    Living at the edge of life: metallophytes from the most toxic arsenic-thallium tailings in the world (Allchar, North Macedonia)

    Background and aims: The abandoned Allchar Mine in the Republic of North Macedonia is a globally unique deposit with historical tailings from processing ore containing extremely toxic metal(loid)s concentrations, with up to 76.1 g kg−1 and 6.5 g kg−1 of As and Tl, respectively. Methods: Five pseudo-metallophyte species which naturally colonise the Allchar tailings were sampled: Alyssum kavadarcensis, Knautia caroli-rechingeri, Minuartia verna, Plantago lanceolata, and Silene vulgaris. Rhizosphere soils, roots and leaves were collected for each individual. The samples were subjected to elemental analysis to determine macro and micro-elemental concentrations. Results: The results show that the studied species exhibit different strategies in relation to the dominant elements in the substrate, with exclusion being the most common among them. Hyperaccumulation of Tl was found in M. verna and P. lanceolata (with 1770 and 1030 mg kg−1 as the mean concentration, respectively), whereas As was found in concentrations above the threshold for hyperaccumulation and leaf-to-root ratio > 1 in M. verna, S. vulgaris, and A. kavadarcensis (up to 1340, 2870 and 1330 mg kg−1, respectively).     Exploring In Vitro Immunomodulatory Properties of Moss Atrichum undulatum Extracts

    Bryophytes are rich sources of diverse secondary metabolites with a wide range of biological activities, including anti-inflammatory, antitumor and antimicrobial effects. The aim of this study was to investigate the chemical composition of extracts from two different genotypes (Serbian and Hungarian) of the axenic moss Atrichum undulatum and evaluate the immunomodulatory potential of the prepared extracts in vitro. Both genotypes of moss samples were cultivated in vitro and subsequently extracted in a Soxhlet apparatus with methanol or ethyl acetate. The highest concentration of total phenolic compounds was found in the methanolic extract of the Serbian genotype (54.25 mg GAE/g extract), while the ethyl acetate extract of the Hungarian genotype showed the highest concentration of phenolic acids (163.20 mg CAE/extract), flavonoids (35.57 mg QE/extract), and flavonols (2.25 mg QE/extract). The extracts showed anti-neuroinflammatory properties by reducing the production of reactive oxygen species, nitric oxide, and tumor necrosis factor alpha by lipopolysaccharide-stimulated microglial cells. Moreover, they mitigated the cytotoxic effects of the pro-inflammatory mediators produced by activated microglia on neurons. The data obtained suggest that extracts from A. undulatum moss have promising anti-neuroinflammatory and neuroprotective properties, making them interesting candidates for further research to combat neuroinflammation.M214.51349131

    The immune system in Parkinson's disease: what we know so far

    Parkinson's disease (PD) is characterised neuropathologically by the degeneration of dopaminergic neurons in the ventral midbrain, the accumulation of α-synuclein (α-syn) aggregates in neurons, and chronic neuroinflammation. In the past two decades, in vitro, ex vivo and in vivo studies have consistently shown the involvement of inflammatory responses mediated by microglia and astrocytes, which may be elicited by pathological α-syn or signals from affected neurons and other cell types, and are directly linked to neurodegeneration and disease development. Besides the prominent immune alterations seen in the central nervous system (CNS), including the infiltration of T-cells into the brain, more recent studies have demonstrated important changes in the peripheral immune profile within both the innate and adaptive compartments, particularly involving monocytes, CD4+ and CD8+ T-cells. This review aims to integrate the consolidated understanding of immune-related processes underlying the pathogenesis of PD, focusing on both central and peripheral immune cells, neuron-glia crosstalk as well as the central-peripheral immune interaction during the development of PD. Our analysis seeks to provide a comprehensive view of the emerging knowledge of the mechanisms of immunity in PD and the implications of this for better understanding the overall pathogenesis of this disease.M21a14.
