    Computational insights into biomolecule dynamics

    The translation of proteins in cellular environments and the diffusion coefficientsof particles play crucial roles in biological processes. However, experimental investigations to determine these coefficients are challenging, requiring time-consuming procedures, expensive high purity samples, and specialized instrumentation. To overcome these issues, computational estimations of diffusion coefficients offer a promising solution. To achieve this computationally, all-atom molecular dynamics simulations are used. These calculations are computationally demanding, requiring long-time simulations of biomolecules with thousands of atoms in aqueous environments. To address both experimental and computational challenges, a new approach is taken, focusing on the fundamental building blocks of protein and polysaccharide structures and how they collectively produce the properties and behavior of larger assemblies. By linking the local chemistry of a solvent-exposed patch of a molecule to its contribution to the overall hydrodynamic radius, an accurate determination of the experimentally comparable diffusion coefficient is achieved. The resulting predictions for diffusion coefficients in various protein structures are comparable to state-of-the-art statistical methods, which may heavily rely on limited training data. This research not only applies to large protein structures but also provides predictive insights into the dynamics of oligosaccharides and larger dendrimers. By understanding the fundamental aspects of molecular diffusion through this approach, the study opens up possibilities for bypassing experimental complexities and enhancing our understanding of biological processes at a deeper level

    Reporting guideline and clinical trial registration adherence in nephrology journals: Results of a preliminary systematic review

    Background: Clinicians rely on relevant, high-quality research to inform their decisions regarding patient care. This research is held to a higher standard when journals implement reporting guidelines and clinical trial registration into article submission requirements. Due to the small number of nephrology journals — and the growing yet still limited research in the field — it is of the utmost importance for these journals to apply stringent guidelines to ensure the publication of limited bias and high quality research. However, the extent of reporting guideline adoption and clinical trial registration policies among nephrology journals is unknown. Therefore, the purpose of this study is to examine the recommendation or requirement of reporting guidelines and clinical trial registration in nephrology journals.Methods: The 2021 Scopus CiteScore Tool was used to identify 62 journals in the “Nephrology” subcategory. In a masked, duplicate fashion, we examined the “Instructions for Authors'' webpage of each included journal to determine whether the following reporting guidelines — outlined by the Enhancing the Quality and Transparency of health Research (EQUATOR) Network — were recommended or required: PRISMA, CONSORT, PRISMA-P, STARD, TRIPOD, MOOSE, ARRIVE, CHEERS, QUOROM, STROBE, CARE, SRQR, SPIRIT, and COREQ. Clinical trial registration statements were investigated in a similar fashion. Journal statements were documented as “Not mentioned”, “Recommended”, “Required”, or “Does Not Require”. Stata 17.0 was used to analyze the data. To minimize bias, all journals were contacted to confirm their accepted article types.Results: The most frequently mentioned guidelines were CONSORT, STROBE, and PRISMA. Of the 62 nephrology journals investigated, CONSORT was required by 11 (18%) and recommended by 34 (55%) journals. Furthermore, STROBE was required by 7 (11%) and recommended by 27 (44%) journals, and PRISMA was required by 8 (13%) and recommended by 18 (29%) journals. The least frequently mentioned guidelines were QUOROM (0/62, 0%), PRISMA-P (24/60, 40%), and MOOSE (26/62, 42%). Finally, 32 (52%) of journals required and 19 (31%) of journals recommended clinical trial registration.Discussion: These findings illuminate the variable adoption of reporting guidelines and inconsistent clinical trial registration policies across nephrology journals. We recommend that journal editors in this field require more author adherence to guidelines to improve the quality of research submitted to and published by their journals

    Assessment of the harms reporting in systematic reviews focused on hallucinogens: A cross-sectional study

    Objective: To investigate the reporting of harms in systematic reviews (SRs) focused on hallucinogen use.Methods: A search was conducted in May 2022 using MEDLINE, Embase, Epistemonikos, and Cochrane databases to retrieve SRs focused on the use of hallucinogens. Investigators screened the titles and abstracts from the search for study inclusion in a masked, triplicate fashion. Investigators analyzed the included SRs for reported harms linked to hallucinogen use via a pre-established harms reporting assessment. Methodological quality of SRs were graded using the A MeaSurement Tool to Assess Systematic Reviews-2 (AMSTAR-2) in a masked, duplicate manner. Study characteristics for each review were extracted in duplicate. Corrected covered area (CCA) was measured for SR dyads.Results: Our search returned 908 articles, and 32 SRs met eligibility criteria for final harms reporting analysis. Of the included reviews, 28 SRs (56.2%) indicated harms as a primary or secondary outcome, 2 SRs (6.3%) reported predetermined methods to grade, collect harms data, or statistically analyze harms. A significant relationship was found between completeness of harms reporting and whether harms were listed as a primary or secondary outcome.Conclusion: Harms were largely underreported in scientific literature regarding hallucinogen use, despite many studies designating them as a primary or secondary outcome. Inadequate reporting is unlikely to provide credible evidence used to evaluate the benefit-harm trade-off. Therefore, steps should be taken to improve reporting of harms in studies concerning hallucinogen use

    Structure fire response under active threat: A critical analysis of urban fire service strategies in South Texas

    Structure fire response involving an active threat creates a unique hazard for first responders, and the challenge is manifested in the varying missions and authorities of responding agencies. Specifically, when fire is combined with an active threat, the lines between law enforcement and the fire service become blurred. Moreover, history has demonstrated that structure fires involving active threats are deadly. However, fire service strategies for these events are not well understood. Still, the potential for events involving fire and active attackers continues to exist. Therefore, this study aims to develop a theoretical analysis of structure fire response under active threat conditions to inform policy and practice.This study employs the constructivist form of grounded theory to investigate the research question, “What strategies are needed by Incident Commanders to support safe and effective structure fire response while under active threat?” To answer the question, study participants were selected from the fire service and emergency management disciplines. Specifically, data was collected from a large urban-based all-hazards fire department in South Texas with a history of high-threat response, and regional support. Also, supporting data was collected from local and regional All-Hazards Incident Management Teams (AHIMT) and emergency management coordinators in South Texas.This study focuses on the perceptions of Incident Commanders and includes 31 semi-structured interviews and three focus groups. The combination of semi-structured interviews and focus group participants totals a sample size of 47. The data revealed five themes, supported by 15 categories and 103 concepts. The themes constructed from the data are Interoperability Needs, Incident Command System Components, Risk-based Strategies, Resource Management, and Support Functions. Finally, recommendations are provided based on the findings. The recommendations are intended to improve fire service strategies for structure fire under active threat response

    Methane dynamics in agricultural cropping systems and near source wastewater treatment lagoon and process evaluation of incubation method for methane oxidation

    The three chapters included in this dissertation center on the dynamics of soil methane fluxes and the potential of soil as a methane sink. Soil CH₄ consumption is a natural CH₄ mitigation strategy, and understanding how it works can aid in the development of strategies to reduce CH₄ emissions from soil and support soil health. The first study quantified CH₄ fluxes at the soil surface interface near a dairy lagoon, which served as the CH₄ point source. Methane gas samples were collected using a close vented gas chamber, injected into pre-vacuumed vials, and quantified for CH₄ concentration using gas chromatograph. Net CH₄ influx were greater in chambers that were installed closest to the lagoon than in chambers further away from the lagoon. The second study evaluated a process of incubation method that uses headspace analysis of incubation chambers of CH₄ concentrations quantified by gas chromatograph (GC) using soil soils from different land management. The first phase of incubation created conditions that exposed the microbes to high levels of CH₄ and may have increased the microbes’ methanotrophic potential. The derived calibration curve generated through known concentration of standard CH₄ gas can be used as a simple and practical tool in quantifying and estimating concentrations of CH₄ during the CH₄ oxidation process in a laboratory setting. The last study utilized a large data set of CH₄ that was a result of studies conducted to assess N₂O emissions. The different field experiments generated 11,837 individual measurements that were taken in 362 sampling events of which only 21% or 2,082 manifested a significant methane flux. Methane consumption is a frequent result of the CH₄ dynamics between the atmosphere and soil. The flux average value was -0.0016 mg CH₄-C m⁻² hr⁻¹ and if this flux rate accurately represents the average consumption of CH₄-C in cropland soils of the Central Great Southern Plains, it would remove 55,950.40 Metric tons CO₂ eq yr⁻¹ from the 8.8 million acres of cropland in Kansas

Pari plans on graduating in Fall 2023 with a bachelor’s degree in business management and marketing while embarking on her MBA with a graduate certificate in entrepreneurship in the following year. Following that, she plans on becoming a registered nurse and acquiring all the certifications needed to start her spa. Pari hopes to combine her passion for beauty and business through her medical spa.Angel Skin and Aesthetics is a luxury medical spa that specializes in aesthetic treatment and provides high quality skincare products. The purpose for Angel Skin and Aesthetics is to provide an unmatched experience through various self-care and holistic wellness treatments. The mission at ASA is to offer clients personalized aesthetic treatments to help them achieve skin goals and feel their very best. ASA strives to build trust with clients through expert medical professionals and a safe environment. The vision is to become the designated location for medical spa products and services. The professionals at ASA envision a world where self-care is a top priority, and everyone is able to receive aesthetic services that will aid in overall wellbeing.The ideal target demographic would be clients aged between 25 to 55. This allows ASA to cater to a wide yet specialized clientele that will understand the benefits of the spa’s offerings. The goal at ASA is to offer clients a wide range of treatments with the goal of enhancing their beauty along with fostering self-confidence. ASA will have a marketing manager who will be in charge of the marketing operations for the spa and a receptionist who will be the first point of contact at the spa.ASA will include a rigorous zero-to-five-year plan that will focus on developing their strategic goals, initial market development, and creating a competitive advantage. This annual growth projection will allow ASA to analyze data to create future goals and meet consumer demand.ASA will utilize a comprehensive marketing strategy and encompassing sales strategy to promote ASA’s brand loyalty and customer retention.In conclusion, Pari plans to cultivate her knowledge with spa luxury through Angel Skin and Aesthetics.     Depressive symptoms in individuals who use cannabis as compared to psychotropic medications

    Marijuana legalization throughout the U.S. has increased over the years; however, research has been limited due to government restrictions at the federal level. This study's purpose is to use the National Survey on Drug Use and Health to investigate the relationship between marijuana use and depressive symptoms as compared to those taking prescribed medications for depression. Data were utilized from participants within the age range of 18-64 years old who endorsed experiencing depressive symptoms within the past year. Using SPSS, we investigated the relationships using an independent samples t-test to find any significant differences between the two variables. Results revealed a significant difference between depressive symptoms between marijuana users and psychotropic medication users, showing those medicated for these feelings scored higher on average. These findings indicate that marijuana does not seem to increase depressive symptoms and may in fact lower them
