    Induced photosynthetic mixotrophy in Dunaliella salina: yield and biosynthetic pathways of isoprenoids, β-carotene and carotenogenesis

    La microalga Dunaliella salina es la fuente natural superlativa de β-caroteno que tiene propiedades nutricionales y terapéuticas valiosas. Estos atributos posicionan a esta especie como una fuente deseable para la producción masiva y sostenible de este isoprenoide C40. Sin embargo, la producción fotoautotrófica clásica aún enfrenta desafíos que limitan tanto su crecimiento asi como la posibilidad de obtener rendimientos y productividades mayores. En tanto que los cultivos promisorios hetero y mixotroficos aun requieren de optimización para mejorar los rendimientos celulares y producción de compuestos de alto valor agregado en esta clorofita. El objetivo de este trabajo fue investigar la capacidad de D. salina para crecer en condiciones heterotroficas y mixotroficas asi como producir rendimientos en biomasa y carotenoides (principalmente β-caroteno) de manera práctica, sustentable, de bajo costo y con rendimientos superiores a los reportados en esta microalga. Los modos nutricionales fueron investigados optimizando la relación C/N, la fuente de carbono organico, la intensidad luminosa y la salinidad. El sistema de cribado realizado demostró las preferencias metabólicas y fisiológicas de D. salina por el glicerol sobre la glucosa, acetato, xilosa y sacarosa para lograr rendimientos óptimos en biomasa celular y acumulación de carotenoides especialmente β-caroteno bajo mixotrofia fotosintética. Esta clorofita creció deficientemente en condiciones de cultivo heterotrófico. La combinación de glicerol 12.5 mM, salinidad 3.0 M NaCl y 50 μmol de fotones m2 s-1 de intensidad de luz mejoró la absorción y asimilación del glicerol por D. salina y, en consecuencia, aumentó el crecimiento (2.1 x 106 células mL-1) y la síntesis de β-caroteno (4.43 pg célula-1) en comparación con cultivos fotoautotróficos (0.9 x 106 células mL-1 y 1.37 pg célula-1, respectivamente). El estrés salino y lumínico estimuló la absorción de glicerol por esta microalga. En la etapa final de crecimiento, el aumento de la intensidad de la luz (de 50 a 300 μmol de fotones m2 s-1) permitió que el β-caroteno alcanzara valores superiores a 30 pg célula-1 y excediera en tres veces el nivel de β-caroteno producido por los cultivos fotoautotróficos bajo la misma intensidad de luz. El aumento de la salinidad de 1.5 a 3.0 M NaCl (salinidad no isosmótica) mejoró el crecimiento y el β-caroteno algal en comparación con la salinidad isosmótica de 3.0 M NaCl. Today, this species is a desirable source for the massive and sustainable production of this isoprenoid.C40. However, classic photoautotrophic production still faces challenges that limit both its growth and the possibility of obtaining higher yields and productivities. While heterotrophic and mixotrophic promoter cultures still require optimization to improve cell yields and production of high value-added compounds in this chlorophyta. The goal of this thesis was to investigate the ability of D. salina to grow in heterotrophic and mixotrophic conditions as well as to produce yields of biomass and carotenoids (mainly β-carotene) in a practical, sustainable, low-cost and higher yields than those reported in this microalga. The screening analysis carried out demonstrated the metabolic and physiological preferences of D. salina for glycerol over glucose, acetate, xylose, and sucrose for the achievement of optimal yields in cell biomass and carotenoids accumulation especially β-carotene under photosynthetic mixotrophy. This chlorophyte exhibited a deficient growth under heterotrophic conditions. The combination of 12.5 mM glycerol, 3.0 M NaCl salinity, and 50 μmol photons m2 s−1 light intensity improved absorption and assimilation of glycerol by D. salina and consequently this increased growth (2.1 x 106 cell mL-1) and β-carotene synthesis (4.43 pg cell-1) compared to photoautotrophic cultures (0.9 x 106 cell mL-1 and 1.37 pg cell-1, respectively). The saline and light stress stimulated the uptake of glycerol by this microalga. At the final stage of growth, the increase in light intensity (from 50 to 300 μmol photons m2 s−1) allowed the β-carotene to achieve values higher than 30 pg cell−1 and exceeding by three times the β-carotene level produced by photoautotrophic cultures under the same light intensity. Increasing salinity from 1.5 to 3.0 M NaCl (non-isosmotic salinity) improved the growth and microalgal β-carotene than the isosmotic salinity of 3.0 M NaCl. In this research, the effect of different organic carbon substrates has revealed that D. salina photosystems can choose nutritional options other than obligated photoautotrophy.     Design of Isoflux type radiation systems for satellite applications

    Las telecomunicaciones constituyen un factor de gran importancia en temas tecnológicos, económicos y de servicios a nivel mundial. Por tal motivo los satélites de nueva generación son un requerimiento primordial en el proceso de globalización. En dicha comunicación satelital a fin de difundir y propagar información de forma rápida y segura, se propone una radiación global del tipo Isoflux, necesaria para evitar los efectos que son causados por la distancia y otros factores. Un sistema de radiación tipo Isoflux proporciona una iluminación de la superficie de la Tierra dentro de su margen de visión en el polo ecuatorial. En este caso, un sistema de radiación convencional genera pérdidas de potencia en las partes lejanas del nadir (por los efectos causados por las longitudes distintas que recorre la señal radiada), poco tiempo de visibilidad en satélites MEO (se pierde la señal) y atención a pocas estaciones en Tierra para satélites GEO (mismo caso de satélites MEO). Un sistema de radiación tipo Isoflux evita las pérdidas de potencia en puntos lejanos del nadir iluminando la Tierra de manera uniforme siguiendo la cobertura de la Tierra. Por lo tanto, esta tesis tiene principalmente el objetivo de diseñar arreglos no uniformes o aperiódicos con la finalidad de mejorar el desempeño radiante del sistema y simplificar el sistema de antenas mediante una reducción de hardware, reduciendo el número de excitaciones de antena para simplificar el hardware del sistema de antena en un satélite creando así el patrón de radiación Isoflux. En este trabajo de tesis este diseño se plantea como un problema de optimización usando algoritmos genéticos.Telecommunications are a factor of great importance in technological, economic and service issues worldwide. For this reason, new generation satellites are a primary requirement in the globalization process. Satellite communication in order to spread and spread information quickly and safely, a global radiation of the Isoflux type is proposed, necessary to avoid the effects that are caused by distance and other factors. An Isoflux-type radiation system provides illumination of the earth's surface within its range of vision at the equatorial pole. An Isoflux type radiation system avoids power losses at points far from the nadir by illuminating the earth evenly. Therefore, this thesis mainly has a function in the design of non-uniform or aperiodic arrangements in order to improve the performance of the system and simplify the antenna system through a hardware reduction, reducing the number of antenna excitations to simplify the hardware of the antenna system on a satellite thus creating the Isoflux radiation pattern. This design is posed as an optimization problem using genetic algorithms

    Biostratigraphy and paleoecology of dinoflagellates from the Upper Cretaceous in tropical areas

    Resultados de análisis estadísticos y probabilísticos de una base de datos de quistes de dinoflagelados tropicales del Cretácico Superior de Venezuela permiten reconocer 21 especies en la secuencia estratigráfica óptima. La recopilación de los quistes de dinoflagelados reportados en estudios de diez localidades tropicales del Cretácico Superior del norte de América del Sur y el Golfo de Guinea permite el reconocimiento de un conjunto rico (262 especies). Este conjunto tropical está dominado por taxones gonyaulacoides (129 spp), seguido por protoperidinioides (82 spp), areoligeroides (29 spp), gymnodinioides (12 spp) y ceratioides (10 spp). Setenta y tres especies en la base de datos tropical se reportan en ambas regiones, lo que permite la calibración de edad de la secuencia óptima de Venezuela. La secuencia final propuesta de bioeventos permite el reconocimiento de la mayoría de las etapas en los estratos tropicales del Cretácico Superior. La alta diversidad del grupo Spiniferites-Achomosphaera y la escasez de Impagidinium indican ambientes de depósito marinos poco profundos para la mayoría de las localidades tropicales del Cretácico Superior incluidas. El predominio gonyaulacoides tropicales sugiere que estos fueron los productores primarios dominantes en los océanos tropicales del Cretácico superior. Datos pertenecientes a cuencas de Baja California en México, atribuidas a una provincia subtropical del hemisferio norte, poseen una alta coincidencia de los taxas marcadores de la parte alta del Cretácico Superior, esto permite inferir ambientes depositacionales semejantes en condiciones tectónicas diferentes.Results from statistical and probabilistic analyses of an of Upper Cretaceous, tropical dinoflagellate cyst database from Venezuela allow us to recognize 21 species in the optimal stratigraphic sequence. Compilation of the dinoflagellate cysts reported in ten studies of Upper Cretaceous tropical localities from northern South America and the Gulf of Guinea allow the recognition of a rich assemblage (262 species), dominated by gonyaulacoid (129 spp), followed by protoperidinioid (82 spp), areoligeroid (29 spp), gymnodinioid (12 spp) and ceratioid (10 spp) taxa. Seventy-three species in the tropical database are reported in both regions, allowing the age calibration of the optimal sequence from Venezuela. The proposed final sequence of bioevents allows recognition of most stages in tropical Upper Cretaceous strata. The high diversity of the Spiniferites-Achomosphaera group and scarcity of Impagidinium indicates shallow marine depositional environments for most of the Upper Cretaceous tropical localities included. The dominance of gonyaulacoid species in the tropical database suggests that these were the dominant primary producers in the tropical Late Cretaceous oceans. Data belonging to basins of Baja California in Mexico, attributed to a subtropical province of the northern hemisphere, have a high coincidence of the marker taxa of the upper part of the Late Cretaceous; this allows inferring similar depositional environments in different tectonic conditions

    Flow topology in asymmetric coaxial helical vortices and in the wake of wind turbines

    Los vórtices helicoidales son uno de los fenómenos fundamentales en la estela de turbinas eólicas. Su interacción, inestabilidad y efecto sobre la energía cinética en la estela aún están bajo investigación activa. Esta tesis trata de los vórtices helicoidales en tres casos de realismo creciente: vórtices paralelos, hélices infinitas y hélices formadas y desintegradas en la estela de la turbina. En el primer tema, estudiamos la topología de flujo inducida por tres y cuatro vórtices puntuales. Estos vórtices generan un flujo estacionario cuando se localizan en los vértices de un polígono regular; de lo contrario, generan un flujo periódico con regiones caóticas. Cuantificamos el mar caótico y el intercambio de masa para tres y cuatro vórtices puntuales y encontramos que el mar caótico crece linealmente a medida que los polígonos del vórtice son perturbados y se alejan de su configuración estacionaria. También estudiamos la evolución de tres y cuatro vórtices de Rankine localizados inicialmente en los vértices de un polígono regular y encontramos que se fusionan o interactúan elásticamente (a diferencia de dos vórtices, que también pueden intercambiar masa sin fusionarse). La topología de flujo de los vórtices que interactúan elásticamente es similar a la de los vórtices puntuales con la misma configuración geométrica; sin embargo, los vórtices de Rankine generan un flujo no estacionario debido a que su interacción los deforma con el tiempo. En el segundo tema, estudiamos el movimiento y el flujo inducido por dos vórtices helicoidales coaxiales e iguales. Determinamos la estabilidad de vórtices simétricos, que se mueven de manera estacionaria, en función de su grosor y paso. Los vórtices asimétricos muestran cuatro tipos de movimiento periódico que, a su vez, determinan la topología del flujo que inducen. En el tercer y último tema, estudiamos la estela detrás de una turbina eólica de dos aspas, caracterizada por vórtices helicoidales. Hay fenómenos en la estela, actualmente no comprendidos del todo, que afectan al rendimiento de los parques eólicos. Uno de ellos es la inestabilidad de los vórtices que desprenden las aspas. Encontramos que el inicio de la inestabilidad depende de la intensidad de la turbulencia corriente arriba. Además, la eficiencia de una segunda turbina en la estela depende de su distancia al punto donde los vórtices se vuelven inestables.Helical vortices are one of the fundamental phenomena in the wake of wind turbines. Their interaction, instability, and effect on the kinetic energy in the wake are still under active research. This thesis deals with helical vortices in three cases of increasing realism: parallel vortices, infinite helices, and helices formed and disintegrated in the wake of the turbine. In the first topic, we studied the flow topology induced by three and four point vortices. These vortices generate a steady flow when they are located on the vertices of a regular polygon; otherwise, they generate a periodic flow with chaotic regions. We quantified the chaotic sea and the mass exchange for three and four point vortices and found that the chaotic sea grows linearly as the vortex polygons are perturbed away from their steady configuration. We also studied the evolution of three and four Rankine vortices initially located on the vertices of a regular polygon and found that they either merge or interact elastically (unlike two vortices, which also can exchange mass without merging). The flow topology of elastically-interacting vortices is similar to that of point vortices with the same geometric configuration; however, Rankine vortices generate unsteady flow because their interaction deforms them over time. In the second topic, we studied the motion and flow induced by two equal coaxial helical vortices. We determined the stability of symmetric, steadily-moving vortices as a function of their thickness and pitch. Asymmetric vortices display four types of periodic motion which in turn determine the flow topology they induce. In the third and final topic, we studied the wake behind a two-bladed wind turbine, characterized by helical vortices. There are wake phenomena that affect wind farms’ performance that are not currently fully understood. One of them is the instability of the vortices shed by the blades. We found that the instability onset depends on the upstream turbulence intensity. Furthermore, the efficiency of a second turbine in the wake depends on its distance to the point where the vortices become unstable

    Spatio-temporal distribution of the blue whale (Balaenoptera musculus) according to the habitat off the western coast of the Baja California peninsula

    La península de Baja California (PBC) es una zona importante de alimentación y tránsito a las zonas de crianza para grandes misticetos migratorios como el rorcual azul (Balaenoptera musculus), especialmente de febrero a julio. Esta especie se caracteriza por sus altos requerimientos energéticos durante todo el año, por lo que se espera que presente una distribución predecible hacia regiones de afloramiento con alta producción primaria estacional a lo largo de la costa occidental de la PBC. En esta región, se llevaron a cabo 26 prospecciones sistemáticas aéreas y marítimas entre 1994 y 2020, registrando 162 grupos y 243 individuos de rorcual azul, desde la costa hasta 200 km mar adentro, durante las distintas estaciones del año. Se exploraron potenciales variables predictoras del hábitat de la especie, como distancia a la costa, perfil batimétrico, temperatura superficial del mar (TSM) y concentración superficial de clorofila-a, así como sus anomalías espaciales, para construir modelos aditivos generalizados (GAMs) del número de grupos observados, ponderados por el esfuerzo lineal de muestreo (i.e., tasas de encuentro). El mejor modelo incluyó la TSM y el gradiente batimétrico como covariables ambientales, representada como una relación no lineal, incluyendo un componente estacional y efectos aleatorios espaciales. El modelo mostró una alta variabilidad estacional en la distribución espacial del hábitat idóneo. Se evidenció una preferencia de la especie por la zona oceánica al sur de la PBC durante el verano, otoño e invierno; mientras que, en primavera, la especie usa gran parte la costa peninsular. El modelo de hábitat predijo zonas de alta agregación de rorcuales azules que coinciden con las observaciones, lo que destaca la habilidad del modelo para predecir la tasa de encuentro y la preferencia del hábitat de la especie. Estos “puntos calientes” fueron la zona de frentes térmicos próxima a Bahía Magdalena y Cabo San Lucas, Punta Eugenia y a lo largo del frente de Ensenada. Las áreas mencionadas son de vital importancia para la conservación del rorcual azul y deben tenerse en cuenta durante la planeación de la instalación de infraestructura para obtener energía del océano, entre otras actividades humanas.The Baja California Peninsula (BCP) is an important feeding and transit area for large migratory baleen whales such as the blue whale (Balaenoptera musculus), especially from February to July. The species is characterized by its high energy requirements throughout the year, so it is expected to exhibit a predictable distribution towards upwelling regions with highly seasonal primary production along the western coast of the BCP. In this region, 26 systematic aerial and maritime surveys were carried out between 1994 and 2020. We registered 162 groups and 243 individuals of blue whales, from the coast up to 200 km offshore with a seasonal coverage. Several potential predictive variables of the species’ habitat were explored as predictors, such as distance from shore, bathymetric profile, sea surface temperature (SST) and sea surface chlorophyll-a concentration, as well as their spatial anomalies, to build generalized additive models (GAMs) of the number of groups observed, weighted by the linear effort (i.e., encounter rates). The best model had SST and bathymetry gradient as covariates, represented as a nonlinear environmental relation, including a seasonal component and spatial random effects. Model results indicated a highly seasonality in the spatial distribution of habitat suitability of the species’ preference for the oceanic zone south of the BCP during summer, autumn and winter, while, in spring, the species uses a large part of the peninsular coast. The habitat model predicted areas of high blue whale aggregation that matched spatially the observations, highlighting the model's ability to predict encounter rate and habitat preference for the species. These hotspots were the oceanic fronts off Bahía Magdalena and Cabo San Lucas, Punta Eugenia, and along the Ensenada Front. These areas are therefore of vital importance for the conservation of the blue whale, and for upfront planning the installation of infrastructure to obtain energy from the ocean, among other human activities

    Cleaning interactions and structure of the fish community in rocky reefs of the Revillagigedo archipelago

    Los monitoreos de la ictiofauna en los arrecifes brindan información relevante de las condiciones en las que se encuentra el ecosistema, y ayudan a detectar sus respuestas ante acciones de manejo y conservación. En las comunidades ícticas, las interacciones de limpieza favorecen la salud de los organismos mediante la remoción de ectoparásitos y piel muerta, y podrían aumentar la biodiversidad y abundancia local. El archipiélago de Revillagigedo alberga una alta riqueza de especies ícticas endémicas, protegidas y en peligro de extinción, aunque debido a su lejanía, los trabajos ictiológicos en la zona son escasos. El propósito de este trabajo fue describir las interacciones de limpieza que ocurrieron en Revillagigedo, caracterizar la estructura de la comunidad íctica y evaluar la relación entre la diversidad de especies y la ocurrencia de interacciones. Se realizaron censos visuales con buceo autónomo empleando las técnicas de transectos de banda (peces arrecifales) y transectos errantes (peces pelágicos) en las islas Socorro, San Benedicto y Roca Partida durante cuatro temporadas turísticas (noviembre-junio) desde 2017 hasta 2021. Se registraron 277 interacciones de limpieza en las que participaron siete especies de limpiadores y 23 especies como clientes (15 peces óseos y ocho elasmobranquios). En el caso de los limpiadores, Johnrandallia nigrirostris tuvo la mayor cantidad de clientes (n=16), mientras que Mobula birostris y Carcharhinus albimarginatus fueron los clientes que se relacionaron con un mayor número de limpiadores (n=cuatro). La mayor frecuencia de interacción limpiador-cliente se obtuvo para Holacanthus clarionensis-M. birostris en isla Socorro y San Benedicto, y J. nigrirostris-Paranthias colonus en Roca Partida. La riqueza y diversidad fueron estadísticamente diferentes entre las islas. Las especies dominantes en el archipiélago pertenecieron a las familias Carangidae, Labridae y Serranidae. Las especies pelágicas fueron más abundantes en Roca Partida y las especies arrecifales en Socorro, islas con el mayor valor de disimilitud entre ellas. La diversidad de especies pelágicas tuvo una influencia positiva en el número de interacciones de limpieza registradas. Esta información corresponde al primer acercamiento para identificar a las especies que participan en las interacciones de limpieza, así como su posible relación con características de la comunidad íctica de Revillagigedo.The monitoring of the ichthyofauna in the reefs can provide relevant information on the conditions in which the ecosystem is found or detect possible responses to management and conservation actions. Fish cleaning interactions promote the health of organisms by removing ectoparasites and dead tissue on the skin and could increase local biodiversity and abundance. The Revillagigedo archipelago is inhabited by a high richness of endemic, protected, and endangered fish species, although due to its remoteness, ichthyological studies in the area are scarce. This work aimed to describe the cleaning interactions that occurred in Revillagigedo, characterize the structure of the fish community, and evaluate the relationship between species diversity and the occurrence of interactions. Visual censuses were carried out with scuba diving using the techniques of belt transects (reef fish) and roving diver transect (pelagic fish) in the Socorro, San Benedicto, and Roca Partida islands during four tourist seasons (November-June) from 2017 to 2021. A total of 277 cleaning interactions involving seven species of cleaners and 23 species as clients (15 bony fishes and eight elasmobranchs) were recorded. In the case of cleaners, Johnrandallia nigrirostris had the highest number of clients (n=16), while Mobula birostris and Carcharhinus albimarginatus were the clients interacting with the largest number of cleaners (n=four). The highest frequency of cleaner-client interaction was obtained for Holacanthus clarionensis-M. birostris in Socorro and San Benedicto islands, and J. nigrirostris-Paranthias colonus in Roca Partida. Richness and diversity were statistically different between islands. The dominant species in the archipelago belonged to the Carangidae, Labridae, and Serranidae families. Pelagic species were more abundant in Roca Partida, while reef species were more abundant in Socorro, islands with the highest dissimilarity value. Pelagic species diversity positively influenced the recorded number of cleaning interactions. This information corresponds to the first approach to identifying the species that participate in cleaning interactions, as well as their possible relationship with characteristics of the Revillagigedo fish community

    Seismic Attenuation Study in the Northern Gulf of California Region

    La atenuación de las ondas sísmicas depende las propiedades físicas de las rocas, edad, contenido de fluidos, temperatura y presión. Por lo general, los cambios en el estado físico de las rocas se pueden monitorear mejor evaluando cambios en el factor de calidad que con cambios en la velocidad de propagación de las ondas. Para conocer los mecanismos físicos que afectan la atenuación de las ondas sísmicas en la zona norte del Golfo de California, se determinó el factor de calidad de las ondas de cuerpo (y ) con sismogramas de la Red de banda ancha del Golfo de California (RESBAN) operada por el CICESE. La base de datos consistió en 64 sismos con magnitud que varía entre 4.5 y 6.6, y con distancia hipocentral entre 30 km y 350 km. Se determinaron funciones de atenuación a partir de un modelo no paramétrico que consiste en invertir las amplitudes espectrales observadas de 25 frecuencias comprendidas entre 0.1 Hz y 25.12 Hz, distribuidas en escala logarítmica. Para estimar y se definieron tres funciones de dispersión geométrica: una dependiente de la frecuencia, ()1 y dos independientes de la frecuencia, ()2 y ()3; donde ()2 es 1⁄ para ≤ 100 km y 1⁄√100para > 100 km, mientras que ()3= 1⁄ para todo el rango de distancia. Encontramos que las estimaciones de dependen de manera importante de la función de dispersión geométrica que se adopte. Las estimaciones de usando ()1 indican que la atenuación de las ondas P es mayor ( = 224.5 1.10) que la atenuación de las ondas S ( = 244.5 1.17). Análogamente, usando ()2, la atenuación de las ondas P es mayor ( = 146.5 0.70) que la atenuación de las ondas S ( = 170.7 1.01), y similarmente, las estimaciones de usando ()3 también indican que la atenuación de las ondas P es mayor ( = 179.3 0.60) que la atenuación de las ondas S ( = 233.6 1.03). En general, los valores de son altos ( > 100) lo que sugiere que la región contiene rocas con bajo contenido en fluidos (⁄) y la alta presión pudiera generar una disminución de la atenuación por el cierre de los poros en las rocas.The attenuation of seismic waves depends on rock physical properties, age, fluid content, temperature and pressure. In general, changes in the physical state of the rocks can be monitored better by evaluating changes in the quality factor than by changes in the velocity of wave propagation To understand the physical mechanisms affecting the attenuation of seismic waves in the northern Gulf of California, the quality factor of body waves ( y ) was determined with seismograms from the broadband seismological network of the Gulf of California (RESBAN) operated by the CICESE. Attenuation functions were determined from a nonparametric model which consists of inverting the observed spectral amplitudes of 25 frequencies between 0.1 Hz and 25.12 Hz with a logarithmic spacing. To estimate and , three geometric dispersion functions were defined: one frequency dependent, ()1 and two frequency independent, ()2 and ()3; where ()2 is 1⁄ for ≤100 km and 1⁄√100 for r > 100 km, while ()3 = 1⁄ for the entire distance range. We find that the estimates of depend strongly on the geometric dispersion function adopted. Estimates of using ()1 indicate that the P-wave attenuation is larger ( = 224.5 1.10) than the S-wave attenuation ( = 244.7 1.17). Similarly, using ()2 ,the P-wave attenuation is larger ( = 146.5 0.70) than the S-wave attenuation ( = 170.7 1.01), and similarly, estimates of using ()3 also indicate that P-wave attenuation is larger ( = 179.3 0.60) than S-wave attenuation ( = 233.61.03). In general, Q values are high (>100) suggesting that the region contains rocks with low fluid content (⁄) and the high pressure could generate a decrease in attenuation due to pore closure in the rocks

    La enfermedad de Alzheimer (EA) es un proceso neurodegenerativo, crónico, progresivo e irreversible en adultos mayores de 65 años. La EA se desencadena por la acción de β-secretasa (BACE1), la cual ocasiona daño neuronal. Actualmente, los tratamientos contra la EA tienen poca eficacia en llegar a cerebro y tiene una alta toxicidad hepática en su administración oral. Para superar estos retos, sintetizamos un nanocompuesto con hidrogeles (G) y puntos cuánticos de carbono (CD), con potencial aplicación transdérmica. Los CD se sintetizaron a partir de ácido cítrico por método solvotermal y los hidrogeles de gelatina metacrilada (GelMA) se obtuvieron por fotoentrecruzamiento con luz UV. Los CD se conjugaron con transferrina (CD-T) para cruzar la barrera hematoencefálica (BHE), con un péptido inhibidor de BACE1 (CD-iβ) y con ambos (CD-Tiβ). Los CD emitieron fluorescencia a una longitud de onda de 470.4 nm (cian), se observaron estructuras esféricas y con tamaños desde 8.3 nm (CD) hasta 16.9 nm (CD-T) en promedio, respectivamente. Además, la cantidad de proteínas conjugadas a los CD fue de 8.5 μg/mL (CD-iβ) y 10.5 μg/mL (CD-T y CD-Tiβ). La microestructura en todos los nanocompuestos se conservó y se observaron poros de 51-100 μm de diámetro. En PBS, los nanocompuestos comenzaron a degradarse a las 96h y mostraron una degradación máxima entre las 120-144 h. Los nanocompuestos de G-CD-iβ y G-CD-Tiβ mostraron un mayor hinchamiento y una degradación más rápida, en comparación con los otros nanocompuestos. Los nanocompuestos en medio de Eagle modificado de Dulbecco (DMEM) tuvieron una cinética de liberación de orden cero, mostrando una liberación controlada por difusión. El 50% de los conjugados de CD se liberaron desde los nanocompuestos G-CD-Tiβ a las 96 h, mientras que desde los G-CD-T lo hicieron a las 144 h. Los ensayos de viabilidad celular de las células endoteliales (HBEC 5i) y células de glioblastoma (U-87 MG) mostraron que todos los conjugados de CD eran citocompatibles. Los distintos CD conjugados lograron cruzar un modelo in vitro de BHE e inhibieron >50% de la actividad de BACE1 en un modelo in vitro de enfermedad de Alzheimer inducido con escopolamina en las células U-87 MG.Alzheimer’s disease (AD) is a neurodegenerative, chronic, progressive and irreversible process in adults older than 65 years. AD is triggered by the action of β-secretase (BACE1), causing neuronal damage. Currently, AD treatments have shown little efficacy in reaching the brain and liver toxicity in oral administration. To overcome these challenges, we synthesized a nanocomposite with hydrogels (G) and quantum carbon points (CD), with the potential transdermal application. CDs were synthesized from citric acid by solvothermal method and gelatin methacryloyl hydrogels (GelMA) were photocrosslinked by UV irradiation. CDs were conjugated with transferrin (CD-T) to pass through the blood brain barrier (BBB), with a BACE1-inhibitor peptide (CD-iβ) and both (CD-Tiβ). CDs showed fluorescence emission at a wavelength of 470.4 nm (cyan), were observed spherical and with sizes from 8.3 nm (CD) to 16.9 nm (CD-T) average, respectively. Moreover, the amount of proteins conjugated to CDs was 8.5 μg/mL (CD-iβ) and 10.5 μg/mL (CD-T and CD-Tiβ). Microstructure in all nanocomposites was preserved and pores of 51-100 μm in size were observed. In PBS, nanocomposites degradation began at 96h and showed maximum degradation between 120-144 h. G-CD-iβ and G-CD-Tiβ nanocomposites showed greater swelling and faster degradation compared to others. Nanocomposites in Dulbecco’s Modified Eagle’s Medium (DMEM) had zero-order release kinetics, showing a controlled release by diffusion. 50% of CDs conjugates were released from G-CD-Tiβ nanocomposites at 96 h, while from G-CD-T were at 144 h. Cell viability tests on endothelial cells (HBEC 5i) and glioblastoma cells (U-87 MG) showed that all CDs conjugates were cytocompatible. All CD conjugates succeeded in crossing an in vitro model of BBB and inhibited >50% BACE1 activity in an in vitro model of scopolamine-induced Alzheimer’s disease in U-87 MG cells

    La pared celular de hongos se compone principalmente de quitina y glucanos. La quitina, a pesar de no ser el componente mayoritario de la pared celular, es responsable de la plasticidad y conservación de la integridad celular. La síntesis de quitina en hongos se lleva a cabo por las quitina sintasas (CHS), una familia de proteínas transmembranales que son transportadas dentro de la célula en microvesículas acarreadoras llamadas quitosomas. Los mecanismos que involucran la biogénesis y el transporte de CHS aún conservan múltiples incógnitas. En Saccharomyces cerevisiae, la chaperona Chs7 es requerida para la salida de la quitina sintasa de clase IV, Chs3, del retículo endoplasmático y se transporta en conjunto con la microvesícula hasta la membrana plasmática para llevar a cabo la biosíntesis de quitina. En Neurospora crassa, se identificaron dos proteínas ortólogas a la Chs7 de S. cerevisiae: CSE-7 y CSE-8. La CSE-7 fue caracterizada por ser imprescindible en la secreción y biogénesis de la quitina sintasa de CHS-4 (clase IV). El papel de CSE-8 en el tráfico y biogénesis de quitosomas en N. crassa aún es desconocido. Sin embargo, en este estudio se caracterizó el fenotipo de la cepa ∆cse-8, que en comparación con las cepas de tipo parental y silvestre, presentó una reducción del 40% en el contenido de N-acetilglucosamina total de la célula. Adicionalmente, ∆cse-8 mostró una intensidad de fluorescencia de blanco de calcoflúor en septos disminuida y un menor grosor en hifas principales. Estos resultados, sugieren que CSE-8 podría jugar un papel en la síntesis de la pared celular. Para analizar el papel de CSE-8 en la ruta secretora de quitina sintasas de clase III (CHS-1) y V (CHS-5) en hifas de N. crassa, se llevaron a cabo cruzas genéticas para obtener cepas de chs-1::gfp y chs-5::gfp con fondo mutante ∆cse-8. Los resultados obtenidos permitieron corroborar que CSE-8 no participa en el tráfico de CHS-1 y CHS-5 a septos ni al Spitzënkorper (SPK). Sin embargo, su localización en la región central del SPK, septos, red de cisternas endomembranales y zona periférica nuclear, sugiere una posible participación de CSE-8 en el transporte de otras CHS.The cell wall of filamentous fungi and yeasts is mainly composed of chitin and glucans. Chitin, although not a major component of the cell wall, is responsible for plasticity and preservation of cell integrity. Chitin synthesis in fungi is carried out by chitin synthases (CHS), a family of transmembrane proteins that are transported into the cell in microvesicle carriers called chitosomes. The mechanisms involving CHS biogenesis and trafficking remain largely unknown. In Saccharomyces cerevisiae, Chs7 is required for the exit of class IV chitin synthases from the endoplasmic reticulum and is transported together with the microvesicle to the plasma membrane, where chitin biosynthesis takes place. Therefore, it is considered that Chs7 must be involved in the trafficking of chitin synthase-carrying chitosomes. In Neurospora crassa, two proteins orthologous to S. cerevisiae Chs7p were identified: CSE-7 and CSE-8. CSE-7 was characterized as having a key role in the secretion and biogenesis of the class 4 chitin synthase CHS-4. The role of CSE-8 in chitosome trafficking and biogenesis in N. crassa is unknown. However, previous studies suggest that it may represent an element of the machinery involved in the trafficking of one of the other chitin synthases, whose secretory pathway has not been described. In this study, we characterized the phenotype of a ∆cse-8 strain, which, compared to the parental and wild-type strains, presented a 40% reduction in the total N-acetylglucosamine content of the cell. Additionally, ∆cse-8 showed decreased fluorescence intensity of calcofluor white in septa and reduced thickness in main hyphae. These results suggest that CSE-8 may play a role in cell wall synthesis. To analyze the role of CSE-8 in the secretory pathway of class III (CHS-1) and V (CHS-5) chitin synthases in N. crassa hyphae, genetic crosses were carried out to obtain chs-1::gfp and chs-5::gfp strains with a ∆cse-8 mutant background.. The results corroborated that CSE-8 is not involved in the trafficking of CHS-1 and CHS-5 to septa and to the Spitzenkörper (SPK). However, its localization in the central region of the SPK, septa, network of endomembranous cisternae (NEC), and nuclear envelope suggests a possible involvement of CSE-8 in the transport of other CHS

    Sediment dynamics in valleys and ridges of the bathymetry of the Upper Gulf of California

    Los esteros representan áreas de gran importancia ambiental y socio económica para las regiones aledañas de donde se ubican. Este es el caso del Alto Golfo de California, el cual es considerado un estuario inverso. Dentro de sus características más representativas, se distinguen una batimetría compleja, poca profundidad (menos de 50 m), rangos grandes de marea (∼ 7 m durante mareas vivas) y corrientes rápidas de mareas (∼ 1 m s ·1). Esta serie de características distintas a otros entornos en México permiten el desarrollo de estudios para comprender la dinámica de los procesos que ocasionan cambios en un ecosistema. La batimetría del Alto Golfo presenta una serie de barras orientadas a la circulación principal y que forman una serie de crestas y valles con diferencias de profundidad importantes. En este trabajo se realizó una comparación entre dos sitios para conocer el transporte del sedimento en una cresta y un valle en condiciones de corrientes y oleaje. El objetivo fue conocer la distribución y características del sedimento en cada uno de los sitios y analizar la importancia de los sedimentos cohesivos. Además, se utilizó el modelo GOTM para obtener la variabilidad temporal de los modos de transporte de sedimento en los dos sitios. Los resultados indican para la cresta (∼ 10 m de profundidad), un aumento del material partículado en suspensión debido principalmente al oleaje. Mientras que para el valle (∼ 25 m de profundidad), la corriente de marea es el principal forzante para la resuspensión del sedimento. En cuanto al transporte de sedimentos se establecieron dos modos de transporte, siendo para la cresta la carga de fondo el principal modo y para el valle, de carga de fondo y carga suspendida. En ambos sitios los flóculos se clasificaron como microflocs (diámetros < 125 μm). En la cresta se registraron las concentraciones más bajas, con valores máximos de hasta 54 μ/ y en el valle con valores de hasta 90 μ/. El corte turbulento fué mayor en la cresta, favoreciendo la agregación de los flóculos y la ruptura debido a la intesidad de la turbulencia. En el valle, se registraron valores de corte turbulento bajos al inicio de las observaciones, aumentando la probabilidad de mayor agregación durante ese período. La microescala de Kolmogorov fué más grande que el tamaño de los flóculos, indicando intensidad de turbulencia suficiente para ocasionar el rompimiento de flóculos grandes.The estuaries represent areas of great environmental and socio-economic importance for the surrounding regions where they are located. This is the case of the Upper Gulf of California, which is considered an inverse estuary. Among its most representative characteristics, it is distinguished by a complex bathymetry, shallow water (depth < 50 m), large tidal ranges (∼ 7 m during spring tides) and fast tidal currents (∼ 1 m · s−1 ). These characteristics, which are different from other environments in Mexico, allow the development of studies to understand the dynamics of the processes that cause changes in an ecosystem. The bathymetry of the Upper Gulf presents a series of bars oriented to the main circulation, forming series of crests and valleys with significant differences in depth. In this work, a comparison was made between two sites to determine the sediment transport in a crest and a valley under current and wave conditions. The objective was to determine the distribution and characteristics of the sediment in each of the sites and to analyze the importance of cohesive sediments. Furthermore, the GOTM model was used to obtain the temporal variability of the sediment transport modes at the two sites. The results indicate for the crest (10 m deep), an increase in suspended particulate matter mainly due to waves. While for the valley (25 m deep), the tidal current is the main forcing for the resuspension of the sediment. In terms of sediment transport, two transport modes were established, with bottom load being the main mode for the crest and bottom load and suspended load for the valley. In both sites the flocs were classified as microflocs (diameters < 125 μm). The lowest concentrations were recorded at the crest, with maximum values of up to 54 μ/, while in the valley with values of up to 90 μ/. The turbulent shear was greater at the crest, favoring floc aggregation and breakup due to the intensity of the turbulence. In the valley, low turbulent shear values were recorded at the beginning of the observations, increasing the probability of greater aggregation during that period. The Kolmogorov microscale was larger than the size of the flocs, indicating sufficient turbulence intensity to cause the breakup of the large flocs
