Echinococcus spp.in golden jackals (Canis aureus)
The golden jackal (Canis aureus) is a confirmed definitive host for tapeworms of the genus Echinococcus. As Serbia has one of Europe’s largest resident populations of golden jackals, investigating their role in the transmission and distribution of Echinococcus spp. is of interest to public health. To analyze the population genetics of Echinococcus spp. circulating in golden jackals, the project WORM_PROFILER, which started in March of 2024, is collecting gastrointestinal (GI) tracts of legally hunted animals from different areas of Serbia. For this study, GI tracts of 33 animals were processed. After defrosting, the intestines were cut open longitudinally and feces were transferred into 50 mL centrifuge tubes. The mucosa was scraped and analyzed microscopically to isolate parasites. Approximately 3 g of feces was processed by ZnCl2 flotation and sequential mesh filtration to collect taeniid eggs. The DNA from adult Echinococcus spp. and taeniid eggs was extracted using quick boiling in 0.02 M NaOH and screened by multiplex PCR designed to detect E. multilocularis, E. granulosus and E. canadensis. Examination of the mucosa yielded several gravid Echinococcus tapeworms from the small intestine of one animal from western Serbia and eggs were collected from the feces. PCR identified the tapeworm species as E. multilocularis. Single egg picking and sequencing of the cox1 and nad1 genes is underway. Taeniid eggs were additionally collected from another jackal from the same area, but could not be identified as Echinococcus spp. by PCR. These early findings suggest that E. multilocularis is present in western Serbia and sampling in the same geographical area will be intensified. Population-genetics analysis of the brown trout broodstock in the “Panjica” hatchery (Serbia) and its conservation applications
Artificial propagation and stocking of brown trout is a standard practice in recreational fishery management. In recent decades, the importance of maintaining intraspecific diversity and protecting locally adapted lineages has been recognized for the species' long-term survival. The first step in selecting donors for stocking involves distinguishing native trout from non-native and introgressed individuals. The established method for discerning Atlantic hatchery strains from the wild populations involves genetic screening of individual diagnostic SNPs and microsatellite assignment tests. This study, using Serbia's Panjica hatchery as an example, illustrates the proper conduct of routine genetic screening for identifying suitable donors for supportive stocking. The broodstock and reference populations were screened using mtDNA control region, LDH nuclear gene, and 12 microsatellite loci to assess the origin, diversity, and inbreeding levels. The analysis revealed only moderate contamination with Atlantic trout and showed the regional origin of the Danubian genes – over 50% of the broodstock was composed of non-introgressed Danubian individuals tracing their origin to the Zapadna Morava River system. Additionally, the study highlighted a considerable discordance between LDH locus and microsatellites in identifying introgressed individuals, raising concerns about the sole reliance on LDH locus for the identification of Atlantic genetic origin in nuclear DNA.M221.81942
Phloretin inhibits the growth of Arabidopsis shoots by inducing chloroplast damage and programmed cell death
Phloretin is a key secondary metabolite produced by apple trees. Known for its strong antioxidant properties, this dihydrochalcone has been extensively studied in animals but less so in plants. Recently, we identified phloretin as a phytotoxic allelochemical that inhibits growth in the model plant Arabidopsis by disrupting auxin metabolism and distribution in the roots. In this study, we found that phloretin significantly hinders the growth of Arabidopsis seedlings' aerial parts after a short-term treatment (10 days) and causes their decay after long-term exposure (28 days). These effects result from ultrastructural damage in the mesophyll cells of the leaves, including chloroplast displacement and swelling, lesions, and alterations in thylakoid and cell wall organization. Interestingly, phloretin-treated plants showed a decrease in malondialdehyde levels and antioxidant enzyme activities, while hydrogen peroxide and proline levels remained unchanged. This suggests that phloretin-induced chlorosis and seedling decay are not due to oxidative stress but rather to severe chloroplast structural damage, leading to inefficient photosynthesis, starch degradation, starvation, and activation of micro- and macroautophagic processes for self-preservation. Ultimately, these processes result in programmed cell death. These new insights into the phytotoxic effects of phloretin on Arabidopsis shoots could pave the way for future research into phloretin as a potential multitarget bioherbicide and enhance our understanding of autoallelopathy in apple trees.M214.315435430
Cumulative effect of arsenic on the number of mouse offspring and the female reproductive hormones in mice
In this study, we evaluated the cumulative effects of arsenic (III) oxide on the number of mouse offspring over three consecutive generations and monitored changes in levels of the reproductive hormones, oestradiol and progesterone in female mice during the dioestrus phase of the cycle. The control group received water from the mains. In two experimental groups, mice were given drinking water containing dissolved arsenic (III) oxide at concentrations of 10.6 mg L-1 and 106 mg L-1, respectively. These concentrations represent the values converted from a human model to an animal model (mice) thus correspond to the arsenic content of the groundwater in the southern part of the Pannonian Basin, in the province of Vojvodina, in the Banat region, in particular in the town of Zrenjanin. The average number of newborn mice in both experimental groups decreased for three consecutive generations. The total arsenic content of day-old mice did not show significant differences between the experimental groups. Arsenic (III) oxide affected the reproductive hormone levels of female mice at both concentrations.M230.9334072
