    Synchrotron μXRF imaging reveals elemental distribution in the nickel hyperaccumulator Odontarrhena muralis (Brassicaceae) from Serbia

    Odontarrhena muralis (Brassicaceae) is a nickel hyperaccumulator species from the Balkans used as a “metal crop” in nickel phytomining. This study aimed to determine the elemental distribution, focusing on nickel, in fresh-hydrated plant tissue (stems, leaves and inflorescences), to clarify where nickel is localized at the tissue and cellular scale-level and to infer the physiological response to its hypertolerance and hyperaccumulation. For the analysis, intact plant organs of O. muralis were subjected to elemental imaging using synchrotron-based micro-X-ray fluorescence (μXRF). The predominant distribution of nickel occurs in the epidermal tissue and at the base of the trichomes, which are also the main sinks for calcium deposition. The obtained results represent a further contribution to the knowledge of the physiological characteristics of this hyperaccumulating “metal crop” species and, consequently, to its application in sustainable metal extraction using phytomining.M232.

    Growth, feed utilization, and quantitative histological assessment of the distal intestine and liver of common carp (Cyprinus carpio L.) fed formulated diets containing grains of different soybean cultivars

    A 12-week feeding trial with common carp (Cyprinus carpio) was conducted to test the hypothesis that compound diets formulated on the basis of different soybean cultivars will have effects on growth parameters, feed utilization, and digestive system histology. Soybean grains were from the following cultivars: Alisa (SB-A), Balkan (SB-B), and Galeb (SB-G). The fourth tested diet contained grains from a mixture of different SB cultivars (SB-M). The results confirmed the tested hypothesis, as the fish from the group SB-A showed higher average weight gain, specific growth rates, and feed efficiency, as well as lower feed intake and feed conversion ratio, compared to the groups SB-B and SB-M. On the other hand, diets were expected to cause inflammation in the distal intestine of the fish, but this did not occur. Histological examination of the intestine and liver, performed at the end of weeks 1, 3, 6 and 12, showed no pathological changes. Most of the differences between the groups were found at the end of the trial. The group SB-A had a higher surface area of different intestinal layers compared to the groups SB-B and SB-G. The surface area of the goblet cells was greatest at most of the time points in SB-M. In the liver, the evaluation of the surface area of hepatocytes and their nuclei showed no significant differences between the groups. The differences in final body mass, which showed a maximum value of 18% between the groups, could be of significant importance for culture of this freshwater species.M212.

    Experimental Models to Study Immune Dysfunction in the Pathogenesis of Parkinson's Disease

    Parkinson's disease (PD) is a chronic, age-related, progressive multisystem disease associated with neuroinflammation and immune dysfunction. This review discusses the methodological approaches used to study the changes in central and peripheral immunity in PD, the advantages and limitations of the techniques, and their applicability to humans. Although a single animal model cannot replicate all pathological features of the human disease, neuroinflammation is present in most animal models of PD and plays a critical role in understanding the involvement of the immune system (IS) in the pathogenesis of PD. The IS and its interactions with different cell types in the central nervous system (CNS) play an important role in the pathogenesis of PD. Even though culture models do not fully reflect the complexity of disease progression, they are limited in their ability to mimic long-term effects and need validation through in vivo studies. They are an indispensable tool for understanding the interplay between the IS and the pathogenesis of this disease. Understanding the immune-mediated mechanisms may lead to potential therapeutic targets for the treatment of PD. We believe that the development of methodological guidelines for experiments with animal models and PD patients is crucial to ensure the validity and consistency of the results.M215.6433025

    Ketamine’s Amelioration of Fear Extinction in Adolescent Male Mice Is Associated with the Activation of the Hippocampal Akt-mTOR-GluA1 Pathway

    Fear-related disorders, including post-traumatic stress disorder (PTSD), and anxiety disorders are pervasive psychiatric conditions marked by persistent fear, stemming from its dysregulated acquisition and extinction. The primary treatment for these disorders, exposure therapy (ET), relies heavily on fear extinction (FE) principles. Adolescence, a vulnerable period for developing psychiatric disorders, is characterized by neurobiological changes in the fear circuitry, leading to impaired FE and increased susceptibility to relapse following ET. Ketamine, known for relieving anxiety and reducing PTSD symptoms, influences fear-related learning processes and synaptic plasticity across the fear circuitry. Our study aimed to investigate the effects of ketamine (10 mg/kg) on FE in adolescent male C57 BL/6 mice at the behavioral and molecular levels. We analyzed the protein and gene expression of synaptic plasticity markers in the hippocampus (HPC) and prefrontal cortex (PFC) and sought to identify neural correlates associated with ketamine’s effects on adolescent extinction learning. Ketamine ameliorated FE in the adolescent males, likely affecting the consolidation and/or recall of extinction memory. Ketamine also increased the Akt and mTOR activity and the GluA1 and GluN2A levels in the HPC and upregulated BDNF exon IV mRNA expression in the HPC and PFC of the fear-extinguished mice. Furthermore, ketamine increased the c-Fos expression in specific brain regions, including the ventral HPC (vHPC) and the left infralimbic ventromedial PFC (IL vmPFC). Providing a comprehensive exploration of ketamine’s mechanisms in adolescent FE, our study suggests that ketamine’s effects on FE in adolescent males are associated with the activation of hippocampal Akt-mTOR-GluA1 signaling, with the vHPC and the left IL vmPFC as the proposed neural correlates.M214.666917

    Comparison of the ABC and ACMG systems for variant classification

    The ABC and ACMG variant classification systems were compared by asking mainly European clinical laboratories to classify variants in 10 challenging cases using both systems, and to state if the variant in question would be reported as a relevant result or not as a measure of clinical utility. In contrast to the ABC system, the ACMG system was not made to guide variant reporting but to determine the likelihood of pathogenicity. Nevertheless, this comparison is justified since the ACMG class determines variant reporting in many laboratories. Forty-three laboratories participated in the survey. In seven cases, the classification system used did not influence the reporting likelihood when variants labeled as "maybe report" after ACMG-based classification were included. In three cases of population frequent but disease-associated variants, there was a difference in favor of reporting after ABC classification. A possible reason is that ABC step C (standard variant comments) allows a variant to be reported in one clinical setting but not another, e.g., based on Bayesian-based likelihood calculation of clinical relevance. Finally, the selection of ACMG criteria was compared between 36 laboratories. When excluding criteria used by less than four laboratories (<10%), the average concordance rate was 46%. Taken together, ABC-based classification is more clear-cut than ACMG-based classification since molecular and clinical information is handled separately, and variant reporting can be adapted to the clinical question and phenotype. Furthermore, variants do not get a clinically inappropriate label, like pathogenic when not pathogenic in a clinical context, or variant of unknown significance when the significance is known.M215.

    FIRST RECORD OF HUCHEN Hucho hucho (Linnaeus 1758) IN THE GRZA RIVER: HUMAN-CAUSED INTRODUCTION FROM ITS NATIVE HABITAT IN SERBIA

    Huchen or Danube salmon Hucho hucho (Linnaeus 1758) is one of the largest salmonid species in the world and the largest species native to the Danube basin in Europe. In Serbia, this species inhabits the Drina river system, as well as the upper reaches of the River Ibar. It has already been introduced into the rivers Moravica and Đetinja, which are part of the Zapadna Morava catchment area. Most recently, huchen was stocked from its native habitat into the rivers Jerma and Nišava in the Južna Morava river system, and into the River Mlava which flows into the Danube. Huchen is listed as an endangered (EN) species on the IUCN Red List of Threatened Species and is protected in Serbia, and a Conservation Action Plan has already been adopted. In this report, we present the first record of this species in the River Grza, which is a part of the Velika Morava river system, outside its native range. The ichthyofauna of this recipient river consists of brown trout Salmo trutta L. and Eurasian minnow Phoxinus phoxinus L. In a small river of a fragile ecosystem such as this, the introduction of huchen as an apex predator can seriously harm the species of the native fish community, which warns of the illegal stocking measures currently in process.M2482

    Uncovering Norway: Descriptions of Four New Aphidiinae Species (Hymenoptera, Braconidae) with Identification Key and Notes on Phylogenetic Relationships of the Subgenus Fovephedrus Chen

    With only 33 reported species, Norway ranks among the European countries with the lowest documented diversity of parasitoids from the subfamily Aphidiinae. The “MUST Malaise” project, carried out by Museum Stavanger in Norway, aimed to assess insect abundance and biodiversity and create a reference base for future studies. The preliminary results of our study revealed four species new to science, indicating that the current number of recorded species in Norway is significantly lower than the actual diversity. All species possess unique combinations of morphological characters, distinguishing them from other known Aphidiinae species. Molecular analysis of the barcoding region confirmed that these specimens all belong to the previously undescribed species. In this study, we describe Aphidius norvegicus sp.n., Praon breviantennalis sp.n., Ephedrus gardenforsi sp.n., and Ephedrus borealis sp.n., all collected in Norway. We also provide an identification key and discuss the phylogenetic relationships within the subgenus Fovephedrus Chen, 1986.M213.051815
