Suppressing the effect of lithium dendritic growth by the addition of magnesium bis(trifluoromethanesulfonyl)amide
Practical applications of Li-S and Li-air batteries require the morphology of the Li metal negative electrode during charge/discharge (i.e., Li-deposition/dissolution) cycling to be precisely controlled. Herein, we used magnesium bis(trifluoromethanesulfonyl)amide [Mg(TFSA)(2)] as an electrolyte additive to suppress the growth of Li dendrites, utilizing the occurrence of an alloying reaction between the initially substrate-deposited Mg and the subsequently deposited Li. Notably, no metallic Mg formation and no change in Li deposition morphology were observed at an electrolyte composition of 0.1 M Mg(TFSA)(2) + 0.9 M LiTFSA/triglyme, irrespective of the applied potential. In contrast, increasing the Mg salt concentration to 0.5 M resulted in the deposition of interconnected granules, reflecting a dramatic morphology improvement. X-ray diffraction analysis revealed the occurrence of the abovementioned alloying, which finally afforded a deposit composition of Li0.9Mg0.1 via the formation of an intermediate Li0.14Mg0.86 phase. Efficient generation of an oxidopyrylium ylide using a Pd catalyst and its [5+2] cycloadditions with several dipolarophiles
An efficient method for the generation of an oxidopyrylium ylide from 6-acetoxy-6-acetoxymethyl-2H-pyran-3(6H)-one using a Pd catalyst and [5+2] cycloadditions of the resulting ylide are described. Among substituted styrene derivatives as dipolarophiles, electron-rich styrenes showed higher yield (up to 80%). The [5+2] cycloaddition reactions can also be applied to exo-methylene cyclic compounds, and an improved method for the synthesis of polygalolide intermediate has been demonstrated.ArticleCHEMICAL COMMUNICATIONS.54(9):1109-1112(2018)journal articl
Targeted mutagenesis in chicken using CRISPR/Cas9 system
The CRISPR/Cas9 system is a simple and powerful tool for genome editing in various organisms including livestock animals. However, the system has not been applied to poultry because of the difficulty in accessing their zygotes. Here we report the implementation of CRISPR/Cas9-mediated gene targeting in chickens. Two egg white genes, ovalbumin and ovomucoid, were efficiently (> 90%) mutagenized in cultured chicken primordial germ cells (PGCs) by transfection of circular plasmids encoding Cas9, a single guide RNA, and a gene encoding drug resistance, followed by transient antibiotic selection. We transplanted CRISPR-induced mutant-ovomucoid PGCs into recipient chicken embryos and established three germline chimeric roosters (G0). All of the roosters had donor-derived mutant-ovomucoid spermatozoa, and the two with a high transmission rate of donor-derived gametes produced heterozygous mutant ovomucoid chickens as about half of their donor-derived offspring in the next generation (G1). Furthermore, we generated ovomucoid homozygous mutant offspring (G2) by crossing the G1 mutant chickens.
