Mikrobielle Gemeinschaften auf Pflanzen und wie ihre Mitglieder dem Wirt dienen
Healthy plants in nature are colonized by multi-kingdom microbial communities including bacteria and fungi, termed the plant microbiota, which promote plant growth and health. Our work focuses on the root microbiota and aims at a deeper understanding of the mechanisms underlying the establishment of root-associated microbial communities and their beneficial functions for the host. We have established systematic culture collections of the root microbiota of the crucifer Arabidopsis thaliana and the legume Lotus japonicus, allowing us to culture the majority of bacterial and fungal taxa associated with healthy roots of these two plant hosts. We then compose defined consortia, called synthetic communities (SynComs), from the isolated root commensals and reconstitute the root microbiota in co-cultivation experiments with germ-free plants to study physiological microbial community functions in laboratory environments. This enabled us to demonstrate that the bacterial root microbiota is crucial for the survival of the plant in natural soil by protecting the host against harmful fungal root endophytes. In iron-limiting soils, we show that the bacterial root microbiota is important for plant iron nutrition via a mechanism dependent on the secretion of the plant-derived coumarin fraxetin into the rhizosphere. We propose that the bacterial root microbiota, stimulated by secreted coumarins, is an integral mediator of plant adaptation to iron-limiting soils. Our experiments have also revealed an unexpected host specificity of the root microbiota.Gesunde Pflanzen in der Natur werden von mikrobiellen Gemeinschaften einschließlich Bakterien und Pilzen besiedelt, die als Pflanzenmikrobiota bezeichnet werden und das Pflanzenwachstum und die Pflanzengesundheit fördern. Unsere Arbeit konzentriert sich auf die Wurzel-Mikrobiota und zielt auf ein tieferes Verständnis der Mechanismen, die der Etablierung von wurzelassoziierten mikrobiellen Gemeinschaften und ihren nützlichen Funktionen für den Wirt zugrunde liegen. Wir haben systematische Stammsammlungen der Wurzel-Mikrobiota des Kreuzblütlers Arabidopsis thaliana und der Hülsenfrucht Lotus japonicus angelegt, die es uns ermöglichen, die Mehrzahl der bakteriellen und pilzlichen Taxa zu kultivieren, die mit gesunden Wurzeln dieser beiden Pflanzenwirte assoziiert sind. Aus den isolierten Wurzelkommensalen stellen wir dann definierte Konsortien, sogenannte synthetische Gemeinschaften (SynComs), zusammen und rekonstituieren die Wurzel-Mikrobiota in Co-Kultivierungsexperimenten mit keimfreien Pflanzen, um physiologische mikrobielle Gemeinschaftsfunktionen in Laborumgebungen zu untersuchen. So konnten wir zeigen, dass die bakterielle Wurzelmikrobiota für das Überleben der Pflanze in natürlichen Böden entscheidend ist, indem sie den Wirt vor schädlichen pilzlichen Wurzelendophyten schützt. In eisenlimitierten Böden zeigen wir, dass die bakterielle Wurzelmikrobiota für die mineralische Eisenernährung der Pflanze mittels eines Mechanismus wirksam wird, der von der Sekretion des pflanzlichen Cumarins Fraxetin in die Rhizosphäre abhängt. Wir schlagen vor, dass die bakterielle Wurzelmikrobiota, stimuliert durch pflanzliche Cumarine, ein integraler Vermittler der pflanzlichen Anpassung an eisenlimitierende Böden ist. Unsere Experimente haben auch eine unerwartete Wirtsspezifität der Wurzelmikrobiota aufgedeckt
Complex Structures Determine the Consequences of Global Change for Biodiversity
Die Ökosysteme der Welt sind derzeit einem globalen Wandel der Umweltbedingungen ausgesetzt, der an den meisten Orten zu einer drastischen Umgestaltung der Biodiversität und der Ökosystemfunktionen führt. Unter diesem anhaltenden Wandel bleibt es eine wissenschaftliche Herausforderung zu verstehen, welche Ökosysteme am anfälligsten sind und wie die Folgen menschlicher Einflüsse auf die biologische Vielfalt gemildert werden können. Ein wichtiger Aspekt der Anfälligkeit der Lebensgemeinschaften gegenüber dem globalen Wandel ist die komplexe Struktur ihrer Interaktionsnetzwerke, die alle koexistierenden Populationen durch ihre Räuber-Beute-Interaktionen miteinander verbinden. Im Allgemeinen sind nicht alle Arten gleichermaßen anfällig für externe Störungen. Zum Teil lassen sich diese Unterschiede durch ihre Position im trophischen Netzwerk erklären. So nehmen Arten auf hohen trophischen Ebenen viele aggregierte Effekte wahr, die sich kaskadenartig durch die Nahrungsnetze ziehen, und sind am anfälligsten für ein sekundäres Aussterben, selbst wenn ihre direkte Reaktion auf die Störung schwach war. Diese Folge des globalen Wandels ist besonders problematisch, da die Arten der hohen trophischen Ebene auch eine starke Kontrolle über die multitrophische Ökosystemfunktion natürlicher Gemeinschaften ausüben. Verschiedene Treiber des globalen Wandels haben unterschiedliche Auswirkungen auf die Biodiversität und das Funktionieren von Ökosystemen. Zum Beispiel stellt die globale Erwärmung eine starke Bedrohung für den Fortbestand von Karnivoren mit hohem trophischem Level dar. Interessanterweise untergräbt die Eutrophierung ebenfalls den Fortbestand dieser Arten mit hohem trophischen Level, aber die Prozesse unterscheiden sich, was zu der überraschenden Erkenntnis führt, dass die globale Erwärmung und die Eutrophierung als zwei der Haupttreiber des globalen Wandels sich teilweise gegenseitig aufheben könnten. Die Forschung zu komplexen Artengemeinschaften unter dem globalen Wandel zeigt neue Herausforderungen auf und bietet Lösungen, um die Folgen des globalen Wandels für die Biodiversität und das Funktionieren von Ökosystemen zu beheben.The world’s ecosystems are currently undergoing global environmental change that is dramatically reshaping biodiversity and ecosystem functions in most places. Under this ongoing change, understanding which ecosystems are most vulnerable and how to mitigate the effects of human impacts on biodiversity remains a scientific challenge. An important aspect of the vulnerability of communities to global change is the complex structure of their interaction networks, which link all coexisting populations through their predator-prey interactions. In general, not all species are equally vulnerable to external disturbances. In part, these differences can be explained by their position in the trophic network. For example, species at high trophic levels perceive many aggregate effects cascading through food webs and are most vulnerable to secondary extinction, even if their direct response to the disturbance was weak. This consequence of global change is particularly problematic because high trophic level species also exert strong control over the multitrophic ecosystem function of natural communities. Different drivers of global change have different impacts on biodiversity and ecosystem functioning. For example, global warming poses a strong threat to the persistence of high trophic level carnivores. Interestingly, eutrophication also undermines the persistence of these high trophic level species, but the processes differ, leading to the surprising finding that global warming and eutrophication may partially cancel each other out as two of the main drivers of global change. Lokales Saatgut und globale Bedürfnisse: Ethnobotanik, Agrarökologie und die Geschichte der In-situ-Erhaltung der Nutzpflanzenvielfalt
Various disciplines have sought in recent decades to develop accounts of the agricultural practices of Indigenous peoples. These include ethnobotany, agroecology, and agricultural anthropology. Since the 1980s, this research has often been linked to conservation work, for example the promotion of local farming methods or tools as means of maintaining biodiverse forests or preventing soil erosion. In this presentation, I explore the influence of research on indigenous agriculture on the conservation of global crop diversity. I discuss how social scientists developed new narratives about the past and present of indigenous cultivation, and used these to sustain arguments about the most desirable futures for farming, both within and beyond Indigenous communities. These researchers’ accounts of both the past and the future of agriculture typically centered on particular crop varieties: the locally adapted varieties, or landraces, thought to have been traditionally cultivated but widely understood to be endangered as a result of agricultural intensification. As a result, the research of ethnobotanists and agroecologists contributed to new interest in and approaches to protecting those same varieties, and ultimately forged a lasting link between local acts of cultivation and global conservation concerns.Verschiedene Fachgebiete haben in den vergangenen Jahrzehnten versucht, ein Bild von den landwirtschaftlichen Praktiken der indigenen Völker zu schaffen. Dazu gehören die Ethnobotanik, Agrarökologie und Agraranthropologie. Seit den 1980er-Jahren wurde diese Forschung oft mit Naturschutzarbeit verbunden, zum Beispiel mit der Förderung von lokalen Anbaumethoden oder Geräten als Mittel zur Erhaltung artenreicher Wälder oder zur Vermeidung von Bodenerosion. In diesem Vortrag betrachte ich den Einfluss der Forschung zur indigenen Landwirtschaft auf die Erhaltung der globalen Pflanzenvielfalt. Ich erörtere, wie Sozialwissenschaftler neue Narrative zur Vergangenheit und Gegenwart des indigenen Anbaus entwickelten und diese nutzten, um Argumente über die wünschenswerteste Zukunft der Landwirtschaft zu unterstützen, sowohl innerhalb als auch außerhalb der indigenen Gesellschaften. Die Erklärungen dieser Forscher über die Vergangenheit und die Zukunft der Landwirtschaft konzentrierten sich typischerweise auf bestimmte Pflanzensorten: lokal angepasste Sorten oder Landsorten, die vermutlich traditionell angebaut wurden, aber allgemein durch die Intensivierung der Landwirtschaft als gefährdet betrachtet wurden. Infolgedessen trugen die Forschungen von Ethnobotanikern und Agrarökologen zu einem neuen Interesse am Schutz dieser Sorten bei und schufen letztlich eine dauerhafte Verbindung zwischen lokalen Anbaupraktiken und globalen Naturschutzbelangen
International Sharing of Personal Health Data for Research: the ALLEA, EASAC and FEAM joint initiative on resolving the barriers of transferring public sector data outside the EU/EEA
Personal health data provide a vital resource for research to save and improve lives, reduce health inequalities and benefit society. Research data should be regarded as a global public good. Sharing of data is an essential part of public sector medical research for improved health care and disease prevention, for example to ensure sufficiently large sample sizes, identify complex pathways, and compare the determinants and outcomes of disease in different settings, thereby making the most of the contribution by patients and volunteers to research. It is important for EU citizens that their data are shared for health research, to ascertain whether research results from elsewhere are relevant to their particular genetic makeup and risk factors. At the same time, it is essential to provide appropriate protections for personal data privacy. The General Data Protection Regulation (GDPR) addresses the protection of personal data in the European Union (EU) and European Economic Area (EEA) and the international transfer of data to areas outside the region. It has become apparent that the implementation of the GDPR has introduced impediments to this international transfer of data to outside the EU/EEA, creating problems for academic researchers, healthcare professionals and others in the public sector. These problems affect patients and all citizens who are the ultimate beneficiaries of public sector health research
Economic consequences of the COVID-19 pandemic: analysis and possible courses of action
The COVID-19 pandemic has brought about new medium- and long-term challenges that will need to be addressed by economic and social policy. It has also exposed problems that already existed. This statement presents a discussion of the economic consequences of the pandemic. The editors adress the topics of structural changes and economic growth, distribution and ineaquality, the capacity of government organisations and the sustainability of public fiances. Each of these four substantive sections starts out by providing an analysis of the status quo of a particular area, followed by suggestions for specific political courses of action

Data, Data Science and Artificial Intelligence in Medicine
Due to the rapid development of sensor technologies, vital data can be recorded on a completely new scale with high temporal resolution even over long periods of time. This applies both to the clinical care context, but also to all other areas of daily life. As these amounts of data were historically difficult to manage, the technological evolution in information technology and computer science now offers new possibilities. Big data and artificial intelligence are increasingly finding a wide application in life sciences. The effects on medical care and research are not fully foreseeable as of today. As the necessary algorithms and infrastructures are more easily accessible, their use in a wide variety of areas becomes easier. Technological advances in information technology will, for the foreseeable future, continue to follow a slightly weaker form of Moore’s law. This evolves the miniaturization of IT accompanied by an increase in cost efficiency and performance. Taking these developments into account, an estimation can be made of the extent to which medical data can be recorded and analyzed at affordable costs. From this follows the creation of a qualitatively ever better personal digital twin, whose 24/7 monitoring of health data allows completely new possibilities regarding the detection and prediction of abnormalities. In addition to the individual perspective, this provides new possibilities for research to carry out analysis on a new scale based on large populations. Various projects are already working on the exchange, merging and analysis of clinical care data for research. The development of common data formats, semantic interoperability and data protection-compliant consent procedures are essential steps. However, these developments also have to be considered in relation to ethical, social and economic issues. It requires a joint effort by different disciplines and stakeholders to leverage the potential of these technologies.Durch die rasante Entwicklung von Sensortechnologien können Vitaldaten in einem völlig neuen Maßstab mit hoher zeitlicher Auflösung auch über lange Zeiträume hinweg erfasst werden. Dies gilt sowohl in der klinischen Versorgung als auch in allen anderen Bereichen des Alltags. Früher waren diese Datenmengen kaum handhabbar. Nun bieten Entwicklungen in der Informationstechnologie und Informatik neue Möglichkeiten. Große Datenmengen und die künstliche Intelligenz finden zunehmend eine breite Anwendung in den Biowissenschaften. Die Auswirkungen auf die medizinische Versorgung und Forschung sind bis heute nicht vollständig absehbar. Da die notwendigen Algorithmen und Infrastrukturen leichter zugänglich sind, wird ihr Einsatz in den verschiedensten Bereichen erleichtert. Die technologischen Fortschritte in der Informationstechnologie folgen auf absehbare Zeit weiterhin dem Mooreschen Gesetz in abgeschwächter Form. Damit wird die Miniaturisierung von IT weiterentwickelt, begleitet von einer Steigerung der Kosteneffizienz und der Leistungsfähigkeit. Unter Berücksichtigung dieser Entwicklungen lässt sich abschätzen, in welchem Umfang medizinische Daten zu erschwinglichen Kosten erfasst und analysiert werden können. Daraus folgt die Erzeugung eines qualitativ immer besseren, persönlichen digitalen Zwillings, dessen Rund-um-die-Uhr Überwachung von Gesundheitsdaten völlig neue Möglichkeiten zur Erkennung und Prognose von Anomalien ermöglicht. Neben der individuellen Perspektive bietet es der Forschung neue Möglichkeiten, Analyse in einem neuen Maßstab auf Basis großer Bevölkerungsgruppen durchzuführen. Verschiedene Projekte befassen sich bereits mit dem Austausch, der Zusammenführung und der Analyse von klinischen Versorgungsdaten für Forschungszwecke. Wesentliche Fortschritte werden durch die Entwicklung gemeinsamer Datenformate, der semantischen Interoperabilität und datenschutzkonformen Einwilligungsverfahren gemacht. Bei diesen Entwicklungen müssen jedoch auch ethische, soziale und wirtschaftliche Fragen in Betracht gezogen werden. Die Ausnutzung dieser Technologie erfordert gemeinsame Anstrengungen verschiedener Disziplinen und Interessengruppen
AWMF: Is The Transformation of Evidence through Digitalization Possible?
The digital transformation promises disruptive developments to improve medical knowledge management and medical care. Digital transformation as a future development and the exponentially growing applications in this context must, however, prove their benefit for patients and citizens and, to this end, be integrated into the principle of evidence-based medicine. Medical science is called upon to take a leading role in shaping the digital trans-formation and develop concepts for its evaluation. In this context, the idea of a “Digital Trustworthy Evidence Ecosystem” is being promoted internationally. The core of this idea is interoperability as a prerequisite for the accelerated transfer and critical evaluation of medical knowledge on the basis of uniform and publicly accessible data models, terminologies and interfaces. However, the digital transformation in the healthcare system also requires a consideration of changes at the societal level with regard to medical knowledge management and a changed role of patients and citizens as owners of their health data and partners in shared decision-making. In this context, the AWMF pursues a comprehensive project on the digitalization of quality-assured guideline knowledge as a partial aspect of the overall idea of the “Digital Trustworthy Evidence Ecosystem”.Die digitale Transformation verspricht bahnbrechende Entwicklungen zur Verbesserung des medizinischen Wissensmanagements und der medizinischen Versorgung. Die digitale Transformation als zukünftige Entwicklung und die in diesem Zusammenhang exponentiell wachsenden Anwendungen müssen jedoch ihren Nutzen für Patienten und Bürger unter Beweis stellen und zu diesem Zweck in das Prinzip der evidenzbasierten Medizin integriert werden. Die Medizinwissenschaft wird aufgefordert, die digitale Transformation federführend mitzugestalten und Konzepte für ihre Bewertung zu entwickeln. In diesem Zusammenhang wird das Konzept eines „digitalen vertrauenswürdigen Beweissystems“ (Digital Trustworthy Evidence Ecosystem) international gefördert. Im Mittelpunkt dieser Idee ist die Interoperabilität als Voraussetzung für den beschleunigten Transfer und die kritische Bewertung medizinischen Wissens auf der Grundlage einheitlicher und öffentlich zugänglicher Datenmodelle, Terminologien und Schnittstellen. Der digitale Wandel im Gesundheitswesen erfordert jedoch auch eine Berücksichtigung der Veränderungen auf gesellschaftlicher Ebene im Hinblick auf das medizinische Wissensmanagement und eine veränderte Rolle der Patienten und Bürger als Eigentümer ihrer Gesundheitsdaten und Partner bei der gemeinsamen Entscheidungsfindung. In diesem Zusammenhang befasst sich die AWMF in einem umfassenden Projekt mit der Digitalisierung von qualitätsgesichertem Richtlinienwissen als Teilaspekt der Gesamtidee des „digitalen vertrauenswürdigen Beweissystems“

It is the aim of the Leopoldina Symposium “Mission-Innovation” to discuss interdisciplinary possibilities, challenges and responsibilities of telematik, eHealth and High Definition Medicine, as well as the resulting innovation potential in acute medicine so that we can apply it for the benefit of patients. In order to be able to evaluate the potential and the benefits for our patients, it is crucial to engage with the patient perspective
