrial, tales como el aumento en la producción de ATP, disminución en la cantidad de especies reactivas de oxígeno (ROS), así como en la expresión del ADN mitocondrial (mtDNA). Estas evidencias en conjunto, sugieren que los efectos del E2 son mediados por los receptores a estrógenos mitocondriales (mtERs), sin embargo, este mecanismo no ha sido descrito por completo. Dado que la evidencia sugiere que los efectos del E2 sobre la función mitocondrial pudieran ser mediados por los mtERs, este trabajo tiene como objetivo evaluar la unión del E2 a los mtERs. Para esto se corroboró la presencia de los ERs en la mitocondria y se cuantificó el E2 presente en las mitocondrias y el citosol; así como el E2 unido a los mtERα en condiciones basales y de tratamiento con E2 1 y 100 nM. Además, con estos datos, se estableció por primera vez un método para la cuantificación del E2 unido a mtERs. Estos resultados son de gran importancia para el desarrollo de compuestos mitodirigidos que puedan modular la función mitocondrial y aplicarse en el área biomédica.The mitochondria is one of most important targets in biomedicine. This organelle is the site of a variety of biochemical processes such as cellular respiration, glucose and amino acids metabolism, synthesis of ATP, and synthesis of hormones, e.g., estrogens and androgens. Estrogens, mainly estradiol (E2), has been showed to have protective effects on neurovascular and cardiovascular systems. The majority of these estrogenic effects are mediated by estrogen receptors (ERs). There are three types of ERs: estrogen receptor alpha (ERα), estrogen receptor beta (ERβ), and G protein-coupled estrogen receptor (GPER). ERα and ERβ, as well as their respective isoforms, have been detected in different cell compartments like the nucleus, cytosol, cell membrane, and mitochondria. Several studies have suggested that beneficial estrogenic effects fall upon mitochondria directly. The positive effect of E2 on mitochondrial function, such as increases in ATP production, decrease in the amount of reactive oxygen species (ROS), and induction of mitochondrial DNA expression have been reported. The evidence suggests E2 effects are mediated by mitochondrial ERs (mtERs). The main objective of this thesis is to evaluate the binding of E2 specifically to mtERs. The presence of ERs in mitochondria was corroborated by Western blot; E2 was quantified in the cytosol and mitochondria; and E2 binding to mtERα in basal and treatment conditions. Most importantly, a protocol for quantifying E2-bound to mtERs was established. These results are essential for the development of a mitochondria-targeted treatment that can modulate mitochondrial function with biomedical applications
Elastic waves simulation in media with connected fractures
La presencia de fracturas es común en formaciones geológicas y tienden a dominar las propiedades hidráulicas y mecánicas en las rocas, por lo que el flujo de fluidos a través de estas no puede ser entendido sin un detallado conocimiento de la conectividad y geometría de las redes de fractura que las componen. El rango de potenciales aplicaciones abarca la exploración y explotación de hidrocarburos, almacenamiento de desechos nucleares, geohidrología, almacenamiento de CO2, entre otras, por lo que existe un gran interés en mejorar las técnicas para detectar y caracterizar redes de fracturas. Con ayuda de modernas computadoras y el desarrollo de nuevos métodos numéricos, se han impulsado nuevas metodologías para estudiar mejor el efecto de los medios fracturados en el campo de ondas sísmico. El método de Galerkin Discontinuo de Penalización Interior es un esquema numérico que permite simular la ecuación de onda elástica y evaluar modelos representativos de rocas fracturadas con diversas geometrías. Se construyeron volúmenes representativos con fracturas conectadas y no conectadas, para estudiar, no solo el efecto que tiene la conectividad en el campo de ondas, sino también la longitud de fracturas y la frecuencia de la fuente. Se implementó el medio equivalente llamado T-Matrix, que es una solución analítica que describe el comportamiento elástico de un medio con inclusiones, en este caso las fracturas. La principal característica de T-Matrix es que permite caracterizar la interacción de fracturas y su efecto en el campo de ondas. Este método busca replicar los modelos con fracturas discretas y así evaluar su efectividad en la caracterización de medios fracturados. Los análisis espectral y de coherencia en un plano tiempo-frecuencia muestran un efecto considerable producto de la conectividad, sugiriendo la posibilidad de incluir este efecto en los esquemas de evaluación de yacimientos fracturados.The presence of fractures is common in geological formations and tends to dominate their mechanical and hydraulic properties, so the flow of fluids through them cannot be understood without a detailed knowledge of the connectivity and geometry of the rocks. Potential applications include the exploration and exploitation of hydrocarbons, nuclear waste storage, CO2 sequestration, geohydrology, among others, so there is a great interest in improving techniques to detect and characterize fracture networks. With the help of modern computers and the development of new numerical methods, new methodologies have been promoted to better study the effect of fractured media on the seismic wavefield. The Interior Penalty Discontinuous Galerkin method is a numerical scheme that allows us to simulate the elastic wave equation and evaluate representative models of fractured rocks with various geometries. Representative volumes with connected and disconnected fractures were constructed to study, not only the effect of connectivity on the wavefield, but also the fractures’ length and the source frequency. The T-Matrix equivalent medium was implemented. This is an analytical solution that describes the elastic behavior of a medium with inclusions, in this case, fractures, which allowed characterizing the interaction of fractures and their effect on the wavefield. This method seeks to replicate the models with discrete fractures, and thus evaluate their effectiveness in the characterization of fractured media. Spectral and coherence analyses in the time-frequency plane, show a considerable effect as a result of connectivity, suggesting the possibility of including this effect in reservoir evaluation schemes
Multitemporal analysis of land cover and its association with water erosion rates in the Acoculco, Puebla
Las zonas de interés geotérmico son modelos interesantes en el diseño de políticas de ordenamiento territorial, debido a las iniciativas de sustentabilidad y análisis de impacto ambiental por parte de la Comisión Federal de Electricidad (CFE), con la finalidad de preservar el ecosistema y los recursos naturales del país fue analizado el comportamiento histórico de la cobertura del terreno, y la relación que esta tiene, con las tasas de erosión hídrica de la zona, a una escala territorial de microcuenca, las cuales presentan condiciones de aprovechamiento distintas (microcuenca 1 (C1), uso forestal; microcuenca Cuenca 2 (C2), uso agrícola). C1 y C2 se ubican en el municipio de Chignahuapan, en el Estado de Puebla. El análisis de la cobertura del terreno se realizó por medio de una clasificación supervisada de imágenes satelitales (5) de resolución media y baja de los satélites Landsat 5TM (3), Landsat 7 ETM+ (1), Landsat 8 OLI (2) para los años 1986, 1996, 2000, 2013, y 2018. Para la cuantificación de la pérdida del suelo por erosión hídrica, se utilizó la Ecuación Revisada de la Pérdida del Suelo (RUSLE, por sus siglas en inglés), el cual expresa su resultado en t año-1. En ella participan cinco variables: La agresividad de la lluvia (Factor R ), la erodabilidad del suelo (Factor K), la longitud e inclinación de la pendiente (Factor LS), la cobertura del terreno (Factor C) y las prácticas de manejo y conservación del suelo (Factor P). Para calcular la agresividad de la lluvia, fueron utilizados sistemas satelitales de la NASA para la medición de la precipitación, para corroborarlos, fueron correlacionados con datos de precipitación obtenidos en tierra por medio estaciones climatológicas automatizadas de la Comisión Nacional del Agua (CONAGUA). La erodabilidad del suelo fue calculada a través del análisis de la textura del suelo en laboratorio, por lo que se realizaron dos campañas de muestreo en las zonas estudiadas. La cobertura del terreno fue obtenida a través del promedio del Indice de Vegetación de Diferencia Normalizada (NDVI) de 3 imágenes (Temporada de lluvias, secas, e Intermedia) por año (los mismos que los de la cobertura). The analysis of the land cover was carried out by means of a supervised classification of satellite images (5) of medium and low resolution of the satellites Landsat 5TM (3), Landsat 7 ETM + (1), Landsat 8 OLI (2) for the years 1986, 1996, 2000, 2013, and 2018. For the quantification of soil loss due to water erosion, the Revised Soil Loss Equation (RUSLE) was used, which expresses its result in t yr-1. Five variables participate in it: the aggressiveness of the rain (Factor R), the erodibility of the soil (Factor K), the length and inclination of the slope (Factor LS), the coverage of the land (Factor C) and the management practices and soil conservation (Factor P). The land cover was obtained through the average of the Normalized Difference Vegetation Index (NDVI) of 3 images (Rainy, dry, and Intermediate season) per year (the same as those of the cover). The length and inclination of the slope was performed through a Digital Elevation Model of 30 m of spatial resolution, and the management and conservation practices were taken as 1.
