    HERBAL PRODUCT USE FOR ORAL HEALTH AMONG PATIENTS AND THEIR COMPANIONS ATTENDING THE FACULTY OF DENTISTRY

    Background and Aim: Many people tend to use herbal products without consulting a healthcare provider for various purposes including oral and dental problems. There is a significant lack of research focused on dental patients’ use of herbal products for oral and general health. This study aimed to determine the status and reasons for herbal product use among patients attending the Faculty of Dentistry at Hacettepe University. Materials and Methods: Data were collected via a selfadministered questionnaire applied under supervision. The questionnaire was developed by the researchers and consisted of 20 questions in total. A total of 300 questionnaires were evaluated. Descriptive statistics are given by number and percent distributions. Results: The mean age of participants was 37.86 years, ranging from 18 to 74. The proportion of female participants was 66.2%, while males constituted 33.8%. 35.5% (n=106) of participants were using herbal products. The most commonly selected reason for herbal product use was immune support, followed by skin care, pain relief, weight control, and wound or burn healing. It was observed that only 9.4% of the reported uses were directly related to oral health. Conclusion: Although general interest in herbal products appears strong, this interest seems to be directed more toward systemic and cosmetic applications rather than oral and dental health. There is a clear need to improve public awareness and professional guidance related to the use of herbs. It is imperative that the dentist be familiar with herb-drug interactions, as clearly a large number of patients are using these products

    Cytotoxic and HPLC-based analysis of eleven<i> Rumex</i> extracts: Anthraquinone profiling and synergistic potential with doxorubicin in cancer cells

    The genus Rumex is a rich source of bioactive anthraquinones with known pharmacological properties, including anticancer potential. In this study, dichloromethane extracts of eleven Rumex species were tested on MCF-7, SK-BR-3, and MDA-MB-231 breast cancer cell lines for the evaluation of their cytotoxic properties. Among the tested species, the dichloromethane extract of Rumex crispus roots (DERCR) demonstrated the most potent cytotoxicity, particularly towards SK-BR-3 cells, with a survival rate of 54.56 % at 30 mu g/mL and was taken to the further studies in order to determine its modes of action. The DERCR did not induce a significant change in the cell cycle phases of SK-BR-3 cells while generating reactive oxygen species (ROS) leading to apoptosis and necrosis. Furthemore, the combination regimens of the DERCRs with doxorubicin were also applied to the cancer cells to see if there is a possible synergistic effect in cancer cell lines. No synergistic effects were observed. High-performance liquid chromatography (HPLC) was used to identify and quantify major anthraquinone compounds in the composition of each extract. Thus, the Rumex species based on the substantial anthraquinone derivatives in their composition were assessed in terms of their cytotoxicity. The findings supported R. crispus including anthraquinones as a complementary agent in breast cancer therapy

    Metabolomic Profiling of Leukemic Hematopoiesis: Effects of BNT162b2 mRNA COVID-19 Vaccine Administration

    Background: Leukemia is marked by clonal hematopoietic stem cell expansion and metabolic reprogramming. The BNT162b2 mRNA COVID-19 vaccine has been proven effective, though questions remain about its broader physiological effects. This study investigates metabolomic alterations in leukemic bone marrow potentially associated with BNT162b2 vaccination. Objective: To compare the bone marrow metabolomic profiles of leukemia patients with and without BNT162b2 vaccination, and healthy unvaccinated controls, to explore potential metabolic differences. Methods: Bone marrow samples were obtained from three groups: vaccinated leukemia patients (n=7), unvaccinated leukemia patients without COVID-19 history (n=2), and unvaccinated healthy controls (n=7). Untargeted metabolomics was performed using LC-QTOF-MS. Data were analyzed using XCMS and MetaboAnalyst 5.0 to identify statistically significant metabolite differences and affected pathways. Fold change >1.5 and p<0.05 were considered significant. Results: Distinct metabolic profiles were observed between the leukemia and control groups. Increased glycolysis, pentose phosphate pathway activity, and altered tryptophan, lipid, and heme metabolism were noted in leukemia samples. Metabolic changes in vaccinated patients (ASL) were more similar to unvaccinated leukemia patients (LO) than to healthy controls, with minor vaccine-associated variations. Notable metabolites included 5-methoxyindoleacetate, phosphorylcholine, and tetrahydrofolic acid. Conclusion: This preliminary study identified altered metabolic pathways in leukemia bone marrow and suggests metabolomic differences associated with BNT162b2 vaccination. While the findings do not support a causal link between mRNA vaccination and leukemia development, they highlight the need for further studies to understand vaccine-induced metabolic modulation in hematological contexts

    GLP-1 Receptor Analogs: Evidence Linking to Effect on Metabolic and Reproductive Functions in Patients with PCOS and Obesity

    Background: Polycystic ovary syndrome (PCOS) is a multifaceted hormonal disorder that impacts both metabolic functions and reproductive health in women. PCOS pathophysiology is characterized by insulin resistance and chronic inflammation, which contribute to disrupted gonadotropin secretion, elevated androgen levels, and irregular ovulation. There is a bidirectional relationship between obesity and PCOS. Obesity contributes to the pathophysiology of PCOS, particularly by exacerbating insulin resistance - a core feature of the condition. Insulin resistance can lead to further weight gain and hinder successful weight loss. Summary: According to the latest international guidelines, improving insulin sensitivity and promoting weight loss are key strategies in the effective management of PCOS. GLP-1 receptor analogs (GLP-1 RAs) are a group of agents that are used in type 2 diabetes mellitus to regulate blood glucose levels, in addition to being indicated for weight loss in obesity. Thus, GLP-1 RAs may play a role in the management of PCOS, both due to weight loss effect and through pleiotropic effects like ameliorating insulin resistance, decreasing inflammation, and modulation of adiposity. Key Messages: In light of these effects, this review aimed to perform a systematic search and narrative synthesis on the impact of GLP-1 receptor agonists (GLP-1 RAs) on the metabolic and reproductive outcomes in PCOS, with the goal of elucidating the mechanistic link between the action of GLP-1 RAs and the underlying pathophysiology of the condition

    The Impact of an EMDR-Focused Psychoeducation Program for Women Who Cannot Have Children Naturally on Infertility Stress

    This research aims to examine the impact of an EMDR-focused Psychoeducation program developed for women experiencing infertility on the stress experienced by women. A quasi-experimental design was employed, incorporating a pre-test, post-test, and follow-up test with a control group. Fourteen people, 7 in the experimental group and 7 in the control group, aged between 25 and 40, living in Istanbul, who could not have children naturally, participated in this study. An EMDR-focused Psychoeducation program was applied to the participants in the experimental group; no treatment was applied to the participants in the control group. Pre-test and post-test data were collected using the Infertility Stress Scale. Mann-Whitney U test and Wilcoxon Signed Ranks test, which are nonparametric tests, were used to analyze the data. The data obtained from the research were analyzed using the SPSS 29.00. According to the results, the infertility stress experienced by women participating in the psychoeducation program in the personal area is lower than that of the control group. Post-test scores in the individual domain of infertility stress were lower than pre-test scores among participants in the experimental group. In this case, the EMDR-focused Psychoeducation program may effectively reduce infertility stress experienced in the personal domain
